建立了分光光度法测定田七花中黄酮含量的方法.采用分光光度法,以芦丁为对照品,在波长510nm处对样品中的总黄酮进行含量测定.样品总黄酮在浓度0~50μg/mL范围内线性关系良好,其回归方程为A=11.069C-7.143×10-4;R=0.9993.该方法可作为田七花中总黄酮含量的测定的方法之一.  相似文献

比较了微波辅助提取、超声提取、索氏提取3种方式对穿心莲中总黄酮的提取效率.确定以微波辅助提取法最佳.实验结果表明:用60%乙醇在固液比为1:15的条件下提取6 min提取效率最高.用NaNO2-硝酸铝-NaOH-黄酮体系分光光度法测定总黄酮含量,平均回收率为103.00%,RSD=1.37%(n=5),证实了可以用微波辅助提取法提取,紫外可见分光光度法检测穿心莲中总黄酮的可行性.  相似文献

为比较分析紫苏叶与白苏叶的总黄酮和微量元素的含量差异,用可见分光光度法测定了两者总黄酮的含量,用电感耦合等离子体原子发射光谱法(ICP-AES法)测定了两者微量元素的含量。结果表明,测得紫苏叶与白苏叶中总黄酮的含量分别为7.49%和5.69%,两者均含有Cu、Mn、Zn、Fe等多种微量元素,但含量存在一定的差异。白苏作为紫苏的同种植物资源,有待进一步的开发和利用。  相似文献

采用超声波提取法提取蛇床子不同炮制品中总黄酮,以芦丁为对照品,紫外分光光度法测定总黄酮的含量,标准曲线在2.1~11.0μg/mL范围内,吸光度与浓度的线性关系良好(R~2=0.999 7),实验的精密性和稳定性RSD分别为1.13%和2.15%,平均回收率为105.81%,RSD为2.98%。结果表明紫外分光光度法测定蛇床子不同炮制品中的总黄酮含量操作简便、结果准确、重现性好,实验方法切实可行。  相似文献

研究鸡足山耳蕨中总黄酮的提取及其抗氧化性.采用70%乙醇提取鸡足山耳蕨中总黄酮,用NaNO2Al(NO3)3-NaOH分光光度法测定黄酮含量,将提取液采用Fenton体系、普鲁士蓝法进行体外抗氧化活性研究,用硫代巴比妥酸(TBA)分光光度法研究其对羟自由基·OH引发DNA氧化损伤的抑制作用.结果表明样品中总黄酮含量为4.98%,回收率99.78%( RSD=1.06%,n=5).总黄酮浓度为90μg/mL时,对·OH的清除率可达36.2%;浓度为87.5μg/mL时,对羟自由基引发DNA损伤的抑制率可达93.0%.说明鸡足山蕨中总黄酮对羟自由基有较好的清除能力,对DNA氧化损伤有显著抑制作用.  相似文献

从花生壳中提取总黄酮物质,并用紫外分光光度法对其进行测定。建立了一种以无水乙醇为溶剂,检测波长为359nm的测定花生壳总黄酮含量的新方法。结果表明,总黄酮含量在0.005~0.025mg/mL范围内与吸光度线性关系良好,相关系数r=0.9995,平均加标回收率为98.5%,测定结果的相对标准偏差为2.1%(n=5)。该法适用于花生壳总黄酮的定量测定。  相似文献

Summary As a part of an investigation of the content of herbal drug preparations and herbal medicinal products, we have investigated tinctures (prepared with 40:60 and 60:40 (%,v/v) ethanolwater) of calendular flower, milk-thistle fruit, and passion flower, which are, respectively, widely used for their anti-inflammatory properties, to treat hepatic injuries, and to treat tension and difficulty falling asleep. The aim of this work was to evaluate the flavonoid content, because flavonoids are the active constituents or markers of these herbal drugs, and to establish the best solvent for the extraction of the constituents. The findings reported herein both confirm the presence of several flavonoids previously identified in these herbal drugs and report the presence of others not previously described and identified here for the first time. In general the flavonol content was highest in 60% tinctures. A rapid, reversed-phase (RP) HPLC assay was developed and validated and enabled good separation of all classes of flavonoids including flavones, flavonols, flavanonols, and flavanolignans. Aglycones and mono, di, and triglycosides (bothO-andC-derivatives) of the flavonoids are also easily and satisfactorily separated. This method is thus proposed for the analysis of other herbal drugs or herbal drug preparations in which flavonoids are the active constituents or markers.  相似文献

Twenty herbs, showing medicinal benefits on human life, like Taraxacum officinale, Plantago lanceolata, Hypericum perforatum, Ocimum basilicum, Melissa officinalis, Mentha piperita, and others, were tested. The percentage content of pigments, i.e. chlorophylls, carotenoids, and flavonoids, in the chosen herbs was determined. The spectrophotometric method, which is quick and readily available, was used. Measurements required pigment extraction with a solvent. Ethanol was used for the determination of chlorophylls and carotenoids, and acetone for the determination of flavonoids. Hierarchical agglomerative cluster analysis was performed in order to confirm the experimental results. The study of flavonoids content showed an interesting regularity which can be used for the classification of herbs. Plants belonging to the Lamiaceae family showed the flavonoids content in the range from 0.18 % to 0.68 %, whereas those belonging to the Apiaceaehas family showed the flavonoids content in the 0.75–1.20 % range.  相似文献

