应用壳聚糖将葡萄糖氧化酶固定于鸡蛋膜上,结合氧电极制得葡萄糖传感器.实验表明,壳聚糖比戊二醛能更好地固定葡萄糖氧化酶,最佳条件为壳聚糖浓度0.3%、固定化酶量0.8 mg、 pH 7.0、缓冲溶液浓度300 mmol/L和温度25 ℃.本葡萄糖传感器的线性范围为0.016~1.10 mmol/L;检出限为8.0 μmol/L(S/N=3), 响应时间<60 s,有很好的稳定性,寿命>3个月.同一个传感器重复使用以及同方法制作的不同传感器之间都有很好的重现性,RSD分别为2.5%(n=10)和4.7%(n=4).实际样品中可能存在的烟酰胺、 VB6、 VB12、 VE、Ca2+、 Mg2+、 K+和Zn2+等对葡萄糖的测定不产生干扰.本传感器已成功地应用于市售饮料中葡萄糖含量的测定. 相似文献

The direct electrochemistry of glucose oxidase (GOD) was achieved based on the immobilization of GOD on a natural nano‐structural attapulgite (ATP) clay film modified glassy carbon (GC) electrode. The immobilized GOD displayed a pair of well‐defined quasi‐reversible redox peaks with a formal potential (E0′) of ?457.5 mV (vs. SCE) in 0.1 mol·L?1 pH 7.0 phosphate buffer solution. The peak current was linearly dependent on the scan rate, indicating that the direct electrochemistry of GOD in that case was a surface‐controlled process. The immobilized glucose oxidase could retain bioactivity and catalyze the oxidation of glucose in the presence of ferrocene monocarboxylic acid (FMCA) as a mediator with the apparent Michaelis‐Menten constant Kappm of 1.16 mmol·L?1. The electrocatalytic response showed a linear dependence on the glucose concentration ranging widely from 5.0×10?6 to 6.0×10?4 mol·L?1 (with correlation coefficient of 0.9960). This work demonstrated that the nano‐structural attapulgite clay was a good candidate material for the direct electrochemistry of the redox‐active enzyme and the construction of the related enzyme biosensors. 采用浸渍法结合KBH4还原法制备了Ru-B/SiO2非晶态催化剂(3.14%Ru),并将其用于液相葡萄糖加氢制山梨醇反应. 结果表明,该催化剂具有良好的热稳定性,且其催化活性远高于Raney Ni和Ni-B/SiO2非晶态催化剂,山梨醇的选择性接近于100%,显示出良好的工业化应用前景. 通过与晶态Ru-B/SiO2及Ru/SiO2的催化活性进行比较,并结合ICP,XRD,DSC,SEM,XPS及氢吸附等表征结果,初步讨论了非晶态合金结构和表面电子态对催化剂活性的促进作用. 相似文献

The scarcity and expense of access to L ‐sugars and other rare sugars have prevented the exploitation of their biological potential; for example D ‐psicose, only recently available, has been recognized as an important new food. Enzyme‐mediated self‐healing of dynamic covalent bond‐driven protein hydrogels was realized by the synergy of two enzymes, glucose oxidase (GOX) and catalase (CAT). The reversible covalent attachment of glutaraldehyde to lysine residues of GOX, CAT, and bovine serum albumin (BSA) led to the formation and functionalization of the self‐healing protein hydrogel system. The enzyme‐mediated protein hydrogels exhibit excellent self‐healing properties with 100 % recovery. Glucose is directly related to brain activity and to diabetes.Therefore,developing a rapid and sensitive method for glucose detection is essential.Here,label-free glucose detection at attomole levels was realized by detecting the average diameter change of gold nanoparticles(AuNPs)utilizing dynamic light scattering(DLS).Single-strand DNA(ssDNA)adsorbed into the AuNPs’surfaces and prevented them from aggregating in solution that contained NaCl.However,ssDNA cleaved onto ssDNA fragments upon addition of glucose,and these fragments could not adsorb onto the AuNPs’surfaces.Therefore,in high-salt solution,AuNPs would aggregate and their average diameter would increase.Based on monitoring the average diameter of AuNPs with DLS,glucose could be detected in the range from 15 pmol/L to 2.0 nmol/L,with a detection limit of 8.3 pmol/L.Satisfactory results were also obtained when the proposed method was applied in human serum glucose detection. 相似文献

In general, lignocellulosic biomass contains three major components, namely lignin, hemicellulose and cellulose which are the polymers of C5 and C6 sugars. Thus, there is potential to utilize of this biomass for bioethanol production. The hydrolysis of cellulose into glucose was difficult due to the more fibrous nature and thus inhibit enzyme penetration into the cellulose. In order to solve this problem, hydrothermal pretreatment can be used for breaking the bonds within the lignin structure and increase the accessibility of enzyme into the cellulose. In this study, the effect of chemical addition, sodium hydroxide (NaOH) and calcium oxide (CaO) in hydrothermal pretreatment at 180 °C and 30 minutes reaction time of palm oil empty fruit bunches (EFB) on the enzymatic hydrolysis efficiencies was investigated. The enzymatic hydrolysis of hydrothermally pretreated EFB give the highest concentration of glucose at 0.67 g/L while the hydrothermally pretreated of EFB in the presence of NaOH gives the lowest glucose concentration 0.45 g/L. 相似文献
