高效液相色谱-荧光检测法测定环境水中的苯胺和苯酚   总被引:9,自引:0,他引:9  
建立了用高效液相色谱荧光检测法同时测定环境水中苯胺和苯酚的分析方法。色谱柱为EclipseXDB C8(4.6mmi.d.×150mm,5μm),流动相为甲醇 磷酸盐缓冲液(0.1mol/L磷酸二氢钾 0 1mol/L磷酸氢二钠,pH6.87)V(甲醇)∶V(磷酸盐缓冲液)=50∶50,流速1 0mL/min,柱温25℃,检测波长0minλex/λem=230/340nm(测定苯胺),3.5minλex/λem=215/300nm(测定苯酚)。测定苯胺的线性范围0.2~120ng,r=0.9999,检出限0.01ng;测定苯酚的线性范围0.4~500ng,r=0.9998,检出限0.02ng,回收率98.1%~101.2%。该方法已用于对环境水中苯胺和苯酚的测定。  相似文献

建立了高效液相色谱-荧光检测法同时测定人体尿液中的酪氨酸(Tyr)和色氨酸(Typ)。采用Diamonsil C18柱(250×4.6mm,5μm),以水-乙腈(80∶20,V/V)为流动相,流速1.0mL/min,荧光检测波长为λex=260nm、λ_(em)=340nm。Tyr和Typ的含量分别在0.10~6.0μg/mL及0.050~6.0μg/mL范围内与色谱峰面积呈良好的线性关系(r=0.9993),检测限分别为0.05μg/mL和0.02μg/mL。在三个添加水平的回收中,两种氨基酸的平均回收率在86.3%~122.3%之间,相对标准偏差(RSD)小于8.51%。将所建立的方法应用于健康人、普通病人和癌症患者晨尿检测,结果发现:癌症患者尿样中色氨酸的水平与健康人和普通病人相比偏低。  相似文献

用高效液相色谱法(HPLC)测定多环芳烃时,因芴、苊和菲,茚(1,2,3-cd)芘和苯并(g,h,i)苝的色谱峰严重重叠而影响测定结果。本研究用高效液相色谱-二极管阵列检测器(DAD)和荧光检测器(FLD)测定多环芳烃,在激发波长λex=230nm,发射波长λem=300~500nm范围内采集重叠峰的HPLC-FLD二维色谱数据,再用直观推导式演进特征投影法(HELP)解析它们的重叠色谱峰,分辨结果令人满意。该方法对重叠组分的分辨下限为0.02mg/L。结果表明,用二维色谱荧光数据解析色谱重叠峰,灵敏度更高,可用于环境样品中多环芳烃的测定。  相似文献

马兜铃酸(aristolochic acid)是马兜铃科植物所含的主要药物成分,属硝基菲类衍生物.但是,近几年有许多报道马兜铃酸具有致癌和致突变作用.动物实验和临床研究表明,马兜铃酸进入人体后,在机体组织中形成马兜铃酸-DNA加合物,进而引起致癌作用.因此,马兜铃酸-DNA加合物现已被用作检测马兜铃酸中毒的生物标记物.本文介绍了当前马兜铃酸-DNA加合物主要的检测方法:32P标记法,高效液相色谱-紫外检测法,高效液相色谱-放射检测法,高效液相色谱-荧光检测法,高效液相色谱-质谱联用法和超高效液相色谱-质谱联用法.本研究也通过实验表明,超高效液相色谱-质谱联用法对马兜铃酸-DNA加合物的检测具有快速、灵敏、专一性和重现性强、定量准确的特点,是马兜铃酸-DNA加合物研究的有力工具.本文最后总结了在不同实验对象和给药剂量的马兜铃酸研究中,马兜铃酸-DNA加合物的检测数据,对马兜铃酸-DNA加合物的研究具有参考价值.  相似文献

