结合单因素试验和响应面法优化苦槛蓝总生物碱的最佳提取工艺条件,并对其抑菌活性进行研究。以苦参碱为对照品,采用酸性染料比色法,以总生物碱含量为指标,分别考察乙醇体积分数、提取温度、提取时间和料液比4个因素对醇提法提取苦槛蓝总生物碱的影响,在此基础上通过Box-Behnken设计-响应面法确定最佳提取工艺,并进行验证。并检测其大肠杆菌和金黄色葡萄球菌的抑菌活性。苦槛蓝总生物碱的最佳提取工艺为:乙醇体积分数86.18%、提取温度75.42℃、提取时间2.622h、料液比0.080。验证实验下总生物碱提取率为(0.400 6±0.009 2)%,与模型结果接近。苦槛蓝提取物对大肠杆菌的抑菌直径达到33.17±0.53mm,金黄色葡萄球菌抑菌直径达到32.62±0.19mm。Box-Behnken设计响应面法能够有效地优化苦槛蓝总生物碱的提取工艺,并且苦槛蓝总生物碱提取物对大肠杆菌和金黄色葡萄球菌有明显的抑菌效果,这为苦槛蓝生物碱类化学成分的分离及其药理活性的深入研究提供了实验依据。  相似文献

按《中华人民共和国药典》分别对净制后的小茴香进行清炒、盐制、醋制、酒制和姜汁制,并提取出活性挥发油,采用气相色谱-质谱联用仪和薄层色谱对提取物进行分析。小茴香经不同辅料炮制后挥发油含量显著降低;与生品相比,炮制品均含有相同的主要活性成分,其中反式-茴香脑含量最高,顺式-番桧烯水合物、顺式-罗勒烯、α-松油烯、4-松油烯、对位-烯丙基茴香醚和枯敬醛为首次在小茴香药材中检测到;鉴定出的24种化合物经不同方法炮制后含量均发生了明显变化,且发生了相应的转化,这是导致不同炮制品具有不同药性和功效的重要原因。  相似文献

建立了高效液相色谱(HPLC)同时测定知母不同炮制品中新芒果苷、芒果苷、知母皂苷BIII、知母皂苷I和知母皂苷AIII含量的方法,探讨了不同炮制方法对知母化学成分的影响,为通过采用不同炮制方法来改变知母药性提供了依据。采用AlltimaTM C18色谱柱(250 mm×4.6 mm, 5 μm)分离,所用流动相为乙腈(A)和0.1%甲酸水溶液(B),梯度洗脱,流速为0.8 mL/min;用紫外检测器检测新芒果苷与芒果苷,检测温度为室温,检测波长为265 nm。用蒸发光散射检测器检测知母皂苷BIII、知母皂苷I和知母皂苷AIII,漂移管温度为50 ℃,气体压力为179.1 kPa(26 psi)。知母经炮制后,5种化合物含量均发生了不同程度的改变,表明炮制方法的不同对知母化学成分的含量影响较大。该方法对进一步研究炮制方法对知母的药效学影响提供了一定的参考依据。  相似文献

本文优化了蒙药漏芦花多糖的提取工艺,并评价其抗氧化和抗菌活性。以超声功率、超声温度、超声时间和浸泡时间为影响因素,多糖提取率为评价指标,在单因素实验的基础上利用正交试验优化提取工艺,测定漏芦花多糖对DPPH自由基、羟基自由基清除效果,检测其抗氧化活性,涂布平板法测定多糖抗菌活性。超声波辅助法提取蒙药漏芦花多糖的最佳工艺条件为超声功率180W,超声温度70℃,超声时间30min,浸泡时间40min,在此条件下多糖得率为1.125%。抗氧化实验表明,漏芦花多糖浓度在0.5~2.5mg·mL~(-1)范围内,随着漏芦花多糖浓度增加,总还原能力不断增强;其体外清除DPPH自由基能力也随浓度增大而增强,清除率最高达72.98%;当多糖浓度为1.5mg·mL~(-1)时,漏芦花多糖体外清除羟基自由基能力最强,为7.24%。漏芦花多糖浓度为12.8mg·mL~(-1)时,对沙门氏菌的抑菌圈直径为9.72mm。超声法提取蒙药漏芦花多糖操作简便、准确、灵敏、重现性好,漏芦花多糖抗氧化能力良好,且对沙门氏菌有一定的抑菌效果。  相似文献

