本文介绍通过相同组分、不同配比冲洗剂等度和梯度冲洗样品的实验,首次发现在液相色谱中,溶质的色谱峰半宽度与保留值间存在同一线性关系;在梯度冲洗时流出组分的色谱峰半宽度与虚拟保留值间同样存在线性关系。该结果证明,我们提出的组分在柱末端有相近的以长度为量纲的谱带半宽度的正确性,同时该结果对梯度最佳分离条件的预测亦很有益。  相似文献

〕提出了采用线性梯度洗脱求取未知样品中溶质保留值方程的原理及其计算方法,用光二极管阵列扫描检测器解决了梯度洗脱条件下色谱峰的归宿。实验证明:用所得的保留值方程预测等度洗脱下容量因子k'的相对误差不大于3%,而用梯度洗脱下的半峰宽规律可以估算未知样品容量因子k的分布范围,以解决对未知样品是采用等度或梯度洗脱方式。最后,借助计算机仿真方法用于样品的优化,获得了满意的结果。  相似文献

提出了测量气相色谱中吸附等温线的新方法,用不同进样量下测定的容量因子计算平衡时气-固两相组分浓度。利用建立的方法分别测量了正-己烷、正-壬烷和正-十一烷在邻苯二甲酸二辛酯(DOP)、阿皮松-L(Apiezon)和β,β′-氧二丙腈(ODPN)柱上的吸附等温线,讨论了色谱峰形状与吸附等温线的对应关系。研究了气相色谱中进样量对保留值的影响,利用佛伦德利希(Freundlich)吸附等温式导出了描述保留值与进样量关系的数学表达式。理论和实验证明,保留值与进样量具有良好的对数线性关系。  相似文献

建立了使用氢氧化钠淋洗液时用计算机模拟离子色谱梯度淋洗分离阴离子的新方法,研究了在线性梯度淋洗过程中不同时间阴离子在色谱柱中的位置及所在位置的淋洗液浓度,得出阴离子在不同时间的容量因子、迁移速度,通过积分得到保留时间。再根据离子色谱峰峰形变化的规律,得到色谱峰峰形的参数。模拟色谱图保留值与实验值的相对误差小于5%,模拟色谱图的色谱峰峰形与实验所得到的色谱图的色谱峰峰形也非常接近。  相似文献

根据毛细管电色谱中溶质保留值与有机改性剂种类和浓度之间的相互关系,首次实现了毛细管电色谱有机流动相双二元双台阶分离。利用此方法分离含12种芳香族化合物的混合样品,各组分在六次连续进样中保留时间的RSD(%)值均在1.65%以内,并将样品的分离时间缩短到等度洗脱方式下的三分之一。这说明该方法分离复杂样品快速、可靠。此外还推导了双二元双台阶分离方式下溶质保留值的预测公式,并对该公式的准确性进行了检验,预测值与实测值间的相对误差在5.73%以内。  相似文献

发展了一种大体积循环进样方法,用于富集低丰度蛋白质。在优化的色谱分离条件下,通过增加蛋白质样品的上样体积提高低丰度蛋白质的绝对含量;进一步采用增加样品进样循环次数的方法提高蛋白质的富集效率。以猪肝提取蛋白质为样品,每次上样量500 μL的大体积11次循环进样。根据色谱峰的信号强弱,选择了在原始谱图中看不到色谱峰、有较少小峰和有较多小峰出现的时间段等有代表性的馏分进行研究。在中等极性的组分中,保留时间为11.38 min和12.58 min组分的富集效率分别提高了52倍和61倍,实验结果与理论富集效率相近。所发展的方法为生物蛋白质样品研究提供了一种新的富集制备及检测方法。  相似文献

 根据溶质在柱内的迁移规律 ,建立了一种利用线性梯度实验快速获得溶质保留值方程系数 ,然后以串行响应函数为优化指标进行多台阶梯度分离条件优化的方法。与利用等度实验获得保留值方程的方法相比 ,该法可以大大缩短优化时间。通过该方法对芳香胺和衍生化氨基酸样品进行了分离 ,获得了满意的分离度 ,表明该方法的预测精度很好。  相似文献

基于溶质在径向色谱柱内输运的质量平衡方程, 在线性分配条件下, 得到了描述分离柱效和流出曲线形状各参数的理论表达式, 也对柱效和流出曲线对称性的变化趋势加以系统讨论. 结果表明: 径向色谱中, 柱效与体积流速之间的关系与轴向色谱中柱效与流动相线速度的关系在趋势上相同; 在较高流速下运行时, 径向色谱仍可以得到高柱效. 随着溶质容量因子、进样时间的增加, 柱效单调降低. 柱直径和柱长对柱效的影响存在交叉, 设计半径较大而长度较短的色谱柱将更有利于提高分离柱效. 径向色谱适宜于大分子样品的稳定分离方法建立, 也预示其对于蛋白、DNA等样品的制备分离具有明显优势.  相似文献

