This paper describes the development of organic fluorescence nanoparticles. The nanoparticles have a narrow, tunable, symmetric emission spectrum and a broad, continuous excitation spectrum. The nanoparticles have high room-temperature fluorescence quantum yields and long fluorescence lifetime. They are also photochemically stable and water-soluble. They were used as fluorescence biosensor in the determination of proteins, which was proved to be a simple, rapid and specific method. In comparison with single organic fluorephores, these nanoparticles are brighter, more stable against photobleaching, and do not suffer from blinking. Under optimal conditions, the linear ranges of the calibration curves were 0.1-4.5 microg ml(-1) for human serum albumin (HSA), 0.2-3.5 microg ml(-1) for bovine serum albumin (BSA) and 0.04-0.8 microg ml(-1) for gamma globulin (gamma-IgG), respectively. The detection limits were 0.062 microg ml(-1) for HSA, 0.036 microg ml(-1) for BSA and 0.022 microg ml(-1) for gamma-IgG, respectively. However, when the content of HSA is lower than 0.8 microg ml(-1), HSA makes little contribution to the fluorescence quenching. So, the method was applied to direct selective quantification of gamma-IgG in human blood serum without separation of HSA. The results were in good agreement with these reported by the hospital, indicating that the method presented here is not only sensitive, selective and simple, but also reliable and suitable for practical applications.  相似文献

本文采用层层自组装法合成了具有良好水溶性的功能化磁性荧光Fe3O4/Py(芘)/PAM(聚丙烯酰胺)纳米粒子.利用其磁性,能够对该粒子进行简单有效的分离纯化,并可以对待测粒子进行富集以提高其检测灵敏度.利用Cr(VI)对该复合粒子水溶液的荧光猝灭,建立了测定Cr(VI)的荧光分析法,讨论了反应机理.在最佳实验条件下,该方法的线性区间为0.1~14.0μgmL-1,检测限为0.02μgmL-1.常见的共存离子不干扰测定,该方法可用于环境废水中Cr(VI)的测定.  相似文献

The polystyrene-acrylic acid (PS-AA) nanoparticles have been prepared by ultrasonic polymerization, characterized by FT-IR and TEM. It is the first report on the determination of proteins with macromolecules nanoparticles of PS-AA by resonance light-scattering (RLS). At pH 6.9, the RLS of macromolecules nanoparticles of PS-AA can be enhanced by proteins. Based on this, a novel quantitative assay of proteins at the nanogram levels has been proposed. At pH 6.9, the RLS signals of PS-AA were greatly enhanced by proteins in the region of 250-700 nm characterized by the peak at 342 nm. Under optimal conditions, the linear ranges of the calibration curves were 0.02-11.0 microgml-1, 0.04-10.0 microgml-1 and 0.03-10.0 microgml-1 for gamma-globulin (gamma-IgG), bovine serum albumin (BSA) and human serum albumin (HSA), respectively. The detection limits were 16.0 ngml-1, 19.0 ngml-1, and 15.0 ngml-1 for gamma-IgG, BSA and HSA, respectively. The method has been applied to the analysis of total proteins in human serum samples collected from the hospital and the results were in good agreement with those reported by the hospital, which indicates that the method presented here is not only sensitive, simple, but also reliable and suitable for practical application.  相似文献

The manganese-tetrasulfonatophthalocyanine (MnTSPc) catalyzed luminol-hydrogen peroxide chemiluminescence (CL) systems can be quenched in the presence of proteins. A highly sensitive CL quenching method has been developed for the determination of proteins. Under optimum conditions, the linear ranges of the calibration curves were 0.1-20 microg/mL for human serum albumin (HSA), 0.2-20 microg/mL for human gamma-IgG, and 0.5-50 microg/mL for the bovine serum albumin (BSA) with the corresponding detection limits were 1.9 ng/mL, 2.7 ng/mL, and 3.4 ng/mL. The method has been applied to the analysis of total proteins in human serum samples and the results were in good agreement with clinical data provided.  相似文献

