质子辐射生物学效应是太空放射生物学和质子束放疗研究的重要基础,可为空间环境下人员的危险性评估以及质子治疗优化设计提供科学依据。依托加速器建立相应的生物样本辐照技术是开展此类研究的前提条件。中国原子能科学研究院最近建立的100 MeV强流质子回旋加速器提供的中能质子束流为目前国内能量最高,特别适合用于太空放射生物学和质子治疗相关研究。本研究中,利用在束和离线等多种手段建立了中能质子束流诊断和剂量测量方法,对加速器引出的100 MeV质子照射野大小、均匀性等束流品质以及剂量测量系统准确性进行了分析和评估。结果表明,对光子剂量响应好的LiF(Mg, Ti)热释光探测器,对90 MeV质子同样具有良好的剂量响应关系,可作为中能质子剂量准确性评估的手段之一。在5.0 cm×5.0 cm照射野范围内,加速器引出的100 MeV质子束流的均匀性好于90%,在线剂量测量系统准确性好于93%,束流品质和剂量测量条件基本满足辐射生物学的要求,可为质子辐射生物学效应研究的开展提供可靠保障。  相似文献

Choosing ^7 Li and ^12C heavy ions respectively with different linear energy transfer (LET) values, purified plasmid DNA samples in aqueous solution are irradiated with various doses. The atomic force microscopy (AFM) is used for analysis of DNA fragments induced by both the kinds of heavy ions. There is a change of three forms of DNA, i.e. supercoiled, open circular and linear form, as the dose is observed. The distribution function of DNA fragment length is obtained for the first time and fitted with the Tsallis entropy statistical theory. The result indicates that AFM is a useful tool for analysis of the short fragment of DNA, high-LET heavy ion radiation induces DNA double strand breaks (DSBs) more effectively, and the distributions of the DSBs are more local and dense in comparison with low-LET radiation.  相似文献

利用HI-13串列加速器提供的19 MeV 质子照射麻醉小鼠2, 4, 8和16 Gy , 照射后不同时间麻醉处死小鼠进行病理组织学观察。 结果显示, 照射后各组小鼠腰背部开始环状脱毛, 病理改变为表皮细胞坏死、 脱落, 皮下水肿, 心、 肝出现小灶性坏死或伴出血, 肝细胞明显再生, 肺组织可出现充血及肺泡间隔明显增厚, 肾小球出现明显固缩、 坏死及消失。 这些病理改变在大剂量照射组尤为明显。 通过观察质子照射小鼠组织病理损伤效应, 为质子损伤的防治研究提供参考依据。Space radiation come from galactic cosmic rays and solar particle events which contain considerable protons is an invisible killer to astronauts. Furthermore, many wounded persons were injured by proton and other heavy ion radiation in a nuclear warhead or nuclear terrorism attack. However, the damage effect of proton radiation is known little in our country. In this study, the proton induced histopathologic changes of mice were observed primarily. naesthetic mice were irradiated with the 19 MeV protons dili vered by HI 13 tandem accelerator at CIAE in different doses(2, 4, 8, 16 Gy). In order to observe the da mage effect, mice were killed by anaesthetic at different time point after radiation, and the tissue sections were analyzed by a histopathologic method. The results show that all of the mice’s backs began displaying ringed depilation at post irradiation day 7. Many epidermal cells necrosis and exfoliation happened, and subcutaneous dropsy was observed. There were some focal necrosis or even followed with bleeding in heart and liver. Hepatocyte evidently regenerated. A hyperaemia was seen in lung tissue and alveolar septum was obviously thickening. Acinus renis represented notable pyknosis, necrosis and disappearance. Especially, these pathologic alteration clearly displayed in high dose groups. In conclusion, proton radiation can induce different grade injury to skin, heart, liver, lung, kidney and other organs. Further studies should be made to deeply understand the mechanism about the proton radiation damage.  相似文献

