A novel method of on-line pyrolytic methylation–gas chromatography was developed for the rapid analysis of fatty acid composition in Polysorbate 80 without any tedious pre-treatment steps. Fatty acids in Polysorbate 80 were converted into their corresponding fatty acid methyl esters in the presence of trimethylsulfonium hydroxide with a vertical microfurnace pyrolyzer at 300 °C. The premixing procedure of sample and organic alkali reagent was necessary before the on-line pyrolytic methylation to improve the repeatability. The relative standard deviations for peak areas of fatty acids in Polysorbate 80 were over the range of 0.3–9.1% (n = 5). Six Polysorbate 80 samples, consisting of three samples of pharmaceutical grade and three samples of non-pharmaceutical grade, were analyzed to evaluate the feasibility of the proposed method. The relative percentages (%) of fatty acids for samples of pharmaceutical grade meet the Chinese Pharmacopoeia requirements with the amount of oleic acid varying from 78.4 to 89.3%. On the other hand, the relative percentages (%) for palmitic acid and stearic acid in samples of non-pharmaceutical grade were out of the specification limits, with the amount of oleic acid varying from 62.0 to 63.5%. The quantitative results determined by on-line pyrolytic methylation were in agreement with those obtained by off-line methylation. The result proved that gas chromatography with on-line pyrolytic methylation technique is of great value for rapid screening analysis of Polysorbate 80 samples in bulks.  相似文献

A novel methodology in on-line pyrolysis-gas chromatography (Py-GC) for the fast analysis of fatty acids in vegetable oils with minimal sample treatment and the use of non-toxic reagents is described. Pyrolysis at 500 degrees C for 10 s of sub-microgram quantity of vegetable oil dissolved in dimethyl carbonate (DMC) and in the presence of nanopowder titanium silicon oxide resulted in the production of fatty acid methyl esters (FAMEs) as unique products. Pyrolysis performed by means of a resistively heated filament pyrolyser interfaced to a GC-MS apparatus enabled the direct analysis of evolved FAMEs. The DMC/Py-GC-MS analysis was tested on soybean, coconut, linseed, walnut and olive oil and the results compared to the classical BF(3)-methanol as reference methodology. The DMC method exhibited a lower precision and was biased towards lower levels of polyunsaturated fatty acids (PUFA) in comparison to the BF(3)-methanol method, but was more advantageous in terms of reduced sample treatment, waste generation and risk factors of employed chemicals.  相似文献

建立了分析棉籽仁中脂肪酸组成的在线热辅助甲基化-气相色谱法。将0.3 mg棉籽仁样品与2 μ L三甲基氢氧化硫(0.2 mol/L)加入裂解器,在350℃下进行甲基化反应,通过气相色谱仪进行分离分析,共检测到8种脂肪酸甲酯成分,分别为亚油酸(C18:2)、油酸(C18:1)、棕榈酸(C16:0)、硬脂酸(C18:0)、肉豆蔻酸(C14:0)、棕榈油酸(C16:1)、花生酸(C20:0)和二十二酸(C22:0),不饱和脂肪酸的相对含量为66.30%~72.54%,其中亚油酸的相对含量为43.20%~53.61%,相对峰面积的相对标准偏差(RSD)小于10%(n=5)。通过分析5组棉籽仁样品与3种食用油中的脂肪酸组成,结果表明不同产地的棉籽仁中的脂肪酸组成差异不明显,且棉籽仁中的脂肪酸组成与玉米油最为接近,相似度为0.960~0.992。该方法简单、快速、准确,适合分析棉籽仁中的脂肪酸组成。  相似文献

Methanolic extracts of paint samples of different composition and age were qualitatively investigated by GC–MS using an on-column injector after off-line methylation or trimethylsilyl derivatisation, and on-line thermally assisted (trans)methylation with tetramethylammonium hydroxide using Curie-point pyrolysis–GC–MS. The combination of these three analytical strategies led to the identification of typical oxidation products of unsaturated fatty acids by interpretation of their mass spectrum. Some of the identified compounds have not been reported before. Both the off-line and on-line GC–MS strategy show series of short-chain fatty (di)acids and C16 and (oxidised) C18 fatty acids. The major advantage of the on-line pyrolysis–GC–MS approach is that chemical work-up is minimal and very quick. With this technique both the carboxylic acid functionalities, and hydroxyl groups are methylated. Young paint films are shown to contain relatively more oxidised C18 fatty acids and less diacids compared to older paints, which is indicative for the on-going oxidation processes within the paint. After trimethylsilylation, monoacylglycerols are detected indicative for hydrolytic processes, which reflect the relative distribution of the most prominent silylated fatty acids present. Relatively more C16 and C18 monoacylglycerols are found in young paints, whereas older paints contain higher amounts of monoacylglycerols of diacids.  相似文献