A novel composite nanoparticle has been prepared by an in situ polymerization method and applied as a protein fluorescence probe. The nano-CdS has been prepared, then the polymerization of acrylic acid (AA) was carried out by initiator potassium persulfate (KPS) under ultrasonic irradiation. The surface of the composite nanoparticles was covered with abundant carboxylic groups (--COOH). The nanoparticles are water-soluble, stable, and biocompatible. The synchronous fluorescence intensity of the composite nanoparticles is significantly increased in the presence of trace protein at pH 6.90. Based on this, a new synchronous fluorescence scan (SFS) analysis was developed for the determination of proteins including BSA, HSA, and human gamma-IgG. When Delta lambda = 280 nm, maximum synchronous fluorescence is produced at 290 nm. Under the optimum conditions, the response is linearly proportional to the concentration of proteins. The linear range is 0.1-10 microg ml(-1) for HSA, 0.09-8.0 microg ml(-1) for BSA, and 0.08-15 microg ml(-1) for human gamma-IgG, respectively. The method has been applied to the determination of the total protein in human serum samples collected from the hospital and the results are satisfactory.  相似文献

A novel luminescent and magnetic Fe(3)O(4)/pyrene/polyacrylamide (Fe(3)O(4)/Py/PAM) nanocomposite has been prepared under ultrasonic radiation. This magnetic nanocomposite combined with pyrene would lead to a special functional magnetic luminescent composite that enjoys both the advantages of magnetic nanoparticles of Fe(3)O(4) and fluorescence nanoparticles of pyrene. Taking advantage of the magnetic property of Fe(3)O(4) nanocomposites, we can separate Fe(3)O(4)/Py/PAM nanocomposites from solution easily just by using a permanent magnet. Based on the fluorescence quenching of Fe(3)O(4)/Py/PAM nanocomposites by Cr(VI), a method for the selective determination of Cr(VI), without separation of Cr(III) in water, was developed. Under optimal experimental conditions, a limit of detection of 0.01 microg mL(-1) was achieved. The calibration curve was linear over the concentration range of 0.1-14.0 microg mL(-1) with a correlation coefficient of 0.9975. The proposed method has been applied to the selective quantification of Cr(VI) in synthetic samples and wastewater samples with the satisfactory results.  相似文献

The binding equilibrium between phosphotungstic acid (H7[P(W2O7)6] · XH2O;PTA) and human serum albumin (HSA) or bovine serum albumin (BSA) has been studied by UV-Vis, fluorescence spectroscopies and equilibrium dialysis. It has been observed that UV absorption enhanced and the fluorescence quenched as the PTA binding to HSA or BSA at physiological pH 7.43(?.02). The Scatchard analysis indicated that there exists a strong binding site of PTA in both HSA and BSA, and the successive stability constants of these two systems are obtained by nonlinear least-squares methods fitting Bjerrum formula.  相似文献

