采用反相加压毛细管电色谱与紫外检测联用技术,建立了一种高效、简便的糖皮质激素分析方法,适用于头发中糖皮质激素的检测。使用C18反相色谱柱,流动相为pH 8.0, 1.5 mmol/L的Tris-乙腈(65:35, v/v),检测波长为245 nm、分离电压为~10 kV、反压为10.5 MPa、泵流速为0.05 mL/min,进行等度洗脱,倍他米松、地塞米松、泼尼松、泼尼松龙、醋酸泼尼松龙、醋酸氢化可的松、醋酸可的松、皮质脂酮等8种激素在20 min内实现快速分离。各组分的质量浓度线性范围达到3个数量级,检出限(S/N=3)在μg/g水平,迁移时间和峰面积的相对标准偏差(RSD)分别小于4.8%和7.4%。将所建立方法应用于头发样品分析,检测前采用蛋白酶水解提取和净化处理样品,不同浓度糖皮质激素的回收率为71%~85%。该研究为糖皮质激素药物暴露监测以及压力检测提供了新手段,有望用于滥用药物的控制和临床诊断。  相似文献

A novel method was developed for the simultaneous determination of glucocorticoid residues such as triamcinolone (TR), prednisolone (PR), hydrocortisone (HC), cortisone (CO), methylprednisolone (MP), dexamethasone (DE) and triamcinolone acetonide (TA) by high-performance liquid chromatography (HPLC) coupled with chemiluminescence (CL) detection. The procedure was based on the enhancement effect of glucocorticoids on the chemiluminescence reaction between luminol and the complex of trivalent copper and periodate ([Cu(HIO6)2]5-), which was on-line electrogenerated by constant current electrolysis. The HPLC separation used a Nucleosil RP-C18 column (250 mmx4.6 mm i.d., 5 microm, pore size, 100 A) with a mobile phase consisting of acetonitrile and 1.0 mmol L(-1) ammonium acetate (pH 6.8, 40:60,v/v) at a flow rate of 0.8 mL min(-1). The effects of several parameters on the HPLC resolution and CL emission were studied systematically. Liver samples were hydrolyzed with Helix pomatia juice followed by a solid-phase extraction procedure. Under optimum conditions, the limits of detection (LOD) at a signal-to-noise of 3 ranged from 0.08 to 1.0 ng g(-1) and the limits of quantification (LOQ) at a signal-to-noise of 10 ranged from 0.27 to 3.33 ng g(-1) for seven glucocorticoids. The relative standard deviations (RSD) of intra- and inter-day precision were below 6.8%. The average recoveries for glucocorticoids (spiked at the levels of 5-50 ng g(-1)) in pig liver ranged from 88 to 106%, and the relative standard deviations of the quantitative results were from 2.0 to 6.9%. The proposed method had been successfully applied to the determination of glucocorticoid residues in pig liver.  相似文献

建立了以离子液体和β-环糊精为添加剂的反向微乳毛细管电泳(MEEKC)法分离测定化妆品中丙酸氟替卡松、曲安奈德、醋酸可的松、地塞米松、氢化可的松和泼尼松6种激素的方法.在优化的实验条件下,6种物质于12min内达到基线分离,丙酸氟替卡松在2.0~200mg/L范围内线性关系良好,其它组分在2.0 ~ 500mg/L范围内线性关系良好,检出限(S/N =3)分别为0.75、0.81、0.66、0.68、0.74、0.86 mg/L.方法用于化妆品样品测定,加标回收率在93.5%~105%之间,RSD均小于4.3%(n=3).  相似文献

A method is described for extraction, separation and quantification of 3 corticosteroids in tablets (triamcinolone, prednisolone and dexamethasone). The extraction was achieved using a solid phase extraction (SPE) on RP-18 cartridges. The steroids eluates were examined by HPLC. The chromatographic separation was carried out on Chromolith® Performance RP-18e column, a new packing material consisting of monolithic rods of highly porous silica, using isocratic binary mobile phase of MeOH and H2O in the ratio 1: 1. A diode array detector was used at 254 nm for detection. The method was validated for system suitability, linearity, precision, limits of detection, specificity, stability and robustness. The limits of detection were 6.25, 6.25 and 12.50 ng per 25 L injected volumes for them respectively. The recovery values of this method were between 89 and 101% and the reproducibility was within 6.86.  相似文献