利用25 MeV/u 40Ar+Se中能重离子核反应, 制备包含24种元素在内的多重示踪剂, 通过对元素γ谱的分析, 研究了正常鼠和肿瘤鼠体内微量元素的分布及代谢。Mg, Sr, Ga, As, Sc, V, Cr, Mo, Co, Fe, Y, Zr, Nb和Ru等元素在肿瘤鼠的粪便和尿液中的排泄时间与正常鼠不同。 肿瘤鼠与正常鼠相比, Na, Mn, Fe和Co的排泄变化较大, 肿瘤鼠的排泄率较正常鼠高; 而Ca, Y, Zr和Mo的排泄率肿瘤鼠则较正常鼠低; 在正常鼠脏器、 组织和血液样品中测出了Na, Rb, Ga, Sc, As, Zn, V, Cr, Mn, Co, Fe、Y, Zr, Tc, Ru, Ag和In等17种元素, 肿瘤鼠检测出了除Zn和As之外的其它15种元素。 几乎所有的元素在正常鼠中的吸收均比肿瘤鼠高。 大部分元素分布在肝、 肾和毛皮以及实体瘤中, 而Fe和Na在肿瘤组织则没有检测到。 A radioactive multitracer solution of the 24 elements, e.g. Be, Na, K, Rb, Mg, Ca, Sr, Ga, As, Sc, V, Cr, Mn, Co, Fe, Zn, Y, Zr, Mo, Nb, Tc, Ru, Ag and In, was obtained from the nuclear reaction of 25 MeV/u 40Ar+Se with a series of chemical process. The multitracer solution was orally administered to normal and muscular tumour bearing mice of male Balb/c mice. Urine and faeces samples of mice were collected. The two group mice were sacrificed after 96 h. The uptake of 17 elements, Na, Rb, Ga, As, Sc, V, Cr, Mn, Co, Fe, Zn, Y, Zr, Tc, Ru, Ag and In, were simultaneously detected in normal mice while 15 elements, Na, Rb, Ga, Sc, V, Cr, Mn, Co, Fe, Y, Zr, Tc, Ru, Ag and In, were simultaneously detected in tumour bearing mice. Our results indicate that the majority of the detected elements were distributed in liver, kidney, pelt, tumour while a small fraction of the biotrace elements were distributed in heart and spleen (tumour bearing mice) in the two groups of mice. The higher concentrations of Fe, Na, Mn were detected in heart or kidney of normal mice. Na, Mn, Fe and Co showed better absorption in most tissues in the normal mice, except for Na and Mn in heart.  相似文献

介绍了经辐射损伤后细胞失活或存活的理论模型及相关问题的一些研究工作。重点考虑了径迹结构和能量沉积特点的新的细胞失活模型。在径迹结构模拟的基础上, 计算并分析了DNA损伤谱, 同时把径迹结构得到的损伤谱与细胞存活联系起来, 初步在分子水平上理解了细胞失活或存活的机制。The several theoretical models of cell inactivation or cell survival fraction are introduced. Emphasis is placed on a new model——Ionization Ionization Clustering Cluster model. In order to understand some of the mechanisms of DNA damage related to cell killing and their dependence on radiation quality, the yields of single track lethal events are compared with the estimated yields of different classes of complex DNA lesions calculated by a fast Monte Carlo methods.  相似文献

