Semens of Astragali Complanati own anti-erectile dysfunction effect; however, the components which contribute to the anti-erectile dysfunction effect remain unclear. This work raised a strategy that integrates liquid chromatography coupled mass spectrometry-based quantitative analysis, anti-erectile dysfunction assessment on impotent rats, and their relationship analysis for pinpointing anti-erectile dysfunction components from semens of Astragali Complanati. For simultaneous quantification of seven major components in raw and salt-processed semens of Astragali Complanati, an accurate and reliable liquid chromatography–mass spectrometry method was developed under multiple reaction monitoring mode. Of note, chloramphenicol was employed as the internal standard. The method showed good linearity and repeatability, where the recovery rates of each component ranged from 98.1 to 104.7%, and the precisions of intra- and interday were all within 3.4%. The method has been used for quantification of the seven major components in 10 batches of raw and salt-processed semens of Astragali Complanati. Then, the anti-erectile dysfunction effects of raw and salt-processed semens of Astragali Complanati were evaluated on impotent rats. Gray relationship analysis and partial least squares regression were combined for elucidating the relationship. As a result, complanatuside, astragalin, complanatoside B, and kaempferol were found to be responsible for anti-erectile dysfunction effect of Astragali Complanati. 相似文献

Some medicines are poorly soluble in water. For tube feeding and parenteral administration, liquid formulations are required. The discovery of natural deep eutectic solvents (NADES) opened the way to potential applications for liquid drug formulations. NADES consists of a mixture of two or more simple natural products such as sugars, amino acids, organic acids, choline/betaine, and poly-alcohols in certain molar ratios. A series of NADES with a water content of 0–30% (w/w) was screened for the ability to solubilize (in a stable way) some poorly water-soluble pharmaceuticals at a concentration of 5 mg/mL. The results showed that NADES selectively dissolved the tested drugs. Some mixtures of choline-based NADES, acid-neutral or sugars-based NADES could dissolve chloral hydrate (dissociated in water), ranitidine·HCl (polymorphism), and methylphenidate (water insoluble), at a concentration of up to 250 mg/mL, the highest concentration tested. Whereas a mixture of lactic-acid–propyleneglycol could dissolve spironolacton and trimethoprim at a concentration up to 50 and 100 mg/mL, respectively. The results showed that NADES are promising solvents for formulation of poorly water-soluble medicines for the development of parenteral and tube feeding administration of non-water-soluble medicines. Honey has good antimicrobial properties and can be used for medical treatment. The antimicrobial properties of unifloral honey varieties are different. In this study, we evaluated the antimicrobial and antioxidant activities of nine kinds of Chinese monofloral honeys. In addition, headspace gas chromatography-ion mobility spectrometry (HS-GC-IMS) technology was used to detect their volatile components. The relevant results are as follows: 1. The agar diffusion test showed that the diameter of inhibition zone against Staphylococcus aureus of Fennel honey (21.50 ± 0.41 mm), Agastache honey (20.74 ± 0.37 mm), and Pomegranate honey (18.16 ± 0.11 mm) was larger than that of Manuka 12+ honey (14.27 ± 0.10 mm) and Manuka 20+ honey (16.52 ± 0.12 mm). The antimicrobial activity of Chinese honey depends on hydrogen peroxide. 2. The total antioxidant capacity of Fennel honey, Agastache honey, and Pomegranate honey was higher than that of other Chinese honeys. There was a significant positive correlation between the total antioxidant capacity and the total phenol content of Chinese honey (r = 0.958). The correlation coefficient between the chroma value of Chinese honey and the total antioxidant and the diameter of inhibition zone was 0.940 and 0.746, respectively. The analyzed dark honeys had better antimicrobial and antioxidant activities. 3. There were significant differences in volatile components among Fennel honey, Agastache honey, Pomegranate honey, and Manuka honey. Hexanal-D and Heptanol were the characteristic components of Fennel honey and Pomegranate honey, respectively. Ethyl 2-methylbutyrate and 3-methylpentanoic acids were the unique compounds of Agastache honey. The flavor fingerprints of the honey samples from different plants can be successfully built using HS-GC-IMS and principal component analysis (PCA) based on their volatile compounds. Fennel honey, Agastache honey, and Pomegranate honey are Chinese honey varieties with excellent antimicrobial properties, and have the potential to be developed into medical grade honey. 相似文献

