月季花黄色素清除超氧自由基和羟自由基作用研究   总被引:4,自引:1,他引:3  
月季花(rosa chinensis)是一种常见中草药,为蔷薇科植物.月季花味甘,性温,有活血调经散瘀止痛的功能[1].月季花色素属花青苷类色素[2],花期长,花量大,花色多,分布广泛,是一种较理想的食用天然色素资源.常新丽等对月季花色素的稳定性及对亚硝酸盐的清除作用、提取工艺、理化性质、光谱性质及稳定性和抗油脂过氧化性等进行了研究[3-8],而有关超声波提取的月季花黄色素对超氧阴离子自由基(O2-·)和羟自由基(OH·)的清除作用未见报道.作者曾经对超声波提取月季花红色素工艺条件进行了研究[9],本文进一步研究了月季花黄色素对OH·和O2-·自由基的清除作用[10-14].  相似文献

对福建产花生壳进行了微波萃取天然黄色素及其稳定性的研究 .研究表明微波萃取的工艺条件是 :以pH =3,体积分数 70 %的乙醇水溶液作提取剂 ,原料与提取剂配比为 1g∶5mL ,微波辐射功率为 12 0W ,辐射时间2 4 0s.对该色素稳定性研究结果表明 ,该色素为水溶性色素 ,属黄酮类色素 ,适用pH值范围比较宽 ,尤其碱性状态效果最佳 ,对光、热稳定性好 ,大多数食品添加剂对色素稳定性影响不大 ,对氧化剂H2 O2 的耐受能力较差 ,对还原剂Na2 SO3 的耐受能力强  相似文献

研究了食品中常见的几种金属离子Na^ 、Cu^2 、Al^3 、Fe^3 、Sn^2 和维生素C、苯甲酸等食品添加剂对食用天色色素密蒙黄稳定性的影响,并对色素的耐碱性和耐热性进行了探讨,结果表明其性能稳定。本研究为密蒙黄色素的开发应用提供了依据。  相似文献

研究了微波提取野菊花 (Chrysanthemumindicum)黄色素的新工艺并确定了最佳工艺条件 :原料为 2 0 0 0 0g野菊干花 ,提取剂为无水乙醇 ,提取剂比例为 1 /70 ,微波功率为 80 0W ,提取时间为 45 0s,提取次数为 3次 ,最佳工艺条件下色素的提取率为 91 1 % ,产率为 1 4 6% ,色价E ( 1 % ,3 2 1nm)为 42 2 ,产品pH值为 6 5。与溶剂浸提法相比 ,微波提取野菊花黄色素的每次提取时间由 1 2h减小为 45 0s,提取率从 88 6%增加到 91 1 %。  相似文献

以粗梗稠李为研究对象,采用超声波为辅助手段、水为溶剂提取其色素,通过单因素实验和正交试验探究出色素最佳提取工艺,研究了光照、温度、酸碱度、金属离子、食品添加剂、氧化剂、还原剂对粗梗稠李色素稳定性的影响。通过实验得出,粗梗稠李最佳提取工艺为料液比1∶55、温度60℃、时间为15 min、功率为144 w。在此条件下粗梗稠李色素溶液吸光度为2.120。通过稳定性实验,粗梗稠李色素在4~60℃、避光及弱光、强酸性条件下色素较稳定,在氧化剂(KMnO_4、H_2O_2)、还原剂(亚硫酸钠、抗坏血酸)条件下,色素不稳定。常见金属离子(Fe~(3+)、Ca~(2+)、Fe~(2+)、Cu~(2+)、Mn~(2+)、Al~(3+)、Zn~(2+)、Pb~(2+)、Na~+、Mg~(2+)、K~+)对色素稳定性影响较小。常见食品添加剂对色素稳定性影响较大,硫酸铝钾、醋酸、柠檬酸、山梨酸、苯甲酸对粗梗稠李色素具有增色作用,蔗糖、葡萄糖具有一定的护色作用,碳酸氢钠对粗梗稠李色素具有减色作用。  相似文献

研究了应用微波技术从金银花(Lonicera japonicaThunb)中提取黄色素的新工艺,并确定了最佳工艺条件:提取剂为无水乙醇,原料用量(g)与提取剂用量(mL)比为1∶60,提取时间为50 s,微波功率为560 W,提取次数为3次。最佳工艺条件下的色素提取率为83.40%,产品pH值为6。与溶剂浸提法相比,微波法提取金银花黄色素的每次提取时间由1 h减少到50 s,提取率从52.21%增加到83.40%,效果明显优于常规的溶剂浸提法。  相似文献

