提出一类红外光谱分析数据特征指纹的可视化表达方法。该方法采用基于贝叶斯准则的多元统计方法对原始数据进行投影变换,再以地维灰度图对变换后数据进行可视化表征,形成基于计算机图像的虚拟化学指纹图谱。将其用于鉴别3种不同产地中药当归样品的结果表明,它能有效提取红外光谱分析数据的特征指纹,实现以虚拟指纹图谱对药材产地的分类鉴别,从而为辨识复杂化学物质体系提供了新的技术手段。  相似文献

提出了一种快速无损的化学指纹图谱分析新方法. 根据小波变换多分辨分析的特点, 对近红外(NIR)光谱进行小波分解, 从中提取被测样品的化学特征信息, 并作数字信息可视化处理, 构建形成可直观识别样品模式特征的NIR指纹图谱. 将该方法用于中药材丹参的质量检测, 检测结果与色谱指纹图谱检测结果相符, 能快速有效地识别丹参质量模式间的差异, 有望发展成为一种快速分析检测天然产物质量的方法.  相似文献

中药药对是复方的最小组成单位,具有中药配伍的基本特点. 药对化学是复方化学的核心内容. 联用色谱和化学计量学方法为中药复杂体系的分离与分辨提供了强有力的工具. 采用GC-MS法分离测定中药药对川芎-赤芍、 单味药川芎和赤芍的挥发油成分,并对其中的重叠色谱峰采用化学计量学解析方法(CRM)进行分辨,得到药对和各单味药的纯色谱曲线和质谱图,药对川芎-赤芍、 单味药川芎和赤芍分别分辨出82,78和57个色谱峰,通过质谱库对分辨的纯组分进行定性,分别得到61,52和33个定性结果,占总含量的90.18%,95.14%和95.82%.  相似文献

建立了川芎挥发油的全二维气相色谱/飞行时间质谱(GC×GC/TOF MS)指纹图谱,并结合聚类分析评价了川芎药材的质量。4种萜类和苯酞类物质在DB-Petro×DB-17柱系统上实现了明显的族组分分离。从新都市的1个样品中分离出375种组分,其中相似度、反相似度皆大于800的化合物215个;根据质谱库检索和保留指数验证,或参照标准化合物及文献报道,从中定性了43个化合物。结合偏最小二乘法-判别分析,可以很好地将4个产区的挥发油样品加以区分,并找出20种差异最大的化合物,其中包括4种苯酞类物质。在此基础上,进一步应用正交偏最小二乘法,关联化学成分和抗氧化活性,结果发现,苯酞类物质(如藁本内酯、川芎内酯A和新蛇床内酯)对川芎挥发油样品地区差异的影响最大,其中彭州产川芎样品中苯酞类物质的含量最高。本文通过高分辨色谱技术研究了相邻产地的药物化学组成差异,并将化学指纹及生物学活性相关联,建立了系统的中药质量评价及活性化合物筛选研究模式。  相似文献

在运用神经元计算技术对高维小样本复杂化学模式进行分类时,通过模式特征提取,降低输入变量维数,能使复杂的模式分类问题比较容易解决。根据模式类别相关分步分析思路,提出复杂化学模式特征分步提取法,可将原始模式数据中与类别指标相关较大的特征量有效地提取出来。应用于天然植物组效关系辨识结果表明,这种化学模式特征提取方法比经典主成分分析法更为实用可靠。  相似文献

采用自行研制的表面解吸常压化学电离质谱(DAPCI-MS),无需样品预处理,对硫磺熏蒸八角和未熏八角直接进行正、负离子模式检测,获得其化学指纹图谱,并通过主成分分析(PCA)及聚类分析(CA)方法对所获指纹谱图信息进行分析,进而对不同样品进行鉴别。结果表明,在正、负离子模式下,DAPCI-MS都可对八角表面多种特征化学成分进行分析,快速获得八角的化学指纹谱图,并能够对目标组分进行多级串联质谱鉴定,结合PCA及CA方法可对八角是否经硫磺熏蒸进行快速鉴别。本方法无需样品预处理,灵敏度高,分析速度快,无污染,可望应用于市场上硫熏制品的快速鉴别。  相似文献

