Abstract

This paper describes a very versatile laminar flowthrough cell designed to perform enthalpimetric determinations of biochemical substrates in the field of clinical and biochemical analysis. The performance of the apparatus is demonstrated for the glacose/glucose-oxidase system instrument. Sensitivity allows determination of glucose concentrations in the range of 10?3 Moles/litre. Other enzyme-substrate exothermic reactions can be used without any modification of the instrument.  相似文献

The power–time curves of micellar formation of two anionic surfactants, sodium laurate (SLA) and sodium dodecyl sulfate (SDS), in N,N-dimethyl acetamide (DMA) in the presence of various long-chain alcohols (1-heptanol, 1-octanol, 1-nonanol and 1-decanol) were measured by titration microcalorimetry at 298 K. The critical micelle concentrations (CMCs) of SLA and SDS under various conditions at 298 K were obtained based on the power–time curves. Thermodynamic parameters (, and ) for micellar systems at 298 K were evaluated according to the power–time curves and the mass action model. The influences of the number of carbon-atom and the concentration of alcohol were investigated. Moreover, combined the thermodynamic parameters at 303, 308 and 313 K in our previous work and those of 298 K in the present work for SLA and SDS in DMA in the presence of long-chain alcohols, an enthalpy–entropy compensation effect was observed. The values of the enthalpy of micellization calculated by direct and indirect methods were made a comparison.  相似文献

建立灵敏表征鹿茸中具有补益活性的4个部位对两种肠道特征菌群生长代谢影响的评价方法.采用微量热法,在不同给药条件作用下,以产热功率-时间曲线(热谱曲线)的特征参数(生长速率常数(k)、最大产热功率(Pmax)、达峰时间(tp)及有效率(E)等)为指标,对肠道主要有益菌(青春双歧杆菌)生长代谢程度进行客观地量化评价.挑选鹿茸促菌最强的活性部位,进一步考察该部位对病原微生物(金黄色葡萄球菌)生长抑制的作用规律.结果表明,鹿茸不同活性部位促进青春双歧杆菌生长代谢强弱顺序为:正丁醇乙醚氯仿乙酸乙酯.活性最强的正丁醇萃取液对病原微生物(金黄色葡萄球菌)抑制影响的研究表明,正丁醇萃取液在浓度大于200μg·mL-1时,对金葡菌生长具有抑制作用.且tp随药液浓度的增加而相应地延长;k,Pmax均随浓度的增加而相应地减小.微量热法具有灵敏准确、实时在线、重现性好等特点,可用于名贵药材——鹿茸的活性部位的筛选,也为药效物质的研究提供了一个新的研究思路和方法.  相似文献

Stability research on gun propellants has been widely performed by microcalorimetry since the 1980s. TNO Prins Maurits Laboratory has already a broad experience since the early 1970s. In the past many studies were performed, to investigate the influence of oxygen, humidity etc. Less attention was paid to two other important aspects, namely the sample geometry and the filling degree during the microcalorimetric test.A statement in the old Dutch military literature presents the following “It is a well-known fact that the free surface influences the decomposition rate of the gun powder, i.e. unground propellant decomposes slowly in comparison to ground propellant”. This is the same for all types of propellant (Amsterdam, February 1924), which implies research on this topic, related to the stability prediction measured by microcalorimetry is important.Since the decomposition of nitrocellulose is influenced by the amount of oxygen and the surface area, the best way to investigate the stability of gun propellants is to measure it ‘ammunition like’. This means that a combination of the propellant grain size and the filling degree of the ammunition should be used for investigation. For small caliber ammunition the filling degree is close to 95%, and for large caliber bag ammunition (e.g. 155 mm) it is around 60%. As a result on these important aspects, TNO has developed five different sizes of sample vessels, to investigate the propellant grains in the most ‘ammunition like’ condition.In this paper, an overview is given of the influences of relative humidity, different grain sizes and available oxygen. The available oxygen is adjusted by changing the oxygen content of the air or by changing the filling degree.  相似文献

