以5,10,15,20-四(4′-吡啶基)卟啉为原料,合成了一系列季铵盐型金属卟啉1a,1b和2a~2c.使用LKB2277热活性监测器测定了大肠杆菌在不同浓度的金属卟啉1a,1b和2a~2c作用下的产热曲线,得到了大肠杆菌生长速率常数k,最大发热功率Pmax和最大发热功率的出现时间tp,发现季铵盐型金属卟啉2c对大肠杆菌生长代谢的抑制活性明显大于其它季铵盐型金属卟啉.  相似文献

合成并表征了一系列水溶性和非水溶性金属卟啉3a~3e,5a~5f和8a~8f;测试了这些金属卟啉作为超氧化物歧化酶(SOD)和过氧化氢酶(CAT)的模拟物催化歧化O2-和催化分解H2O2等有毒氧自由基的活性及抗脂质过氧化性质.结果表明,含有吡啶溴化盐的水溶性金属卟啉8a~8f的仿酶活性明显大于含有羟基的非水溶性金属卟啉3a~3e和含有酯基的金属卟啉5a~5f.  相似文献

使用LKB 2277热活性监测器测定了大肠杆菌在几种含吡啶双酰胺基双Schiff碱作用下的生热曲线,得到了不同Schiff碱在不同浓度下大肠杆菌生长代谢的生热速率常数k、传代时间、半抑制量浓度。结果发现,含有2,4-二羟基苯甲醛双Schiff碱(Ⅱa和Ⅱg)对大肠杆菌生长代谢的抑制活性要明显大于含其他Schiff碱,而杂环Schiff碱对大肠杆菌抑菌活性顺序为吡啶噻吩呋喃。  相似文献

低聚壳聚糖负载金属卟啉配合物的制备及生物活性研究   总被引:1,自引:0,他引:1  
将低聚壳聚糖(COS)分别与不同卟啉金属(MTPPS4,M=Cu,Co,Zn)配合物结合,制备了水溶性低聚壳聚糖负载金属卟啉配合物(COS-MTPPS4),并采用红外光谱和紫外-可见光谱对其结构进行了表征。采用SRB细胞染色法,研究了壳聚糖负载金属卟啉(COS-MTPPS4)对人体肝癌细胞Bel-7402的抗肿瘤细胞活性。结果表明,金属离子配位到卟啉环中,使系列化合物对Bel-7402有较强的抑制生长活性,IC50值均小于100μg/mL,在10~20μg/mL范围内。水溶性低聚壳聚糖金属卟啉配合物作为抗肿瘤药物有很好应用前景。  相似文献

近年来, 卟啉及金属卟啉与DNA的相互作用已成为研究的热点. 通过对卟啉及金属卟啉与DNA相互作用的研究将有助于更深地认识DNA本身的结构和功能, 有助于卟啉及金属卟啉在医学上的应用, 对认识很多疾病的治病机制和药物的设计也起到非常重要的作用. 本文阐述了卟啉及金属卟啉与DNA相互作用的方式和影响因素, 并将水溶性卟啉及金属卟啉分为三类: 具有吡啶基或铵基的阳离子型卟啉及金属卟啉, 具有磺酸基或羧基的阴离子型卟啉及金属卟啉, 以及非离子型的卟啉及金属卟啉. 重点论述了这三类水溶性卟啉及金属卟啉与DNA之间的相互作用, 以及这三类水溶性卟啉及金属卟啉在DNA中的应用研究进展.  相似文献

采用Alder法合成四羟基苯基卟啉,并用四种醋酸盐:醋酸锌,醋酸镍,醋酸铜,醋酸钴与原卟啉合成了相应4种金属卟啉(MTHPP).采用金属卟啉为敏化剂,并与蒸汽热法制备的纯锐钛矿型TiO2作用,得到相应的金属卟啉敏化TiO2复合光催化剂.利用红外、紫外、SEM、和XRD对所得金属卟啉以及金属卟啉-TiO2复合光催化剂进行了表征和分析.结果表明,所合成的金属卟啉均为目标化合物,金属卟啉负载于TiO2表面,未改变TiO2的晶型和形貌.金属卟啉与TiO2之间存在氢键的作用力.可见光降解亚甲基蓝(MB)实验结果表明,用锌卟啉(ZnTHPP)敏化的复合催化剂性能最好,复合催化剂性质稳定,可重复使用多次.  相似文献

