6-O-(2-hydroxybutyl)-β-CD, as a new β-cyclodextrin (β-CD) derivative, was successfully synthesized. It was used as a chiral selector to separate six chiral drugs, including propranolol, anisodamine, promethazine, ketoconazole, benzhexol, and fenfluramine in nonaqueous capillary electrophoresis (NACE). The effects of the organic solvent, the electrolytes, the concentrations of cyclodextrin derivatives, and the pH of the buffer on the chiral resolution (Rs) were investigated. The baseline separation of enantiomers, including propranolol (Rs = 2.26), anisodamine (Rs = 2.31), promethazine (Rs = 2.42), ketoconazole (Rs = 2.56), benzhexol (Rs = 3.38), and fenfluramine (Rs = 3.04), could be achieved using a buffer of 100 mmol · Lt−1 citric acid and 50 mmol · Lt−1 Tris (hydroxymethyl) aminomethane (Tris) at pH 4.6 containing 100 mmol · Lt−1 6-O-(2-hydroxybutyl)-β-CD in formamide (FA).  相似文献

The resolving ability of 2-O-(2-hydroxybutyl)-beta-CD (HB-beta-CD) with different degrees of substitution (DS = 2.9 and 4.0) as a chiral selector in CZE is reported in this work. Fourteen chiral drugs belonging to different classes of compounds of pharmaceutical interest such as beta-agonists, antifungal agents, ageneric agents, etc., were resolved. The effects of the DS of HB-beta-CD on separations were also investigated. The chiral resolution (R(s)) was strongly influenced by the concentrations of the CD derivative, the BGE, and the pH of the BGE. Under the conditions of 50 mmol/L Tris-phosphate buffer at pH 2.5 containing 5 mmol/L HB-beta-CD, all 14 analytes were separated. The very low concentration necessary to obtain separation was particularly impressive. The DS had a significant effect on the resolution of the chiral drugs and the ionic strength of the separation media; hence, the use of a well-characterized CD derivative is crucial.  相似文献

A charged highly water-soluble CD derivative, 6-O-(2-hydroxy-3-trimethylammoniopropyl)-beta-CD (herein noted as 6-HPTMA-beta-CD) was synthesized and successfully used as a chiral selector for enantiomeric separation of some acidic compounds by CZE in an uncoated capillary. Substitution with 2-hydroxy-3-trimethylammoniopropyl groups at the primary hydroxyl group of the CD was aimed at influencing the magnitude and selectivity of analyte-CD interactions. The behavior of 6-HPTMA-beta-CD was compared with that of the commercially available quaternary ammonium-beta-CD (QA-beta-CD) under the same separating conditions. The experiments were carried out using a BGE consisting of 50 mM phosphate in the pH range of 4-6 by adding a relatively low concentration of chiral selector (less than 10 mM). The effects of the concentration of CD and the pH of the electrolyte on the resolution of these compounds were studied.  相似文献

Determination of enantiomeric purity is most often done under overload conditions, which leads to deformed peaks. In general, the best resolutions are obtained when the small peak appears before the large peak in the electropherogram. To be able to determine the R(+)-impurity in the S(-)-form as well as the S(-)-impurity in the R(+)-form the elution orders have to be reversed. The present paper describes reversal of enantiomeric elution order for the basic analyte propranolol and the acidic analyte ibuprofen. For propranolol, a charged heptakis-(6-sulfo)-beta-cyclodextrin (CD) is used in the background electrolyte. For ibuprofen, a mix of the charged heptakis-(6-sulfo)-beta-CD and the uncharged heptakis-(2,3,6-tri-O-methyl)-beta-CD is used in the background electrolyte. The use of a coated capillary and reversal of the polarity shift the elution order, buffer composition is unchanged in both cases. The enantiomers of propranolol and ibuprofen are well separated on both the coated and uncoated capillaries. Detection limits of enantiomer impurities are investigated using spiked samples of both propranolol and ibuprofen.  相似文献

