This paper reviews the recent developments in bioanalysis sample preparation techniques and gives an update on basic principles, theory, applications and possibilities for automation, and a comparative discussion on the advantages and limitation of each technique. Conventional liquid-liquid extraction (LLE), protein precipitation (PP) and solid-phase extraction (SPE) techniques are now been considered as methods of the past. The last decade has witnessed a rapid development of novel sample preparation techniques in bioanalysis. Developments in SPE techniques such as selective sorbents and in the overall approach to SPE, such as hybrid SPE and molecularly imprinted polymer SPE, have been addressed. Considerable literature has been published in the area of solid-phase micro-extraction and its different versions, e.g. stir bar sorptive extraction, and their application in the development of selective and sensitive bioanalytical methods. Techniques such as dispersive solid-phase extraction, disposable pipette extraction and micro-extraction by packed sorbent offer a variety of extraction phases and provide unique advantages to bioanalytical methods. On-line SPE utilizing column-switching techniques is rapidly gaining acceptance in bioanalytical applications. PP sample preparation techniques such as PP filter plates/tubes offer many advantages like removal of phospholipids and proteins in plasma/serum. Newer approaches to conventional LLE techniques (salting-out LLE) are also covered in this review article.  相似文献

To overcome the limitations of the detection systems associated with gas or liquid chromatography, a sample pretreatment is required with the objective to provide a sample fraction enriched with all the target analytes and as free as possible from other matrix components. There is now no doubt that solid-phase extraction (SPE) has now become the method of choice for carrying out simultaneously the extraction and concentration of many compounds in aqueous samples. Many recent applications of SPE to multiresidue analysis are reviewed with an emphasis on the importance of the choice of the sorbent and of the sample volume. SPE is particularly well adapted to multiresidue analysis including compounds from a wide range of polarity or characterized by various physico-chemical properties. However, SPE is not completely free from practical problems inherent to the nature of the compounds or to the coupling to the chromatographic systems. Many examples are reported to illustrate these problems which can in most cases be circumvented. New developments in SPE are also reviewed.  相似文献

A review with 136 references on the principles and recent developments in the solid-phase extraction based on polymer sorbents is presented. New polymer-based materials, chromatographic modes, experimental configurations are described and their advantages for a rapid sample preparation of certain classes of compounds with different functional groups are discussed and compared to silica-based sorbents.  相似文献

This paper highlights recent advances in the solid-phase extraction (SPE) of quaternary ammonium herbicides in water, soil, plant and biological samples. After a brief introduction summarizing the properties of quaternary ammonium herbicides and the difficulties involved in measuring them, attention is paid primarily to solid supports used for isolation and concentration, pre-treatments required for the different matrices, and eluents applied for quantitative desorption of these analytes. The determination techniques used after SPE and applications of the proposed SPE methodology are also briefly discussed.  相似文献

Sorbent preconcentration offers good strategies to overcome the poor detection limits of capillary electrophoresis (CE). The present review focuses on the recent trends of the coupling between sorbent preconcentration techniques, namely solid-phase extraction (SPE) and solid-phase microextraction (SPME), to capillary electrophoresis (CE). Special attention is given to their environmental and biological application. We also discuss the most important advantages and disadvantages of the different methodologies and briefly outline the new trends of the coupling between sorbent preconcentration and CE.  相似文献

Molecularly imprinted polymers (MIPs) are tailor-made synthetic materials possessing specific cavities designed for a target molecule. Since they recognise their target analyte with affinities and selectivities comparable to those of antibody–antigen, enzyme–substrate and ligand–receptor interactions, they are often referred to as synthetic receptors or plastic antibodies. In this review, we describe the great potential and recent developments of MIPs in affinity separations, with emphasis on their application to the solid-phase extraction (SPE) of analytes from complex matrices. Research efforts made in this field to obtain water-compatible polymers for their applicability in aqueous environments are described. We particularly discuss problems encountered in the use of MIPs in SPE and the attempts carried out to improve their efficiency.  相似文献

Fluorescence correlation spectroscopy has been increasingly used in polymer science. In the present perspective, the principles of the method are briefly reviewed, and the temporal and spatial resolutions are critically discussed. Examples of recent findings are summarized, focusing on polymer solutions, environmental parameters, combination with other techniques, near-interface measurements, simulations, and modeling. Sample preparation for phenol analysis using solid-phase extraction (SPE) is reviewed. The scope of the review has been restricted to the literature dealing with the analysis of phenols as the main objective. The use, advantages and disadvantages of silica sorbents, polymeric, functionalized, carbon-based and mixed available sorbents, when applied to the separation and preconcentration of phenols, as well as the available experimental devices, are discussed. Other aspects such as phenol derivatisation prior to SPE, solid-phase microextraction, matrix effects and the storage of phenols in SPE cartridges, have been also discussed.  相似文献

