利用超声波辅助萃取(USE)技术,联合固相萃取(SPE)净化浓缩以及液相色谱-电喷雾串联质谱(LC-ESI-MS/MS)技术,建立了城市污泥和河流底泥中微量-痕量大环内酯类、磺胺类抗生素、甲氧苄胺嘧啶和氯霉素的多目标定量分析方法.样品经USE提取,SPE净化后,用LC-ESI-MS/MS检测.选择C18为分析柱,甲醇、5 mmol/L醋酸铵和0.1%甲酸混合溶液为流动相,选择在ESI正电离源下多反应监测(MRM)模式检测.对比了USE和加速溶剂萃取(ASE)对抗生素的萃取效率,优化了萃取条件.结果表明: USE和ASE对所选抗生素的萃取效率相当; 而50%甲醇溶液在酸性条件下(pH 2)萃取效率最好.抗生素的方法检出限为2.2~66.9 ng/g(干重,下同); 回收率介于74.7%~111.8%之间; 相对标准偏差小于10.6%.应用此方法在广州某污水处理厂脱水污泥及某河涌底泥中检测到磺胺二甲基嘧啶、克拉霉素、脱水红霉素及罗红霉素等多种抗生素,含量为6.8~125.6 ng/g.  相似文献

建立了用于检测牛奶和奶粉中拉沙洛菌素、莫能菌素、尼日利亚菌素、盐霉素、甲基盐霉素和马杜霉素铵6种聚醚类抗生素残留量的超高效液相色谱-串联质谱分析方法.用乙腈提取样品中的聚醚类抗生素,提取液经HLB固相萃取柱净化,采用超高效液相色谱分离,以电喷雾离子源正离子多反应监测模式进行质谱分析.6种抗生素在0.5~100.0 μg/L范围内均呈线性,相关系数r>0.99.在空白样品中添加6种聚醚类抗生素的回收率均在74.0%~98.5%之间; 精密度(RSD)4.8%~17.2%.牛奶中6种聚醚类抗生素检出限均为0.2 μg/L;奶粉中6种聚醚类抗生素检出限均为1.6 μg/kg.  相似文献

Brefeldin A(布雷菲德菌素甲)是一种天然抗生素, 最早发现于1958年, 许多种微生物中都能产生该化合物. 由于其有趣的结构和显著的抗真菌、抗病毒、抗肿瘤等生物活性, 该化合物很早就引起合成化学家们的注意. 到目前为止, 文献中有报道的全合成和表观合成已有30多种. 对近十年来的全合成及部分表观合成进行了综述, 对各路线的主要优缺点也进行了简要的分析. 一些类似物的合成也一并予以简介.  相似文献

The widespread occurrence of antibiotics as contaminants in the aquatic environment has increased attention in the last years. The concern over the release of antibiotics into the environment is related primarily to the potential for the development of antimicrobial resistance among microorganisms. This article presents an overview of analytical methodologies for the determination of quinolone (Qs) and fluoroquinolone (FQs), macrolide (MLs), tetracycline (TCs), sulfonamide (SAs) antibiotics and trimethoprim (TMP) in different environmental waters. The analysis of these antibiotics has usually been carried out by high-performance liquid chromatography (HPLC) coupled to mass spectrometry (MS) or tandem mass spectrometry (MS/MS) and to a lesser extent by ultraviolet (UV) or fluorescence detection (FD). A very important step before LC analysis is sample preparation and extraction leading to elimination of interferences and prevention of matrix effect and preconcentration of target analytes.  相似文献

The confirmatory LC-MS/MS method for the determination of residues for twelve coccidiostats including ionophore antibiotics (lasalocid, maduramycin, monensin, narasin, salinomycin, semduramycin) and chemical coccidiostats (clazuril, decoquinate, diclazuril, halofuginone, nicarbazin and robenidine) in poultry liver has been developed. The sample preparation was based on extraction with acetonitrile, defatting with Alumina columns and clean-up on Oasis HLB spe. The separation of analytes was performed on PhenylHexyl column with a gradient of acetonitrile, methanol and the ammonium formate pH 4.0. For all analytes, at least 2 diagnostic fragmentation ions were monitored. The validation, performed according to the CD 2002/657/EC, proved the suitability of the method for the confirmatory analysis of the coccidiostats. For lasalocid, however, low reproducibility was observed and the proper quantification could not be performed with this method.  相似文献

A highly stereocontrolled, convergent total synthesis of kendomycin [(?)‐TAN2162], an ansa‐macrocyclic antibiotic, is reported. The key of the strategy is an unprecedented Tsuji–Trost macrocyclic etherification, followed by a transannular Claisen rearrangement to construct the 18‐membered carbocyclic framework. The oxa‐six‐ and five‐membered rings were also stereoselectively constructed respectively by a cascade oxidative cyclization at an unfunctionalized benzylic position and using a one‐pot epoxidation/5‐exo‐tet epoxide opening.  相似文献

Graphene-like layered hexagonal boron nitride (g-BN) was prepared and characterized. The performance of using g-BN as an adsorbent for removal of fluoroquinolone antibiotic gatifloxacin (GTF) from aqueous solution was evaluated. g-BN showed an excellent adsorption capability with notable GTF adsorption ratio of more than 90%. Data of equilibrium adsorption of GTF onto g-BN at different temperatures were represented by Langmuir, Freundlich and Tempkin isotherm models, and Langmuir exhibited the best fitting with the maximum adsorption capacity of 88.5 mg·g?1 at 288 K. GTF adsorption was insignificantly affected by solution pH. Competitive role of Na+ and Ca2+ in the solution inhibited the adsorption of GTF and decreased the adsorption capacity a bit. The adsorption process was spontaneous and exothermic. The adsorption was probably governed by π–π interaction between GTF and g-BN, and electrostatic interaction may also exist in the adsorption process.  相似文献

Pharmaceutical antibiotics, as emerging contaminants, are usually composed of several functional groups that endow them with the ability to interact with adsorbents through different interactions. This makes the preparation of adsorbents tedious and time‐consuming to screen appropriate functionalized materials. Herein, we describe the synthesis of clickable SBA‐15 and demonstrate its feasibility as a screening material for the adsorption of antibiotics based on similar adsorption trends on materials with similar functional groups obtained by a click reaction and cocondensation/grafting methods.  相似文献

Twenty-two novel, variously substituted nitroazetidines were designed as both sulfonamide and urethane vinylogs possibly endowed with antimicrobial activity. The compounds under study were obtained following a general procedure recently developed, starting from 4-nitropentadienoates deriving from a common β-nitrothiophenic precursor. While being devoid of any activity against fungi and Gram-negative bacteria, most of the title compounds performed as potent antibacterial agents on Gram-positive bacteria (E. faecalis and three strains of S. aureus), with the most potent congener being the 1-(4-chlorobenzyl)-3-nitro-4-(p-tolyl)azetidine 22, which displayed potency close to that of norfloxacin, the reference antibiotic (minimum inhibitory concentration values 4 and 1–2 μg/mL, respectively). Since 22 combines a relatively efficient activity against Gram-positive bacteria and a cytotoxicity on eucharyotic cells only at 4-times higher concentrations (inhibiting concentration on 50% of the cultured eukaryotic cells: 36 ± 10 μM, MIC: 8.6 μM), it may be considered as a promising hit compound for the development of a new series of antibacterials selectively active on Gram-positive pathogens. The relatively concise synthetic route described herein, based on widely available starting materials, could feed further structure–activity relationship studies, thus allowing for the fine investigation and optimization of the toxico-pharmacological profile.  相似文献