The complex stability constants (K(S)) and thermodynamic parameters (DeltaG degrees, DeltaH degrees, and TDeltaS degrees ) for 1:1 inclusion complexation of beta-cyclodextrin (beta-CD) derivatives, 6-O-phenyl-beta-CD (2) 6-O-(4-formyl-phenyl)-beta-CD (3), 6-O-(4-nitrophenyl)-beta-CD (4), 6-O-(4-bromophenyl)-beta-CD (5), 6-O-(4-chlorophenyl)]-beta-CD (6), and 6-O-(4-hydroxybenzoyl)-beta-CD (7) with representative guest molecules, cyclic alcohols (cyclopentanol, cyclohexanol, cycloheptanol, cyclooctanol), (+/-)-borneol, and (+/-)-camphor, have been determined by means of titration microcalorimetry in an aqueous phosphate buffer solution (pH = 7.20) at 298.15 K. The results obtained indicate that the introduction to beta-CD of an aromatic ring bearing different substituent groups significantly enhances the molecular binding ability and moderately alters the chiral discrimination ability for the guests examined here, displaying the highest enantioselectivity of up to 4.01 for the inclusion complexation of 6 with (+/-)-camphor. The enhanced molecular/chiral discrimination ability caused by derivatization is attributed solely to increased positive entropy changes due to the expanding hydrophobic interaction and desolvation effects. The binding modes of host-guest interactions derived from ROESY spectroscopy data show that the resulting complex of 4 and (+)-borneol possesses better induced-fit interaction as compared to (-)-borneol, which is responsible for the enhanced molecular/chiral recognition ability.  相似文献

The chiral separation of simendan enantiomers using capillary electrophoresis was studied with beta-cyclodextrin (beta-CD) as chiral selector. The influences of the concentration and pH of borate buffer solution, beta-CD concentration and methanol content in the background electrolyte were investigated. These factors were compared with those in an HPLC with beta-CD as chiral mobile phase additive (CMPA-HPLC). The quantification properties of the developed CE method were examined. A baseline separation of simendan enantiomers was achieved in the background electrolyte of 20 mmol/L borate buffer (pH 11.0) containing 12 mmol/L beta-CD-methanol (50:50 in volume ratio). The CE method is comparable with CMPA-HPLC in chiral resolution, although the optimal pH in CE (11.0) is much higher than that (6.0) in CMPA-HPLC. This chiral CE method is applicable to the quantitative ananlysis and enantiomeric excess value determination of L-simendan.  相似文献

The potential of tert.-butylcarbamoylquinine as chiral selector (SO) added to a non-aqueous background electrolyte for the capillary electrophoretic separation of the enantiomers of N-derivatized amino acids (selectands, SAs) is evaluated. Separation is based on different ion-pair formation equilibrium constants of (R) and (S) enantiomers of the negatively charged chiral analytes with the positively charged quinine-derived chiral SO and on mobility differences of free and complexed SAs, so that differences in the overall migration behavior of the SA enantiomers result. To suppress problems associated with the high UV absorption of the chiral SO and thus the high detector background in the 'total filling technique', the 'partial filling technique' has been adopted. Several parameters including filling time and length of SO zone, respectively, SO concentration, type of background electrolyte, have been evaluated. Using such an optimized method, for example, (R) and (S) enantiomers of 2,4-dinitrophenyl (DNP)-protected proline could be separated with alpha=1.08, R(S)=6.60, and N=130,000 theoretical plates within 15 min. Similar alpha values, resolution, and efficiencies were observed for other DNP-protected, as well as for diverse, N-derivatized amino acids like N-benzoyl, N-9-fluorenylmethoxycarbonyl, N-3,5-dinitrobenzyloxycarbonyl amino acids. A repeatability study clearly validated the robustness of the method and revealed its practical applicability.  相似文献