为了研究灵香草浸膏的热裂解行为,采用在线有氧热裂解-冷阱捕集-气相色谱-质谱联用技术,模拟卷烟燃吸状态对灵香草浸膏进行了热裂解分析,并对灵香草浸膏热裂解前后的挥发性成分进行了比较分析。从灵香草浸膏的热裂解产物中共鉴定出64种成分,占总峰面积的88.27%,主要成分为高级脂肪酸及其酯类;灵香草浸膏热裂解后的挥发性成分数量多于裂解前(45个),说明灵香草浸膏经热裂解生成了新的化合物。热裂解前后共有的化合物有20个,主要是高级脂肪酸及其酯类、新植二烯、5-(羟甲基)-2-呋喃甲醛、3-羟基-4,5-二甲基-2(5H)-呋喃酮等化合物。在线有氧裂解模式更接近烟用添加剂样品的真实裂解状态,操作简单、快捷,结果准确。  相似文献

A mild procedure for the purification of methyl esters of the fatty acid components of cholesterol esters, from interfering free cholesterol and other contaminating residues, is described. Methyl esters and free cholesterol are formed during the methylation of cholesterol esters. When co-extracted, cholesterol and other contaminating residues interfere with the methyl esters because minute proportions of these residues tend to elute at the same retention times as palmitoleic and stearic acids, to yield unreliable but significantly higher values for palmitoleic (p less than 0.001) and stearic acids (p less than 0.0001), and correspondingly lower values for oleic acid (p less than 0.0001). Purification of methyl esters by thin-layer chromatography eliminates this problem and yields reliable analysis of cholesterol ester fatty acids, without measurable oxidation of unsaturated fatty acids.  相似文献

利用衰减全反射(ATR)傅立叶红外光谱(FTIRS)法对市售优质品牌的8种植物油(转基因豆油、非转基因豆油、葵花籽油、花生油、玉米油、红花籽油、调和油、橄榄油)未加热及反复高温加热后的红外二阶导数光谱进行比较,发现复热食用油在反复高温加热下会使不饱和脂肪酸亚油酸、亚麻酸的含量降低,饱和脂肪酸棕榈酸和硬脂酸的含量增加;顺式脂肪酸的含量下降,反式脂肪酸的含量明显增加。确定以二阶导数光谱中3 009、988、966 cm-13处特征吸收峰的峰高判定是否为反复加热用油。此方法样品用量少、分析速度快,可从整体上了解油品的质量及成分,并可作为市场复热食用油的快速筛查方法,也可为地沟油检测方法的建立提供理论依据。  相似文献

Methyl ester derivatives of fatty acids were analyzed for the determination of the constituents of Datura alba seed oil. Gas chromatography coupled to mass spectrometer was used for these analyses. Results delivered that there were saturated as well as unsaturated fatty acids in Datura alba seed oil. Total of 15 different fatty acid components were identified and quantified. Methyl linoleate was found in highest concentration (16.22%) among the identified analytes of interest. In addition methyl esters of Palmitic acid (6.59%), Oleic acid (5.41%) and Stearic acid (1.35%) were found. Concentrations of rest of the detected fatty acids were less than 1%. From the literature it appears that no such work has been performed for the determination of fatty acids in Datura alba seed oil.  相似文献