毛细管电动力学色谱(MEKC)中通过“扫”的技术可以使样品组分在线富集。本研究基于柱分离过程弛豫理论的基本方法,对溶质在MEKC中柱内输运过程加以研究,得到了描述进样长度及样品区带和运行区带性质差别对富集效果影响的理论表达式。通过对进样长度及溶质在两区带中的容量因子与富集效果之间关系的进一步探讨,证实溶质在胶束中的溶解度是影响富集的最重要因素;在基本不影响分离效果的情况下,适当加大进样长度一般对富集有利。  相似文献

Abstract

The injected volume of a sample dissolved in the mobile phase of an HPLC system must be maintained as small as possible so as to minimize the loss in efficiency. Generally this requirement limits the sensitivity of HPLC methods devoted to trace quantity determinations of drugs in biological fluids. In order to avoid this limitation and to increase the effective sensitivity of HPLC methods for determination of drugs such as antrafenine, nifuroxazide and cipropride, the samples were dissolved in a non-eluting solvent and a large volume (> 100 μl) was injected on to the chromatographic column.

The above-mentioned compounds and their internal standards were dissolved in a series of eluting and non-eluting solvents and increasing volumes (5 to 1000 μl) were injected. Peaks corresponding to injections made in an eluting solvent showed retention times independent of the injection volume but their variances increased with the volume injected. In contrast, peaks corresponding to injections made in a non-eluting solvent, similar to the mobile phase, had a variance independent of the injection volume but their retention times increased linearly with the injection volume. The repeated injection of such non-eluting solvents had no influence on chromatographic behaviour. Peaks corresponding to compounds injected in a non-eluting solvent made with components different from those of the mobile phase had a variance independent of the injection volume but their retention times varied both with the injection volume and with the interval between injection.

The application of non-eluting solvents has been defined theoretically and it has been demonstrated that solutions composed of 25% of the mobile phase diluted with the least eluting of its components act as non-eluting solvents and can be injected in large volume without loss in efficiency. This feature could be used to inject all the samples volume or only part of it, manually or automatically, since any automatic injector can be used with large volumes.

Thus, using the relatively simple procedure of making injections with a non-eluting solvent it is possible to increase both sensitivity and the rate of sample analysis.  相似文献

To circumvent the detrimental effects of large‐volume injection with fixed‐loop injector in modern supercritical fluid chromatography, the feasibility of performing multiple injection was investigated. By accumulating analytes from a certain number of continual small‐volume injections, compounds can be concentrated on the column head, and this leads to signal enhancement compared with a single injection. The signal to noise enhancement of different compounds appeared to be associated with their retention on different stationary phases and with type of sample diluent. The diethylamine column gave the best signal to noise enhancement when acetonitrile was used as sample diluent and the 2‐picolylamine column showed the best overall performance with water as the sample diluent. The advantage of multiple injection over one‐time large‐volume injection was proven with sulfanilamide, with both acetonitrile and water as sample diluents. The multiple injection approach exhibited comparable within‐ and between‐day precision of retention time and peak area with those of single injections. The potential of the multiple injection approach was demonstrated in the analysis of sulfanilamide‐spiked honey extract and diclofenac‐spiked ground water sample. A low-volume automated injection system for the analysis of chemically complex, amino acid samples is presented. This system utilizes submicroliter sample volumes stored on a 75-μm inner diameter capillary. A pulse of positive pressure (82 kPa) is used to load nanoliter sample volumes into an in-house fabricated interface and onto a separation capillary. Residual sample solution in the interface is immediately washed away by a continuous transverse flow through the injection interface, yielding a sharp and reproducible sample plug. By performing multiple injections of a static sample, one may average the signals to yield a signal-to-noise ratio improvement of up to 4.07-fold for 20 injections compared with a theoretical maximum of a 4.47-fold improvement. Without interruption of the applied voltage, injections performed every 150 s were used to monitor the progress of the reaction of multiple amino acids with the fluorogenic dye 3-(4-carboxybenzoyl)quinoline-2-carboxaldehyde. Analysis of dialyzed clinical vitreous samples demonstrates the resolution and quantitation of arginine, lysine, leucine, glutamine, and glutamate. Observed levels are comparable with those of nonautomated injection methods and reports by others.  考察了填充柱超临界流体色谱法 (SFC)中的样品溶剂及连续进样等因素对化合物保留行为变化的影响规律。以超临界 CO2 或含低体积分数甲醇的 CO2 为流动相时 ,氨基柱上组分的保留时间随着样品溶剂的极性增大而增大 ,而溶剂对 C1 8柱上组分的保留时间影响不大 ;在 C1 8柱上 ,溶剂对连续进样的后续效应不强 ;而在氨基柱上 ,甲醇溶液的后续效应比丙酮、氯仿溶液的后续效应强。当甲醇的体积分数大于 1 .0 %时 ,溶剂的效应明显减弱。这种变化规律对填充柱 SFC的合理进样并获得重现性良好的色谱数据具有实际意义。  相似文献