The aim of this work was to optimize conditions for capillary electrophoresis separation of different neurotransmitters (serotonin, phenylalanine, dopamine, adrenaline, ephedrine, propranolol and DOPA) in a single run, including separation of existing enantiomers. As chiral selectors added to the borate background, electrolyte unsubstituted alpha-, beta- and -gamma-cyclodextrins (CDs), methyl-, dimethyl-, and trimethyl-substituted beta-CDs, and hydroxypropyl-substituted alpha-, beta- and gamma-CDs were examined. Also carboxymethyl-beta-CD and succinyl-beta-CD were used for this purpose. In addition to the kind and concentration of chiral selector, some other experimental factors also have been optimized, such as concentration of borate buffer, content of methanol, pH of electrolyte, method of sample introduction into the capillary and washing procedure between consecutive runs. The best results were obtained using 20 mM carboxymethyl-beta-CD in borate buffer of pH 7.5 as running electrolyte and hydrostatic injection. The obtained sensitivity of response (peak height) varied from 0.4 for adrenalines to 2.3 mAU mM(-1) for propranolols. The concentration detection limits (S/N=3) were in the range from 0.04 mM for propranolols to 0.2 mM for adrenalines. The resolution obtained in optimized conditions in a single run was from 0.75 for adrenalins and 1.0 for propranolols up to 2.0 for ephedrines. The developed method was employed for determination of these analytes in brain tissue extracts.  相似文献

采用高效毛细管区带电泳法,以β-环糊精及其衍生物作为手性选择剂,对外消旋N-(2-甲基-6-乙基苯基)丙氨酸(EMPA)的两个对映体进行了手性分离,比较了环糊精种类、环糊精浓度、电解质溶液pH值、温度和电场强度对分离的影响.实验结果表明,采用2,6-O-二甲基-β-环糊精为手性选择试剂,环糊精浓度为40mmol/L、电解质溶液pH=5.5及温度为20℃时分离效果最佳,对映体基本达基线分离,线性范围为20~200mg/L,最低检测限为10mg/L.  相似文献

采用C18反相色谱柱,利用在流动相中加入手性选择剂的方法实现酮康唑对映体的拆分。研究了手性选择剂的种类及浓度、流动相pH值、甲醇比例和柱温等因素对酮康唑手性分离的影响,结果表明磺丁基-β-环糊精可以使酮康唑对映体完全分离,最后选择的流动相组成为甲醇-0.02 mol/L磷酸二氢钠(体积比为60∶40,含0.02%三乙胺和1.0 mmol/L磺丁基-β-环糊精,用稀磷酸调节pH值到3.00)。酮康唑对映体在6 min内得到基线分离,分离度为2.05。方法简便,分离效果好,对酮康唑对映体的拆分具有应用价值。  相似文献

The capillary electrochromatographic separations of three acidic enantiomers (carprofen, coumachlor and warfarin) were studied on a capillary column packed with 5 microm (3R,4S)-Whelk-O 1 chiral stationary phase. The influence of several experimental parameters (mobile phase pH, type of background electrolyte, acetonitrile ratio, temperature, applied voltage and ionic strength) on electroosmotic flow velocity, retention factor, selectivity factor, efficiency, resolution and effectiveness of chiral separation was evaluated. It was notable that the optimum resolution of the acidic enantiomers was achieved at pH 3.0 phosphate buffer, suggesting that capillary electrochromatography in the ion-suppressed mode can be applied for chiral separations of a range of acidic compounds.  相似文献

EKC using a neutral CD as chiral selector was applied in this work to the development of a method enabling the enantiomeric separation of ketoconazole and terconazole antifungals. The influence of different experimental conditions such as temperature, CD concentration, pH, and nature and concentration of the buffer on the enantiomeric resolution of the compounds studied was investigated. The use of 10 mM heptakis-(2,3,6-tri-O-methyl)-beta-CD in a 100 mM phosphate buffer (pH 3.5) with a temperature of 15 degrees C allowed the separation of the enantiomers of ketoconazole and terconazole with high resolution (R(s) > 2.0). The rapid separation of ketoconazole enantiomers with an analysis time less than 3 min was carried out after fitting some experimental parameters. The developed method was applied to the determination of ketoconazole in different pharmaceutical formulations.  相似文献

