The goal for the treatment of type-1 diabetes mellitus is the development of an artificial pancreas to regulate the blood glucose level in a closed loop design. A system of nonlinear differential equations having five variables (glucose, insulin, β-cell mass, α-cell mass, and glucagon) with seventeen parameters is considered. The Lyapunov function is used to check the stability analysis of the model. Controllability and observability of the linearized model are discussed under two different cases: in case 1 insulin is taken as an input, and in case 2 insulin and glucagon are taken as inputs for the system. This model played an important role in the development of a fully automatic artificial pancreas by stabilizing the control loop system for the glucose-insulin glucagon pump. A proportional integral derivative (PID) controller is designed for an artificial pancreas by using the transfer function. According to the desired value, the algorithm of an artificial pancreas measures the glucose level in the blood of a patient by using a glucose sensor that sends a signal to an insulin glucagon pump to adjust the basal insulin. A closed-loop system is tested in the Simulink environment, and the simulation results show the performance of the designed controller.  相似文献

Diabetes is a major public health problem. Development of new therapies that are able to improve glycemia management, cure diabetes, and can even protect from it, are of great interest. This study investigated the protective effect of sodium tungstate against STZ-induced beta-cell damages by means of stereological methods. Sixty rats were divided into six groups: control (C), tungstate-treated control (TC), STZ-induced diabetic (D), STZ-induced diabetic rats were treated by sodium tungstate from 1 week before STZ injection (TDB), food-restricted diabetic (FRD), and diabetic rats treated with sodium tungstate 1 week after STZ administration (TDA). Stereological estimation of pancreas volume, islets volume density, volume-weighted mean islets volume and mass of beta cells, islets, and pancreas and total number of islets were done. Islets volume density, volume-weighted mean islets volume, and mass of beta cells, islets, and pancreas of TDB group was significantly higher than D, FRD and TDA groups (P < 0.001) and was comparable to controls (C and TC groups). Total number of islets, pancreas wet weight and volume did not show any significant changes between these groups (P > 0.05).Results suggested that sodium tungstate preserves pancreatic beta cells from STZ-induced damages and diabetes induction in rats.  相似文献

Artificial pancreas is one of the solutions to control the type1 diabetes mellitus now days. Efforts are being made to develop the artificial pancreas. The controllability and observability of the glucose insulin glucagon dynamical model is calculated which is the modified form of composite model of glucose insulin glucagon dynamics for type 1 diabetes mellitus is considered. The concept of controllability and observability for the linearized control system is used to design a feedback control. This model completely describes the glucagon affects the safety of artificial pancreas to overcome the risk of hyperglycemia. Composite model of glucagon-glucose dynamics and its extension is treated for type 1 diabetes mellitus to check the linear controllability and observability. Two cases are discussed on the basis of input of the system. For case I, insulin as the only input and glucose as an output and in case II insulin and glucagon as inputs and glucose as only output. The model is observable but not controllable in each case. This may show vital part in the progress of fully automatic artificial pancreas and stabilize the control loop system for the glucose insulin glucagon (GIG) pump to control the type 1 diabetes mellitus.  相似文献

Islets of Langerhans were isolated from the Sprague Dawley rat pancreas digested by injected collagenase, and purified by Ficoll density gradient centrifugation. In order to make smaller and more uniform microencapsulated islets, we designed a special high-voltage electrostatic microcapsule generator. The effects of operational parameters of the generator on the size and the uniformity of microcapsules were analyzed, such as the voltage, the plunger speed of suspension delivery to the needle tip, the distance between needle tip and solution surface. The optimal parameter combinations for making microcapsules are: 5 kV of voltage, 50 mm/h of the plunger speed, and 20 mm distance. The high-voltage electric system can produce uniform microcapsules with diameters ranging from 0.3 to approximately 0.5 mm, which are smaller and more uniform than those produced by air-jet system. A comparison of the cryopreservation effects between microencapsulated islets and unencapsulated islets showed that the microcapsules can protect the fragile islets from freezing damage, and increase the retrieval rate from 68.5% to 92.6%. Xenotransplantation of the cryopreserved rat islets resulted in the normalization of the metabolic blood glucose of the diabetic mice for 90 days, whereas the unencapsulated islets were easily fragmented and lost during the freezing process. They only reversed hyperglycemia for less than 3-5 days.  相似文献

