Improvement in enzyme activity and stability by addition of low molecular weight polyethylene glycol to sodium bis(2-ethyl-L-hexyl)sulfosuccinate/isooctane reverse micellar system

The activity and stability of Chromobacterium viscosum lipase (glycerolester hydrolase, EC 3.1.1.3)-catalyzed olive oil hydrolysis in sodium bis (2-ethyl-1-hexyl)sulfosuccinate (AOT)/isooctane reverse micelles is increased appreciably when low molecular weight polyethylene glycol (PEG 400) is added to the reverse micelles. To understand the effect of PEG 400 on the phase behavior of the reverse micellar system, the phase diagram of AOT/PEG 400/water/isooctane system was studied. The influences of relevant parameters on the catalytic activity in AOT/PEG 400 reverse micelles were investigated and compared with the results in the simple AOT reverse micelles. In the presence of PEG 400, the linear decreasing trend of the lipase activity with AOT concentration, which is observed in the simple AOT reverse micelles, disappeared. Enzyme entrapped in AOT/PEG reverse micelles was very stable, retaining>75% of its initial activity after 60 d, whereas the half-life in simple AOT reverse micelles was 38 d. The kinetics parameter maximum velocity (V _max)exhibiting the temperature dependence and the activation energy obtained by Arrhenius plot was suppressed significantly by the addition of PEG 400.     

On Solutions of the q-Hypergeometric Equation with q N = 1

We consider the q-hypergeometric equation with q ^N = 1 and , , . We solve this equation on the space of functions given by a power series multiplied by a power of the logarithmic function. We prove that the subspace of solutions is two-dimensional over the field of quasi-constants. We get a basis for this space explicitly. In terms of this basis, we represent the q-hypergeometric function of the Barnes type constructed by Nishizawa and Ueno. Then we see that this function has logarithmic singularity at the origin. This is a difference between the q-hypergeometric functions with 0 < |q| < 1 and at |q| = 1.     

Determination of overt carnitine palmitoyltransferase by reversed-phase high-performance liquid chromatography

A method for the determination of carnitine palmitoyltransferase I (CPT I; EC 2.3.1.19) in isolated rat liver mitochondria by reversed-phase high-performance liquid chromatography is described. Enzyme activity is assayed by direct determination of coenzyme A (CoA) released from palmitoyl-CoA within 60 min by a linear gradient system. CPT 1 in rat liver mitochondria can be assayed from only 30 micrograms of mitochondrial protein per millilitre of assay mixture. The changes in the kinetic parameters of CPT I, including Ki for malonyl-CoA, resulting from the fasting-feeding cycle are also discussed.     

Enzyme immunoassay of somatostatin (SS)-like immunoreactive substance in bovine milk

A sensitive and specific enzyme immunoassay (EIA) for somatostatin (SS)-like immunoreactivity (SS-LI) was developed with the use of beta-D-galactosidase labeled antigen. The minimum amount of SS-like immunoreactive substance (SS-IS) detectable by this method was 1.0 fmol/well (25 pmol/l). The level of SS-IS in bovine foremilk was about 20 pmol/l, and the level was unchanged after delivery. On the other hand, the levels of gastrin releasing peptide (GRP)-IS and vasoactive intestinal polypeptide (VIP)-IS in bovine foremilk were very high, but fell during 1 week after delivery to about 10% of those in foremilk.     

Enzyme immunoassay of thyrotropin releasing hormone (TRH).

A sensitive and specific double-antibody enzyme immunoassay (EIA) for a thyrotropin releasing hormone (TRH)-like immunoreactive substance has been developed. In order to synthesize TRH-labeled beta-D-galactosidase (beta-gal), a newly devised TRH derivative, pGlu-His-Pro-NH-(CH2)6-NH2 (TRH-Hex), was employed. TRH-Hex was linked to beta-gal by the N-(epsilon-maleimidocaproyloxy) succinimide coupling procedure. For competitive reactions, the TRH antibody was incubated with standard TRH and TRH-Hex-beta-gal (delayed addition). Free and antibody-bound enzyme hapten were separated by using an anti-rabbit immunoglobulin G coated immunoplate. Activity of the enzyme on the plate was fluorometrically determined. The present immunoassay allows detection of 0.8 to 100 pmol/well of TRH.     

Electromagnetic moments of the doubly closed shell ±1 nucleon nuclei and on-line nuclear polarization

Electromagnetic moments of doubly closed shell ±1 nucleon nuclei were studied. Recent technical developments of the β-NMR was shown that was applicable for the polarized β emitters. Also, the implication of those new moments were discussed especially on the nuclear interactions and non nucleonic degrees of freedom.     

Cadmium recovery by a sulfate-reducing magnetotactic bacterium,Desulfovibrio magneticus RS-1, using magnetic separation

Cadmium recovery by a sulfate-reducing magnetotactic bacterium, Desulfovibrio magneticus strain RS-1, was investigated. D. magneticus precipitated >95% of cadmium at an initial concentration of 1.3 ppm in the growth medium. Electron microscopic analysis revealed that D. magneticus formed electron-dense particles on its surface when cultivated in the presence of cadmium ions (Cd²⁺). Sulfide was also found in the precipitate, and the composition ratio of sulfide/cadmium was 0.7. Sixty percent of viable RS-1 cells was recovered by a simple magnetic separation revealing the removal of 58% cadmium from the culture medium.     

Spectroscopic study of a brush cathode plasma in a magnetic field

The brush cathode helium discharge in the magnetic field has been operated stably at discharge currents larger than those without magnetic field. The diameter of the plasma column has been determined by the configuration of the magnetic field. The measurements of the spectral intensities of the recombination continuum followed by the 2³S-n³P series reveals that the electron density is 1·8 × 10¹³ cm^-3 and the electron temperature is 0·17 eV at a discharge current of 500 mA and a pressure of 0·9 torr for a magnetic flux density of 1·3 kG. The principal quantum number for line merging is 20.     

掺杂ZnSe/BeTe Ⅱ型量子阱结构中带电激子的磁场效应

报道了n型掺杂ZnSe/BeTe/ZnSe Ⅱ型量子阱(type-Ⅱ QW)在极低温 (5—10 K)条件下的各种光学性质. 磁场中(Farada配置)ZnSe层的反射光谱展示了一个典型的负的带电激子(X^-)的跃迁特征. 对于空间间接光致发光(spacially indirect PL)光谱,它的主发光峰显示了一个反玻尔兹曼分布的非对称性,并且在磁场中(Voigt配置)它的峰值能量随磁场的增加而降低. 这些实验结果显示了该掺杂样品的空间间接PL是来自Ⅱ型QW结构所特有的带电激子的跃迁. 关键词： 光致发光 二维电子气 带电激子 Ⅱ型量子阱