The enantiomeric separation of ofloxacin enantiomers (OFLX) was achieved by using capillary electrophoresis partial-filled with Escherichia coli, Pseudomonas aeruginosa (Gram-negative), and Staphylococcus aureus (Gram-positive) as chiral selectors. Experimental parameters, including the concentration of background electrolyte, applied voltage, length of the filled bacteria plug, and pH of the buffer, were intensively investigated. Baseline separation of OFLX could be achieved within 7 min by using E. coli and P. aeruginosa as chiral selectors under the following conditions: electrophoretic buffer composed of 10 mM phosphate buffer at pH 7.4, applied voltage at 15 kV, and the bacteria (6.0 × 10(8) cells/mL) were injected into the capillary by gravity with injection height of 17.5 cm for 180 s (E. coli), 300 s (P. aeruginosa), and 300 s (S. aureus), respectively. E. coli and P. aeruginosa had better chiral selectivity for OFLX than S. aureus, which was in good agreement with OFLX having better antimicrobial activity on Gram-negative rather than Gram-positive bacteria. A novel method was developed for the enantioselective separation of enantiomers using bacteria as chiral selectors, which provides a new approach for antimicrobials enantioselective analysis, chiral pharmacodynamics, and chiral pharmacokinetics studies.  相似文献

A selective capillary electrophoresis method using sulfobutyl ether-β-cyclodextrin as a chiral selector was developed and validated for the determination of the enantiomeric impurity of (R)-modafinil, i.e., armodafinil. Several parameters were optimized for a satisfactory enantioresolution, including the type and concentration of chiral selector and organic modifier, pH of background electrolyte (BGE), capillary temperature. The finally adopted condition was: 20 mmol/L phosphate buffer at pH 7.5, containing 20 mmol/L sulfobutyl ether-β-cyclodextrin and 20% methanol, at temperature of 25 °C. A good resolution of 3.3 for the two enantiomers of modafinil was achieved by applying the optimal conditions. The limit of detection (LOD) and limit of quantification (LOQ) of (S)-modafinil were 1.25 μg/mL and 2.50 μg/mL, respectively. The established method was also proven to display good selectivity, repeatability, linearity and accuracy. Finally, the method was used to investigate the enantiomeric purity of armodafinil in bulk samples.  相似文献

To date, a series of chiral selectors have been utilized successfully in capillary electrophoresis (CE). Among these various chiral selectors, macrocyclic antibiotics have been demonstrated to represent powerful enantioselectivity towards many chiral compounds. Differing from macrocyclic antibiotics, the use of lincosamide antibiotics as chiral selectors has not been reported previously. In our recent work, clindamycin phosphate belonging to the group of lincosamides has been first used as a chiral selector in capillary zone electrophoresis (CZE). In this paper, a micellar electrokinetic chromatography (MEKC) method has been developed for the evaluation of enantioseparation capability of this novel chiral selector towards several racemic basic drugs. As observed during the course of this work, clindamycin phosphate allowed excellent separation of the enantiomers of nefopam, citalopram, tryptophan, chlorphenamine, propranolol and metoprolol, as well as partial enantioresolution of tryptophan methyl ester and cetirizine. In this MEKC chiral separation system, different types of anionic surfactants, organic additives and background electrolytes were tested, and satisfactory enantioseparations of basic drugs above-mentioned were achieved using sodium dodecyl sulfate (SDS) as the surfactant, isopropanol as the organic additive, and phosphate as the background electrolyte. Furthermore, both migration times and enantioseparation of the analytes were influenced by several experimental parameters such as pH of the BGE, clindamycin phosphate and SDS concentrations, phosphate and isopropanol concentrations, and applied voltage. Consequently, the effects of these factors on enantioseparations of the studied basic drugs were systematically investigated in order to evaluate the stereoselectivity of clindamycin phosphate in MEKC.  相似文献

