建立了气相色谱-串联质谱(GC-MS/MS)同时测定6种乳制品中232种农药残留的检测方法。样品经1%乙酸乙腈提取后,加入无水醋酸钠和无水硫酸镁液液分离,然后加入无水MgSO4、C18和N-丙基乙二胺(PSA)进行分散固相净化,上清液经氮吹浓缩复溶后,结合气相色谱-串联质谱测定,用多反应离子监测(T-MRM)模式进行检测,基质匹配内标法定量。结果显示:232种农药线性关系良好,相关系数均大于0.99,方法的定量下限为0.000 4 ~ 0.05 mg/kg,在0.01、0.02、0.10 mg/kg加标水平下的平均回收率为65.2% ~ 110%,相对标准偏差(RSD)为3.6% ~ 13%。该方法样品处理简单快速,相比其他多残留分析方法净化效果好,灵敏度和选择性高,适用于日常检测工作。  相似文献

氯霉素是一种广谱抗生素,化妆品卫生规范规定氯霉素等抗生素为禁用物质。关于氯霉素的检测多采用高效液相色谱(HPLC)和高效液相色谱-质谱(HPLC-MS)测定,或者经N,O-双三甲摹硅烷基-三氟乙酰胺(BSTFA)、三甲基氯硅烷(TMCS)等硅烷化后进行气相色谱(GC)、GC-MS测定。本文建立了采用较为普通的乙酸酐试剂进行衍牛化,然后用GC-MS测定的方法。将该法应用于化妆品中氯霉素的检测,取得了较好的效果。  相似文献

采用电喷雾串联质谱技术对两组单核苷酸多态性(SNP)样品进行了研究,初步研究结果显示应用串联质谱可快速鉴定SNP分型以及确定SNP位点。采用16.3 mmol/L三乙胺(TEA)-400 mmol/L六氟异丙醇(HFIP)缓存溶液,电喷雾负离子模式下,长度为20 bp的寡核苷酸的MS扫描谱图显示其母离子呈较宽的多电荷态分布。选择低电荷态[M-4H]4-母离子,碰撞能为54 eV时,MS/MS扫描得到丰富的碎片离子,通过鉴定特征离子碎片(w和a-B离子系列)可确定其序列。  相似文献

建立了高效液相色谱-串联质谱(HPLC-MS/MS)测定化妆品中 8-甲氧基补骨脂素和 5-甲氧基补骨脂素含量的方法。以甲醇提取样品,以甲醇和水为流动相,经HPLC分离后以多级反应监测(MRM)测定。两种补骨脂素在2.0~100.0 μg/L范围内线性良好,检出限均为0.10 mg/kg,回收率分别在96.88%~101.81%和94.25%~101.26%之间,相对标准偏差分别在0.40%~6.02%和0.76%~4.26%之间。  相似文献

建立了气相色谱-串联质谱同时测定食用油中对苯二甲酸二(2-乙基己基)酯和偏苯三甲酸三(2-乙基己基)酯两种非邻苯二甲酸酯类增塑剂的方法。样品采用乙腈提取,于-20℃冷冻除脂净化,用气相色谱-串联质谱选择反应监测模式测定。两种化合物的检出限为0.03 mg/kg,定量限为0.1 mg/kg,线性范围为0.1~10 mg/kg。3个添加水平(0.1、0.3和1.0 mg/kg)下的回收率为81.04%~108.31%,相对标准偏差为0.70%~9.91%。该方法简便、准确,适用于食用油中对苯二甲酸二(2-乙基己基)酯和偏苯三甲酸三(2-乙基己基)酯的检测。  相似文献

目前,毛细管电色谱(CEC)在中草药分析中显示出一些独特的优势,如:CEC流动相相对简单、用量少、峰容量大、更易与质谱(MS)联用。特别是植物提取物中活性成分复杂,需要对色谱峰进行精确的指认并控制峰纯度;采用CEC—MS方法可在线提供中药中各种化合物的质谱图。此外,有许多天然产物在中药中的含量极低,MS的高灵敏度可弥补紫外检测器灵敏度较低的缺陷。因此,研究CEC分离中药成分的选择性问题具有重要的实际意义。蝉翼藤(Securida cainappendiculata Hassk.)为远志科远志族蝉翼藤属攀援灌木,其根茎叶有多种药效,其化学成分主要为黄酮、山酮(XAN)和皂甙。从蝉翼藤中已分离出的10个XAN类成分(结构见图1)。具有显著的抗氧化和保护神经细胞作用。我们曾用毛细管区带电泳(CZE)、高效液相色谱(HPLC)、毛细管胶束电动色谱(MEKC)和毛细管微乳电动色谱(MEEKC)对这10个XAN进行分离,并就电泳条件的变化对分离选择性的影响进行系统研究。本文采用CEC法对这10个XAN成分进行分离,并对其色谱分离条件进行优化研究,以期为今后CEC与MS联用分析中药成分打下基础。  相似文献