以超声波提取法对蒌叶中抑菌活性成分进行提取,考察不同溶剂、料液比、提取时间、提取温度、提取功率对提取工艺的影响,同时采用96孔板法和倾注平板法测定了提取物对枯草杆菌、金黄色葡萄球菌、大肠杆菌、毛霉、黑曲霉、青霉的最小抑菌浓度;并考察了温度、光照、放置时间对提取物抑菌活性稳定性的影响。结果表明:蒌叶抑菌活性成分提取的最佳工艺条件为,无水乙醇为溶剂、m_(蒌叶干粉)∶V_(乙醇)=1.75∶10(g·mL~(-1))、提取时间为60 min、提取功率为192 W、提取温度为50℃时,提取物的抑菌效果最好。对大肠杆菌、枯草杆菌、金黄色葡萄球菌、黑曲霉、毛霉、青霉最小抑菌浓度分别为5.00、2.50、2.50、1.25、1.25、2.50 mg·mL~(-1)。同时,提取物的石油醚部位和氯仿部位富集有抑菌活性很强的物质。在设定的温度范围、光照条件下,蒌叶提取物的抑菌活性较稳定。因此,蒌叶乙醇提取物的抑菌活性具有一定的光热稳定性。  相似文献

在单因素试验的基础上,利用星点设计-响应面法来优化酶提取法提取麦冬总黄酮的提取工艺,同时利用DPPH法和Fenton法测定总黄酮提取液清除1,1-二苯基-2-三硝基苯肼自由基和羟基自由基的能力。结果表明麦冬总黄酮酶提取法的最佳提取工艺条件:酶用量为46.61 mg,酶解时间为4.14 h,酶解温度为55.56℃,提取率为0.283%。在此条件下,麦冬总黄酮提取液对DPPH自由基和羟基自由基有较强的清除作用,随着总黄酮浓度的升高,其抗氧化能力逐渐增强,其IC_(50)分别为7.33 mg·L~(-1)和14.08 mg·L~(-1)。该工艺稳定可行,可为麦冬黄酮类化合物的开发利用提供参考。  相似文献

中药刺五加叶中有效成分的几种微波辅助提取方法研究   总被引:13,自引:0,他引:13  
利用高压微波提取法、常压回流微波提取法、流动微波提取法和索氏提取法提取中药刺五加中黄酮和皂苷类化合物,考察各种提取方法提取效率随时间的变化。结果表明,高压微波提取法在压力为300kPa,完全提取的时间为10min;常压回流微波提取法完全提取的时间为14min;流动微波提取法完全提取的时间为10min;索氏提取法需8h尚能完成。各种提取方法总黄酮的提取率分别为43.7、48.2、42.7和34.7mg/g;总皂苷的提取效率分别为60.2、61.5、59.8和46.5mg/g。与索氏提取法比较,微波提取法提取黄酮和皂苷,提取时间大大缩短,且提取效率显著提高。  相似文献

对阿尔泰狗哇花总皂苷的提取与纯化工艺进行了研究.利用正交设计确定了提取温度、乙醇浓度、溶剂用量、时间及次数等提取工艺参数.比较了五种大孔吸附树脂和聚酰胺树脂对阿尔泰狗哇花总皂苷的吸附与脱附性能.结果表明,在优化的提取条件下提取,经AB-8大孔吸附树脂柱层析分离纯化,制得总皂苷产品,其含量比原药材提高约7倍多,进一步完善后可适用于工业化生产.  相似文献

对虎耳草全草中总黄酮物质进行有效提取,并将蒸发浓缩后的虎耳草提取液分别进行了体外抗氧化、抗炎、抑菌性能以及美白功效等研究。结果表明,虎耳草提取液在较高浓度时的抗氧化功效与维生素C相当;具有良好的抑制炎症性能,其乙酸乙酯萃取物消炎性能更优;虎耳草提取物能够抑制常见的普通细菌的生长,其正丁醇萃取物抑菌性能最佳;在美白功效测试实验中提取液浓度为4000μg/mL时对酪氨酸酶活性的抑制率为79.25%。  相似文献