Enantioseparations of five chiral phenothiazines in CD-modified CZE using the single isomer sulfate-substituted beta-CD (heptakis(2,3-dihydroxy-6-O-sulfo)-beta-CD, SI-S-beta-CD) and dual CD systems consisting of SI-S-beta-CD and a neutral CD as chiral selectors in a citrate buffer at pH 3.0 were investigated. The results indicate that SI-S-beta-CD is an excellent chiral selector for enantioseparation of promethazine. The enantiomers of trimeprazine were well separated, while those of ethopropazine could also be baseline-resolved with SI-S-beta-CD. With dual CD systems, especially with hydroxypropyl-beta-CD (HP-beta-CD) as neutral CD, the enantioselectivity of thioridazine and ethopropazine was considerably enhanced. Effective enantioseparation of phenothiazines, except for methotrimeprazine, could thus be favorably and simultaneously achieved. Moreover, reversal of the enantiomer migration order of ethopropazine and thioridazine occurred by varying the concentration of gamma-CD in the presence of SI-S-beta-CD. These phenomena may be attributable to the opposite effects of sulfated beta-CD and gamma-CD on the mobility of the enantiomers of ethopropazine and of thioridazine. Comparative studies on the enantioseparations of phenothiazines with single CD and dual CD systems containing SI-S-beta-CD and randomly sulfate-substituted beta-CD (MI-S-beta-CD) were made.  相似文献

The enantiomers of D- and L-tryptophan were separated by capillary electrophoresis, using alpha-cyclodextrin as a chiral active component in the background electrolyte. The separation of (-) and (+) epinephrine was achieved by supplementing the background electrolyte with Heptakis (2,6-di-O-methyl-beta-cyclodextrin). This paper reports a new chiral separation technology—biphasic recognition chiral extraction for the separation of aromatic acid enantiomers such as α-cyclohexyl-mandelic acid (CHMA) and naproxen (NAP). The biphasic recognition chiral extraction system was established by adding hydrophobic d(l)-isobutyl tartrate in 1,2-dichloroethane organic phase and hydrophilic β-cyclodextrin (β-CD) derivative in aqueous phase, which preferentially recognize the (R)-enantiomer and (S)-enantiomer, respectively. These studies involve an enantioselective extraction in a biphasic system, where aromatic acid enantiomers form complexes with the β-cyclodextrin derivative in the aqueous phase and d(l)-isobutyl tartrate in the organic phase, respectively. Factors affecting the extraction mechanism are analyzed, namely the influence of the concentrations of the extractants and aromatic acid enantiomers, the types of the extractants, pH, and temperature. The experimental results show that the biphasic recognition chiral extraction is of much stronger chiral separation ability than the monophasic recognition chiral extraction, which utilizes the cooperations of the forces of the tartrate and the β-CD derivative. Hydroxypropyl-β-cyclodextrin (HP-β-CD), hydroxyethyl-β-cyclodextrin (HE-β-CD), and methyl-β-cyclodextrin (ME-β-CD) have stronger recognition abilities for the (S)-aromatic acid enantiomers than those for (R)-aromatic acid enantiomers, among which HP-β-CD has the strongest ability. d-Isobutyl tartrate preferentially recognizes (R)-CHMA and (S)-NAP, while l-isobutyl tartrate preferentially recognizes (S)-CHMA and (R)-NAP. The maximum enantioselectivities of CHMA and NAP are 2.49 and 1.65, under conditions at which the pH values of the aqueous phases are 2.7 and 2.5, at the ratio of 2:1 of [isobutyl tartrate] to [HP-β-CD].  相似文献
