Development of ELISA and immunochromatographic assay for ofloxacin

Two rapid,sensitive and reliable immunoassay methods,namely competitive indirect enzyme-linked immunosorbent assay(CI- ELISA)and colloidal gold-based immunochromatographic assay(CGIA),were developed to detect ofloxacin(OFL).The linear range of the CI-ELISAwas from 0.5 to 128 ng/mL with a limit of detection(LOD)of 0.35 ng/mL.Good recoveries were obtained in analyzing simulated swine urine samples.The CGIA could accurately estimate OFL at concentrations as low as 10 ng/mL in less than 10 min,and test results were read visually without any instrument.     

Pharmacokinetic study of free mangiferin in rats by microdialysis coupled with microbore high-performance liquid chromatography and tandem mass spectrometry

Mangiferin (2-beta-D-glucopyranosyl-1,3,6,7-tetrahydroxyxanthen-9-one) has been isolated from the herbal root of Anemarrhena asphodeloides Bung showing antioxidative, antiviral, and anticancer effect. An in vivo microdialysis sampling method coupled to microbore high-performance liquid chromatography (HPLC) was employed for continuous monitoring of free mangiferin in rat blood. Microdialysis probes were inserted into the jugular vein/right atrium and brain striatum of Sprague-Dawley rats, and mangiferin at doses of 10, 30 or 100 mg/kg were then administered via the femoral vein. Dialysates were collected every 10 min and injected directly into a microbore HPLC system. Mangiferin was separated by a reversed-phase C18 microbore column (150 x 1 mm) from dialysate within 10 min. The mobile phase consisted of acetonitrile-0.05% phosphoric acid-tetrahydrofuran (10:75:15, v/v/v) with a flow-rate of 0.05 ml/min. The wavelength of the UV detector was set at 257 nm. The limit of quantification for mangiferin was 0.05 microg/ml and in vivo recovery of mangiferin at concentrations of 1, 5 and 10 microg/ml was in range of 37.7-39.8%. The results indicate that the pharmacokinetics of mangiferin at doses of 10-30 mg/kg reveals a linear relation, while doses of 30-100 mg/kg show a nonlinear pharmacokinetic phenomenon. Mangiferin was undetectable in brain dialysate. The proposed method provides a technique for rapid and sensitive analysis of free mangiferin in rat blood and further application in pharmacokinetic study. Furthermore, the metabolites of mangiferin in the rat bile were confirmed by LC electrospray ionization (ESI) tandem mass spectrometry (MS-MS).     

血镁水平与糖尿病肾病

为探讨糖尿病肾病 (DN)患者血镁水平 ,以 1 5 9例 2型糖尿病 (T2DM)患者为研究对象 ,用xylidylblue比色法测定了其正常白蛋白尿期、微量白蛋白尿期 (早期DN)、临床DN及晚期DN血镁水平 ,与 2 0例正常对照组相比较 ;同时将 1 1 0例DN患者分为肾功能不全代偿组、失代偿组和肾功能衰竭组 ,将其血镁水平与 1 0 2例慢性肾小球肾炎 (CGN)患者相比较 ,组间比较采用t检验 ;对 79例T2DM正常白蛋白尿期及微量白蛋白尿期患者尿白蛋白排泄率 (UAER)与血镁水平进行了等级相关分析。结果表明 ,T2DM正常白蛋白尿期、早期DN及临床DN血镁水平降低 ,晚期DN血镁水平升高 ,各组与正常对照组相比存在显著性差异 (P <0 0 5或P <0 0 1 ) ;T2DMDN及CGN肾功能不全代偿期、失代偿期及肾功能衰竭期血镁水平渐升高 ,且肾功能处于同一期的DN和CGN相比 ,前者血镁水平均较后者显著降低 (P <0 0 1或P <0 0 5 ) ;T2DM正常白蛋白尿期和微量白蛋白尿期血镁水平与UAER呈负相关 (r=0 5 47,P <0 0 1 )。提示T2DM患者肾功能正常时存在低镁倾向 ,但晚期DN血镁水平升高 ;随着肾功能不全进展 ,DN和CGN患者血镁水平升高 ,但前者血镁水平仍较后者低 ,镁代谢紊乱与DM及其并发症的相互关系有待进一步研究 ;血镁水平测定可否作为DN的早期诊断指标亦     

有序自组装聚合物纳米结构

采用水辅助方法(water-assisted fabrication method),分别以4-十二烷基苯磺酸掺杂的聚苯胺(PANI-DBSA)和聚2-甲氧基-5-(2′-乙烯基-己氧基)苯乙炔(MEH-PPV)两种功能高聚物为成膜材料,冷凝水滴为模板,利用水滴在聚合物溶液表面的自组装,制备出了两种纳米层次以上的蜂窝状有序多孔聚合物薄膜.通过原子力显微镜和共聚焦荧光显微镜对其形貌、电学性质和荧光图像进行了表征.     