Radix Ophiopogonis is a widely used traditional Chinese medicine. The quality of Radix Ophiopogonis available in the market varies, and some confusing or fake herbs exist. In order to improve the quality control of Radix Ophiopogonis, a novel fingerprinting method was established using HPLC coupled with UV and evaporative light-scattering detectors (ELSDs). Extraction with methanol and liquid-liquid extraction with water-saturated n-butanol were employed for the preparation of the sample solution. Chromatographic separation was performed on a Lichrospher C(18) column (250x4.6 mm id, 5.0 microm particle size) with a linear gradient elution program. UV detection at 280 nm and evaporative light-scattering detection were utilized to obtain two subfingerprinting chromatograms. A novel protocol for data processing was proposed, in order to identify and remove redundant data obtained by the two detectors, and balance the weight of the two subfingerprints on the similarity values. The method was validated and applied to quality evaluation of 16 samples of Radix Ophiopogonis and related herbs.  相似文献

Dioscorea nipponica Makino exhibits many biological activities, including relieving cough, eliminating phlegm and preventing asthma. The present study extensively evaluated the extraction process, major components, antioxidant, antibacterial and anti-inflammatory activities of total saponins extraction from Dioscorea nipponica Makino. In this study, the optimal extraction process of total saponins extract was optimized by single-factor test and response surface methodology as follows: extraction time 25 min, ethanol concentration 50 % and liquid to material ratio 55:1 ml/g, and the extraction rate was 1.72 %. Eighteen components were initially analyzed by UPLC-QTOF-MS method. Although total saponins extract exhibited mild antibacterial activities against Escherichia coli, Salmonella, Staphylococcus aureus and Streptococcus, and antioxidant activities against ferric-ion, ABTS and DPPH radicals, the perfect anti-inflammatory activity of TSE was demonstrated by significantly reducing the content of NO and the phagocytic activity in LPS induced RAW 264.7 cells, which provided a theoretical basis for the research and development of new anti-inflammatory Chinese medicine.  相似文献

This article reports the lipophilic chemical composition of different processed products (Changii Radix, Changii Radix Alba) and parts (root bark, leaf and fruit) of Changium smyrnioides Wolff.. The lipophilic constituents were extracted with petroleum ether in Soxhlet apparatus, subsequently identified and determined by gas chromatography–mass spectroscopy (GC–MS). Yield of lipophilic constituents from Changii Radix (3.65%) was about three times more than Changii Radix Alba's (1.07%), which indicated processing by boiling in water had an impact on the content of lipophilic constituents. Moreover, the major compounds in different processed products and parts were found to be fatty acids and sesquiterpenes. The results are a contribution for the lipophilic chemical composition and can serve as a reference for product development of Changium smyrnioides Wolff..  相似文献

This study developed a robust and reliable approach using liquid chromatography– tandem mass spectrometry for the simultaneous determination of five saponins in rat tissues: β‐ecdysterone, chikusetsusaponin IV, ginsenoside Ro, 25S‐inokosterone and chikusetsusaponin IVa. This is the first report on a comparative tissue distribution study of crude and salt‐processed Radix Achyranthes in rats. After one‐step protein precipitation by acetonitrile, the tissue samples were sent to LC–MS/MS for multiple reaction monitoring. The retention times of the five saponins and internal standard were 1.77, 3.14, 3.01, 1.83, 3.26 and 4.77 min. The standard curves showed good linear regression (r2 > 0.9991) in the range of 10.3–1562.5 ng/mL. The intra‐ and inter‐day accuracy and precision were within 15% of the nominal concentration. The recoveries of the five saponins were 92.0–99.9%. Finally, this approach was successfully applied to tissue distribution analysis of the five saponins after oral administration of crude and salt‐processed Radix Achyranthes in rats. The largest concentration of the five saponins was observed in kidney after salt‐processing, which indicated that processing could enhance the bioavailability.  相似文献

Codonopsis Radix, a popular food homology medicine, is widely used in clinical traditional Chinese medicine and food supplement, raw products and three types of processed products are the main forms of decoction pieces in China. However, there is no scientific basis for comprehensive chemical characterization of raw and three types of processed products. Herein, we investigated qualitatively and quantificationally secondary and primary metabolites in raw Codonopsis Radix and three types of processed products by metabolomics and glycomics employing multiple chromatography-mass spectrometry technology combined with chemometric analysis further to look for differential compounds and propose the processing-induced chemical mechanisms. The results indicated that Codonopsis Radix became dark-colored and the smell of burnt incense odor was observed after processing. The principal component analysis demonstrated that secondary metabolome and glycome were significantly altered between raw and processed products, and 36 differential secondary metabolites and 11 differential primary metabolites were finally screened through orthogonal partial least-squares-discriminant analysis. The main types of compounds are alkaloids, terpenoids, glycosides, amino acids, monosaccharides, oligosaccharides, and furfural derivatives. Meanwhile, Chemical mechanisms could be involved, including oxidation, glycosidic hydrolysis, esterification, dehydration, and Maillard reaction. This work supplies a chemical basis for the application of various types of Codonopsis Radix decoction pieces.  相似文献

