10-羟基喜树碱首先在N,N-二甲基甲酰胺(DMF)中经N-溴代丁二酰亚胺(NBS)溴代得到9-溴-10-羟基喜树碱, 9-溴-10-羟基喜树碱和氯甲酸乙酯反应得到9-溴-10-羟基喜树碱-10,20-双乙氧基碳酸酯(6). 化合物6和烯丙基三正丁基锡通过Stille偶联反应[9]得到关键中间体7, 最后水解化合物7得到目标化合物. 通过柱层析纯化得到纯度大于99.8%, 单杂小于0.1%的吉咪替康(HPLC). 所有中间体及目标产物经1H NMR, 13C NMR, LRMS, HRMS表征确证. 相似文献

一种可循环使用的固相试剂:N-烷基-N-酰基磺酰胺聚苯乙烯基微球(5), 通过对聚苯乙烯磺酰氯微球树脂进行两步功能基化的修饰反应来制备. 制备过程如下:聚苯磺酰氯树脂(1)与伯胺(2)反应得到聚苯乙烯基N-烷基磺酰胺树脂(3), 树脂3用酰氯(4)或酸酐酰化得到N-烷基-N-酰基磺酰胺聚苯乙烯基树脂(5). 酰化的树脂5作为酰基转移试剂与亲核试剂胺反应得到二级酰胺. 根据5上取代基对酰胺生成的程度的影响结果表明, 烷基R1和酰基(R2CO)对酰基转移反应活性的大小依次分别为:苯基>苄基>甲基>正丁基>>H和对硝基苯甲酰基(苯甲酰基>乙酰基. 胺的亲核能力对酰胺的收率也有一定的影响. N-苯基-N-苯甲酰基磺酰胺树脂重复使用3次没有发现活性降低. 相似文献

以二甲酰亚胺钾3a~3g与2-氯-5-氯甲基吡啶的N原子氧化后得到的2-氯-1-氧-5-氯甲基吡啶发生亲核取代反应, 用传统和微波两种方法合成了7种未见文献报到的化合物N-(2-氯-1-氧-5-吡啶甲基)二甲酰亚胺类化合物4a~4g. 对比两种合成方法, 在常压下, 微波辐射作为反应热源具有用时少、环境友好、易纯化和产率高的特点. 这些目标化合物4a~4g的结构经元素分析结果, IR, GC-MS, 1H NMR, 13C NMR确证. 初步的生物活性测定结果表明, N-(2-氯-1-氧-5-吡啶甲基)二甲酰亚胺类部分化合物具有良好的杀虫活性. 相似文献

5-氨基四唑(1)经过重氮化反应得到5-硝基四唑(5), 5与甲醛反应得到2-羟甲基-5-硝基四唑(6), 6与HCl或HBr反应分别得到5-5氯代四唑(7)和5-溴代四唑(8), 采用MS, IR, 1H NMR, 13C NMR等技术对这些化合物进行了表征. 用X射线单晶衍射法测定了化合物5-硝基四唑钠(4), 5和6的晶体结构. 化合物4属于三斜晶系, P-1空间群; 化合物 5和6均属于单斜晶系, P21空间群, 化合物6的晶胞参数a=0.66131(18) nm, b=0.54905(15) nm, c=0.7566(2) nm, Z=2, V=0.27470(13) nm3, Dc=1.754 g/cm3, F(000)=128, μ=0.160 mm-1. 相似文献

以N-保护的氨基酸钾盐与1-(ω-溴丙基)-5-氟尿嘧啶和1-(ω-溴丁基)-5-氟尿嘧啶反应,制备了18种氨基酸的ω-(N1-5-氟尿嘧啶基)-丙醇酯和丁醇酯的盐酸盐,并确定了它们的结构。动物试验的初步结果表明,酪氨酸、苯丙氨酸的3-(N1-5-氟尿嘧啶基)-丙醇酯盐酸盐对小鼠艾氏腹水癌的抑制率分别为88.1%和86.7%。 相似文献

In order to get good antitumor agents especially better than 5-fluorouracil, tegafur and l-hexylcarbamoyl-5-fluorouracil (HCFU), fourty nine 1-carbamoyl-5-fluorouracil having aromatic ring were synthesized from 5-fluorouracil and isocyanates or mines. Antitumor activity was tested in the L1210 tumor system, and 5 compounds gave better value of therapeutic ratio than 5-fluorouracil, tegafur, HCFU. 1-(4-Methoxybenzylcarbamoyl)-5-fluorouracil gave the best result. 相似文献

提出了一种采用谷氨酸席夫碱Ni(II)配合物共保护L-谷氨酸的α-氨基和α-羧基合成γ-L-谷氨酰二肽的新方法. 首先由手性助剂——2-[N-(N-苄基-脯氨酰)氨基]二苯甲酮(1)、六水合氯化镍和L-谷氨酸反应, 得到谷氨酸席夫碱Ni(II)配合物2, 产率为98.2%; 进而采用二异丙基碳二亚胺(DIC)/1-羟基-苯并三唑(HOBt)复合缩合剂法分别与L-氨基酸3a~3h反应, 得到相应的γ-L-谷氨酰二肽席夫碱Ni(II)配合物4a~4h, 产率为93.1%~99.0%; 最后稀酸水解配合物, 得到γ-L-谷氨酰二肽5a~5h, 产率为73.0%~86.4%, 高收率(92.2%~97.4%)回收手性助剂. 中间产物和终产物的结构经由旋光, 1H NMR, 13C NMR和HRMS表征. 相似文献

A series of six novel 5-fluorouracil derivatives 1-6 were synthesized and their structures confirmed by (1)H- and (13)C-NMR, MS and elemental analysis. The preliminary in vitro antitumor activities against B16, K562 and CHO cells and the in vivo inhibitions of liver cancer H(22) demonstrated that some of these compounds effectively inhibit the growth of tumor cells, but the in vivo trials in mice revealed that the compounds also exhibited serious liver and lung tissue toxicity. The hydrolysis experiments indicated that this type of compound did not readily liberate 5-fluorouracil, as expected. 相似文献

1‐Amino‐2‐pyrid‐3‐yl‐5‐(2‐benzoylethylthio)‐s‐triazole ( 1 ) was condensed with 1‐amino‐3‐mercapto‐5‐ [(un)substituted phenyl]‐s‐triazoles and subsequently substituted with chloroacetic acid to afford bis‐s‐triazole sulfanylacetic acid mono‐Schiff bases ( 3a – 3e ), which were condensed with 9‐formylanthracene to produce asymmetric bis(s‐triazole Schiff base) sulfanylacetic acids ( 4a – 4e ).