The liner range is 0.1-15 microgmL(-1) for HSA, 0.2-20 microgmL(-1) for BSA and 0.1-50.0 microgmL(-1) for human gamma-IgG, respectively. The method has been applied to the determination of the total protein in human serum samples collected from the hospital and the results are in good agreement with those reported by the hospital. Three bis-tetradentate acyclic amine ligands differing only in the arm length of the pyridine pendant arms attached to the 4,6-positions of the pyrimidine ring, namely, 4,6-bis[N,N-bis(2'-pyridylethyl)aminomethyl]-2-phenylpyrimidine (L(Et)), 4,6-bis[N,N-bis(2'-pyridylmethyl)aminomethyl]-2-phenylpyrimidine (L(Me)), and 4,6-[(2'-pyridylmethyl)-2'-pyridylethyl)aminomethyl]-2-phenylpyrimidine (L(Mix)) have been used to synthesize nine air-sensitive diiron(II) complexes: [Fe(II)(2)L(Et)(NCS)(4)]·MeOH·(3)/(4)H(2)O (1·MeOH·(3)/(4)H(2)O), [Fe(II)(2)L(Et)(NCSe)(4)]·H(2)O (2·H(2)O), [Fe(II)(2)L(Et)(NCBH(3))(4)]·(5)/(2)H(2)O (3·(5)/(2)H(2)O), [Fe(II)(2)L(Me)(NCS)(4)]·(1)/(2)H(2)O (4·(1)/(2)H(2)O), [Fe(II)(2)L(Me)(NCSe)(4)] (5), [Fe(II)(2)L(Me)(NCBH(3))(4)]·(3)/(2)H(2)O (6·(3)/(2)H(2)O), [Fe(II)(2)L(Mix)(NCS)(4)]·(1)/(2)H(2)O (7·(1)/(2)H(2)O), [Fe(II)(2)L(Mix)(NCSe)(4)]·(3)/(2)H(2)O (8·(3)/(2)H(2)O), and [Fe(II)(2)L(Mix)(NCBH(3))(4)]·(3)/(2)H(2)O (9·(3)/(2)H(2)O). Complexes 3·(5)/(2)H(2)O, 4·(1)/(2)H(2)O, 5, 6·(3)/(2)H(2)O, and 8·(3)/(2)H(2)O were structurally characterized by X-ray crystallography, revealing, in all cases, both of the iron(II) centers in an octahedral environment with two NCE (E = S, Se, or BH(3)) anions in a cis-position relative to one another. Variable temperature magnetic susceptibility measurements showed that all nine diiron(II) complexes are stabilized in the [HS-HS] state from 300 K to 4 K, and exhibit weak antiferromagnetic coupling. M?ssbauer spectroscopy confirmed the spin and oxidation states of eight of the nine complexes (the synthesis of air-sensitive complex 3 was not readily reproduced).  相似文献

A novel quantitative method for the determination of proteins in aqueous solutions has been based on the quenching of the resonance scattering light of colloidal silver chloride in the presence of proteins. The detection limits for eight kinds of proteins (BSA, HSA, egg albumin, human gamma-IgG,alpha-chymotrypsin, E. Coli. alpsase, myoglobin, alpha-casein) were at about 8 ng/mL; the linear ranges of the calibration curves were 10-400 ng/mL under optimal conditions,except for human gamma-IgG (20-400 ng/mL), myoglobin (10-300 ng/mL), and alpha-casein (10-300 ng/mL). Three wavelengths (398 nm, 475 nm, 499 nm) were all suitable for the determination and any acidity from pH 3.0 to pH 9.0 could be chosen. A few non-protein substances at high concentration levels interfered with this method, but this problem could simply be overcome by diluting the samples before the assay. Mechanism studies showed that the quenching effect of proteins on the scattering light of colloidal silver chloride was mainly due to the coagulation of AgCl particles retarded by protein. The method was employed for the determination of total protein in human serum with satisfactory results.  相似文献

Two chiral supramolecules with enantiomeric three-dimensional porous host frameworks, (Delta){[Fe(II)(phen)(3)][Fe(III)Na(C(2)O(4))(3)]}(n) (1) and (Lambda){[Fe(II)(phen)(3)][Fe(III)Na(C(2)O(4))(3)]}(n) (2) (phen = 1,10-phenanthroline), have been synthesized, and their crystal structures have been determined. The structural analysis shows that compounds 1 and 2 are a pair of enantiomers, both consisting of a three-dimensional porous skeleton formed by (Delta)/(Lambda){[Fe(III)Na(C(2)O(4))(3)](2-)}(n) and guest (Delta)/(Lambda)[Fe(phen)(3)](2+) units. The circular dichroism spectrum measurements confirmed the optical activity and the enantiomeric nature of complexes 1 and 2.  相似文献