Various extraction techniques can be combined with column liquid chromatography (LC) and ultraviolet (UV) or mass spectrometric (MS) detection for the determination of synthetic corticosteroids in biological matrices. Target analysis of low concentrations of 25 microg/kg of dexamethasone in feed can be performed by combining immunoaffinity chromatography (IAC) and LC with UV detection. A straightforward multi-analyte procedure is obtained by tandem solid-phase extraction (SPE) and subsequent LC-UV. However, the limits of detection for feed samples are then relatively poor, viz. 100 microg/kg. A multi-analyte method which meets modern demands of about 5 microg/kg detection limit requires one-step SPE combined with LC-MS analysis. As regards urine corticosteroids can be determined down to a level of 0.5 microg/l by either SPE-LC-MS- MS or SPE(IAC)-LC-MS.  相似文献

Automatic SPE has been coupled on-line to CE by a transfer tube and the replenishment system of the CE instrument. The approach allows the target analytes (viz. creatinine, creatine, xanthine, hypoxanthine, uric acid, p-aminohippuric acid and ascorbic acid in urine samples) to be removed from the sample matrix, cleaned up, preconcentrated and injected into the capillary. The detection limits range between 0.14 and 4.50 microg/mL, the quantification limits between 0.45 and 15.0 microg/mL, and linear dynamic ranges - which include the reference healthy human values - from the quantification limits to 1332 microg/mL. The precision, expressed as RSD, ranges between 0.38 and 2.22% for repeatability and between 1.79 and 7.61% for within-laboratory reproducibility. The errors, expressed as RSD for all compounds, range between 0.20 and 6.90%. The time for automatic SPE and that necessary for the individual separation-detection of the target analytes are 13 and 12 min, respectively; the analysis frequency is 5 h(-1). The accuracy of the method and potential matrix effects were studied by using spiked samples and recoveries between 96.00 and 103.07 % were obtained. We have developed a micellar electrokinetic chromatography (MEKC) method using bile salts for the simultaneous determination of six corticosteroids, including betamethasone, cortisone, prednisolone, 6alpha-methylprednisolone, triamcinolone, and prednisone. The separation was performed using borate buffer containing sodium cholate and sodium deoxycholate. Several parameters were studied, including bile salt concentrations, concentrations and pH of borate buffer, and analytical voltages. In method validation, calibration curves were linear over a range of 10-100 microM for each corticosteroid. The RSD (relative standard deviation) and RE (relative error) were all less than 5% for intra- and interday assays. The limit of detection of each analyte was 5 microM. The recoveries were greater than 95%. Application of this method for quality control of commercial tablets also proved to be feasible. All analytical values fall within the labeled amount of 90-110% for betamethasone and prednisolone, and of the labeled amount of 92.5-107.5% for 6alpha-methylprednisolone, as required by the United State Pharmacopeia 25 (USP 25).  相似文献

A novel microemulsion based on a mixture of diethyl L-tartrate (DET) and SDS was developed for the microemulsion EKC (MEEKC) determination of structurally related steroids. The system consisted of 0.5% w/w DET, 1.7% w/w SDS, 1.2% w/w 1-butanol, 89.6% w/w phosphate buffer (40 mM, pH 7.0), and 7% w/w ACN. With an applied voltage of +10 kV, a baseline separation of aldosterone (A), cortisone acetate (CA), dexamethasone (D), hydrocortisone (H), hydrocortisone acetate (HA), prednisolone (P), prednisolone acetate (PA), prednisone (Ps), triamcinolone (T), and triamcinolone acetonide (TA) could be achieved. Under the optimized conditions, the reproducibility of the retention time (n = 4) for most of the compounds was less than +/-0.8% with the exception of A, Ps, and T. The average number of theoretical plates was 18 800 plates/m. The results were compared with those achieved by the modified micellar EKC (MEKC). MEEKC showed obvious advantages over MEKC for the separation of highly hydrophobic substances. To further evaluate the system, we tested the MEEKC method by analyzing corticosteroids in a spiked urine sample.  相似文献

