探讨了在微波加热条件下,芳香醛、取代苯乙酮和尿素的三组分反应在N,N-二甲基甲酰胺(DMF)中制得4,6-二芳基-3,4-二氢嘧啶-2(1H)-酮类化合物,收率为68%~84%.若在反应体系中加入三甲基氯硅烷,该三组分反应则高产率(66%~87%)地生成相应的脱氢产物4,6-二芳基嘧啶-2(1H)-酮类化合物.该反应具有反应条件温和、产物收率高、操作方便等优点,为4,6-二芳基-嘧啶-2(1H)-酮类药物中间体的合成提供了一条全新的路线.  相似文献

With visible light irradiation (λ > 400 nm) of rhenium(I) complexes (P1-P4), a photochemical conversion from 3,4-dihydropyrimidin-2(1H)-ones to pyrimidin-2(1H)-ones at room temperature has been achieved with good to excellent yields in CH(3)CN-H(2)O solution containing CCl(4) and K(2)CO(3). Luminescence quenching study and product analysis reveal that photoinduced electron transfer between rhenium(I) complex P and 3,4-dihydropyrimidin-2(1H)-ones plays an important role in the initial event.  相似文献

喹喔啉酮作为重要的含氮杂环骨架,广泛应用于抗菌化合物、抗肿瘤药物、半导体材料中.因此,开发新方法合成不同官能团化喹喔啉酮以丰富其结构多样性在近年来广受关注.在众多合成方法中,喹喔啉酮的直接C–H官能团化是一类步骤经济性较高、简便高效地构建喹喔啉酮衍生物的重要方法.近年来,光催化、电催化和光电催化技术已发展成为现代有机合成中强有力的绿色合成工具,可以有效利用光能或者电能促进有机化学转化,减少传统有机反应中的高能耗.过去几年,化学工作者在光/电催化喹喔啉酮的直接C–H官能团化反应方面进行了大量研究,开发了系列不同官能团化喹喔啉酮的绿色合成方法.本文系统总结了喹喔啉酮的光/电催化官能团化研究最新进展,重点讨论了光/电催化下自由基历程的喹喔啉酮C–H键直接官能团化反应,反应类型主要包括喹喔啉酮的3-芳基化、3-烷基化、3-氟烷基化、3-烷氧基化、3-酰化、3-氨基化、3-膦酰化、3-巯基化、3-硅烷基化和环化反应等方面,这将有助于理解光/电催化喹喔啉酮官能团化反应.反应在温和条件下可以成功产生各种碳中心或杂原子为中心的自由基,这些自由基进一步与喹喔啉酮的3-位进行加成反应,进而开发出众多有效的方法来合成不同官能团化的喹喔啉酮.这些条件温和的反应方法已经有部分成功应用于合成具有重要生物活性的喹喔啉酮化合物.此外,药物中常见的一些重要官能团,例如三氟甲基和二氟甲基等可以顺利地引入到喹喔啉酮骨架结构,这些研究为喹喔啉酮衍生物的生物活性相关研究奠定了基础.本文还对当前光/电催化喹喔啉酮官能团化反应所面临的挑战和未来发展作了展望和总结,期待在不久的将来,有机合成工作者能基于喹喔啉酮的C–H键直接官能团化开发更多不同类型的新反应.另一方面,需要进一步开发新型的非均相可见光催化剂,实现光催化剂的回收与再利用,进一步降低反应成本,促进其在药物开发、功能材料合成等应用方面的发展.  相似文献