采用微量热法测定了大肠杆菌在左金丸、甘露散、茱萸丸和反左金丸水煮液作用下的生长热谱曲线,获得相应的生物热动力学参数,经主成份分析并结合本草文献报道,综合分析左金丸及类方的药性差异,以从生物物理和生物化学的角度,探讨中医经典类方药性差异的客观真实性.实验发现,左金丸及类方的水煮液对大肠杆菌的生长代谢表现出不同的活性作用.寒凉药方左金丸和甘露散不同程度的抑制细菌的生长代谢.温热药方反左金丸和茱萸丸对细菌的生长代谢有不同程度的促进作用.类方之间存在较稳定的药性差异.类方药性主要受生物热动力学参数:第二指数生长期的生长速率常数k2和最大发热功率Pm2的影响.  相似文献

一种新型希夫碱及其3d,4f配合物的抗菌活性   总被引:4,自引:0,他引:4  
用微量热法研究一种新型希夫碱及其3d,4f配合物(2L, 2LZnYb)对大肠杆菌和金黄色葡萄球菌的抗菌活性, 得到了在它们作用下大肠杆菌和金黄色葡萄球菌生长代谢的产热曲线, 并且基于分析生长代谢和非生长代谢的产热曲线建立的热动力学方程, 获得了它们的抗菌活性. 结果表明, 两种化合物(ZL, 2LZnYb)对大肠杆菌的生长代谢有强的活性(IC50分别为6.1 和5.1 mg·L-1), 但对金黄色葡萄球菌的生长代谢的活性弱得多(IC50分别为310.1 和595.5 mg·L-1). Zn 和Yb的导入使化合物对大肠杆菌生长代谢的抑制作用稍微增加, 但大大降低了对金黄色葡萄球菌的抑制作用. 对于非生长代谢, 两种化合物的活性有很大的差别. 无论对大肠杆菌还是金黄色葡萄球菌, 由于配体2L的导入, 表现出显著的抑制作用, 2L的MSC50为6.4和209.7 mg·L-1. 配体2L可能成为新的抗菌先导化合物.  相似文献

合成了水溶性四-[(4-三甲铵基)苯基]卟啉TAPP及其金属卟啉化合物MTAPP,其中M=Zn(),Cu(),Fe(),Co(),Ni(),Mn().通过元素分析、紫外-可见光谱确定了化合物的组成和结构.以紫外-可见光谱、荧光光谱及粘度法等手段研究了卟啉化合物TAPP和金属卟啉化合物ZnTAPP与小牛胸腺DNA的相互作用,结果表明,TAPP,ZnTAPP与DNA相互作用使紫外-可见光谱发生了8~12nm的红移,并出现了明显的减色效应.荧光光谱强度明显增大,吸收峰位发生变化.以艾氏腹水癌(Ac)和肝癌(Hept)细胞为靶细胞,采用MTT染色法评价了化合物的抗癌活性,部分化合物表现出较高的抗癌活性  相似文献

By using LKB2277 Bioactivity Monitor, we have determined the thermogenesis power curves of four kinds of bacteria: Brucella M5(55010), 83-980, 83-981 and E. coli. We have also derived the thermokinetic equation for the process of bacterial growth, which was untypical "S" , unideal growth, as:
Ln[P•(1-P/Pm)?-1]=ln[P0•(1-P0/Pm)?-1]+kt
From this equation, the rate constants of bacterial growth k wore obtained. This thermokinetic equation is very suitable for cell growth of separated culture, in which the thermogenesis curve is untypical. The model propoted by equationl(1) has been compared with the exponential and logistic models. This thermokinetic equation is very significant for the study of bacterial limited growth and their characteristics.  相似文献