采用气相色谱-四极杆飞行时间串联质谱(GC-QTOF MS)对加拿大一枝黄花中的挥发性成分进行检测。利用气相色谱的保留指数、四极杆飞行时间串联质谱的高分辨多级质谱数据以及谱库检索等对该植物的叶、茎、根三个部位中的挥发性成分进行结构鉴定。共鉴别出挥发性成分54种,其中叶47种、茎35种、根32种。主要成分均为蒎烯、表双环倍半水芹烯、柠檬烯等萜烯类化合物。结果表明,气相色谱的高分离度和四极杆飞行时间串联质谱的高分辨率可以有效地对复杂样品中的挥发性成分进行检测,结合多维数据定性技术能够显著提高未知成分鉴别的准确性。  相似文献

The formation of cyclic dipeptides (DKPs, 2,5-diketopiperazines) from dipeptides having proline (Pro) as the fixed N-terminal amino acid was investigated by analytical pyrolysis with a heated platinum filament coil. Glutamic acid (Glu), aspartic acid (Asp), glutamine (Gln), lysine (Lys) or arginine (Arg) was the terminal amino acid. Products evolved from off-line pyrolysis at 500 °C were analysed by GC–MS as such or after trimethylsilylation. The structure of a novel DKP from the pyrolysis of Pro-Glu, namely hexahydrodipyrrolo[1,2-a:1′,2′-d]pyrazine-3,5,10(10aH)-trione, was established by ESI-MS/MS and extensive NMR analysis. This DKP could be identified in the pyrolysates of dipeptide Pro-Gln, tripeptide Pro-Glu-Leu, collagen and bovine serum albumin (BSA). Pyrolysis of Pro-Lys and Lys-Pro-Leu afforded the cyclo(Pro-Lys) tentatively identified by interpretation of its trimethylsilyl (TMS) derivative mass spectra and the DKPs resulting from the deamination of the lateral chain. The dipeptide Pro-Asp yielded isomeric cyclo(Pro-Asp) revealed as TMS derivatives and cyclo(Pro-Ala) from the decarboxylation of the side chain. Analytical pyrolysis of the above peptides as well as of Pro-Tyr, collagen and BSA enabled the compilation of GC (retention times relative to an internal standard) and MS data (characteristic ions) of over eighty DKPs including their TMS derivatives, the latter ones useful in the identification of thermal degradation products of proteinaceous materials in several matrices.  相似文献

A new complex of cyclic peptide lactone antibiotics from Bacillus subtilis, which we named Maltacines has recently been described. The structure elucidation of three of them is reported in this paper. The amino acid sequences and structures of the peptides were found by MS(n) of the ring-opened linear peptides that gave uninterrupted sequences of Bn and Y'n ions. The identities of four unknown residues in the sequences were solved by a combination of derivatisation with phenylisothiocyanate (PITC), high-resolution mass spectrometry and H/D exchange. The nature and position of the cyclic structure was disclosed by a chemo-selective ring opening with Na18OH and was found to be a lactone formed between a hydroxyl of residue number 4 and the C-terminal amino acid number 12. For verification of the structure of the B2 + ion, peptides with different combinations of P/Q and P/K at the N-terminus were synthesized. The structures of the four peptides is tentatively suggested to be: D1a: cyclo(4,12)-P-Q-Y-Adip-A-E-T-Y-Orn-HGly-Y-I-OH, D1b: cyclo(4,12)-P-Q-Y-Adip-A-E-T-Y-Orn-S-Y-I-OH and D1c: cyclo(4,12)-P-Q-Y-Adip-A-E-T-Y-K-S-Y-I-OH. Porphyrin amino acid conjugates with one or two porphyrin units were analyzed by electrospray ionization tandem mass spectrometry (ESI-MS/MS). The ESI-MS spectra of all the porphyrins studied, obtained in positive ion mode, show the presence of the corresponding protonated molecule [M+H]+; ESI-MS spectra of diporphyrinyl compounds also show the doubly charged ions [M+2H]2+. The fragmentations of these ions induced by collision with argon were studied (ESI-MS/MS). ESI-MS/MS gives detailed structural information about the amino acids associated with the porphyrin. Cleavage of the bonds in the vicinity of the porphyrin moiety and those involving the side chain of amino acid residues gives structural information about this type of association. A fragmentation common to all derivatives corresponds to the cleavage of the phenyl-CO bond. The expected cleavage of the amide bond, that links the porphyrin to the amino acid moiety, is a minor fragmentation, which in some cases is even absent. The MS/MS spectra of the monoporphyrinyl derivatives show product ions characteristic of the amino acid linked to the porphyrin; the fragmentation also indicates when the amino acids has a terminal carboxylic group or a terminal ester group. The fragmentations of the diporphyrinyl compounds occur mainly by the cleavage of the spacer, leading, in the case of the doubly charged ions, to predominantly mono-charged ions, indicating a preferential location of the two protons in separated porphyrinic units.  相似文献

