研究了9种金属离子对一株南海红树内生真菌B77生长代谢的影响。结果表明,质量浓度为0.15 mg/mL的Zn2+可明显促进该真菌的生长,菌丝体增多,并且使代谢时间缩短,代谢产物产量提高。结果表明,适量的金属离子可以对该真菌的生长,次级代谢产物的生产具有明显的作用。  相似文献

枯草芽孢杆菌63501(Bacillus subtilis 63501)培养液在连续培养时分别通入不同强度的直流电场,研究了它们在持续直流电刺激下的生长曲线、pH曲线、ATP酶活性、胞内总蛋白含量、细胞膜通透性以及细菌微观形态变化。 结果表明,适宜的直流电能促进枯草芽孢杆菌增殖;枯草芽孢杆菌的ATP酶活性在0.045 5×10-3 A/cm2电流密度作用下,在对数生长前期、后期和稳定期分别比对照组同期提升1.9、3.5和3.8倍,表明适宜强度的电流增强了细菌细胞的代谢活性。 细菌通透性也有不同程度的提高,与透射电子显微镜观察到的细菌细胞通电后的形态变化具有一致性。  相似文献

建立灵敏表征乌头类双酯型生物碱(DDA)对嗜热四膜虫生长代谢影响的评价方法. 采用微量热法, 在不同给药条件下, 以表达功率-时间曲线(热谱曲线)的特征参数生长速率常数(k)、半数抑制浓度(IC50)、最大输出功率(Pmax)及达峰时间(tp)和总产热量(Qt)为指标, 对嗜热四膜虫生长代谢程度进行客观地量化评价. 结果表明, 随着3种DDA浓度的增加, 达峰时间tp延长, 生长速率常数k、最大输出功率Pmax等随之降低, 且生长速率常数k与相应的浓度间呈现良好的线性关系: r乌头碱=0.9910 (IC50=207.7 μg•mL-1); r新乌头碱=0.9923 (IC50=412.5 μg•mL-1); r次乌头碱=0.9977 (IC50=1497 μg•mL-1). 同时由半抑制浓度IC50得到3种DDA毒性的大小顺序: 乌头碱>新乌头碱>次乌头碱. 初步构效关系研究表明, 在化合物C(3)上引入羟基(OH)以及N原子上引入乙基(CH2CH3)均可明显增加双酯型生物碱对嗜热四膜虫的毒性作用.  相似文献

以5,10,15,20-四(4′-吡啶基)卟啉为原料,合成了一系列季铵盐型金属卟啉1a,1b和2a~2c.使用LKB2277热活性监测器测定了大肠杆菌在不同浓度的金属卟啉1a,1b和2a~2c作用下的产热曲线,得到了大肠杆菌生长速率常数k,最大发热功率Pmax和最大发热功率的出现时间tp,发现季铵盐型金属卟啉2c对大肠杆菌生长代谢的抑制活性明显大于其它季铵盐型金属卟啉.  相似文献

用微量热法研究一种新型希夫碱及其3d,4f配合物(2L, 2LZnYb)对大肠杆菌和金黄色葡萄球菌的抗菌活性, 得到了在它们作用下大肠杆菌和金黄色葡萄球菌生长代谢的产热曲线, 并且基于分析生长代谢和非生长代谢的产热曲线建立的热动力学方程, 获得了它们的抗菌活性. 结果表明, 两种化合物(ZL, 2LZnYb)对大肠杆菌的生长代谢有强的活性(IC50分别为6.1 和5.1 mg·L-1), 但对金黄色葡萄球菌的生长代谢的活性弱得多(IC50分别为310.1 和595.5 mg·L-1). Zn 和Yb的导入使化合物对大肠杆菌生长代谢的抑制作用稍微增加, 但大大降低了对金黄色葡萄球菌的抑制作用. 对于非生长代谢, 两种化合物的活性有很大的差别. 无论对大肠杆菌还是金黄色葡萄球菌, 由于配体2L的导入, 表现出显著的抑制作用, 2L的MSC50为6.4和209.7 mg·L-1. 配体2L可能成为新的抗菌先导化合物.  相似文献

Abstract— We have recently reported that DNA double-strand breaks arise enzymatically during the course of excision repair in uvr + strains of Escherichia coli K-12. Survival curves for ultraviolet (UV) irradiated E. coli K-12 pol+ (JG139) and polA1 (JG138) strains have a pronounced shoulder region. The regions of the survival curves at which killing approaches exponential correspond to the fiuences at which DNA double-strand breaks (assumed to be lethal events) accumulate linearly. Reducing the number of UV photoproducts either by photoreactivation or fluence fractionation results in an increase in survival and a decrease in the yield of DNA double-strand breaks in both strains. These data support the hypothesis that enzymatically-induced DNA double-strand breaks may be the lesion ultimately responsible for UV-induced cell killing in the pol+ strain of E. coli K-12. and perhaps also in the polA1 strain.  相似文献