首次研究了维药细穗柽柳(Tamarix leptostachys Bunge)实中挥发油和脂肪酸的化学成分。 分别采用药典中的挥发油提取法和索式取提法提取柽柳实中的挥发油和脂肪酸,使用气质联用技术获取总离子流图,各色谱峰相应的质谱图经过NIST2011标准谱库检索定性,并采用峰面积归一化法进行定量分析,计算各成分的相对百分含量。 两种方法分别鉴定出48种挥发油和19种脂肪酸。 挥发油主要成分为芳香类化合物(43.71%)、芳香性醛酮类(20.58%)、脂肪酸类(13.03%)、酯类(17.36%)和醇类(4.19%)等。 脂肪酸主要成分为棕榈酸(35.61%)、亚油酸(27.26%)和油酸(11.33%)等,其中不饱和脂肪酸含量占总脂肪酸含量的38.65%。 维药细穗柽柳实中富含丰富的挥发油和不饱和脂肪酸,具有很好的开发利用价值。  相似文献

A new process for enzymatic synthesis of biodiesel at high water content (10–20%) with 96% conversion by lipase from Candida sp. 99–125 was studied. The lipase, a no-position-specific lipase, was immobilized by a cheap cotton membrane and the membrane-immobilized lipase could be used at least six times with high conversion. The immobilized lipase could be used for different oil conversion and preferred unsaturated fatty acids such as oleic acid to staturated fatty acids such as palmitic acid. The changes in concentration of fatty acids, diglycerides, and methyl esters in the reaction were studied and a mechanism of synthesis of biodiesel was suggested: the triglycerides are first enzymatically hydrolyzed into fatty acids, and then these fatty acids are further converted into methyl esters.  相似文献

The production of bio-oil via the slow pyrolysis of dissolved air flotation (DAF) skimmings from poultry processing is described. The raw DAF skimmings were characterized for physicochemical properties and for thermal behavior (TGA). The bio-oil was produced in a batch pyrolysis system at varying temperatures between 400 and 700 °C to study the effect of temperature on product yield. The fatty acids in the bio-oil produced displayed a high degree of saturation that caused the bio-oil to have poor cold flow properties (high cloud point and viscosity) so a solvent extraction scheme was devised to extract a bio-oil fraction rich in unsaturated fatty acids that could be further esterified into a bio-diesel and fatty nitriles that could be further processed into surfactants. This ethyl acetate-soluble fraction demonstrated much improved cold flow properties as well as lower water content and a higher HHV. The esterification of this soluble fraction was performed using methanol and sulfuric acid as an acid catalyst and the formation of fatty acid methyl esters was verified using GC/MS and FT-IR.  相似文献

Supercritical carbon dioxide can be utilized both as an introduction solvent in capillary SFC and as a reaction medium; both possibilities are illustrated in this study. An off-line SFE unit was modified for on-line SFE-SFC. To facilitate rapid depressurization of the extraction cell and to prevent memory effects, a 6-port valve was installed at the outlet line of the extraction unit. An increased background signal was obtained when PEEK polymer was used in the construction of the extraction vessel; when stainless steel was used, the blanks improved. The synthesis of methyl esters and butyl esters of fatty acids from triglycerides in edible fat was examined using an immobilized lipase as a catalyst in on-line SFE-SFC. As a result of 30 minutes reaction – extraction time, high yields of fatty acid esters were obtained at a pressure of 150 bar and a temperature of 50°C.  相似文献

Summary Tetramethylammonium hydroxide in methanol has been proposed as a catalyst for preparing methyl esters of both free fatty acids and glycerides of fats. Nevertheless, procedures normally recognized as suitable can be inadequate when the fat under study contains noticeable amounts of low relative molecular mass esters. The method using tetramethylammonium hydroxide has been examined and modified in order to analyze milk fats by capillary GC. The pyrolysis methylation of free fatty acids in capillary injectors has been also studied. The modified method was satisfactory for both neutral and acid milk fats.  相似文献