The characteristics shown in the electrospray ionization/ion trap mass spectra of ring-opened LI-F antibiotics (cyclic depsihexapeptides with a 15-guanidino-3-hydroxypentadecanoic group as a side-chain) were examined. Collision-induced dissociation (CID) MS of protonated molecules of the depsipeptides produced many fragment ions. Most of these fragment ions contained information for determining the amino acid sequences of antifungal antibiotics. The fragment ions were classified into six groups (b(n'), B(n'), B'(n'), beta(n'), y(n) and Y(n)). According to MS(3) spectra, the B(n'), B'(n) and beta(n) ions can be considered to be derived with a cleavage at each CO--NH in the peptide bonds of [MH--NH(3)](+),[MH--NH(3)--OH](+) and [MH--NH(3)--2H(2)O](+), respectively, in ion trap MS. Losses of NH(3) and H(2)O from the amino acid residues of the depsipeptides in ion trap MS are likely to be smaller than those from the side-chain. The measurements with electrospray ionization (ESI)/ion trap MS of depsipeptides with a side chain containing polar groups may provide useful information for structural determination.  相似文献

High-throughput proteomics aims to investigate dynamically changing proteins expressed by a full organism, specific tissue or cellular compartment under certain conditions. High-sensitivity mass spectrometry has gradually become a significant tool for characterizing peptides. Here, we analyzed angiotensin II using ultra-fast liquid chromatography (UFLC) coupled with matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-ToF MS). First, we applied UFLC in isolating and collecting the angiotensin II, and then Axima-Resonance (MALDI-QIT-ToF MS(5)) was adopted, which enables collision-induced dissociation-MS(5) analysis for fine structural characterization of angiotensin II. Resultant MS, MS(2), MS(3) and MS(4) spectra of interested [M+H](+) ions selected as precursor ions yielded detailed information about the sites of fragmentation as well as the amino acid sequence for angiotensin II; meanwhile, the average deviation between theoretical mass and actually measured mass from MS to MS(5) spectra was only 0.32 Da. It indicated that Axima-Resonance was capable of analyzing the peptide sequence accurately and provide the corresponding fragmentation information thoroughly, thus suggesting a potential strategy involving UFLC assay coupled with MALDI-QIT-ToF MS(5) analysis on high-throughput proteomics study in future.  相似文献

MS/MS is indispensable for the amino acid sequencing of peptides. However, its use is limited for peptides containing disulfide bonds. We have applied the reducing properties of 1,5-diaminonaphthalene (1,5-DAN) as a MALDI matrix to amino acid sequencing and disulfide bond mapping of human urotensin II possessing one disulfide bond, and human guanylin possessing two disulfide bonds. 1,5-DAN was used in the same manner as the usual MALDI matrices without any pre-treatment of the peptide, and MS/MS was performed using a matrix-assisted laser desorption/ionization quadrupole ion trap time-of-flight mass spectrometer (MALDI QIT TOFMS). The results demonstrated that MS/MS of the molecular ions reduced by 1,5-DAN provided a series of significant b-/y-product ions. All 11 amino acid residues of urotensin II were identified using 1,5-DAN, while only 5 out of 11 residues were identified using 2,5-dihydroxybenzoic acid (DHB); similarly 11 out of 15 amino acid residues of guanylin were identified using 1,5-DAN, while only three were identified using DHB. In addition, comparison of the theoretical and measured values of the mass differences between corresponding MS/MS product ions using 1,5-DAN and DHB narrowed down the possible disulfide bond arrangement candidates. Consequently, 1,5-DAN as a reductive matrix facilitates rapid amino acid sequencing and disulfide mapping for peptides containing disulfide bonds.  相似文献