以5,10,15,20-四(对羟基苯基)卟啉(2)为原料,合成并表征了一系列 水溶性和非水溶性的金属卟啉。使用LKB 2277热活性监测器测定了大肠杆菌在金属 卟啉4a ~ 4f和7a ~ 7f作用下的生热曲线,得到了不同金属卟啉在不同浓度下大 肠杆菌生长代谢的生热速率常数k,最大发热功率p_(max)和最大发热功率的出现时 间t。结果发现,含有吡啶溴化盐的水溶性金属卟啉7a ~ 7f对大肠杆菌生长代谢 的抑制活性要明显大于含有酯基的金属卟啉4a ~ 4f。  相似文献

The microcalorimetric method was used to study the antibacterial activity of two newly synthesized Schiff basecompounds(H_2L~(3')and H_2L~3)on Escherichia coli,trying to obtain the action on both of multiplying bacteria andnon-multiplying bacteria at one experiment.The metabolic power-time curves of the bacteria treated with the com-pounds were obtained,and the thermokinetic parameters were analyzed,from which the antibacterial activities ofthese compounds were evaluated.The results showed that both of the two compounds have good activity on aerobicmultiplying metabolism of E.coli,with the value of IC_(50)75.8 and 168.8 mg/L respectively,but have not effectiveaction on fermentation metabolism of E.coli.The action of the compounds on the non-multiplying metabolism wasinvestigated by taking the heat output of E.coli in the stationary phase as the guideline of the activity.The value ofMSC_(50)(minimum stationary-cidal concentration 50)of them is 118 and 187.5 mg/L,respectively.So,H_2L~(3')hasstronger antibacterial action on E.coli than H_2L~3 either for multiplying bacteria or non-multiplying bacteria,andtheir activity on the aerobic multiplying bacteria of E.coli is mainly shown.It does strongly suggest that the calo-rimetric method should play an important role in the fight against the drug-resistant bacteria.  相似文献

Since Schiff base and its metal complexes are of antibacterial and anticancer bioactivity, it has been a research subject of much interest. It is reported that Cu(Ⅱ)-SG could inhibite the synthesis of O2- markedly and could also combine with the salmn sperm DNA. The paper repored that by using microcalorimetric method The growth meloabolism of E.coli at different temperature and reacted upon by Cu(Ⅱ)-SG. The power-time curves of E.coli reacted with Cu(Ⅲ)-SG at different temperature has been determined by LKB2277 Microcalorimetric Monitor and the multiplication rate constant k, generation time G,bacterial growth inhibition ratio I,total thermogenetic quantity Q, the heat quantity of a single bacterium. Q0 and the heat quantity of a single bacterium per minute Q0 have also been calculated. On the basis of k~T data, the formula Lnk~1/T has graphically obtained and activation energy Ea and pre-exponential factor A have been calculated. According to the parameters of the growth metabolism of E.coli,some linear relationships have been derived. The paper provides a discussion about the growth metabolism of E.coli reacted upon by Cu(Ⅱ)-SG at different temperature, and it is found that、and tr can be used to characterize bacterial growth metabolism and the antibacterial activity of Cu(Ⅱ)-SG at different temperature.  相似文献

利用分子生物学方法(SDS-PAGE和酶活检测法)未检测到所克隆的aprE基因在大肠杆菌中的表达产物(碱性蛋白酶), 而微量热法检测结果发现: 重组菌株的生长代谢产热曲线之间存在明显的差异. 根据这些差异, 分析了该基因的上、下游调控序列对该基因在大肠杆菌中表达的重要作用, 从而进一步对该基因进行了亚克隆, 得到了生物学方法可检测到的表达产物. 由此推测, 微量热技术有可能为检测外源基因表达及其调控, 以及为指导进一步基因工程操作提供一种新的快速灵敏的技术和方法.  相似文献

In this study, the effects of Benzoinum and Styrax on Escherichia coli (E. coli) growth were investigated by microcalorimetry. Using a TAM Air Isothermal Calorimeter, ampoule method, the power-time curves of E. coli growth at 37°C affected by Benzoinum and Styrax were measured. By analysing some quantitative thermokinetic parameters, such as growth rate constants k, the maximum heat-out power Pm, the time of the maximum heat-out power tm, the total heat production Qt and inhibitory ratio I, one could find that low concentrations (0-3.9 mg mL(-1)) of Benzoinum and Styrax had stimulation effects on E. coli growth, and high concentrations (7.8-125.0 mg mL(-1)) of these two drugs would inhibit the growth of the bacteria. The antibacterial effects of Benzoinum and Styrax can also be expressed as half inhibitory concentration IC50. The IC50 values for Benzoinum and Styrax are 78.5 and 88.0 mg mL(-1), respectively, which suggests that the antibacterial effect of Benzoinum on E. coli was much stronger than that of Styrax. This study provides a useful method to investigate the effects of herbal medicines on microbes. It also supplies some references for the application of Benzoinum and Styrax in clinical treatment.  相似文献