7Li离子属于高LET辐射,是辐射生物学基础研究中常用的重离子射线之一,它是硼中子俘获治疗癌症中起关键作用的离子,在辐射诱变育种等领域也有较好的应用前景。实验在北京HI-13串列加速器R20支线生物用新终端上进行,对43 MeV 7Li离子束斑大小、均匀性和监测注量的准确性进行测量和评估,结果表明:均匀性好于90%的最大束斑面积为5.0cm×5.0cm;4.2×104 ~1.5×105 particles/cm2/s注量率范围内,两种不同探测器测得的注量率变化呈线性关系,闪烁体探测器监测的注量与CR39探测器测得的绝对注量的误差小于10%。这些束流特性能够满足辐射生物学实验要求,有利于进一步开展与硼中子俘获治疗癌症或辐射诱变育种中7Li离子的辐射生物学相关研究。7Li ion with high linear energy transfer (LET) radiation has been used in radiobiological research, takes main effect in Boron Neutron Capture Therapy and is applied in radiation mutagenic breeding. This experiment was carried on the new terminal established for radiobiological application, located at the end of R20 branch beam line of HI-13 tandem accelerator. The beam qualities of 7Li ions of 43MeV generated by HI-13 tandem accelerator, including spot size and uniformity and particle fluence accuracy were measured using different detection methods. The results showed that beam uniformity was over 90% at 5cm×5cm area, the flux of S1 and M3 had a good linear relationship as particle flux ranged from 4.2×104 particles/cm2/s to 1.5×105 particles/cm2/s, and particle fluence accuracy was better than 90%. All the results showed that the beam qualities of 7Li ions finely met the basic requirements for radiation biological experiment. It provides important method to study biological effects for fundamental research and tumor therapy or radiation mutagenic breeding application associated with 7Li ions.  相似文献

用重离子^7Li和^12C以及^60Co的γ射线分别辐照玉米种子胚,研究重离子束辐照玉米种子M1代的生物学效应.结果表明:^60Co的丫射线使种子的发芽势和发芽率降低,重离子^7Li和^12C对发芽势和发芽率的抑制作用不明显;重离子辐照后,单株之间光合速率差异增大;M1代植株在雄性育性、叶片颜色及株高等方面发生了多种变化,并产生了一株双胚苗.应用随机多态性DNA分析技术,发现双胚苗的大、小苗之间及其与亲本自交系478在随机多态性DNA指纹上存在的差异,从分子水平上初步证实了双胚苗为自交系478的变异类型.In order to study M_(1) biological effects of heavy ion irradiation on maize seeds, the embryos of dry maize seeds are irradiated with~(7)Li ions and~(12)C ions as well as ~( 60)Co gamma rays. The results are as follows: ~( 60)Co gamma rays decrease germination impetus and germination rate of maize seeds, while the inhibitory effects of germination impetus and germination rate induced by the heavy ions~(7)Li and~(12)C are not significant. 在进行辐射致DNA损伤中, DNA浓度和剂量率是两个重要的因素。 利用γ射线在不同的剂量率下对不同浓度的质粒DNA pUC19水溶液进行了辐照, 利用凝胶电泳对辐照后的样品进行分析。 结果表明, 在同一剂量下被辐照的DNA样品, 随浓度的降低DNA损伤越来越严重; 通过分析软件和理论公式得到的每个DNA上的双链断裂数随着DNA浓度的变化呈现为非线性趋势。 As two factors, the changes of concentration of DNA and dose rate are of significance in the study of DNA damage induced by irradiation. The influence of DNA damage induced by γ ray at different concentration of DNA under different dose rate was investigated. The result of gel electrophoresis indicate that DNA molecules were damaged severely as the concentration of DNA decreasing. The number of double strand breakages per DNA were obtained by the software of Alpha Innotech and theoretical analysis. It shows that the numbers of double strand breakages per DNA presented nonlinear feature as the concentration of DNA decreasing, and the dose rate had little influence on DNA damage.  相似文献

为满足质子单粒子效应实验对大面积、均匀化质子束流的需求,针对中国原子能科学研究院100 MeV质子回旋加速器提供的100 MeV质子进行了双环双散射体扩束方案设计.Geant4模拟表明该方案可在2.4 m位置产生一个均匀性为±1.89%、半径为8 cm的照射野,在5 m位置产生一个均匀性为±5.32%、半径为20 cm的照射野.此外,利用Geant4对双环双散射体扩束方法的基本原理进行了进一步探索,并对第二散射体后加速器管道、初始束斑尺寸、照射野形成距离、改变入射质子能量等实际中各种可能因素对该方案扩束效果的影响进行了讨论.  相似文献