建立了用高效液相色谱快速检测反应香精中2-氨基-N-甲基-5-苯基咪唑并吡啶的方法. 反应香精中的2-氨基-N-甲基-5-苯基咪唑并吡啶用二甲基甲酰胺直接超声振荡提取; 高效液相色谱的色谱柱为Eclipse XDB-C18 (250 mm×4.6 mm, 5 μm); 流动相为V(二甲基甲酰胺)∶V(甲醇)∶V(水)=8∶50∶42; 流速为0.5 mL/min; 光电二极管阵列检测器检测波长为UV-327 nm. 10种反应香精中均未检出该种物质, 回收率为78.9%~85.9%, 检出限为19.2 ng/mL.  相似文献

A new and simple synthesis of novel N-protected methyl 5-substituted-4-hydroxypyrrole-3-carboxylates, which exist in equilibrium with their 4-oxo tautomers, has been developed in two steps starting from N-protected α-amino acids. The key intermediates are enaminones, which can also be isolated, characterized, and used for the construction of other functionalized heterocycles, before they spontaneously decompose to pyrrole products. 4-Hydroxypyrroles are prone to partial aerial oxidation but can be efficiently alkylated or reduced to stable polysubstituted pyrrolidine derivatives.  相似文献

The chemoselectivity in the intramolecular CH insertion of various diazosulfonamides has been experimentally studied. The results reveal that the aliphatic 1,4-, 1,5-, or 1,6-C(sp3)?H insertions of diazosulfonamides are not accessible, while the aromatic 1,5-C(sp2)?H insertion can be realized specifically by adjusting the diazo-adjacent group. In addition, the general chemoselectivities in the intramolecular CH insertions of diazosulfonyl compounds are summarized. Generally, diazosulfones undergo both aromatic 1,5-C(sp2)?H and aliphatic 1,5- and 1,6-C(sp3)?H insertions, while diazosulfonates undergo aliphatic 1,5- and 1,6-C(sp3)?H insertions. However, diazosulfonamides only undergo aromatic 1,5-C(sp2)?H insertion.  相似文献

An efficient iodine-mediated oxidative Pictet-Spengler reaction in dimethyl sulphoxide (DMSO) using terminal alkynes as the 2-oxoaldehyde surrogate for the synthesis of aryl (9H-pyrido[3,4-b]indol-1-yl)methanones is described. The scope of the protocol includes the total synthesis of Fascaplysin, Eudistomins Y1 and Y2. The methodology is extended for preparing pyrrolo[1,2-a]-quinoxaline and indolo[1,5-a]quinoxaline derivatives. The utility of 1-aroyl-β-carbolines was demonstrated by performing palladium-catalyzed β-carboline directed ortho-C(sp2)-H functionalization of the phenyl ring with thiomethyl (SMe) group using DMSO as source and for accessing 4-aryl-canthin-6-ones.  相似文献

In this Letter, we described a facile method for constructing fused bicyclic 1-arylpyrazol-5-one ring system. We employed various methylene-containing carboxylic acids as the substrates and proved that the pyrazolone ring closure requires activated methylene group in intermediate II. Accordingly, a series of structurally diversified, fused bicyclic 1-arylpyrazol-5-ones was prepared in moderate to high yields using the requisite substrates.  相似文献

A series of 20 CuAIAC reactions between eight 4-acylamino substituted pyrazolidine-3-one-1-azomethine imines and four terminal ynones were performed using Cu0 as catalyst. The corresponding fluorescent cycloadducts were obtained in very high yields upon simple workup. Thus, Cu-metal turned out to be a better catalyst than CuI in terms of yield and ease of isolation. Availability of azomethine imines, mild reaction conditions, and simple workup enable a “click” access to libraries of densely substituted 2,3-dihydro-1H,5H-pyrazolo[1,2-a]pyrazol-1-ones. Reactivity of differently substituted dipoles was evaluated experimentally and by quantum chemical methods (DFT).  相似文献

(E)-4-(Fullerenopyrrolidin-1-yl)-3-methylbut-2-enoic acid and its corresponding succinimidyl ester, readily obtained through Prato-type modification of C60, were used for the selective N-acylation of polyamines. The thus obtained conjugates were evaluated for their antioxidative and anti-inflammatory activity and their cytotoxicity was determined. Members of this family of compounds showed interesting anti-lipid peroxidation, anti-lipoxygenase and anti-inflammatory activity and comparable cytocompatibility to spermidine.  相似文献
