成纤维细胞基因治疗血友病B的临床Ⅰ期试验

本文首次报道了以皮肤成纤维细胞为靶细胞,对血友病B患者实施基因治疗的监床Ⅰ期试验。首先用构建有人凝血因子Ⅸ cDNA的反转录病毒载体(XL-Ⅸ和N2 CMV-Ⅸ)感染血友病B患者皮肤成纤维细胞(HBSF),选择得到表达有人Ⅸ因子蛋白的细胞,而后进行一系列安全性检测,包括细胞连续传代形态观察;染色体分析,软琼脂试验;裸鼠接种试验;胶原过敏试验;内毒素试验等,在确定表达人Ⅸ因子蛋白的细胞的安全性后,大量扩增细胞,最后,细胞用胶原包埋,直接注射到血友病B患者腹部或背部皮下。患者1体内凝血因子IX浓度从71ng/ml上升到约240 ng/ml,至今维持在220ng/ml,持续表达6个月,凝血因子Ⅸ的凝血活性也从2.9％上升到6.3％,该患者的临床症状改善;患者2体内凝血因子Ⅸ浓度亦从130 ng/ml上升到约280 ng/ml,至今维持在220 ng/ml,持续表达5个多月,但活性增加不稳定。两名受试者至今没有发现任何副作用和危害,正在随访之中。我们认为反转录病毒介导的基因转移自体皮肤成纤维细胞,用胶原包埋,皮下移植是安全可靠的,也是简便易行的,从而成功地完成了血友病B基因治疗的临床Ⅰ期试验。

怎样在課堂上檢查作業

在數学教学中,時間花得較多的就是批改作業,在过去,我們学校曾为这事十分头痛,每日夜深人尽,總是我們組的办公室散射灯光,一直弄到十點,甚至十二點,我們知道,若是大部分時間花在批改作業上,那麼怎样能够提高教学呢?因此,提出了怎样才能从作業中解放出來。从雜誌上及教育学上都可看到,苏联很重視在課堂上檢查家庭作業,那麼,我們不是也可以在課堂上來檢查作業嗎?下面是一年來的一些經驗与体会。在起初,我們曾疑心,在課堂上檢查作業,豈不耽誤教学時間嗎?我們学校是中等技術学校,課程多,時間少,平常上課,常是講得滿堂灌;若再檢查作業,豈不是講不完了嗎?但是通过观摩,也有老師講得輕鬆的,經过研究,認为是重點明確所致,当然,重點明確是每一个老師都知道的;但是,我們若仔細檢查講过的一堂課,總会發現有的地方是不必要的細緻,有的話是廢話;甚至有的老師,不管問題大小,交代得詳詳細細,若用同学的話來說:『听的清楚明就是抓不住重點』,我認为,教課好比喂小孩吃东西白,一样,既不能大塊地拚命往內塞,也不能嚼得細細的,連一點滋味都没有,应提出这样的口号:「不說一句廢話」,在以前,每到期考時,同学就反映:「太多了,那能看得完」,今年期考時,就有同学这样反映:「虽然有好幾本書(代數、三角、高等數学教程),但是總起來問題並不多」,但是这裹不是討論重點教学,

Optical phase shifting with acousto-optic devices

A novel optical phase-shifting method based on a well-known acousto-optic interaction is proposed. By using a pair of acousto-optic modulators (AOMs) and properly aligning them, we construct an optical phase shifter that can directly control the phase of a collimated beam. The proposed phase shifter is insensitive to the polarization of the incident beam when polarization-insensitive AOMs are used, and no calibration is necessary. The proposed approach is confirmed with experimental results.     

Omnidirectional reflector using nanoporous SiO2 as a low-refractive-index material

Triple-layer omnidirectional reflectors (ODRs) consisting of a semiconductor, a quarter-wavelength transparent dielectric layer, and a metal have high reflectivities for all angles of incidence. Internal ODRs (ambient material's refractive index n > 1.0) are demonstrated that incorporate nanoporous SiO2, a low-refractive-index material (n = 1.23), as well as dense SiO2 (n = 1.46). GaP and Ag serve as the semiconductor and the metal layer, respectively. Reflectivity measurements, including angular dependence, are presented. Calculated angle-integrated TE and TM reflectivities for ODRs employing nanoporous SiO2 are R(int)/TE = 99.9% and R(int)/TM = 98.9%, respectively, indicating the high potential of the ODRs for low-loss waveguide structures.