Pancreatic β-cells play a central role in the maintenance glucose homeostasis by secreting insulin, a key hormone that regulates blood glucose levels. Dysfunction of the β-cells and/or a decrease in the β-cell mass are associated closely with the pathogenesis and pathophysiology of diabetes mellitus, a major metabolic disease that is rapidly increasing worldwide. Clarification of the mechanisms of insulin secretion and β-cell fate provides a basis for the understanding of diabetes and its better treatment. In this review, we discuss cell signaling critical for the insulin secretory function based on our recent studies.  相似文献

In present paper series of trimethine cyanines modified in 5,5′- or 6,6′- position with hydroxy- or methoxy- substituents is studied for their ability to interact selectively with fibrillar formations. Processes of dye aggregation that accompany this interaction were also investigated. Meso-methyl trimethynecyanines with 5,5′- methoxy (7519) and hydroxy (7515) substituents strongly (up to 40 times) increase fluorescence intensity in the presence of fibrillar insulin, and also give noticeable fluorescent response on the presence of various aggregated proteins (lysozyme, β-lactoglobulin, α-synuclein A53T). 7519 and 7515 dyes can be used for fluorometric detection of fibrillar insulin at concentrations of approximately 1.5–120 microg/ml. For meso-ethyl substituted dye 7514 the ability to form H- and J-aggregates upon interaction with insulin fibrils was suggested. The model of the H- and J-aggregate packing in the protein fibrillar structure has been proposed.  相似文献

基于核酸对甲基青莲6B的减色效应,以阳离子型染料甲基青莲6B为生色探针,研究该染料与核酸的结合反应,建立了新的核酸测定体系,研究探讨了体系的作用机理。在pH=9.0条件下,hs-DNA,sm-DNA, ct-DNA,yeast RNA的浓度与甲基青莲6B减色效应成线性关系,响应线性范围分别为0.50~4.00 μg·mL-1,0.20~5.00 μg·mL-1,0.20~5.00 μg·mL-1,0.20~4.50 μg·mL-1,检出限(3σ/K)分别为0.082,0.037,0.038,0.041 μg·mL-1。分析核酸合成样品,回收率为93.5%~105.0%。  相似文献

Beta cells in pancreas represent an example of coupled biological oscillators which via communication pathways, are able to synchronize their electrical activity, giving rise to pulsatile insulin release. In this work we numerically analyze scale free self-similarity features of membrane voltage signal power density spectrum, through a stochastic dynamical model for beta cells in the islets of Langerhans fine tuned on mouse experimental data. Adopting the algebraic approach of coherent state formalism, we show how coherent molecular domains can arise from proper functional conditions leading to a parallelism with “phase transition” phenomena of field theory.  相似文献