应用TAM Air微量热系统, 采用微量热法, 考察不同提取方法的板蓝根水煎液及其化学萃取部位在37 ℃时, 对大肠杆菌生长代谢的影响. 结合中医药理论, 分析板蓝根水煎液及其化学萃取部分的药效作用及其差异, 挑选板蓝根药材最佳的提取方法和抑菌最强的活性部位, 并采用试管稀释法求出抑菌活性最强部位的最小抑菌浓度(MIC). 结果表明微量热法可用于常用中药——板蓝根的提取方法和活性部位的挑选, 为进一步研究中药的药性基础提供了一种新的、快速灵敏的技术和方法.  相似文献

Deng‐Zhan‐Sheng‐Mai capsules are a well‐known traditional Chinese patent medicine that was developed in China for the treatment of ischemic stroke. Its quality control focuses on Erigerontis Herba but ignores the contributions of Ginseng Radix et Rhizoma, Schisandrae Chinensis Fructus, and Ophiopogonis Radix. To improve the quality standards for this medicine, this work reports the application of a systematic ultra high performance liquid chromatography tandem mass spectrometric method coupled with chemometrics. Three qualitative and quantitative parameters are established for the evaluation of quality: chemical profiling, the relationship between the contents of 18 compounds and the antioxidant activity, and chemometric analysis. A total of 55 compounds, including 20 phenolic acids, 10 flavonoids, 15 saponins, and 10 lignans, were identified. The method for the quantitative determination of the aforementioned 18 compounds was validated. The limit of quantification ranged from 0.13 to 9.60 ng/mL. The overall recoveries ranged from 95.31 to 103.54%. Hierarchical cluster analysis and principal component analysis were applied to the data of 18 components in ten batches of samples. Nine compounds, including scutellarin, 3,5‐O‐dicaffeoylquinic acid, 4,5‐O‐dicaffeoylquinic acid, ginsenoside Rb1, ginsenoside Re, ginsenoside Rg1, ophiopogonin D, schisandrin, and schisandrol B, are suggested as chemical markers for evaluating the quality.  相似文献

建立了超高效液相色谱-四极杆飞行时间质谱(UPLC-QTOF-MS)结合多元统计分析技术对不同加工何首乌中化学成分差异的分析方法。何首乌样品采用甲醇在室温下超声提取后,采用UPLC-QTOF-MS进行分析,对采集的图谱通过峰匹配、峰对齐、滤噪处理等进行特征峰提取,然后用主成分分析(PCA)和偏最小二乘法-判别分析(PLS-DA)对数据进行分析。结果显示,不同加工何首乌样品间的化学组成存在显著性差异;根据一级质谱精确质荷比和二级质谱碎片信息,结合软件数据库搜索及相关文献进行成分鉴定,初步筛选并鉴定出33种不同加工何首乌间差异显著的化学成分,其中15种为共有差异化学成分,并呈现出不同的变化规律。研究结果可为揭示不同加工方法对何首乌代谢产物差异性的影响规律提供依据。  相似文献

The current study focused on the extraction technology, components analysis, in vitro antioxidant and antibacterial activities of total flavonoids and fatty acids from Tribulus terrestris L. fruits. The extraction process of total flavonoids and fatty acids was optimized by the response surface method, and the compositions were identified from the two extracts by HPLC–DAD–ESI–MS? and GC–MS, respectively. In addition, the antioxidant and antibacterial activities were evaluated by assay of ABTS, DPPH radical scavenging activity, ferric reducing antioxidant power and minimal inhibitory concentration. The yields of total flavonoids and fatty acids were 0.46 and 9.76% under the optimized conditions. Moreover, nine and eight compositions were identified from the two extracts based on the related references, respectively. In addition, total flavonoids and fatty acids extracts both exhibited certain antioxidant and antibacterial activities. The present findings suggest that total flavonoids extracted from T. terrestris L. fruits comprised a more interesting candidate than fatty acids for the research and development of natural and healthy antioxidants and antibacterial agents for the pharmaceutical and food industries.  相似文献