The enantiomeric resolution of N-t-butyloxycarbonyl (N-t-Boc) amino acids D/L isomers by reversed-phase HPLC was investigated using cyclodextrins (CD's) as chiral selectors for the mobile phase. The use of a low pH (pH<4) for the mobile phase enabled the enantioseparation of N-t-Boc amino acids. The opposite elution order of D/L isomers was observed when hydroxypropyl-derivatized beta-CD was used instead of native beta-CD. A computer simulation of the enantioseparation showed that the ratio of the retention factors of the chiral selector and the sample determined the elution order and the resolution. When the retention factor of the chiral selector is smaller than that of the sample, an isomer having larger complex formation constant eluted faster. However, when the chiral selector had a larger retention factor than the sample, an opposite elution order of the isomers was obtained. The large difference in the retention factors between the chiral selector and the sample led to good enantiomeric separation.  相似文献

The experimental technique of mass spectrometric tracer pulse chromatography was used to study the effect of the sorption of eluent components by a C18-bonded silica RPLC packing on the retention of a series of test analytes during isocratic and gradient elution experiments. The analytes of interest were a substituted phenol, a substituted nitroaniline, an anti-malaria drug, tetrahydrofuran, and methanol. The eluent used was a mixture of acetonitrile and water. The solutes and isotopically labeled eluent components were injected at fixed time intervals during each gradient run. The mass specific detector allowed the assignment of individual analyte peaks even when there was overlap in the chromatograms from successive injections. Thus, the retention time of each analyte could be determined as a function of gradient slope and initial eluent composition at the time of each injection. Experimental gradient retention time data were then compared with the calculated results from two theoretical models. The first model assumed the velocity of the mobile phase and eluent were equal. The second and most realistic model assumed the velocity of the eluent was less than the velocity of the mobile phase due to the uptake of eluent by the stationary phase. Gradient retention times predicted by the two models were reasonably accurate with the sorption model giving slightly more accurate values. Inverse calculations, i.e., calculation of isocratic retention factors from gradient elution data were also carried out with very similar results. That is, the model allowing for the uptake of eluent was slightly more accurate than the model assuming no eluent-stationary phase interaction.  相似文献

计算机辅助高效液相色谱(HPLC)分离条件优化可以低成本、快速地得到优化的分离条,因而已较为广泛地用于复杂样品的分离分析。基于移动重叠分离图方法,又发展了一种新型的多台阶梯度分离条件的优化方法可调移动重叠分离图法。该方法通过预测不同流动相条件下各组分的保留时间、峰宽和分离度,绘制出对于样品中各组分的重叠分离区域图。在对当前台阶流动相组成进行优化的同时,考虑其对后面一到两个台阶上流出组分保留的影响,实时地重新绘制对于后面台阶上流出组分的重叠分离区域图。通过观察当前台阶流动相条件对当前台阶和后面台阶上流出组分分离的影响,综合考虑样品中所有组分的分离情况,找到更接近全局最优的分离条件。通过扫描的方法对优化得到的分离条件进行微调,能够进一步提高分离效果。采用文献数据对可调移动重叠分离图法的应用加以说明,在二元流动相体系下,证明了该方法在HPLC方法建立方面的优越性。  相似文献

用脲衍生物型手性固定相直接拆分了药物对映体噻利洛尔。优化的正相色谱流动相的组成为正己烷/1,2-二氯乙烷/乙醇(77∶21∶2,V/V/V)。实验表明,流动相中乙醇含量的改变对分离度产生了较大的影响;不同极性调节剂的使用表现出了不同的拆分效果。最后讨论了异构体的出峰次序,认为异构体与固定相的手性中心之一的氢键作用力是造成异构体分离的主要因素。  相似文献

The first six linear and cyclic oligomers of polyamide-6 can be quantitatively determined in the polymer using HPLC with the sandwich injection method and an aqueous acetonitrile gradient. In this final part of the triptych concerning the determination of the oligomers in polyamide-6, the irregular elution behavior of the cyclic monomer compared to the cyclic oligomers was investigated. We also optimized the separation of the involved polyamide oligomers, with respect to gradient steepness, stationary phase, column temperature and mobile phase pH. The irregular elution behavior of the cyclic monomer could be caused by its relatively large exposed/accessible hydrophobic surface, which permits relatively easy penetration into the hydrophobic stationary phase giving extra retention. The dipole moment of the different oligomers was used as a measure for this exposed/accessible hydrophobic area to correlate the retention factors using quantitative structure-retention relationships. We also studied the retention behavior of the polyamide, which is injected each run directly onto the column and modifies the stationary phase. Using a 250-microl post gradient injection zone of formic acid on a 250x3 mm Zorbax SB-C18 column, the polyamide could be effectively removed from the stationary phase after each separation. The linear solvent strength (LSS) model was used to optimize the separation of the first six linear and cyclic oligomers. As the LSS model assumes a linear correlation between the modifier concentration and the logarithm of the retention factor and the cyclic monomer and dimer show extreme curvation of this relation in the eluting region, we investigated different models to predict gradient elution from isocratic data. A direct translation of the isocratic data to gradient retention times did not yield adequate retention times using the LSS model. It was found that the LSS model worked acceptably if gradient retention times were used as input data. Even for fast non-linearly eluting components, an average error of 0.4 resolution units of 4sigma was obtained. Using the LSS model in combination with different column temperatures and mobile phase pH values, a separation of the first six linear and cyclic oligomers was accomplished.  相似文献
