The potential of non-aqueous capillary electrophoresis (NACE) was investigated for the simultaneous separation of paroxetine, tamoxifen, and their main metabolites. Baseline separation of the studied solutes was obtained on a  μm capillary using a non-aqueous buffer composed of 18 mM ammonium acetate and 1.1% acetic acid in 80:20 (v/v) methanol-acetonitrile, with a temperature and voltage of 22 °C and 15 kV, respectively. Clomipramine was used as internal standard. Aspects such as stability of the solutions, linearity, accuracy, precision and ruggedness were examined in order to validate the proposed method. Detection limits obtained for all the studied compounds ranged between 3.0 and 7.1 μg l−1. The developed method is sensitive and robust and was used to determine paroxetine, tamoxifen, and their metabolites at clinically relevant levels in human urine. Before NACE determination, the samples were purified and enriched by means of an extraction-pre-concentration step with a pre-conditioned C18 cartridge. Determination of these analytes in the urine of four females urines was demonstrated.  相似文献

An Ir-catalyzed asymmetric allylic alkylation using chiral diaminophosphine oxide is described. Asymmetric allylic alkylation of terminal allylic carbonates proceeded using 5 mol % of Ir catalyst, 5 mol % of DIAPHOX 1i, 10 mol % of NaPF6, 10 mol % of LiOAc, and N,O-bis(trimethylsilyl)acetamide (BSA), affording the corresponding branched products in excellent yield and in up to 95% ee. The developed catalytic asymmetric reaction was successfully applied to a formal enantioselective synthesis of (−)-paroxetine.  相似文献

The enantioseparation of trans-3-ethoxycarbonyl-4-(4′-fluorophenyl)-1-methyl piperidine-2,6-dione (3), which is one of the important racemic precursors of trans-(−)-paroxetine, has been investigated using supercritical fluid chromatography on a Daicel Chiralpak AD column. Supercritical CO2 modified with methanol, ethanol and 2-propanol were used as mobile phase. The influence of type and concentration of alcohol modifier on retention factor, enantioselectivity and resolution were studied. Among methanol, ethanol and 2-propanol, 2-propanol was proved to be the most favorable modifier, and 9.5% (v/v) of 2-propanol was the preferred concentration at which racemate 3 could be separated with resolution of 15.86 and retention factor of 6.323. The effects of pressure and temperature were investigated at 9.5% (v/v) of 2-propanol in the pressure range of 12–24 MPa and temperature range of 303.15–318.15 K. It was found that the lower pressure and temperature were favorable to the enantioseparation. Using van’t Hoff plot, the isoenantioselective temperature was calculated to be 410 K. The enantioseparation process was “enthalpically driven” under experimental conditions. Finally, the retention factors were satisfactorily correlated by a simplified lattice–fluid model with average absolute relative deviation (AARD%) of both enantiomers smaller than 1.76%.  相似文献

trans-(-)-Paroxetine is a selective 5-hydroxytryptamine (5-HT) reuptake inhibitor currently used as an antidepressant. trans-(+/-)-3-Ethoxycarbonyl-4-(4'-fluorophenyl)-1-methylpiperidine-2,6-dione is an important intermediate of trans-(-)-paroxetine. It was separated on amylose and tartaric acid-based chiral stationary phases by HPLC. The equilibrium constants and overall mass transfer coefficients together with the axial dispersion coefficients were experimentally determined by moment analysis based on the lumped kinetic model of chromatography. In case of Kromasil CHI-TBB, the equilibrium constants measured were found to be 8.36 and 9.37 for trans-(+) and trans-(-) enantiomers, respectively. For Chiralpak AD-H, the equilibrium constants were 6.68 and 4.13 for trans-(+) and trans-(-) enantiomers, respectively. The axial dispersion coefficients of both enantiomers on Kromasil CHI-TBB column were about one order of magnitude greater than on Chiralpak AD-H. Fast kinetics of mass transfer in both chiral stationary phases was observed. Their overall mass transfer coefficients on Kromasil CHI-TBB and Chiralpak AD-H were 32.12, 33.18, 26.50, 46.85 s(-1) for trans-(+) and trans-(-) enantiomers, respectively. The parameters obtained were utilized to simulate the elution profiles, and the simulated and experimental results match well, which confirmed that the parameters obtained in this study were valid.  相似文献