Calorimetric titration and NMR experiments in aqueous phosphate buffer (pH 7.2) at 298.15 K have been done to determine the binding mode, complex stability constants and thermodynamics (ΔG°, ΔH°, and TΔS°) for 1:1 inclusion complexation of water-soluble calix[n]arenesulfonates (CnAS, n = 4 and 6) and thiacalix[4]arene tetrasulfonate (TCAS) with acethylcholine, carnitine, betaine and benzyltrimethylammonium ion. The results show the inclusion complexations are driven by enthalpy (ΔH° < 0), accompanied by negative entropic changes (ΔS° < 0). The binding affinities (C4AS > C6AS > TCAS) are discussed from the viewpoint of CH-π/π-π interactions, electrostatic interactions and size/shape-fit relationship between host and guest.  相似文献

在有限的生长空间内和营养条件下,细胞是无法实现无限生长的.细胞在发展初期,由于本身还不够强大,增大的速率比较低;而发展到后期,又由于衰老和环境的制约,增长速率降低,甚至停滞下来.只有在发展中期,才处于增殖高峰,增长速率较快.细胞群体增殖速率经历“慢一快一慢”的变化过程,导致了生长曲线是一条S型变化曲线[‘,’].对于细菌的生长,只有局部、短时间内符合指数生长特征*,对于整个生长过程中是不符合的,对干理论的有限生长曲线,可设计三种方程式*,以适合有限生长曲线的特征:单分子、自催化或简单逻辑、Gomner…  相似文献

在0.1mol/L的磷酸盐缓冲溶液(pH7.4)中,用微量热法测定了298.15K时漆树漆酶与邻氨基苯酚反应的摩尔反应焓ΔrHm=252.21±3.59kJ/mol,米氏常数Km=7.77×10-3mol/L和表观一级反应速率常数k1=7.23×10-3s-1,及此条件下邻氨基苯酚作底物时的漆酶活性值EA=0.190(IU)。  相似文献

应用微量热学的方法,我们已能成功地测得细菌生长过程的热谱,这种热谱包含着有关细菌生长代谢过程的丰富信息,例如对热谱曲线的指数生长段进行解析,可得出细菌生长的速率常数、激活能和有关的热力学参数。故采用微量热法测定细菌在不同培养温度下的生长速率常数,利用计算机拟合出相应k(速率常数)和T(培养温度)的线性关系式后,若把生长速率为零的温度定义为临界生长温度时,就可以根据由上述实验所得的k~T线性关系式求得细菌的临界生长温度。本工作仍采用微量热法对福氏志贺氏菌(S. flexneri)和大肠埃希氏菌(E. coli)进行实验测定。按文献的方法求出它们在不同温度下的生长速率常数;对于大肠埃希氏菌还用几种不同的培养基分别进行实验测定。  相似文献

将微量热法和透射电镜技术相结合,研究了温度对人卵巢癌细胞代谢的影响,不同温度下癌细胞超微结构的变化和加抗肿瘤药物(嘧福绿)的协同作用.从宏观到微观,深入探讨了热疗与化疗协同作用对肿瘤细胞杀伤作用的机理.实验结果表明,在高温下细胞代谢降低,肿瘤细胞的形态发生一系列恶性变化,且高热可能诱导肿瘤细胞的凋亡;热疗加化疗的协同作用则能加速肿瘤细胞的凋亡,使细胞的能量代谢更低,随着作用时间的延长,最终导致细胞死亡.  相似文献

以光谱技术与微量热技术相结合的方法研究水溶液中金霉素与牛血清白蛋白分子间结合作用的热力学性质.荧光猝灭法测得该反应的结合常数K=2.09×105L/mol,结合位点数n=1.75,微量法测得反应的焓变△rHm= -17.50 kJ/mol; 依据Forster非辐射能量转移机制,得到授体-受体间的结合距离(r1=1.67 nm, r2=1.46 nm)和能量转移效率(E1=0.41, E2=0.66). 金霉素与牛血清白蛋白分子间有较强的结合作用, 且结合力以疏水作用为主.  相似文献