A new determination method of proteins with the limit of determination at nanogram levels is proposed by using a common spectrofluorimeter to detect intensity of resonance double line scattering (RDLS). Proteins including bovine serum albumin (BSA), human serum albumin (HSA) can combine with morin and cetyltrimethylammonium briomide (CTMAB) in the pH range 7.0-8.0 and produce enhanced RDLS signal at lambda(ex)/lambda(em) 305.0/610.0 nm. Optimization conditions for the morin-protein-CTMAB interaction were tested. In the studied system, BSA/CTMAB/morin = 1:2:3. The association constant of morin with BSA is 5.2 x 10(4). Under the optimum conditions, the linear range is 7.5 x 10(-8)-1.0 x 10(-5) g/ml for BSA, 2.5 x 10(-8)-5.0 x 10(-6) g/ml for HSA. The detection limits (S/N = 3) are 66.0 ng/ml for BSA and 23.0 ng/ml for HSA, respectively. Four synthetic samples were analyzed satisfactorily.  相似文献

在pH4.3的B-R缓冲体系中,用微相吸附-光谱修正技术[1]研究了茜素红(ARS)与牛血清白蛋白(BSA)、人血清白蛋白(HSA)的结合反应。其吸附结合常数分别为:KBSA-ARS=3.950×104,KHSA-ARS=4.377×104。染料与蛋白的最大结合数分别为NARS∶NBSA=9∶1,NARS∶NHSA=7∶1。经光谱修正技术计算结合产物的实际摩尔吸光系数分别为εBSA-ARS(537nm)=2.517×104L.mol-1.cm-1,εHSA-ARS(519nm)=2.051×104L.mol-1.cm-1,检出限BSA为19mg/L,HSA为23mg/L。经探讨该结合反应机理符合Langmuir吸附聚集反应方程。  相似文献

The interactions of two drugs, cryptotanshinone (CTS) and icariin, with bovine serum albumin (BSA) and human serum albumin (HSA) have been investigated using multiple spectroscopic techniques under imitated physiological conditions. CTS and icariin can quench the fluorescence intensity of BSA/HSA by a static quenching mechanism with complex formation. The binding constants of CTS–BSA, CTS–HSA, icariin–BSA and icariin–HSA complexes were observed to be 1.67 × 104, 4.04 × 104, 4.52 × 105 and 4.20 × 105 L·mol?1, respectively at 298.15 K. The displacement experiments suggested icariin/CTS are primarily bound to tryptophan residues of the proteins within site I and site II. The thermodynamic parameters calculated on the basis of the temperature dependence of the binding constants revealed that the binding of CTS–BSA/HSA mainly depends on van der Waals interaction and hydrogen bonds, and yet the binding of icariin–HSA/BSA strongly relies on the hydrophobic interactions. The binding distances between BSA/HSA and CTS/icariin were evaluated by the Föster non-radiative energy transfer theory. The results of synchronous fluorescence, 3D fluorescence, FT-IR and CD spectra indicates that the conformations of proteins were altered with the addition of CTS or icariin. In addition, the effects of some common ions on the binding constants of CTS/icariin to proteins are also discussed.  相似文献

The reaction of [NEt(4)](2)[Fe(2)OCl(6)] with sodium benzoate, 4,6-dimethyl-2-hydroxypyrimidine (dmhp), and 1,1,1-tris(hydroxymethyl)ethane (H(3)thme) gives the undecametallic compound [NEt(4)][Fe(11)O(4)(O(2)CPh)(10)(thme)(4)(dmhp)(2)Cl(4)]. X-ray crystallography, EPR spectroscopy, bulk magnetic susceptibility studies, and low-temperature single-crystal magnetic measurements were used to characterize the compound. Magnetic measurements indicate an S = (11)/(2) ground state with the parameters g = 2.03 and D = -0.46 cm(-)(1). Single-crystal magnetic studies show hysteresis of molecular origin at T < 1.2 K with fast quantum mechanical tunneling at zero field.  相似文献