确定含羞草中总黄酮的最佳提取部位,并对其初步分离纯化。通过正交试验,分别筛选含羞草根部、茎叶及种子中总黄酮的最优提取条件,比较三者黄酮总含量,最终确定提取部位为茎叶,最佳提取条件:以70%乙醇为溶剂,按照1∶8配料比,在55~58℃下超声50 min。提取液依次用石油醚、乙酸乙酯液液萃取,蒸干上样,用乙醇水溶液以4 m L/min梯度洗脱Diaion HP–20大孔树脂,收集洗脱液并用液相色谱监测每个梯度洗脱液的总黄酮含量,得到分离纯化过的黄酮类物质。当40%乙醇洗脱部位总黄酮含量最高,达57.7%。该工艺确定了含羞草中茎叶部位总黄酮含量最高,大孔树脂初步纯化黄酮类物质有效,为含羞草中黄酮类物质的应用提供了依据。  相似文献

Summary The general objective is to provide an alternative methodology based on capillary electrophoresis (CE) to characterize flavonoids from honey and hence determine its botanical origin. The specific objective is to compare the separation of flavonoids by CE with those achieved by HPLC to assess CE as an alternative technique for the determination of honey flavonoids. Fourteen different flavonoids isolated from honey were analysed by MECC and compared to the HPLC separations. It was difficult to find specific experimental conditions to separate all the flavonoids from honey in a single MEKC run. Three chromatographic conditions are optimized and, depending on the flavonoid markers sought in honey, the appropriate detection method should be chosen. Compared to the HPLC results, it is clear that CE could be an alternative technique in honey flavonoids analysis and particularly in the study of its geographical and floral origin.  相似文献

A high-performance liquid chromatography (HPLC) method with photo-diode array (DAD) detection was developed to separate and quantify flavonoids in sugarcane leaves and bagasse (= the crushed sugarcane refuse from juice extraction), and in sugarcane juice. Sugarcane flavonoids consist of a complex mixture of aglycones and glycosides (including flavonolignan glycosides), and the HPLC-UV method herein proposed is suitable for their quantification as total flavonoids. This method was applied to analyze samples of cultivated sugarcane, commercial juice and transgenic sugarcane leaves. Sugarcane leaves proved a promising source of flavonoids: an average of 1.10 mg of total flavonoids/g plant material was found in fresh leaves. Moreover, the flavonoid content of sugarcane juice (0.6 mg/mL) is comparable to other food sources of flavonoids previously reported. Transgenic sugarcane leaves ("Bowman-Birk" and "Kunitz") were compared with non-modified ("control") plant samples using the proposed HPLC-UV method, which indicated that the content of total flavonoids in transgenic plants is different from that in non-modified sugarcane.  相似文献

L. indica L. cv. Mengzao, a medicinal plant of the Ixeris genus, is rich in flavonoids. In order to thoroughly analyze the the distribution and dynamic change of major flavonoids in its various parts from different growth periods, the flavonoids extracted from L. indica L. cv. Mengzao were identified and quantitatively analyzed by ultra-high-performance liquid chromatography mass spectrometer (LC-MS/MS). Results indicated that 15 flavonoids were identified from L. indica L. cv. Mengzao, and rutin, luteolin, luteolin-7-O-glucoside, kaempferol, quercetin, and apigenin are the major flavonoids in L. indica L. cv. Mengzao. In general, the total flavonoids’ content in different parts of L. indica L. cv. Mengzao followed the order flowers > leaves > stems > roots. Flowers and leaves are the main harvesting parts of L. indica L. cv. Mengzao, and the flowering period is the most suitable harvesting period. This study provides valuable information for the development and utilization of L. indica L. cv. Mengzao and determined the best part to harvest and the optimal time for harvesting.  相似文献

The aim of this research was to provide crucial and useful data about the selection of the optimization criteria of supercritical carbon dioxide extraction of alfalfa at a quarter-technical plant. The correlation between more general output, including total phenolics and flavonoids content, and a more specified composition of polar constituents was extensively studied. In all alfalfa extracts, polar bioactive constituents were analyzed by both spectrometric (general output) and chromatographic (detailed output) analyses. Eight specific phenolic acids and nine flavonoids were determined. The most dominant were salicylic acid (221.41 µg g−1), ferulic acid (119.73 µg g−1), quercetin (2.23 µg g−1), and apigenin (2.60 µg g−1). For all seventeen analyzed compounds, response surface methodology and analysis of variance were used to provide the optimal conditions of supercritical fluid extraction for each individual constituent. The obtained data have shown that eight of those compounds have a similar range of optimal process parameters, being significantly analogous for optimization based on total flavonoid content.  相似文献

为了研究明日叶黄酮类化合物对羟基自由基的清除作用,以明日叶(主要取叶片)为原料,用体积分数为65%乙醇提取明日叶总黄酮,测定其总黄酮含量.通过Fenton反应体系产生羟基自由基,利用明日叶提取液中的功能成分黄酮类化合物对羟基自由基的清除作用进行研究.结果表明:明日叶提取物总黄酮质量分数为10.18%,且黄酮类化合物对羟基自由基有较强清除效力,当提取物总黄酮浓度在0.1~1.0 mg/mL范围内,其与清除率呈正相关.明日叶中黄酮类化合物对羟基自由基有较强清除效力,作为天然抗氧化产品开发具有一定价值.  相似文献