以药典规定的水溶性黄色素吸收度为检测指标,考察了红花黄色素的最佳水提提取工艺。研究结果表明,影响红花黄色素提取量的条件由大到小顺序为:提取次数,水用量,提取时间,其中以提取次数影响最大。最佳提取工艺为:采用16倍水,每次提取时间1 h,3次后提取基本完全。  相似文献

用显色反应检测及UV-VIS-分光光度法确定了柿皮黄色素的主要成分为类胡萝卜色素。探讨了从柿子皮中提取天然黄色素的工艺。研究了提取剂的类型、料液比、浸提温度、时间、次数等因素对柿皮黄色素提取的影响。通过正交试验,确定了最佳提取工艺:浸提料液比为1g:20mL,浸提温度60℃,浸提时间2.5h,浸提次数为2次,此条件下提取率为80.74mg/100g。  相似文献

结合内蒙古地区现状,对沙棘的生物学和生态学特性、沙棘属植物化学成分和微量元素及种植沙生植物——沙棘的重要性和必要性进行了详细的研究和探讨。研究表明,沙棘属植物具有极强的生态适应性并富含多种营养成分和生物活性物质,并以耐干旱、耐瘠薄、萌蘖及固氮能力强等特点被称为治理非宜林地水土流失、改善生态环境的先锋树种。种植沙棘是治理内蒙古脆弱生态环境最经济、最有效的措施,是贫瘠的不毛之地发展经济、增加收入的经济树种。另外.种植沙棘的技术简便,容易掌握,投资少,见效快。  相似文献

The present study aims to establish a high-performance thin layer chromatography (HPTLC)-based comparative analysis, directed toward characterization of nucleobases in aqueous and alcoholic extracts of sea buckthorn leaves from three different varieties: Hippophae salicifolia, Hippophae rhamnoides mongolica, and Hippophae rhamnoides turkestanica. The alcoholic and aqueous leaf extracts from these sea buckthorn varieties were prepared using accelerated solvent extraction technique. A novel HPTLC method for separating and identifying six nucleobases, namely, guanosine, guanine, cytosine, adenine, uracil, and thymine were adopted. HPTLC analysis indicated the presence of one or more of these nucleobases in a total of six leaf extracts evaluated, their quantities varying from 0.23 to 7.76?µg nucleobase per mg of extract. Though a typical trend could not be observed in the values obtained, the extracts were found to be considerably rich with respect to nucleobase contents. The results acquired from HPTLC were subsequently validated by hyphenation with mass spectrometry and also by applying chemometric tools in form of heat maps, hierarchical cluster dendrograms, and principal component analysis. The presence of nucleobases in the leaf extracts was confirmed by HPLC as well but HPTLC proved to be a better approach for characterization of nucleobases in plant extracts, than high performance liquid chromatography (HPLC).  相似文献

A commercial model of the analytical high-speed counter-current chromatography instrument was used for separation of flavonoids from a crude ethanol extract of dried fruits of sea buckthorn (Hippophae rhamnoides). Using a two-phase solvent system of chloroform-methanol-water (4:3:2), a five-fold increase in flow-rate of the mobile phase from 60 to 300 ml/h resulted in a rapid separation of five components in less than 15 min without significant loss in peak resolution. Major flavonoid component, isorhamnetin, was identified in its pure state by mass spectrometric analysis.  相似文献

Room-temperature ionic liquids (ILs) are liquids that are constituted entirely of ions and can provide a solvent environment quite unlike any other available at room temperature. They continue to attract considerable interest in the chemistry research community as they are good solvents for a wide range of both inorganic and organic materials. In this study, a CZE method has been established for resolving natural flavonoids, quercetin, kaempferol and isorhamnetin in the Chinese herbal extract from Hippophae rhamnoides and its medicinal preparation (Sindacon Tablet). In this method, 1-alkyl-3-methyl-imidazolium-based ILs are used as the additive, and the effects of the alkyl group, imidazolium counterion (anionic part), along with the concentration of IL are investigated and discussed. Baseline separation, high efficiencies and symmetrical peaks of the three flavonoids were obtained. The separation mechanism seems to be the hydrogen-bonding interaction between the imidazolium cations of IL and the flavonoids.  相似文献

