Tricaine methanesulfonate is one of most commonly used anesthetics in fish during blood sampling, artificial propagation and long‐distance transportation. In this study, an accurate method for the quantitative determination of tricaine in fish samples by a stable isotope dilution assay coupled with high‐performance liquid chromatography–triple quadrupole mass spectrometry was developed. Tricaine‐D5 was synthesized and used as an isotopically labeled internal standard for the determination of tricaine. The analytical performance of the method was validated for tricaine determination in marine fish and freshwater fish. The determination of tricaine was linear in the range of 2.0–200.0 μg L?1. The limit of detection and limit of quantitation for fish muscle tissues were 1.0 and 4.0 μg kg?1, respectively. Good recoveries were obtained in the range of 92.08–97.50%. The inter‐ and intra‐assay relative standard deviations (RSD values) were investigated, and the values were 0.39–3.01 and 0.85–2.77%, respectively. The values of CCα and CCβ were 10.21–10.43 and 10.42–10.87 μg kg?1, respectively. The clearance of MS‐222 from grass carp was further studied using our method. The results demonstrate that MS‐222 could be well absorbed and rapidly eliminated after bath administration.  相似文献

The increasing world population with their growing consumption of goods escalates the issue of sustainability concepts with increasing demands in recycling technologies. Recovery of post‐consumer packaging waste is a major topic in this respect. However, contamination with odorous constituents currently curtails the production of recycling products that meet the high expectations of both consumers and industry. To guarantee odor‐free recyclates, the main prerequisite is to characterize the molecular composition of the causative odorants in post‐consumer plastic packaging waste. However, targeted characterization of odorous trace contaminants among an abundance of volatiles is a major challenge and requires specialized and high‐resolution analytical approaches. For this aim, post‐consumer packaging waste was characterized by sensory analysis and two‐dimensional high resolution gas chromatography coupled with mass spectrometry and olfactometry. The 33 identified odorants represent various structural classes as well as a great diversity of smell impressions with some of the compounds being identified in plastics for the first time. Substances unraveled within this study provide insights into sources of odorous contamination that will require specific attention in the future in terms of screening and prevention in recycling products.  相似文献

Alternaria toxins are emerging mycotoxins whose regulation and standardization are in progress by the European Commission and the European Committee for Standardization. This paper describes a dilute and shoot approach to determine five Alternaria toxins in selected food samples using liquid chromatography–tandem mass spectrometry (LC-MS/MS). The strategy involves sample extraction with acidified aqueous methanol, followed by a solvent change accomplished via sample evaporation and reconstitution. The quantification is based on isotope dilution, applying all corresponding isotopically labeled internal standards to compensate possible matrix effects of the analysis. The main advantages of the present method over other existing methods includes simple and effective sample preparation, as well as detection with high sensitivity. The five-fold sample dilution can decrease matrix effects, which were evaluated with both external and internal standard methods. The results demonstrated a limit of quantification lower than 1.0 µg/kg for all five analytes for the first time. The newly presented method showed acceptable accuracy (52.7–111%) when analyzing naturally contaminated and spiked standard samples at the described levels. The method was validated for tomato-based and flour samples (wheat, rye, and maize). The absolute recovery ranged from 66.7% to 91.6% (RSD < 10%). The developed method could be an alternative approach for those laboratories that exclude sample cleanup and pre-concentration of state-of-the-art instruments with enhanced sensitivity.  相似文献

建立了测定大气中25种有机氯农药(OCPs)的同位素稀释-高分辨气相色谱/高分辨质谱法(ID-HRGC/HRMS)。样品用正己烷/二氯甲烷(1∶1, v/v)进行加速溶剂萃取(ASE)。通过柱洗脱实验、单柱和组合柱净化实验,最终确定样品的净化方案为弗罗里硅土固相萃取柱和石墨化炭黑固相萃取柱组合净化。样品萃取液净化后进行HRGC/HRMS分析。采用平均相对响应因子(RRF)法对样品中目标物进行定量,6点校准溶液RRF的相对标准偏差(RSD)均≤20%。线性范围为0.4~800 μg/L,相关系数R2均>0.992。对空白样品依次进行100 pg、400 pg和15 ng水平下的加标试验,各添加水平下OCPs测定值的RSD为0.64%~16%,加标回收率为67.2%~135%。穿透试验表明,滤膜+聚氨酯泡沫/聚氨酯泡沫作为吸附介质的大体积主动大气采样器(AAS)在采集环境空气时,五氯苯极易发生穿透,有效采样模式待进一步研究。在上述采样模式下,六氯苯的有效采样体积较小,标准状态(101.325 kPa, 273 K)采样体积应≤30 m3,其他OCPs应≤1200 m3。以上述体积计算,25种目标化合物的检出限为0.002~0.7 pg/m3。对北京环境空气样品分析测定,结果显示除反式-环氧七氯、异狄氏剂、顺式-九氯和4,4'-滴滴滴在部分样品中未检出外,其他OCPs均为100%检出;六氯苯浓度在514~563 pg/m3之间,其他OCPs的浓度在0.01~18.9 pg/m3之间;替代标回收率为33.9%~155%。由于现有相关监测标准的仪器灵敏度较低、方法检出限较高,已无法满足目前空气中痕量OCPs的测定需求,因此亟待修订新的高灵敏度监测方法标准。该方法适用于目前大气中OCPs的超痕量水平分析,为新标准的制订奠定基础,也为国家履行相关国际公约提供有力技术指导。  相似文献