Oilseed crop with high oil content and promising ecological adaptability are potential sources for competitive biodiesel production. This study investigates the scope of utilizing biodiesel development through the methyl and ethyl ester from soybean and mustard oil as an alternative fuel. Methyl and ethyl esters of oils having different fatty acids compositions such as soybean (SOME and SOEE) and mustard oil (MUME and MUEE) were prepared by transesterification with methanol and ethanol in the presence of an alkali-KOH catalyst. The gas chromatographic (GC) analysis of oil samples revealed that primary fatty acid composition in soybean oil was linoleic acid (C18:2, 51.93%), followed by oleic acid (C18:1, 22.82%), palmitic acid (C16:0, 11.56%), linolenic acid (C18:3, 5.95%) and stearic acid (C18:0, 4.32%). Whereas, the main components in mustard oil were erucic acid (C22:1, 32.81%), oleic acid (C18:1, 24.98%), eicosenoic acid (C20:1, 10.44%), linolenic acid (C18:3, 8.61%) and palmitic acid (C16:0, 2.80%). The physicochemical properties (acid value, iodine value, calorific value, flash point, pour point etc.) of methyl and ethyl ester samples were estimated and found to be within the acceptable range of ASTM D6751 standards specifications. The prepared esters and oil samples were examined for cold flow properties by differential scanning calorimetry (DSC). Results revealed better cold flow properties for MUME (−2.55 °C) and MUEE (−3.10 °C) than SOME (3.21 °C) and SOEE (1.83 °C) due to more unsaturated fatty acid content in MU. Thermal and oxidative stability of samples was determined by thermogravimetric analysis (TG) and differential thermal analysis (DTA). The thermal and oxidative stability ranking of the samples was in the order of oil > methyl esters > ethyl esters.

Biodiesel oxidation is a complex process widely influenced by the chemical composition of the biofuel and storage conditions. Several oxidation products can be formed from these processes, depending on type and amount of the unsaturated fatty acid esters. In this work, fatty acid methyl and ethyl esters were obtained by base-catalyzed transesterification of soybean oil and physicochemically characterized according to standards from ASTM, EN, and ABNT. The thermal and oxidative stabilities of biodiesel samples were investigated during the storage process by pressure differential scanning calorimetry (PDSC) and by viscosity measurements. Absolute viscosities of biodiesels after accelerated aging were also determined. The viscosity increased as the aging temperature and time were raised. The results showed that oxidation induction can occur during storage, decreasing the biodiesel stability. PDSC analysis showed that during storage under climate simulation the values of high-pressure oxidative induction times (HPOIT) were reduced for both FAEE and FAME.  相似文献

Thermogravimetry technique is useful to determine the thermal stability of vegetable oils. In this paper some vegetable oils obtained from brazilian Cerrado native plants were studied based on their high oleic acid content. Amburana, baru and pequi pulp oils presented higher yield of extraction compared to soybean oil. The thermal stability of oils in nitrogen was very close hence their fatty acid composition was very similar. Amburana and baru oils have major amount of unsaturated fatty acids, especially linoleic acid and pequi pulp oil has the highest short chain fatty acid content which can explain its lowest thermal stability in synthetic air.  相似文献

Presently, dried distiller's grains with solubles (DDGS) are mainly used as the livestock feed. However, the high fiber content in DDGS limits its use as the diet for animals. Therefore, with increasing production of DDGS in recent years, it is desirable to find some new uses of DDGS for fuels and/or for high value chemicals. In this paper, experiments on pyrolysis of DDGS by spouted-entrained bed and by fixed bed are carried out, and the pyrolytic liquids are analyzed by GC/MS. It was found that the composition of the liquid by pyrolysis of DDGS in 490–570 °C by spouted-entrained bed is rather complex, and varies with pyrolytic temperature. However, the pyrolysis of DDGS material is not quite suitable to the process by spouted-entrained bed, due to a severe clogging problem inside the reactor. By fixed bed, the composition of the oil phase of the liquid obtained in 490–610 °C is much simpler, mainly phenol derivatives, fatty acids and their esters. When pyrolyzed at 570 °C with catalyst of CaO, aliphatic and aromatic hydrocarbons are generated more, while fatty acids and their esters are much reduced.  相似文献

Vapors released by the skin in the hand of one human subject are detected in real time by sampling them directly from the ambient gas surrounding the hand, ionizing them by secondary electrospray ionization (SESI, via contact with the charged cloud from an electrospray source), and analyzing them in a mass spectrometer with an atmospheric pressure source (API-MS). This gas-phase approach is complementary to alternative on-line surface ionization methods such as DESI and DART. A dominating peak of lactic acid and a complete series of saturated and singly unsaturated fatty acids (C12 to C18) are observed, in accordance with previous off-line studies by gas chromatography-mass spectrometry. Several other metabolites have been identified, including ketomonocarboxylic and hydroxymonocarboxylic acids.  相似文献