自19世纪末期,指纹鉴定一直是世界上犯罪侦查中最有效的个体识别物证手段之一。但是,指纹遗留时间的确定至今仍是一个相对未开发的研究领域。指纹脊线的物理形貌特征和残留物的化学成分因遗留者、遗留客体和环境的不同而产生显著差异。而且,随着指纹存留条件和老化时间的不同,其物理特征和化学组成的分子种类及含量等也随之变化。研究指纹残留物的物理特征和起始化学组成以及随遗留时间的变化关系是法医科学研究的重点课题,不仅有利于发展新的指纹检测方法和技术,而且可以提升鉴别犯罪现场发现的指纹的相关性。本文主要讨论已发展的各种方法和技术,如液相色谱、荧光光谱、红外光谱、紫外-可见光谱、拉曼光谱、质谱和高分辨率成像等方法,以及这些方法在指纹残留物的物理特征和化学组成及其随遗留时间的变化关系的研究中取得的进展以及局限性。最后,对发展潜在更可靠的指纹遗留时间确定方法中面临的挑战和未来的发展趋势进行展望。  相似文献

采用加速溶剂萃取法提取挥发性成分,气相色谱-质谱(GC-MS)联用分析,并借助化学计量学分辨方法,对川芎的挥发性成分进行了定性定量分析。通过采用子窗口因子分析法对重叠色谱峰进行分辨,从而获得每一组分的纯色谱和质谱。依靠每一组分纯质谱在NIST质谱库进行相似性检索而定性分析,用总体积积分法进行定量分析。在川芎挥发油中共分辨出67个色谱峰,通过质谱库检索得到其中56个组分的定性定量结果,占总含量的87.67%。该方法增加了可鉴定化合物的数目,提高了定性的准确度,能用于川芎的质量控制。  相似文献

用水蒸气蒸馏和超临界CO2提取方法对川芎药材中的化学成分进行提取,用气相色谱-质谱联用技术(GC-MS)对提取物进行分析,分别鉴定出30和34个化合物,测定了其相对含量。 结果表明,水蒸气蒸馏和超临界CO2提取物的主要成分均为(Z)-藁本内酯。 超临界CO2提取方法能提取出较多的川芎药效物质,如丁基酞内酯、丁烯基酞内酯、川芎内酯A、(Z)-藁本内酯、(E)-藁本内酯和川芎内酯I。 而水蒸气蒸馏提取物中除了川芎药效物质外,还含有较多的萜烯类化合物。 结果表明,超临界CO2对川芎药效物质的提取比水蒸气蒸馏法的效率高。 通过对川芎中带不饱和侧链与饱和侧链内酯类化合物质谱裂解规律的解析归纳,鉴别了川芎中不饱和侧链内酯类化合物如(Z)-藁本内酯、(E)-藁本内酯、丁烯基酞内酯和川芎内酯I以及饱和侧链内酯类化合物如丁基酞内酯和川芎内酯A。  相似文献

在恒温恒压条件下,以丙酮和样本中底物作为主要耗散物的不同成分的样本对非线性化学反应机理产生不同影响,从而引起反应体系电位-时间曲线形状不同变化为特征的B-Z化学振荡体系为例,就非线性化学指纹图谱原理进行了详细研究和讨论,并提出了计算非线性化学指纹图谱系统相似度的通用方法.利用系统相似度和欧氏距离、相关系数及夹角余弦对不同生产批次古汉养生精和18种其他样本的非线性化学指纹图谱的相似度进行了计算与分析.结果表明,相关系数和夹角余弦都不能用来作为评价非线性化指纹图谱相似度的指标.利用欧氏距离公式计算指纹图谱的非参数型相似度时,能正确反映指纹图谱的特征差异,但用其计算参数型相似度时,则有时不能正确反映样本非线性化学指纹图谱特征差异的相对程度.系统相似度能最真实反映样本指纹图谱之间差异程度,是4种相似度计算方法中最好的,可用于非线性化学指纹图谱相似度计算与评价.成功提出了一种经济、简便、易行和有效的鉴别样本真伪与评价其质量的科学方法.  相似文献

Our present study constitutes the first successful attempt to employ eight distinctive chemical groups of compounds for the quality evaluation of a complex traditional Chinese herbal medicine (TCHM) material: BaZhen YiMu (BZYM). Due to the complexity of its matrix, which is composed of nine different herbal ingredients, five representative chemical groups encompassing representative bioactive markers were initially chosen as targets for the quality assessment of this preparation. Furthermore, with the aid of LC‐ESI‐MS, three additional chemical groups were characterized. In summary, a total of nineteen markers belonging to eight different chemical groups were selectively displayed in the chromatographic fingerprint of BZYM preparation. With this fingerprint, the overall quality of any BZYM preparation can be comprehensively authenticated. The chromatographic separation was performed on an HP C18 AQ column with a gradient elution of ACN and aqueous solution containing 0.1% phosphoric acid at the optimal detection wavelength of 230 nm. The established method was rigorously validated with respect to the ICH guidelines and represents the most extensive and facile HPLC quality control technique for this formulation. Compared with the conventional method of using a single or only a few markers of the same chemical group, this technique provides a new dimension for TCHM quality control.  相似文献