The effects (chemical, thermal, transport, and radiative) of CO2 added to the fuel side and oxidizer side on the flame temperature and the position of the flame front in a one-dimensional laminar counterflow diffusion flame of methane/N2/O2 were studied. Overall CO2 resulted in a decrease in flame temperature whether on the fuel side or on the oxidizer side, with the negative effect being more obvious on the latter side. The prominent effects of CO2 on the flame temperature were derived from its thermal properties on the fuel side and its radiative properties on the oxidizer side. The results also highlighted the differences in the four effects of CO2 on the position of the flame front on different sides. In addition, an analysis of OH and H radicals and the heat release rate of the main reactions illustrated how CO2 affects the flame temperature.  相似文献

建立了人生长激素( hGH)纯化分析和高效液相色谱( HPLC)与同位素稀释质谱( IDMS)联用高准确度绝对定量方法。采用快速蛋白液相色谱来分离纯化hGH,以傅里叶变换离子回旋共振质谱仪( FTICR-MS)准确地测定蛋白质的分子质量。纯化的hGH经酸水解后,采用KINETEX C18色谱柱为分析柱,以水(含0.1%三氟乙酸)和乙腈为流动相,等梯度分离,流速0.2 mL/min,温度40℃,采用电喷雾正离子模式进行电离,选择多反应监测模式进行检测,内标法定量分析。结果表明,FTICR-MS所测得的分子量实测值与理论值仅相差0.31 Da,脯氨酸、缬氨酸和苯丙氨酸在液相条件下5 min内达到基线分离,在最优条件下,hGH含量测定结果为186.80 mg/g,相对标准偏差为0.52%。利用本方法参加国际比对,比对结果与参考值等效一致。本方法具有简易、实用的特点,并且准确可靠,可作为hGH纯品标准物质的定值方法,为hGH的日常检测提供参考。  相似文献

提出了同位素稀释-气相色谱-质谱法测定白酒中23种邻苯二甲酸酯类化合物含量的方法。样品用正己烷-乙酸乙酯(1+1)混合液提取后,经DB-5MS毛细管色谱柱分离,采用电子轰击离子源选择离子反应监测模式进行质谱测定,同位素内标法进行定量分析。23种邻苯二甲酸酯类化合物的线性范围在0.2~3.0mg·L-1之间,测定下限(10S/N)在0.05~0.1mg·kg-1之间。加标回收率在73.2%~122%之间,相对标准偏差(n=6)在0.49%~11%之间。  相似文献

Some experiments were made for the buoyant jet from a square orifice with a square disc placed on it in static ambient and concentration along the axis in self-similar area behind disc was measured. And at the same time a three-dimensional mathematical model was established to simulate the whole flowing under different conditions. All the results predicted by the numerical calculation were substantiated by the experiments. The results were compared with experiential formula for obstructed round buoyant vertical jets in static ambient and it was found that the two concentration distributions had good accordance. Star shape of temperature isolines on cross-sections in the near areas from the disc was found and it was a very special figure for obstructed square buoyant vertical jets with a square disc. The shape will transform to concentric circles gradually alike to the round buoyant vertical jet in self-similar area with increasing of the distance from the disc.  相似文献

Dexmedetomidine (Dex) is a selective central α2‐agonist with anesthetic properties and has been used in clinical practice for sedation in the intensive care unit (ICU) after operations. In this study, an analytical assay for the determination of Dex in a small amount of plasma was developed for the application to pediatric ICU trials. The quantification of Dex was constructed using the original stable isotope Dex‐d3 for electrospray ionization‐tandem mass spectrometry (ESI‐MS/MS) in the selected reaction monitoring mode. A rapid ultra‐performance liquid chromatography technique was adopted using ESI‐MS/MS with a runtime of 3 min. Efficacious concentration levels (50 pg/mL to 5 ng/mL) could be evaluated using a very small amount of plasma (10 μL) from patients. The lower limit of the quantification was 5 pg/mL in the plasma (100 µL). For sample preparation, a solid‐phase extraction was used along with the OASIS‐HLB cartridge type. Recovery values ranged from 98.8 to 100.3% for the intra‐ [relative standard deviation (RSD), 0.9–1.3%] and inter‐ (RSD, 0.9–1.5%) day assays. A stable test had recovery values that ranged from 97.8 to 99.7% with an RSD of 1.0–1.9% for the process/wet extract, bench‐top, freeze–thaw and long‐term tests. This method was used to measure the Dex levels in plasma from pediatric ICU patients. In the clinical ICU trial, the small amount of blood (approximate plasma volume, 200 μL) remaining from blood gas analysis was reused and targeted for the clinical analysis of Dex in plasma.