1,3-Dicapryloyl-2-linoleoylglycerol (1), a novel triglyceride, was isolated from berries of Hippophae rhamnoides. The structure was elucidated on the basis of MS, 1D and 2D NMR experiments including HMQC and HMBC. The metal chelating, free radical scavenging, and lipid peroxidation inhibiting properties of the compound were also estimated with particular reference to radiation protection. In case of metal chelation and superoxide ion scavenging, 1 showed maximum inhibition at 50 microg/ml (11%) and 100 microg/ml (55%), respectively, whereas in lipid peroxidation, 1 showed maximum inhibition (57%) at 2 mg/ml as compared to quercetin as a control.  相似文献

In our investigation on the chemical constituents of Hippophae rhamnoides L., the chloroform-soluble fraction of the 80% acetone extract of branch bark was observed to inhibit nitric oxide (NO) production in a lipopolysaccharide and recombinant mouse interferon-gamma-activated murine macrophage-like cell line, RAW 264.7 cells. Two new triterpenoids, 2-O-trans-p-coumaroyl maslinic acid (1) and 2-O-caffeoyl maslinic acid (2), and three known triterpenoids, oleanolic acid (3), 3-O-trans-p-coumaroyl oleanolic acid (4), and 3-O-caffeoyl oleanolic acid (5), and 6-methoxy-2H-1-benzopyran (6) and beta-sitosterol (7) were isolated from the branch bark extract. Their inhibitory activities on the production of NO in RAW 264.7 cells and radical-scavenging activities were examined.  相似文献

建立了低速逆流色谱技术(SRCCC)从栀子黄色素中快速分离制备藏花素的方法。两相溶剂系统由叔丁基甲基醚-正丁醇-乙腈-水(2:2.5:1:5, v/v/v/v)组成,以上相为固定相,下相为流动相。在转速为50 r/min和流速为5 mL/min的条件下,从5 g栀子黄色素粗样中分离得到2.47 g藏花素,纯度为96.8%。该方法制备量大、安全、高效,有可能成为工业级分离制备藏花素的一种有效手段。  相似文献

Using high-performance liquid chromatography (HPLC), a chemical fingerprint method was developed for investigating and demonstrating the variance of flavonoids among different origins of sea buckthorn berries. Thirty-four samples were analyzed including 15 RS (Hippophae rhamnoides ssp. sinensis) samples, 7 RY (H. rhamnoindes ssp. yunnanensis) samples, 5 RW (H. rhamnoides ssp. wolongensis) samples, 4 NS (H. neurocarpa ssp. stellatopilosa) samples and 3 TI (H. tibetana) samples. In the HPLC chromatograms, 12 compounds were identified as flavonoids, including quercetin 3-O-sophoroside-7-rhamnoside, kaempferol 3-O-sophoroside-7-O-rhamnoside, isorhamnetin 3-O-sophoroside-7-O-rhamnoside, isorhamnetin 3-O-glucoside-7-O-rhamnoside, quercetin 3-O-rutinoside, quercetin 3-O-glucoside, isorhamnetin 3-O-rutinoside, isorhamnetin 3-O-glucoside, quercetin, kaempferol 7-O-rhamnoside, kaempferol and isorhamnetin. Both correlation coefficient of similarity in chromatograms and relative peak areas of characteristic compounds were calculated for quantitative expression of the HPLC fingerprints. Our results revealed that the chromatographic fingerprint combining similarity evaluation could efficiently identify and distinguish sea buckthorn berries from different species. However, no obvious difference between RS and RY suggested that the two subspecies might have very close relationship in terms of chemotaxonomy. The established method was considered to be suitable for fingerprint analysis to check the genuine origin and control the quality of sea buckthorn berries and extracts.  相似文献

In this study, the combined effect of pH, temperature and agitation speed on yellow pigment production and mycelial growth of Penicillium aculeatum ATCC 10409 was investigated in whey media. Different pH levels (5, 6.5 and 8), temperatures (25, 30 and 35°C) and agitation speed levels (100 and 150 rpm) were tested to determine the best conditions to produce a fungal yellow pigment under submerged fermentation. The best production of yellow pigment (1.38 g/L) was obtained with a pH value of 6.5, a temperature of 30°C and an agitation speed of 150 rpm. In contrast, the maximal biomass concentration (11.12 g/L) was obtained at pH value of 8, a temperature of 30°C and an agitation speed of 100 rpm. These results demonstrated that biomass and yellow pigment production were not directly associated. The identification of the structure of unknown P. aculeatum yellow pigment was detected using UV absorption spectrum and FT-IR spectroscopy.  相似文献

The marine environment is a major source for many novel natural compounds. A new yellow pigment has been isolated from the marine bacterium P. tunicata and identified as a new member of the tambjamine class of compounds. The structural identification was achieved by a combination of 1D and 2D-NMR spectroscopy and high resolution mass spectrometry data.  相似文献