离子液体支载稀土催化剂催化 2,2-二甲基-三亚甲基碳酸酯均聚及其与ε-己内酯的共聚

首次合成了离子液体支载稀土催化剂,并探讨了其催化2,2-二甲基-三亚甲基碳酸酯(DTC)均聚及其与ε-己内酯(ε-CL)的共聚,结果表明该新型催化剂能有效地催化DTC的均聚及其与ε-CL的共聚.研究了不同稀土以及加料顺序等反应条件对聚合的影响,发现轻稀土的催化活性比重稀土高,其中离子液体支载La催化剂的催化活性最高,而通过控制不同的加料顺序可以制备DTC和ε-CL的无规及嵌段共聚物.对共聚物结构进行了表征.     

一测多评方法应用于高效液相色谱法测定防晒产品中15种防晒剂的含量

选用CAPCELL PAK C18色谱柱为分离柱,柱温为30℃,进样体积为10.0μL,并用不同比例的(A)甲醇、(B)四氢呋喃和(C)0.008 2mol·L^-1高氯酸溶液的混合液作为流动相,按程序梯度洗脱模式对市售防晒产品中所用的15种防晒剂的标准品进行色谱分离,并在波长311nm处进行紫外检测。实样分析时,称取样品0.25g,用甲醇、四氢呋喃、水和高氯酸(体积比为250∶450∶300∶0.2)的混合液(以下简称混合溶剂)15 mL超声提取30 min,用混合溶剂定容至25.0mL,离心10min,分取上清液1.0mL,加入混合溶剂定容至10.0mL,经0.45μm滤膜过滤,取滤液按仪器工作条件进行高效液相色谱分析。结果表明:所测定的15种防晒剂在一定的质量浓度范围内与其对应的峰面积呈线性关系。选定其中的二苯酮-3为参照物,并根据其余14种化合物的质量浓度和峰面积计算了各化合物的相对校正因子和相对保留时间等参数,确定了用一测多评法(QAMS)测定防晒产品中15种防晒剂的条件。还对色谱柱的型号、色谱仪器的型号以及柱温、进样量等对相对校正因子和保留时间可能产生影响的因素进行了系统试验。证明了在选定的色谱柱型号的前提下,用QAMS方法可实现防晒产品中15种防晒剂含量的同时测定。应用QAMS方法测定了6个批次防晒产品中的防晒剂含量,所得结果与用标准曲线法计算的结果基本一致,表明QAMS方法在降低检测成本和节省检测时间方面效果显著。     

对叠氮苯甲酸与β-环糊精的包合作用

合成了对叠氮苯甲酸与β-环糊精的包合物，并利用红外、热重、核磁和紫外 等测试方法进行了详细的表征与研究。包合后客体化合物的热稳定性有了明显的提 高，其分解温度提高了+115 ℃；此外还利用紫外光谱的数据计算了包合反应的平 衡常数，为(7933±192)L·mol~(-1)。     

塑料成型工艺实习CAI课件的开发

针对当前学生实习面临的困难，以Authorware为开发工具，编制了塑料成型工艺实习CAI课件，整个软件图文并茂，图像，动画，解说，音乐相结合，形象地介绍了塑料各种成型工艺的主要原理，生产设备，工艺流程和相关产品，应用本软件学生可以进行模拟实习，该软件激发了学生学习的兴趣，提高了实习质量。     

Structural determinants of steroids for cytochrome P450 3A4-mediated metabolism

Cytochrome P450 3A4 (CYP3A4), a major member of cytochrome P450 isoenzymes, metabolizes the majority of steroids in 6β-position. For the purpose of determining requisite structural features of a series of structurally related steroids for CYP3A4-mediated metabolism, three-dimensional pharmacophore modeling as well as electrotopological state map were conducted for 15 steroids. Though prior studies speculated that the chemical reactivity of the allylic 6β-position might have a greater influence on CYP3A4 selective 6-hydroxylation than steric constraints in the enzyme, our results reveal that for CYP3A4 steroidal substrates, it is not the chemical reactivity of atoms at 6β-site, but the pharmacophoric features, i.e. the two hydrophobic rings together with two H-bond donors, that act as the key factors responsible for determining the CYP3A4 selective 6-hydroxylation of steroids.
