考察了大孔苯乙烯-二乙烯苯交联共聚物的交联度、致孔剂理及胺化试剂对载体固定化猪胰脂肪酶的影响。选择出一种最佳载体对猪胰脂肪酶进行固定化,对比了自由酶和固定化酶在有机相中催化丁酸甲酯和正丁醇的酯交换反应。结果表明,酶经固定化后催化反应活力比自由酶提高近1倍。  相似文献

本文研究了不同温度、pH值和储存时间下,游离猪胰脂肪酶与自制碳酸钙固定化猪胰脂肪酶的稳定性,并将固定化猪胰脂肪酶用于催化对二氧环己酮开环聚合。研究结果显示,固定化酶的热稳定性、pH稳定性以及储存稳定性均优于游离酶。当固定化酶用量为单体质量30%,反应温度为80℃,反应时间为72 h时可获得数均分子量为2024 g·mol-1的聚合物。  相似文献

利用X射线微区分析的方法,对吸附交联法得到的固定化脂肪酶的微观活性进行了分析。结果表明:以合成出的大孔吸附树脂为固定化酶栽体,酶活较高,活性脂肪酶分布较均匀。并得到了固定化脂肪酶的活性定位的最佳条件。  相似文献

Porcine pancreas lipase (PPL) and PPL immobilized on narrow distributed micron-sized glass beads wereemployed successfully for the ring-opening polymerization of 5, 5-dimethyl-1, 3-dioxan-2-one (DTC) for the first time.Different polymerization conditions such as enzyme concentration and reaction temperature were studied. Immobilized PPLexhibits higher activity than native PPL. Along wth the increasing enzyme concentration, the molecular weigh of resultingPDTC decreases. PPL immobilized on narrow distributed micron-sized glass beads has outstanding recyclability. For thethird recycle time, immobilized PPL exhibits the highest catalytic activity and with high activity even after the fifth recyletime for the synthesis of PDTC. The ~1H-NMR spectra indicate that decarboxylation does not occur during the ring-openingpolymerization.  相似文献

Silica nanoparticles were first used as the carrier for the porcine pancreas lipase (PPL) immobilization. The result of transmission electron microscopy (TEM) showed that the immobilized lipase was still in nanosize after enzyme immobilization. The ring-opening polymerization of 2,2-dimethyltrimethylene carbonate (DTC) catalyzed by this immobilized PPL (IMPPL) was explored. 1H NMR spectra suggested no evidence of decarboxylation during propagation. Influences of IMPPL concentration and reaction temperature on the molecular weight and yield of poly(DTC) were studied. The recovery and reuse of IMPPL for the ring-opening polymerization of DTC was also investigated. The recycling IMPPL showed even higher catalytic activity and a higher molecular weight of polycarbonate could be achieved.  相似文献

A series of functionalized ionic liquid modified mesoporous silicas SBA-15 (FIL-SBA) were synthesized by modulating the loading and cation/anion ratio of the functionalized ionic liquid (FIL). The prepared materials FIL-SBA were used as a novel carrier system to immobilize porcine pancreas lipase (PPL). Enzymatic activity and reusability of the immobilized enzyme were investigated using the triacetin hydrolysis reaction. The combined advantages of the nano-sized pore diameter, large surface area and high pore volume of SBA-15, and the tunable properties of the FIL for enzymes immobilized in FIL-SBA gave a maximum improvement of 570% in relative activity, with 63% retention of initial activity after five cycles of use. Carriers and immobilized enzymes were characterized using nitrogen adsorption, small-angle X-ray diffraction (SXRD), Fourier transform infrared (FT-IR), elemental analysis, nuclear magnetic resonance (NMR), scanning and transmission electron microscopy (SEM and TEM). It was shown that the introduction of FIL influenced the catalytic behavior of PPL significantly by changing the structure and surface properties of the carriers.  相似文献

Kinetic resolutions of a number of β-hydroxy selenides promoted by enzymes were performed using PPL (free Porcine pancreatic lipase), PSL (Amano PS—free Pseudomonas sp. lipase) and CALB (NOVOZYM 435®—immobilized Candida Antarctica lipase type B) with (RS)-1-phenylselanyl-propan-2-ol. CALB gave the best results and provided both (R)- and (S)-enantiomers in high enantiomeric purity. A comparative study of the effect of temperature, solvent, enzyme immobilization and the structure of the substrates on the resolution is presented.  相似文献

Chiral furyl substituted carbinols were prepared from the related carbonyl compounds and their enzymatic kinetic resolution was studied by using porcine pancreatic lipase (PPL) in transesterification reaction under solvent free condition. This study revealed that carbinols having one less bulky group on the chiral center are good substrates for PPL and the resolution proceeds with higher ee,s and less reaction time.  相似文献

研究了14元环的碳酸丁二酯二聚体在固定化脂肪酶Novozym-435催化下的开环聚合反应制备聚碳酸丁二酯.聚合在常压,75℃的甲苯溶液中进行,反应条件温和.详细探讨了反应条件诸如单体浓度,酶浓度对于聚合的影响.结果显示Novozym-435具有与异辛酸亚锡可比拟的高催化活性,同时可以回收重复使用.聚合动力学研究表明碳酸丁二酯的酶促甲苯溶液开环聚合和环状内酯的酶促甲苯溶液聚合有所不同,没有表现出活性聚合的特征.  相似文献

The results of validation of a method for the analysis of free and bound phenolics in wine and grapes are presented. Wine and grape extracts are fractionated by automated solid-phase extraction on Bond Elut PPL cartridges to give free and bound phenolic fractions. Bound fractions are subjected to acid hydrolysis, and the phenolics released are recovered by solid-phase extraction on Bond Elut PPL cartridges. The fractions are further purified by automated solid-phase extraction on Bond Elut silica cartridges. After derivatisation to form trimethylsilyl ethers, the phenolics are determined by gas chromatography–mass spectrometry with selected ion monitoring. The method is suitable for robust, high-throughput monitoring of the concentrations of phenolics that can affect the palatability of wine.  相似文献