Secured principal component regression is modified for the qualitative analysis of chromatographic fingerprint data sets of herbal samples with residual concentrations. After chromatographic shift-correction and autoscaling are performed on the data, this modified secured principal component regression (msPCR) can detect unexpected chromatographic features in various herbal fingerprints. The successful application of msPCR to two real herbal medicines of Erigeron breviscapus from different geographical origins and Ginkgo biloba from various sources or vendors demonstrates that the proposed method can detect reasonably unexpected features differing from the regulars or not being modeled. From a chemical point of view, the causes have also been explained to corroborate the results. Moreover, it presents a viable approach for the qualitative evaluation of diverse herbal objects with a regular class of chromatographic fingerprints.  相似文献

Recently, chromatographic fingerprinting has become one of the most powerful approaches to quality control of herbal medicines. However, the performance of reported chromatographic fingerprinting constructed by single chromatogram sometimes turns out to be inadequate for complex herbal medicines, such as multi-herb botanical drug products. In this study, multiple chromatographic fingerprinting, which consists of more than one chromatographic fingerprint and represents the whole characteristics of chemical constitutions of the complex medicine, is proposed as a potential strategy in this complicated case. As a typical example, a binary chromatographic fingerprinting of “Danshen Dropping Pill” (DSDP), the best-sold traditional Chinese medicine in China, was developed. First, two HPLC fingerprints that, respectively, represent chemical characteristics of depsides and saponins of DSDP were developed, which were used to construct binary chromatographic fingerprints of DSDP. Moreover, the authentication and validation of the binary fingerprints were performed. Then, a data-level information fusion method was employed to capture the chemical information encoded in two chromatographic fingerprints. Based on the fusion results, the lot-to-lot consistency and frauds can be determined either using similarity measure or by chemometrics approach. The application of binary chromatographic fingerprinting to consistency assessment and frauds detection of DSDP clearly demonstrated that the proposed method was a powerful approach to quality control of complex herbal medicines.  相似文献

A fingerprint method was developed and combined with chemometrics for quality evaluation of Tianma Toutong tablets, which are herbal medicine tablets used to treat migraine. Samples were analyzed by high‐performance liquid chromatography, where five single‐wavelength profiles (203, 232, 254, 280 and 310 nm) were fused to generate a five‐wavelength fusion fingerprint and were also used for the quantitative analysis of seven chemical markers (gastrodin, caffeic acid, hesperidin, isoimperatorin, chlorogenic acid, ferulic acid and imperatorin). A systematic quantitative fingerprint method and principal component analysis were used to analyze the generated data. Samples could be well distinguished from different manufacturers by analyzing the chromatographic data sets. In addition, the partial least squares model can serve as an antioxidant activity evaluation of Tianma Toutong tablets, as well as a reference for the selection of active constituents to analyze the spectrum–activity relationship. In summary, the integrated use of the fingerprint and chemometric analysis provides a reliable method for the identification of markers and the quality control of Tianma Toutong tablets.  相似文献

A signal-processing method known as spectral correlative chromatography (SCC) for two-dimensional data obtained from hyphenated chromatography is developed and applied to chemical chromatographic fingerprint data sets of herbal medicine under specific experimental conditions. The method can judge the presence or absence of a spectral correlative peak among the spectrochromatograms. A local least squares regression model (LLS) is constructed in a piecewise manner to correct the shifts of retention time of some peaks of interest in the chromatograms of various test samples. The results compare favorably with those obtained by a two-point calibrated algorithm. It is shown that performing SCC and LLS on the piecewise clusters of various chromatographic fingerprints is more helpful in practice in revealing their common nature and for characterizing the chemical constituents. This approach holds great potential for facilitating quality control of herbal medicines.  相似文献

A novel, simple and accurate fingerprint method was developed using high-performance liquid chromatography-photodiode array detection (HPLC-DAD) for the quality control of Hypericum japonicum thunb (Tianjihuang), a Chinese herbal medicine used for the treatment of several bacterial diseases, infectious hepatitis, gastrointestinal disorder, internal hemorrhage and tumors. For the first time, the feasibility and advantages of employing chromatographic fingerprint were investigated and demonstrated for the evaluation of Tianjihuang by systematically comparing chromatograms with a professional analytical software recommended by State Food and Drug Administration (SFDA). Our results revealed that the chromatographic fingerprint combining similarity evaluation could efficiently identify and distinguish raw herbs of Tianjihuang from different sources. The effects resulted from collecting locations, harvesting time and storage time on herbal chromatographic fingerprints were also examined.  相似文献