双酚类化合物作为一类内分泌干扰物广泛存在于环境介质中,经过多种途径迁移至人体后,可对人体产生内分泌毒性、细胞毒性、基因毒性、生殖毒性、二噁英毒性和神经毒性,已被加拿大政府风险评估识别为进一步优先控制名录。随着环境领域对双酚类化合物的广泛关注,相关研究工作逐渐向水、沉积物、灰尘和生物样品等多介质开拓。但是,由于不同环境样品在基质复杂性和污染物浓度水平等方面存在显著差异,开发提取效率高、净化选择性好、普适性强、操作简单、高通量的提取和净化方法,有助于实现环境介质中双酚类化合物的高灵敏、批量检测。近年来,新型前处理技术发展迅速,尤其是固相萃取技术,在双酚类化合物提取与净化方面取得了长足的发展,不仅在一定程度上克服了传统提取净化方法存在的耗时、耗力和耗溶剂等不足,而且为新型污染物分析提供了更多的技术支持。该文简述了典型双酚类化合物的理化性质、用途用量和环境危害,重点围绕新型固相萃取吸附剂开发和固相萃取模式转变两个方面,总结了固相萃取在双酚类化合物提取净化方法方面取得的进展。商品化固相萃取产品普适性强,在环境监测领域应用范围较广,适用于双酚类化合物的产品种类有限;新型吸附剂研发聚焦吸附容量(如介孔硅材料、碳纳米材料、金属-有机框架材料、环糊精)和选择性(如分子印迹聚合物和混合模式离子交换聚合物)两个方面,种类多样化可满足不同检测需求;越来越多的高灵敏分析仪器不断推向市场,为适应新的发展形势,固相萃取模式正逐渐向微型化、自动化、简易化等方向发展,如QuEChERS、固相微萃取、磁固相萃取等。  相似文献

Solid-phase extraction (SPE) for two groups of polyaromatic compounds--polycyclic aromatic hydrocarbons and naphthalenesulfonates--with completely different problems in the extraction process are reviewed. The sorbents used in each case and the different steps of SPE are studied. Particular problems encountered in the SPE of each group are described. Adsorption problems of PAHs which require an organic solvent or a surfactant to be added to the sample are explained. The need of ion-pair solid-phase extraction for extracting naphthalenesulfonates and the influence of the inorganic species in the extraction are discussed. The use of an appropriate sample handling technique is a must in an analysis of organic micropollutants in water. The efforts to use a solid phase for the recovery of analytes from a water matrix prior to their detection have a long history. Since the first experimental trials using activated carbon filters that were performed 50 years ago, solid-phase extraction (SPE) has become an established sample preparation technique. The initial experimental applications of SPE resulted in widespread use of this technique in current water analysis and also to adoption of SPE into standardized analytical methods. During the decades of its evolution, chromatographers became aware of the advantages of SPE and, despite many innovations that appeared in the last decade, new SPE developments are still expected in the future. A brief overview of 50 years of the history of the use of SPE in organic trace analysis of water is given in presented paper.  相似文献

A review with 75 references is presented that deals with the reported methods for analysis of some important central nervous system (CNS) drugs in biological fluids utilizing stir bar sorptive extraction (SBSE) technique covering the years from 2000 to 2008. The theoretical aspects of SBSE, as well as an significant number of applications have been published, showing the advantages of this technique over the classical extraction techniques (liquid–liquid extraction (LLE) and solid-phase extraction (SFE). This article reviews some of the recent developments in the extraction and clean-up areas of biological samples dedicated to dioxin and related compound analysis. A brief introduction on the major dioxin contamination events, which have occurred in the food chain, is given to illustrate the need of fast high throughput methods in case of crises. The emphasis of this paper is the method development based upon reliable instrumental extraction techniques for rapid sample processing and automation such as; supercritical fluid extraction (SFE), microwave-assisted extraction (MAE), pressurized liquid extraction (PLE) and, solid-phase extraction (SPE). The PLE and SPE are also discussed in conjunction with the use of a multi-column automated clean-up system that can accommodate up to 5 g of extracted lipids. The fractionation in sub-groups of analytes during the clean-up process allows the isolation of various types of toxicants from a single sample and illustrates the versatility of the system. An integrated extraction and clean-up instrument is finally presented in terms of feasibility and attainable sample turnover for the parallel processing of liquid and solid biological samples.  相似文献