Protein complexes that mediate secretion and adhesion are located on the plasma membrane of pancreatic β cells. Neuroligins and their binding partners, the neurexins, are among these complexes. β cell maturation and physiologically regulated insulin secretion, as a response to high levels of blood glucose, are dependent on their three-dimensional (3D) arrangement. Both insulin secretion and the proliferation rates of β cells dramatically increase when β cells are co-cultured with clusters of a member of the neuroligin family: NL-2. A membranal protein, such as NL-2, has very limited drugability owing to its low biostability and bioavailability. Thus, based on in silico modeling, a short NL-2 peptide (HSA-28), which was able to mimic NL-2-positive effects on β cells, was designed, as we described in previous publication. However, the peptide was active only as a cluster, created by the covering the maghemite (γ-Fe2O3)-based nanoparticles (NPs) with limited biocompatibility. In this brief communication, we will show that conjugation of HSA-28 to biocompatible hydrogel NPs exhibits an impressive protective effect on INS-1E β cells under oxidative stress and induces their proliferation rate via augmentation of PDX1 nuclear translocation. The diameter of coated by the peptide NPs was 206?±?63 nm (DLS) and 114?±?27 nm (cryo-TEM). This significant change in size can be explained by the very hydrophilic character of the proteinoid NPs, inducing adsorption of many water molecules on their surface, which are accounted only by the DLS. The ability of biocompatible hydrogel NPs to prevent apoptosis and increase β cell mass might be used for developing novel β cell protective therapies.

Background  

The peptide CART is widely expressed in central and peripheral neurons, as well as in endocrine cells. Known peripheral sites of expression include the gastrointestinal (GI) tract, the pancreas, and the adrenal glands. In rodent pancreas CART is expressed both in islet endocrine cells and in nerve fibers, some of which innervate the islets. Recent data show that CART is a regulator of islet hormone secretion, and that CART null mutant mice have islet dysfunction. CART also effects GI motility, mainly via central routes. In addition, CART participates in the regulation of the hypothalamus-pituitary-adrenal-axis. We investigated CART expression in porcine pancreas, GI-tract, adrenal glands, and thyroid gland using immunocytochemistry.  相似文献

The extra chromosome 21 of Down syndrome (DS) or trisomy 21 patients contains an average of 40 extra copies of rRNA genes and the in vivo/in vitro regulation of the activity of these genes is not fully understood. The objective of this work was to compare the NORs expression pattern in interphase lymphocytes of DS patients with regular trisomy 21 and control individuals according to phytohemagglutinin (PHA) concentration (0.37, 0.75, 1.48 and 2.21 ml) per 100 ml of medium. Because the AgNOR staining is an indicator of the active rRNA genes, comparison of the image analysis values of the AgNOR area in 72 h cultivated lymphocytes for each concentration of PHA between DS patients (N=30) and controls (N=24) provided a plausible conclusion on the regulation of the extra rRNA genes in DS lymphocytes. The nucleolus organizer regions area/total nuclear area (NORa/TNa) was calculated using an in-house computer program. Fifty consecutive interphases per PHA concentration were analysed for each individual, for determination of the NORa/TNa. In contrast to healthy controls, NORa/TNa of lymphocytes from DS patient babies/children (0-8 years old) increased gradually in parallel with the PHA concentration in the culture medium: 10.44+/-1.72% for 0.37 ml of PHA, 11.74+/-1.93% for 0.75 ml of PHA, 13.25+/-2.03% for 1.48 ml of PHA and 13.43+/-2.08% for 2.21 ml of PHA per 100 ml of medium. Contrary to control cells (in which the NORa/TNa ratio according to PHA concentration in the culture medium remains constant), DS interphase lymphocytes in culture do not down-regulate their NOR expression. These results obtained from interphase NORs are consistent with the previous results obtained by evaluating the mean of AgNOR+ chromosome number in metaphase cells, also in relation to the mitogen concentration in the culture medium.  相似文献

研究了在微乳液(溴化十六烷基三甲基胺(CTMAB)/正戊醇/正庚烷/水)介质中,痕量铬(Ⅵ)催化过氧化氢氧化茜素红的褪色反应,建立了测定痕量铬的催化动力学新方法。在微乳液体系中,方法的灵敏度比在表面活性剂CTMAB体系中提高了72.5%,催化反应的表观活化能为55.7 kJ·mol-1。水浴温度升高,灵敏度显著提高,校准曲线在不同的反应温度下线性范围不同,在80 ℃反应温度下,线性范围为2.4~75 μg·L-1,检出限达4.27×10-10g·mL-1,相对标准偏差为1.84%(n=11)。该法已应用于电镀废水中铬(Ⅵ)的测定,结果令人满意。  相似文献