Abstract

Recently, the fingerprinting approach using chromatography has become one of the most potent tools for quality assessment of herbal medicine. Due to the complexity of the chromatographic fingerprint and the irreproducibility of chromatographic instruments and experimental conditions, several chemometric approaches such as variance analysis, peak alignment, correlation analysis, and pattern recognition were employed to deal with the chromatographic fingerprint in this work. To facilitate the data preprocessing, a software named Computer Aided Similarity Evaluation (CASE) was also developed. All programs of chemometric algorithms for CASE were coded in MATLAB5.3 based on Windows. Data loading, removing, cutting, smoothing, compressing, background and retention time shift correction, normalization, peak identification and matching, variation determination of common peaks/regions, similarity comparison, sample classification, and other data processes associated with the chromatographic fingerprint were investigated in this software. The case study of high pressure liquid chromatographic HPLC fingerprints of 50 Rhizoma chuanxiong samples from different sources demonstrated that the chemometric approaches investigated in this work were reliable and user friendly for data preprocessing of chromatographic fingerprints of herbal medicines for quality assessment.  相似文献

Herbal injection is one of the most important preparations of traditional Chinese medicine. More than 130 types of herbal injections are used clinically for 400 million patients annually with total sales of over four billion US dollars per year. However, the current quality control (QC) methods relying mainly on chemical fingerprints (CF) can hardly ensure quality and safety of the herbal injections with complex chemical composition and have resulted in an increase in serious adverse drug reactions. In this study, a comprehensive approach for the QC of a controversial herbal injection Shuang-Huang-Lian lyophilized powder (SHL) was established based on the quality fluctuation detection by a combination of CF and biological fingerprint (BF). High-performance liquid chromatography and the impedance-based xCELLigence system were applied to establish the CF and BF, respectively. In addition, multivariate analysis was performed to evaluate the discriminant ability of the two methods. The results showed that being subjected to environmental influence like oxygen/air, high temperature, and extreme illumination could lead to quality fluctuation of SHL. The combination of chemical and biological fingerprint method is a more powerful tool for the QC of SHL because it can clearly discriminate different groups of abnormal samples. This method can be used for the detection of quality fluctuation of SHL and can provide reference for the quality control of other herbal injections.  相似文献

Si-Mo-Tang(SMT) oral liquid preparation, a traditional Chinese medicine, was prepared from four crude herbal drugs, Fructus Aurantii Submaturus, Radix Aucklandiae, Semen Arecae and Radix Linderae Aggregatae. A combinative method using HPLC fingerprint and quantitative analysis was developed and validated for quality consistency evaluation of SMT. Individual HPLC chromatograms were evaluated against the mean chromatogram generated via a similarity evaluation computer program. Data from chromatographic fingerprints were also processed with principal component analysis(PCA) and hierarchical cluster analysis(HCA). Additionally, six components (naringin, isonaringin, hesperidin, neohesperidin, norisoboldine and potassium sorbate) in SMT were simultaneously determined to interpret the quality consistency. For fingerprint analysis, 20 peaks were selected as the characteristic peaks to evaluate the similarities of 26 SMT collected from different manufacturers. Among the 20 characteristic peaks, 10 peaks were assigned to be naringin, hesperidin, neohesperidin, isonaringin, neoeriocitrin, tangeretin, nobiletin, norisoboldine, 5-(ethoxymethyl)furan-2-carbaldehyde and potassium sorbate, respectively. The results of similarity analysis, PCA and HCA, indicate that the samples from different manufacturers were consistent with each other in composition. The results from the quantitative data show that the contents of six compounds were significantly different in SMT oral liquid preparations from different manufacturers. The combinative method of chromatographic fingerprint with quantitative analysis developed here offered an efficient way for the quality consistency evaluation of the traditional Chinese medicine SMT.  相似文献

The use of chromatographic fingerprints from herbal products where the whole chromatographic profile is applied to evaluate the quality of the investigated product. In this paper, recent developments in the set-up and data analysis of chromatographic fingerprints for herbal products are discussed. First different set-ups for fingerprint development are reviewed. Prior to fingerprint development, a suitable sample preparation, e.g. extraction, should be considered. In a second instance, this review focuses on the data analysis with regards to the different applications of fingerprints. Usually, chemometric data pretreatment is necessary. This is discussed first, followed by a short overview of the data handling techniques used in the two main application areas of herbal fingerprints, i.e. quality assurance and classification or calibration. The quality assurance, which involves the identification and quality control of the herbal products, is reviewed, followed by the use of fingerprints in classification or modelling. The different application areas are illustrated and discussed with several case studies.  相似文献