The effect of magnetic field on the growth of bacteria was studied with the series piezoelectric quartz crystal (SPQC) sensing technique. The growth situations of Escherichia coli (E. coli) in the absence and presence of different intensities of static magnetic fields were examined and analyzed. The results showed that the growth of E. coli was inhibited due to the presence of magnetic fields. By fitting frequency shift (deltaD) versus time curves according to the frequency shift response equation of SPQC, the relationships between three kinetic growth parameters, i.e., the asymptote A, the maximum specific growth rate mu(m) and lag time lambda, and magnetic field intensity were established. Based on these results, a new response model containing the magnetic field intensity was derived as: delta(f) = 167.7 (7.25 - 7.11B)/[1 + exp[4 x 2.46e(-3.97B)/(7.25 -7.1 IB)] x (4.42 + 16.46B - t) + 2]] The kinetic parameters of bacterial growth obtained from this model are close to those obtained from the logistics popular growth model, in which the concentration of the bacteria was determined by the traditional pour plate count method.  相似文献

以广西的优势资源松香为原料, 脱氢松香酸与亚硫酰氯、硫氰化钾分别在回流条件下反应6 h和1.5 h, 得到脱氢松香酰异硫氰酸酯, 产率52%; 然后与胺在加热回流条件下反应1.5 h, 得到11种1-脱氢松香酰基-3-取代硫脲4, 产率63%~94%; 4a~4f 分别与水合肼在搅拌下回流反应3~6 h, 得到6种5-(脱羧脱氢松香-4-基)-3-芳氨基-1H-1,2,4-三唑化合物5, 产率70%~94%; 所有化合物的结构均经IR, 1H NMR, 13C NMR和元素分析确认. 初步生物活性测试表明, 4e, 4f, 4j, 5b对枯草杆菌抑菌率较高, 特别是4j在浓度为50 mg/L时就达到较好效果; 4b, 4h, 4i, 5e在100 mg/L时对大肠杆菌的抑菌效果较好.  相似文献

We report a cell-phone based Escherichia coli (E. coli) detection platform for screening of liquid samples. In this compact and cost-effective design attached to a cell-phone, we utilize anti-E. coli O157:H7 antibody functionalized glass capillaries as solid substrates to perform a quantum dot based sandwich assay for specific detection of E. coli O157:H7 in liquid samples. Using battery-powered inexpensive light-emitting-diodes (LEDs) we excite/pump these labelled E. coli particles captured on the capillary surface, where the emission from the quantum dots is then imaged using the cell-phone camera unit through an additional lens that is inserted between the capillary and the cell-phone. By quantifying the fluorescent light emission from each capillary tube, the concentration of E. coli in the sample is determined. We experimentally confirmed the detection limit of this cell-phone based fluorescent imaging and sensing platform as ~5 to 10 cfu mL(-1) in buffer solution. We also tested the specificity of this E. coli detection platform by spiking samples with different species (e.g., Salmonella) to confirm that non-specific binding/detection is negligible. We further demonstrated the proof-of-concept of our approach in a complex food matrix, e.g., fat-free milk, where a similar detection limit of ~5 to 10 cfu mL(-1) was achieved despite challenges associated with the density of proteins that exist in milk. Our results reveal the promising potential of this cell-phone enabled field-portable and cost-effective E. coli detection platform for e.g., screening of water and food samples even in resource limited environments. The presented platform can also be applicable to other pathogens of interest through the use of different antibodies.  相似文献

Since Schiff base and its metal complexes are of antibacterial and anticancer bioactivity, it has been a research subject of much interest. It is reported that Cu(Ⅱ)-SG could inhibite the synthesis of O2- markedly and could also combine with the salmn sperm DNA. The paper repored that by using microcalorimetric method The growth meloabolism of E.coli at different temperature and reacted upon by Cu(Ⅱ)-SG. The power-time curves of E.coli reacted with Cu(Ⅲ)-SG at different temperature has been determined by LKB2277 Microcalorimetric Monitor and the multiplication rate constant k, generation time G,bacterial growth inhibition ratio I,total thermogenetic quantity Q, the heat quantity of a single bacterium. Q0 and the heat quantity of a single bacterium per minute Q0 have also been calculated. On the basis of k~T data, the formula Lnk~1/T has graphically obtained and activation energy Ea and pre-exponential factor A have been calculated. According to the parameters of the growth metabolism of E.coli,some linear relationships have been derived. The paper provides a discussion about the growth metabolism of E.coli reacted upon by Cu(Ⅱ)-SG at different temperature, and it is found that、and tr can be used to characterize bacterial growth metabolism and the antibacterial activity of Cu(Ⅱ)-SG at different temperature.  相似文献