The standard method for determining the 15N abundance of total dissolved nitrogen (TDN) in aqueous samples (e.g., soil leachate, sewage, urine) is currently Kjeldahl digestion followed by steam distillation or diffusion to isolate the ammonium, and then 15N measurement using IRMS. However, this technique is both time-consuming and laborious. One way of overcoming these disadvantages could be to couple a TOC analyser to determine the TDN with a sufficient quadrupole MS to determine the 15N abundance. The high TOC analyser (Elementar Analysensysteme Hanau, Germany), which catalytically oxidises the sample's total nitrogen with a high, constant yield to nitrogen monoxide (NO), appeared particularly suitable. The quadrupole-MS ESD 100 (InProcess Instruments Bremen, Germany) proved to be a suitable mass spectrometer for the 15N determination of NO. This combination of instruments was found to provide a workable method in numerous measurements of standard and actual samples. The detection limit concerning the N amount required per analysis is 2 microg, corresponding to an N concentration of 0.7 mg/l in a maximum sample volume of 3ml. Depending on the N concentration, 15N abundances starting from 0.5 at.% can be measured with the required precision of better than 3% (simple standard deviation). For example, measuring the abundance of 0.5 at.% requires about 50 microg N, whereas for 1 at.% or more only about 5 microg N is needed per analysis.  相似文献

基于SO2在微碱性条件下使孔雀绿褪色的反应原理, 结合顺序注射进样技术建立了光度分析测定大气中SO2的方法. 对试剂加入顺序、各种试剂的浓度、注入体积、反应系统流速等实验参数进行了优化. 采用顺序注射进样, 极大地节省了试剂和试样的用量, 显著地提高了分析速度, 可达到60样·h-1. 方法的线性响应范围0.25~4.0 μg·mL-1, 检出限为0.15 μg·mL-1, 11次重复测定的相对标准偏差为1.5%. 对实际样品的回收率为94%~108%.  相似文献

Although the large use of diethylcarbamazine (DEC), as the major anti-filaricide drug, its mechanism of action remains a matter of controversy. Several authors defend the hypothesis that DEC has no direct effect on nematodes. This study demonstrated that infective larvae (L3) of Wuchereria bancrofti treated in vitro with DEC presented several behaviour and morphological changes. The first alteration produced by treatment for 2 h with 3, 5, 10 microg/ml of DEC was the reduction of motility. Larvae treated with 5, 10 microg/ml DEC showed severely affected organelles, formation of several vacuoles, mainly in neurocytes and in the muscle cells, and dissolution of cytoplasm. Some larvae showed extreme cellular disorganization with abundance of large and dense mitochondria and numerous large vacuoles containing residual organelles. Lamellar bodies, probably related to an assembly of hipodermal membranes, were also observed in some damaged larvae. Thus, undoubtedly in vitro treatment with concentrations of DEC similar to therapeutic conditions, which are 1-5 microg/ml (Hawking, 1979), had a direct effect on infective larvae of W. bancrofti by causing, primarily neuromuscular alterations with subsequent damage to organelles.  相似文献

应用偏最小二乘(PLS)算法对同步荧光光谱严重重叠的诺氟沙星、盐酸洛美沙星和乳酸左氧氟沙星药物三组分混合体系进行波谱解析,建立了该混合体系含量同时测定的新方法。在pH 2.87 B-R缓冲溶液中,波长差Δλ=190 nm 时,诺氟沙星、盐酸洛美沙星和乳酸左氧氟沙星的测量线性范围分别为0.016~0.40 μg·mL-1,0.01~0.336 μg·mL-1和0.01~0.336 μg·mL-1;检出限分别为0.012 6,0.006和0.007 2 μg·mL-1。采用PLS法结合同步荧光法对鳗鱼样品进行测定,结果令人满意。  相似文献
