毛细管电泳具有分析时间短,分离效率高,样品消耗量少等优点,在生物样品分离,特别是蛋白质分析领域有重要应用。然而,毛细管内壁硅羟基的解离给分离结果带来诸多不良影响。聚合物涂层能够抑制蛋白质在毛细管内壁的吸附以及调控电渗流,故对毛细管内壁进行有效修饰能够提高其对蛋白质的分离效率及分离稳定性。该文主要综述了动态及静态聚合物涂层毛细管的最新研究进展,并概述了近些年基于多巴胺/聚多巴胺发展起来的涂层毛细管的研究进展,最后展望了聚合物涂层毛细管的发展趋势。 相似文献

万古霉素作为一种大环抗生素,具有复杂的分子结构。在充分考虑万古霉素分子结构特征的情况下,采用戊二醛间隔臂法制备了万古霉素键合固定相,在反相、亲水、离子交换等分离模式下研究了其色谱分离性能。结果表明,当流动相中有机调节剂含量较低时,该色谱柱表现出典型的反相色谱分离模式特征;随着有机调节剂含量的增加,逐渐转变成亲水模式,分离特性发生明显改变。由于万古霉素分子结构中含有可以解离的氨基,因此该固定相也能够用于阴离子交换模式下的分析方法的发展。分别在反相、亲水和阴离子交换模式下,将其应用于扑尔敏等多种非对映体药物和新型甜味剂甜菊糖的高效液相色谱分离;仅通过改变分离条件,即可在3种不同分离模式下完成分离。这些结果可以为新型色谱固定相的设计,以及发展采用特殊结构改性基团的色谱固定相在相应分离模式下的分析方法提供指导。 相似文献

Hybrid separation processes are becoming more and more important in the practice if membrane technologies are also involved.
In this work, a systematic investigation is completed for three sequence alternatives of distillation and pervaporation. These
are the following: pervaporation followed with distillation (PV+D), distillation followed with pervaporation (D+PV), two distillation
columns and a pervaporation unit between them (D+PV+D). The hybrid separation process alternatives are evaluated with rigorous
modelling tools, but first, a rigorous simulation algorithm is determined for the pervaporation. The three hybrid separation
processes are rigorously modelled with CHEMCAD, and optimized with the dynamic programming optimization method for the case
of the separation of ethanol-water mixture. The objective function is the total annual cost (TAC). The energy consumption
is also investigated. The selection of the ethanol-water mixture has two motivations: (i) it is quite often studied and well
known, and (ii) to make biofuel (ethanol) production more economical, membrane technologies might also be applied. The results
are compared with each other and with the classical separation completed with heteroazeotropic distillation. The optimized
TAC shows that the distillation column followed with pervaporation is the most economical hybrid separation process alternative.
Its TAC is about 66% of that of the classical separation.

Summary A new chiral ligand exchange selector for hydrophobic stationary phase modification has been synthesized by selective alkylation of L-histidine at the pyrrolic nitrogen atom in its imidazolic ring. Its performance was then tested on RP-HPTLC plates treated with copper acetate. Significant selectivity towards aromatic amino acid enantiomers, was observed. Chromatographic retention data were compared to available thermodynamic complex formation parameters for the relevant model systems in aqueous solution.Stationary and mobile phase effects on retention were studied by using different RP-HPTLC plates and various binary aqueous solvent mixtures.Optimized separation conditions for aromatic amino acid sample class are given. 相似文献