建立了鸡肉中3种氯霉素类抗生素残留的高效液相色谱-电喷雾电离三级四极杆质谱(HPLC-ESI-MS-MS)测定法。该方法采用多反应监测(MRM)负离子模式,可一次对鸡肉中的氯霉素、甲砜霉素和氟甲砜霉素进行定性和定量。该方法仅需1 g样品,并省去固相萃取步骤,具有操作简便、有机试剂消耗量少、测定周期短等优点。方法的检出限为0.010 μg/kg,测定低限为0.100 μg/kg,线性范围为0.050~1.00 μg/L,加标回收率为69.0%~92.8%,相对标准偏差为6.3%~12.9%。  相似文献

A multiresidue method for the determination of 12 glucocorticoids (clobetasol propionate, budesonide, triamcinolone, triamcinolone acetonide, fludrocortisone acetate, flumethasone, beclomethasone, prednisone acetate, 6‐α‐methylprednisolone, hydrocortisone, cortisone, and prednisone) in bovine milk was developed using liquid chromatography with tandem mass spectrometry. Isoflupredone was used as an internal standard. Milk samples were treated with ethyl acetate to extract glucocorticoids and were frozen at −20°C for 6 h to precipitate fat. The extract was dried under nitrogen, and residues were dissolved in an acetonitrile/water solution. A further clean‐up step was used by dispersive solid‐phase extraction, with octadecyl silica and primary secondary amine as the absorbents. The recoveries of glucocorticoids spiked at 0.5, 1.0, 10.0 μg/kg ranged from 75.7 to 117.3%, except for clobetasol propionate and budesonide (16.1–49.5%). The limits of quantification were 0.01–0.5 μg/kg in milk. This method has been successfully applied in real samples. The results demonstrated that this method is simple, robust, and suitable for identification of glucocorticoid residues in milk.  相似文献

A multiresidue method was developed and validated to screen bovine urine samples for 10 beta-2-adrenergic agonistic drugs--brombuterol, cimaterol, clenbuterol, clenpenterol, isoxsuprine, mabuterol, ractopamine, ritodrine, salbutamol, and tulobuterol--at the 2 microg/L level. The method is also quantitative in the range of 1 to 4 microg/L for all analytes except salbutamol. The procedure uses enzymatic digestion, liquid-liquid extraction, and cleanup on solid-phase extraction columns, followed by detection using a liquid chromatograph-tandem quadrupole mass spectrometer operated in the positive-ion atmospheric pressure chemical ionization multiple-reaction monitoring mode. Method validation included assessment of recoveries, repeatabilities, linearity of responses, decision limits, and detection capabilities. Overall average recoveries ranged from 70-91%; recoveries were generally lower for salbutamol. The decision limits ranged from 0.4-1.0 microg/L, and detection capabilities from 0.6-1.7 microg/L.  相似文献

采用了基质固相分散萃取(MSPD)和固相萃取(SPE)技术分别对奶制品(奶粉和牛奶)中6种雌激素进行提取和净化。结果显示,MSPD适用于固体奶粉的处理,而SPE则适用于液体牛奶的处理。基于优化结果,利用高效液相色谱-三重四极杆-复合线性离子阱质谱(HPLC-Q-TRAP-MS)建立了在不同奶制品中同时测定6种雌激素含量的方法。方法学考察结果显示,建立的分析方法符合含量测定要求,在0.1~200 mg/L(雌三醇为0.1~20 mg/L)范围内线性关系良好(相关系数(R2)>0.99);检出限(LOD,S/N=3)和定量限(LOQ,S/N=10)分别为0.01~0.05 mg/L和0.05~0.10 mg/L。在添加水平分别为1.0、5.0和10 mg/kg时,固态奶粉经MSPD处理后,6种雌激素的平均回收率为71.8%~106.0%(RSD为1.6%~9.2%,n=3);液态牛奶经SPE处理后,6种雌激素的平均回收率为70.3%~108.4%(RSD为2.0%~11.0%,n=3)。该方法灵敏度和重复性高,适于分析复杂基质中雌激素的痕量残留。  相似文献