Biologically active 3-amino-4-arylquinolin-2(1H)-ones and 3-alkenyl-4-arylquinolin-2(1H)-ones were synthesized in an efficient and concise manner, utilizing readily available 4-hydroxyquinolin-2(1H)-one as starting material. Thermal-induced dimerization cyclization of ethyl N-(styrylcarbamoyl)acetate derivatives has been investigated, leading to 4-hydroxy-2(1H)-pyridone-3-carboxamide derivatives with good yields in diphenyl ether on 200–210 °C. Ethyl N-(styrylcarbamoyl)acetate derivatives readily provided the intermolecular cyclization products 4-hydroxy-2(1H)-pyridone-3-carboxylates on reflux in xylene. In addition, several related 3-acetyl-4-hydroxy-5-phenylpyridin-2(1H)-ones have been prepared. It provided an efficient preparation of 4-hydroxy-2(1H)-pyridone-3-carboxamide derivatives.  相似文献

Key steps in the synthesis include the derivatization of the quinolin-2(1H)-one cores using palladium-catalyzed Suzuki and Heck reactions, installing the 4-aryl and 3-alkenyl substituents. All synthetic transformations (six steps) required for the synthesis of the desired target quinolin-2(1H)-one were carried out using controlled microwave-assisted organic synthesis.  相似文献

The reactions of (2S)-2-amino-2-substituted-N-(4-nitrophenyl)acetamides 16a-c, succindialdehyde (13), and benzotriazole afforded enantiopure (3S,5R,7aR)-5-(1H-1,2,3-benzotriazol-1-yl)-3-substituted-1-(4-nitrophenyl)tetrahydro-1H-pyrrolo[1,2-a]imidazol-2-ones 17a-c, which were converted by sodium borohydride into (3S,7aR)-3-substituted-1-(4-nitrophenyl)tetrahydro-1H-pyrrolo[1,2-a]imidazol-2-ones 18a-c. Chiral (2S)-2-amino-2-substituted-N-(4-methylphenyl)acetamides 12a-d, easily prepared in two steps from N-Boc-alpha-amino acids 10a-d, similarly reacted with glutaraldehyde (20) and benzotriazole to generate 5-benzotriazolyl-3-substituted-hexahydroimidazo[1,2-a]pyridin-2(3H)-ones 21a-d, which were converted by sodium borohydride directly into optically active 3-substituted-hexahydroimidazo[1,2-a]pyridin-2(3H)-ones 22a-d.  相似文献

ZrOCl2·8H2O catalyzed environmentally benign synthesis of isobenzofuran-1(3H)-ones are described.ZrOCl2·8H2O appeared to be an excellent catalyst for the condensation and reactions.Reaction of phthalaldehydic acid(2-carboxybenzaldehyde) with methylaryl and cyclic ketones was initiated by condensation and occurred in one step providing excellent yields(90-98%).  相似文献

<正>A simple and efficient procedure for the synthesis of 2-arylquinazolin-4(3H)-ones has been developed through cyclocondensation of 2-aminobenzamide with aromatic aldehydes using tetrabutylammonium bromide(TBAB) as novel neutral ionic liquid catalyst in the presence of copper(Ⅱ) chloride(CuCl_2) as oxidizing agent under solvent-free conditions at 100℃.In the absence of CuCl_2 and under a nitrogen atmosphere,the unoxidized intermediates,2-aryl-2,3-dihydroquinazolin-4(1H)-ones,were isolated. Treatment of these intermediates with CuCl_2 in TBAB media gave the oxidized products 2-arylquinazolin-4(3H)-ones.On the other hand,cyclocondensation of 2-aminobenzamide with aromatic aldehydes in TBAB under microwave irradiation directly gave 2- arylquinazolin-4(3H)-ones.  相似文献

A series of novel regioisomeric tetrahydrophthalimide-substituted indolin-2-ones has been prepared via the Sommelet-Hauser type cyclization of appropriately substituted anilines as potential herbicides. The resultant indolin-2-ones were then regioselectively alkylated at N-1 and C-3 to give 1,3,3-trisubstituted indolin-2-ones. The most active series was also prepared by the bis-nitration of m-fluorophenylacetic acid followed by reduction and cyclization to give 6-amino-5-fluoroindolin-2-one. Elaboration to the tetrahydrophthalimide-substituted indolin-2-one was followed by C- and N-alkylation to give the desired compounds.  相似文献