A new electrochemical sensor based on a carbon nanotube paste electrode modified with a Santa Barbara Amorphous material (SBA-15) decorated with silver nanoparticles, namely CNT/SBA/Ag-PE, was developed. It was successfully applied for individual and simultaneous determination of both paracetamol (PC) and sulfamethoxazole (SMZ) medicines. The electrode exhibited a linear dynamic range of 0.12–110 μmol L−1 for paracetamol and 0.06–70 μmol L−1 for sulfamethoxazole, and detection limits of 38 and 19 nmol L−1, respectively. The proposed sensor offered high sensitivity, fast response time and the potential for detecting both drugs simultaneously. The CNT/SBA/Ag-PE enabled the simultaneous determination of PC and SMZ in urine samples with high recovery rates. 相似文献

该文基于酶辅助靶标循环信号放大策略构建了用于黄曲霉毒素B1(AFB1)高灵敏检测的化学发光适体传感器。以G-四链体/氯化血红素DNA酶为信号分子设计了免标记的适体探针H1-S1和发夹探针H2。适体探针结合目标AFB1,在核酸外切酶I辅助下,触发靶标循环反应产生发夹H1。发夹H1与H2杂交,释放出完整的G-四链体序列,并进一步与氯化血红素结合形成G-四链体/氯化血红素DNA酶。DNA酶通过催化氧化鲁米诺-H2O2化学发光体系产生化学发光信号,实现AFB1的放大检测。在最优实验条件下,化学发光强度与AFB1质量浓度的对数在0.001~100 ng/mL范围内呈良好的线性关系,相关系数(r2)为0.9955,检出限为0.93 pg/mL,回收率为93.7%~107%。该适体传感器操作简单、灵敏度高、特异性好,在黄曲霉毒素污染检测方面具有良好的应用前景。 相似文献

鉴于传统神经网络和支持向量机机理复杂、计算量大的缺陷,很难实时跟踪磷酸铁锂电池组复杂快速的内部反应,影响电池荷电状态的估算精度,提出应用一种简单、有效的极限学习机对一额定容量为100Ah、额定电压为72V的纯电动汽车磷酸铁锂电池组建模,并分别与BP神经网络、RBF神经网络、支持向量机进行对比。随后,以学习时间和泛化性能为优化目标,应用粒子群方法寻找最佳隐层节点个数。结果表明,基于极限学习机的磷酸铁锂电池组模型的学习时间、泛化性能优于BP神经网络、RBF神经网络、支持向量机;隐层节点优化后,模型的学习时间和泛化性能达到最优。 相似文献

In this work, ultra‐performance LC with ESI quadrupole TOF‐MS (UPLC–ESI‐Q‐TOF‐MS) and automated MetaboLynx analysis was used to rapidly separate and identify the chemical constituents of Danggui San, a traditional Chinese medical formula. The analysis was performed on a Waters UPLC BEH C18 column using a gradient elution system. A hyphenated ESI and Q‐TOF analyzer was used for the determination of the accurate mass of the protonated or deprotonated molecule and fragment ions in both positive and negative modes. Based on retention times, accurate mass, and the mass spectrometric fragmentation characteristics, a total of 47 compounds distributed over the chemical groups of phthalides, flavonoids, monoterpene glycosides, sesquiterpenoids, phenolics, and alkaloids, were simultaneously separated within 18 min and identified or tentatively elucidated in Danggui San for the first time. UPLC–ESI‐Q‐TOF‐MS analysis revealed the complexity of the chemical composition of this formula. The method developed is rapid, accurate, reliable, and highly sensitive to characterize the chemical constituents of Danggui San. 相似文献