A series of layered oxides of nominal composition SrFe(1-x)Mn(x)O(2) (x = 0, 0.1, 0.2, 0.3) have been prepared by the reduction of three-dimensional perovskites SrFe(1-x)Mn(x)O(3-δ) with CaH(2) under mild temperature conditions of 583 K for 2 days. The samples with x = 0, 0.1, and 0.2 exhibit an infinite-layer crystal structure where all of the apical O atoms have been selectively removed upon reduction. A selected sample (x = 0.2) has been studied by neutron powder diffraction (NPD) and X-ray absorption spectroscopy. Both techniques indicate that Fe and Mn adopt a divalent oxidation state, although Fe(2+) ions are under tensile stress whereas Mn(2+) ions undergo compressive stress in the structure. The unit-cell parameters progressively evolve from a = 3.9932(4) ? and c = 3.4790(4) ? for x = 0 to a = 4.00861(15) ? and c = 3.46769(16) ? for x = 0.2; the cell volume presents an expansion across the series from V = 55.47(1) to 55.722(4) ?(3) for x = 0 and 0.2, respectively, because of the larger effective ionic radius of Mn(2+) versus Fe(2+) in four-fold coordination. Attempts to prepare Mn-rich compositions beyond x = 0.2 were unsuccessful. For SrFe(0.8)Mn(0.2)O(2), the magnetic properties indicate a strong magnetic coupling between Fe(2+) and Mn(2+) magnetic moments, with an antiferromagnetic temperature T(N) above room temperature, between 453 and 523 K, according to temperature-dependent NPD data. The NPD data include Bragg reflections of magnetic origin, accounted for with a propagation vector k = ((1)/(2), (1)/(2), (1)/(2)). A G-type antiferromagnetic structure was modeled with magnetic moments at the Fe/Mn position. The refined ordered magnetic moment at this position is 1.71(3) μ(B)/f.u. at 295 K. This is an extraordinary example where Mn(2+) and Fe(2+) ions are stabilized in a square-planar oxygen coordination within an infinite-layer structure. The layered SrFe(1-x)Mn(x)O(2) oxides are kinetically stable at room temperature, but in air at ~170 °C, they reoxidize and form the perovskites SrFe(1-x)Mn(x)O(3-δ). A cubic phase is obtained upon reoxidation of the layered compound, whereas the starting precursor SrFeO(2.875) (Sr(8)Fe(8)O(23)) was a tetragonal superstructure of perovskite.  相似文献

The polarized and nonpolarized fluorescence of bovine serum albumin and human serum albumin in Triton X-100 solutions is studied at different pH values. Analysis of the constants of fluorescence quenching for BSA and HSA after adding Triton X-100 and the hydrodynamic radii of BSA/HSA–detergent complexes show that the most effective complexation between both serum albumins and Triton X-100 occurs at pH 5.0, which lies near the isoelectric points of the proteins. Complexation between albumin and Triton X-100 affects the fluorescence of the Trp-214 residing in the hydrophobic pockets of both BSA and HSA.  相似文献

在十六烷基三甲基溴化胺(CTAB)存在下,将苯胺(An)与吡咯(Py)两种单体在Fe3O4磁流体中原位化学氧化共聚制备了PAn-co-PPy/Fe3O4网状纳米纤维复合材料。通过改变CTAB的浓度、An/Py单体的配比及Fe3O4磁流体的用量获得了形态和电磁性能可调控的纳米纤维复合物。采用SEM、TEM、XRD、TG、电导和磁性能测试对复合物进行了表征,通过矢量网络分析仪获得了试样在2-18 GHz范围的复介电常数和复磁导率,经计算获取相应的微波反射损耗曲线。结果表明共聚复合物具有比单一的聚合物、共聚物、PAn/Fe3O4及PPy/Fe3O4更优越的微波吸收性能,样品(含Fe3O412.4 wt%)在9.0 GHz处具有最大的反射损耗为 -36.5 dB,损耗超过 -10 dB的频带宽度达4.7 GHz。  相似文献
