Quantum dots assisted laser desorption/ionization mass spectrometric detection of carbohydrates: qualitative and quantitative analysis 下载免费PDF全文
A quantum dots (QDs) assisted laser desorption/ionization mass spectrometric (QDA‐LDI‐MS) strategy was proposed for qualitative and quantitative analysis of a series of carbohydrates. The adsorption of carbohydrates on the modified surface of different QDs as the matrices depended mainly on the formation of hydrogen bonding, which led to higher MS intensity than those with conventional organic matrix. The effects of QDs concentration and sample preparation method were explored for improving the selective ionization process and the detection sensitivity. The proposed approach offered a new dimension to the application of QDs as matrices for MALDI‐MS research of carbohydrates. It could be used for quantitative measurement of glucose concentration in human serum with good performance. The QDs served as a matrix showed the advantages of low background, higher sensitivity, convenient sample preparation and excellent stability under vacuum. The QDs assisted LDI‐MS approach has promising application to the analysis of carbohydrates in complex biological samples. Copyright © 2016 John Wiley & Sons, Ltd. 相似文献

We demonstrate the potential of femtosecond two-color pulse interferometry for in vitro optical glucose monitoring, by dispersion of the group refractive index in a glucose solution sample with respect to a red-color
light and a blue-color light. By comparison with femtosecond one-color pulse interferometry, the basic performance of the
present system with regard to sensitivity, quantitativeness, and tolerance to surrounding disturbances, is evaluated. The
resulting accuracy and precision of glucose determination are 77 and 118mg/dl for 10-mm-sample-thickness, respectively. This
near-common-path configuration of the two-color pulse light provides good stability to fluctuations of sample temperature,
which is important in clinical applications. Considering the performance of femtosecond two-color pulse interferometry as

More than 25 sorbents were tested for uptake of succinic acid from aqueous solutions. The best resins were then tested for
successive loading and regeneration using hotwater. The key desired properties for an ideal sorbent are high capacity, complete
stable regenerability, and specificity for the product. The best resins have a stable capacity of about 0.06 g of succinic
acid/g of resin at moderate concentrations (1–5 g/L) of succinic acid. Several sorbents were tested more exhaustively for
uptake of succinic acid and for successive loading and regeneration using hot water. One resin, XUS 40285, has a good stable
isotherm capacity, prefers succinate over glucose, and has good capacities at both acidic and neutral pH. Succinic acid was
removed from simulated media containing salts, succinic acid, acetic acid, and sugar using a packed column of sorbent resin,
XUS 40285. The fermentation byproduct, acetate, was completely separated from succinate. A simple hot water regeneration successfully
concentrated succinate from 10 g/L (inlet) to 40–110 g/L in the effluent. If successful, this would lower separation costs
by reducing the need for chemicals for the initial purification step. Despie promising initial results of good capacity (0.06
g of succinic/g of sorbent), 70% recovery using hot water, and a recovered concentration of >100 g/L, this regeneration was
not stable over 10 cycles in the column. Alternative regeneration schemes using acid and base were examined. Two (XUS 40285
and XFS-40422) showed both good stable capacities for succinic acid over 10 cycles and >95% recovery in a batch operation
using a modified extraction procedure combining acid and hot water washes. These resins showed comparable results with actual

Aim
To study the effect of long-term glucose feeding on aortic lipid composition by using the time of flight-secondary ion mass spectrometry (TOF-SIMS).Method
Rats were divided into two groups, drinking water with or without 10% glucose from birth to 6 months of age. The aortic wall was dissected out, high-pressure frozen, freeze-fractured, freeze-dried and analyzed by TOF-SIMS using a Bismuth cluster ion source. Surface spectra were taken from standardized regions of the vessel wall.Results
Different peaks, such as cholesterol, fatty acids (FAs) and diacylglycerols (DAGs), were identified by the principal component analysis as carries of variance between two groups. These peaks were then compared by conventional t-test. Our data showed that the intensity of cholesterol, but not FAs and DAGs, was significantly decreased in the glucose-drinking rat. Moreover, the long-term glucose intake changed ratios between different FAs in the aorta.Conclusion
The long-term glucose intake led to decreased cholesterol intensity in the aortic wall and this effect was revealed through a global analytical approach with objective selection of significant variables. 相似文献108.

A glucose tolerance test was performed on dogs by injecting glucose intravenously and measuring the plasma glucose and insulin concentrations versus time. A new complex consisting of CdTe quantum dots (QDs) and glucose oxidase (GOx) has been facilely assembled to achieve considerably enhanced enzymatic activity and a wide active temperature range of GOx; these characteristics are attributed to the conformational changes of GOx during assembly. The obtained complex can be simultaneously used as a nanosensor for the detection of glucose with high sensitivity. A mechanism is put forward based on the fluorescence quenching of CdTe QDs, which is caused by the hydrogen peroxide (H2O2) that is produced from the GOx-catalyzed oxidation of glucose. When H2O2 gets to the surface of the CdTe QDs, the electron-transfer reaction happens immediately and H2O2 is reduced to O2, which lies in electron hole traps on CdTe QDs and can be used as a good acceptor, thus forming the nonfluorescent CdTe QDs anion. The produced O2 can further participate in the catalyzed reaction of GOx, forming a cyclic electron-transfer mechanism of glucose oxidation, which is favorable for the whole reaction system. The value of the Michaelis-Menton constant of GOx is estimated to be 0.45 mM L(-1), which shows the considerably enhanced enzymatic activity measured by far. In addition, the GOx enzyme conjugated on the CdTe QDs possesses better thermal stability at 20-80 degrees C and keeps the maximum activity in the wide range of 40-50 degrees C. Moreover, the simply assembled complex as a nanosensor can sensitively determine glucose in the wide concentration range from micro- to millimolar with the detection limit of 0.10 microM, which could be used for the direct detection of low levels of glucose in biological systems. Therefore, the established method could provide an approach for the assembly of CdTe QDs with other redox enzymes, to realize enhanced enzymatic activity, and to further the design of novel nanosensors applied in biological systems in the future. 相似文献