Three types of ion‐selective electrodes: PVC membrane, modified carbon paste (CPE), and coated graphite electrodes (CGE) have been constructed for determining paroxetine hydrochloride (Prx). The electrodes are based on the ion pair of paroxetine with sodium tetraphenylborate (NaTPB) using dibutyl phthalate as plasticizing solvent. Fast, stable and potentiometric response was obtained over the concentration range of 1.1×10?5–1×10?2 mol L?1 with low detection limit of 6.9×10?6 mol L?1 and slope of a 56.7±0.3mV decade?1 for PVC membrane electrode, the concentration range of 2×10?5–1×10?2 mol L?1 with low detection limit of 1.2×10?5 mol L?1 and slope of a 57.7±0.6 mV decade?1 for CPE, and the concentration range of 2×10?5–1×10?2 mol L?1 with low detection limit of 8.9×10?6 mol L?1 and slope of a 56.1±0.1 mV decade?1 for CGE. The proposed electrodes display good selectivity for paroxetine with respect to a number of common inorganic and organic species. The electrodes were successfully applied to the potentiometric determination of paroxetine hydrochloride in its pure state, its pharmaceutical preparation, human urine and plasma.  相似文献

The synthesis of piperidines and piperidines derivatives in enantiopure fashion has been a challenging goal for organic chemists. In this report we developed a nice cascade reaction for piperidine derivatives based in an amidomalonate Michael addition to enals followed by an intramolecular hemiaminal formation with good yields and enantioselectivities. Moreover we studied the ‘in situ’ intramolecular cyclization of this hemiaminals with alcohols forming fused piperidine-oxazolidines.  相似文献

目的 比较度洛西汀与帕罗西汀治疗首发抑郁症性功能的影响。方法 将72 例首发抑郁症患者随机分为度洛西汀组(36 例)和帕罗西汀组(36 例),观察8周。应用亚利桑那性经验量表(ASEX)评定性功能情况,同时用汉密顿抑郁量表(17 项)(HAMD17)观察疗效和副反应量表(TESS)评定不良反应。结果 治疗第2、4、8 周ASEX 评分度洛西汀组得分显著低于帕罗西汀组,自第2周末始,两组与基线时比较ASEX 得分均有显著下降。疗效观察发现,治疗8周末度洛西汀组有效率80.0%,痊愈率51.4%,帕罗西汀组有效率76.5%,痊愈率47.1%,两组比较差异无统计学意义。治疗第1周末,度洛西汀组HAMD17评分显著低于帕罗西汀组,而第2、4、8 周末两组评分比较差异无统计学意义。治疗第2、4、8 周ASEX 评分度洛西汀组得分显著低于帕罗西汀组(均P<0.05),自第2 周末始,两组与基线时比较ASEX 得分均有显著下降(均P<0.05)。度洛西汀组不良反应发生率42.9%,帕罗西汀组44.1%,两组比较差异无统计学意义。结论 度洛西汀对首次发作抑郁症的性功能的改善优于帕罗西汀,或引起性功能障碍少于帕罗西汀。两者 疗效相似,但第1周度洛西汀起效较帕罗西汀快。总体不良反应相似。  相似文献

Abstract

trans‐4‐(4‐Fluorophenyl)‐3‐chloromethyl‐1‐methylpiperidine 3b was subjected to elimination reaction on alumina or KF‐alumina under solvent‐free conditions and microwave irradiation. Compared with the “classical” method of heating in the presence of an organic base, the microwave‐assisted methodology provided higher yields of 4‐(4‐fluorophenyl)‐3‐methylene‐1‐methylpiperidine 7b (65.5–71%) in considerably shorter reaction times (20–40 min). Furthermore, the exocyclic double bond in 7b underwent rearrangement to the endocyclic double bond to furnish compound 8. The effect of alumina and irradiation time on reaction conversion and the extent of isomerization was examined.  相似文献

3,4-Disubstituted piperidines were synthesized through anti SN2′ allylation of 4-substituted 2-cyclopentenyl esters with reagents based on RMgX and CuX, thus allowing equal access to both trans- and cis-isomers. As an application, the paroxetine intermediate was synthesized efficiently. During the investigation, the MeOCH2CO2 group was found to show high reactivity in the pivotal anti SN2′ type reaction using the reagent derived from (i-PrO)Me2SiCH2MgCl and CuCN.  相似文献