建立了采用超高效液相色谱同时测定化妆品中糖皮质激素、雌激素、雄激素、孕激素等共15种激素(曲安西龙、氢化可的松、泼尼松、可的松、甲基泼尼松龙、倍他米松、地塞米松、醋酸泼尼松龙、醋酸氢化可的松、雌三醇、雌二醇、雌酮、己烯雌酚、睾酮、孕酮)的分析方法。不同形态的化妆品样品均以甲醇为提取溶剂进行超声提取,经Oasis HLB固相萃取柱富集净化,以Waters ACQUITY UPLC BEH C18色谱柱(1.7 μm,2.1 mm×50 mm)分离,乙腈和水为流动相梯度洗脱,6 min 内完成了15种激素的分离及检测。在1~25 ng进样范围内,15种激素的工作曲线的线性相关系数r均高于0.9995。在低、中、高(2,10,20 mg/kg)3个添加水平下15种激素的平均回收率为88.2%~102.4%,相对标准偏差为1.6%~7.4%。  相似文献

In this work we have optimized the analysis of 18 human corticosteroids, some endogenous (tetrahydrocortisol, tetrahydrocortisone, cortisol, and cortisone) and others synthetic (betamethasone, budesonide, cortisone acetate, desonide, dexamethasone, dexamethasone acetate, flunisolide, fluocinolone acetonide, halcinonide, methylprednisolone, prednisolone, prednisone, triamcinolone, and triamcinolone acetonide). Three analytical techniques were developed: ELISA, gas chromatography coupled with mass spectrometry (GC–MS), and liquid chromatography coupled with mass spectrometry (LC–MS). Several sample-preparation methods were optimized for each technique and enabled compounds of interest to be extracted from small urine samples (several mL). The results enabled us to assess the possibilities and the sensitivity of each technique for application to doping tests.  相似文献

A capillary gas chromatographic-mass spectrometric method for the simultaneous determination of cortisol, cortisone, prednisolone and prednisone in human plasma is described. [1,1,19,19,19-2H5]Cortisol, [1,1,19,19,19-2H5]cortisone, [1,19,19,19-2H4]prednisolone and [1,19,19,19-2H4]prednisone were used as internal standards. Formation of the bismethylenedioxy-3-heptafluoro-n-butyryl (BMD-HFB) derivatives made possible the separation of the four corticosteroids with good gas chromatographic behaviour. The new double derivatization has been demonstrated to be of value for sensitive and selective quantification by this technique. Detection was performed by monitoring the molecular ion (M+) of the BMD-HFB derivatives for cortisone and prednisolone, the [M - 18]+ ion for cortisol, and the [M - 30]+ ion for prednisone. The method requires no complex corrections for contributions and provides good accuracy and precision.  相似文献

A new synthesized copolymer based on N-vinylimidazole-divinylbenzene (VIm-DVB) was tested as a sorbent for the solid-phase extraction (SPE) of polar analytes. In the on-line SPE, this synthesized sorbent enabled 100 ml of sample to be preconcentrated with recoveries as high as 80% for oxamyl, phenol (Ph) and derivates, bentazone and (4-chloro-2-methylphenoxy)acetic acid (MCPA). For the off-line SPE, 1000 ml of sample was extracted and recoveries were higher than 92% for all compounds with the exception of oxamyl (83%) and methomyl (78%). The VIm-DVB sorbent gives better recoveries than the previously synthesized 4-vinylpyridine-divinylbenzene (VP-DVB) resin and similar to such highly crosslinked commercial sorbents as LiChrolut EN or Oasis HLB. Real water samples were used to validate the on-line SPE method. Linearity was good and detection limits were between 0.1 and 0.2 microg l(-1).  相似文献