Determining the structure and dynamics of large biologically relevant molecules is one of the key challenges facing biology. Although X-ray crystallography (XRD) and nuclear magnetic resonance (NMR) yield accurate structural information, they are of limited use when sample quantities are low. Mass spectrometry (MS) on the other hand has been very successful in analyzing biological molecules down to atto-mole quantities and has hence begun to challenge XRD and NMR as the key technology in the life sciences. This trend has been further assisted by the development of MS techniques that yield structural information on biomolecules. Of these techniques, collision-induced dissociation (CID) and hydrogen/deuterium exchange (HDX) are among the most popular. Despite advances in applying these techniques, little direct experimental evidence had been available until recently to verify their proposed underlying reaction mechanisms. The possibility to record infrared spectra of mass-selected molecular ions has opened up a novel avenue in the structural characterization of ions and their reaction products. On account of its high pulse energies and wide wavelength tunability, the free electron laser for infrared experiments (FELIX) at FOM Rijnhuizen has been shown to be ideally suited to study trapped molecular ions with infrared photo-dissociation spectroscopy. In this paper, we review recent experiments in our laboratory on the infrared spectroscopic characterization of reaction products from CID and HDX, thereby corroborating some of the reaction mechanisms that have been proposed. In particular, it is shown that CID gives rise to linear fragment ion structures which have been proposed for some time, but also yields fully cyclical ring structures. These latter structures present a possible challenge for using tandem MS in the sequencing of peptides/proteins, as they can lead to a scrambling of the amino acid sequence information. In gas-phase HDX of an amino acid it is shown that the structure can be changed from a charge solvated to a zwitterionic structure, thereby demonstrating that HDX can be an invasive technique, in fact changing the structure of the analyte. These results emphasize that more fundamental work is required in order to understand the underlying mechanisms in two of the most important structural techniques in MS.  相似文献

This study compares the main commercial detectors that can detect amino acids in their underivatized form. The detectors tested are: the chemiluminescent nitrogen detector (CLND), the evaporative light scattering detector (ELSD), the nuclear magnetic resonance spectrometer, conductivity detector, refractive index, UV, and electrospray quadrupole mass spectrometry (in simple and tandem MS mode). As ELSD, CLND and MS require a volatile mobile phase, an ion-pair reversed-phase liquid chromatographic system was selected, consisting of an octadecyl column and an aqueous mobile phase containing pentadecafluorooctanoic acid as volatile ion-pairing reagent. Underivatized taurine, hypotaurine, aspartic acid, hydroxyproline, asparagine, serine, glycine, glutamine, cysteine, glutamic acid, threonine and alanine were simultaneously analysed with each detector. In order to test the applicability of these detectors to "real world" samples, the amino acid stoichiometry of the tetrapeptide Gly-Gly-Asp-Ala was determined with each detector after acid hydrolysis. The detectors were compared in terms of linearity, limit of detection, advantages and disadvantages as well as special features (capacity to provide structural information, specificity, quantification with single calibration curve, etc.).  相似文献

Product ions obtained by tandem mass spectrometry (MS/MS) are quite effective for the amino acid sequencing of linear peptides. However, in the case of cyclic peptides, the fragmentation pattern is complicated because the cleavages occur randomly and product ions are generated as a(n), b(n), c(n), x(n), y(n) and z(n) series ions; therefore, the authors have never obtained sufficient sequence information. In order to overcome this problem, we applied ion trap liquid chromatography/multi-stage mass spectrometry (LC/MS(n)) and characterized the product ions obtained from anabaenopeptins and aeruginopeptins as the cyclic peptides. For the anabaenopeptins, MS(2) analysis did not provide sufficient sequence information on the cyclic structure, and MS(3) analysis was applied to sequence the constituent amino acids. Diagnostic product ions were obtained by the MS(3) analysis and were quite effective for obtaining the sequence information of the constituent amino acids. MS(2) analysis was, however, sufficient to obtain the sequence information of the aeruginopeptins. In both cases, the resulting product ions obtained from the cyclic structures were formed by the two-bond fission mechanism of the precursor ion, in which an initial fission of the cyclic structure to a linear one and subsequent fission(s) at the peptide bonds are included. The fragmentations were similar for the structurally related compounds, indicating that the cleavages occurred at definite peptide bonds. In addition, the resulting product ions are generated as b(n) series ions and the mass difference facilitates the amino acid sequencing. Thus, ion trap LC/MS(n) provides sequence information, and the resulting product ions are reproducible among the structurally related compounds and reliable for the sequencing of the constituent amino acids of the cyclic structure.  相似文献