建立了鸡肉和鸡蛋中合成类固醇类激素(睾酮、甲基睾酮、群勃龙、勃地龙、诺龙、美雄酮、司坦唑醇、丙酸诺龙、丙酸睾酮及苯丙酸诺龙)和糖皮质类激素(泼尼松、泼尼松龙、地塞米松、氟氢可的松、甲基泼尼松、倍氯米松及氢化可的松)多残留的液相色谱-串联质谱(LC-MS/MS)检测方法.样品经乙腈超声提取,正己烷脱脂净化,以甲醇-甲酸水溶液为流动相,经C18柱分离后进行LC-MS/MS选择反应监测模式下的定性及定量分析.合成类固醇类激素采用正离子模式检测,糖皮质激素则采用负离子模式检测,正、负离子化模式一次进样同时检测.类固醇类和糖皮质类激素的定量检出限为0.5 μg/kg.在0.5、 1.0和5.0 μg/kg 3种浓度添加水平,上述激素的平均回收率为73.4%~108.9%;相对标准偏差为3.4%~13.4%.可实现样本灵敏、准确地定性定量分析.  相似文献

建立了食品中多杀菌素(spinosyn) A和D残留的液相色谱-串联质谱(HPLC-MS/MS)检测方法。甘蓝等食品样品经乙腈-水(50:50, v/v)提取、HLB固相萃取柱净化后,采用HPLC-MS/MS进行检测分析,利用外标法定量。质谱分析采用电喷雾电离,正离子扫描,多反应监测模式。结果表明,柱净化后无明显的基质效应,多杀菌素在1~20 μg/L内具有较好的线性关系,其相关系数可达0.9999;样品中添加1~10 μg/kg的多杀菌素,其回收率为76.2%~114.0%,相对标准偏差(n=10)小于10%;检出限(信噪比(S/N)=3): 多杀菌素A为0.2 μg/kg,多杀菌素D为0.5 μg/kg;定量限(S/N=10): 多杀菌素A为0.5 μg/kg,多杀菌素D为1.0 μg/kg。在最佳实验条件下,对来自福建厦门的969份食品样品进行了检测,其中15份样品检测结果呈阳性。实验结果表明,该方法提取效果好,具有良好的灵敏度、回收率和重复性。  相似文献

Amphiphilic polymeric micelle, as a novel pseudostationary phase in EKC was used to determine eight kinds of corticosteroids namely hydrocortisone, prednisolone, hydrocortisone acetate, prednisone, cortisone acetate, prednisolone acetate, dexamethasone, and triamcinolone acetonide in cosmetics. Amphiphilic random copolymer poly(methyl methacrylate‐co‐methacrylic acid) (P(MMA‐co‐MAA)) was micellizated via neutralization in alkaline aqueous solution. The influences of the molar ratio of monomer MMA to MAA, the concentration of polymer and pH on the polymeric micelle microstructure and EKC performances were investigated. As molar ratio of MMA to MAA in P(MMA‐co‐MAA) increased, both CMC and environmental polarity of the inner core in polymeric micelle decreased dramatically. With increasing monomer ratio, the size of polymeric micelles increased firstly, and then decreased, finally increased again. ζ potential of the micelle had a slight decline trend. As increment of polymer concentration, the size of the polymeric micelle increased steadily. By optimizing the monomer ratio, the polymer concentration, and pH of the running buffer, as well as operation conditions such as separation voltage and temperature, the eight analytes could be separated within 16.5 min using 7.5 mg/mL polymer with the monomer ratio of 7:3 dissolved in pH 9.2 borax buffer as the running buffer. The method has been used for analysis of corticosteroids in cosmetic samples with simple extraction; the recoveries for eight analytes were between 85.9 and 106%. This method was of accuracy, repeatability, pretreatment simplicity, and could be applied to the quality control of cosmetics.  相似文献
