During the process development for multistep synthesis of Rosuvastatin calcium several impurities were obtained along with the final Rosuvastatin calcium. Out of this; synthesis of impurity A (acetone adduct) a minor impurity of Rosuvastatin calcium (3R,5S,6E)-7-[4-(4-fluorophenyl)-2-[[(2-hydroxy-2-methylpropyl)sulfonyl(methyl)amino]-6-(1-methylethyl)-pyrimidin-5-yl]-3,5-dihydroxyheptenoicacid hemicalcium salt, is described. The synthesis of impurity A has been accomplished in 6 steps; starting from formation of β-hydroxy sulfonamide as the key intermediate and followed by using convenient routes with overall yield of 13.5%. The sulfonamide core in compounds is effective synthons, which offer exciting perspectives in chemotherapeutic and pharmacological research. In this study, we investigated the molecular interaction of potent sulfonamide derivatives (SDs), 2-benzenesulfonamido-3-methylbutyric acid (BSB) and tetraaquabis{3-methyl-2-[(phenylsulfonyl)amino]butanoate}copper(II) (Cu-BSB), with cationic surfactants, cetyltrimethyl ammonium bromide (CTAB) and ethylhexadecyl dimethyl ammonium bromide (EHDAB), using UV-visible spectroscopy and steady-state fluorescence measurements. Various mathematical models were applied to quantify the effect of cationic surfactants on physicochemical characteristics of BSB and Cu-BSB. These interactions were confirmed by estimating the partition coefficient (Kx), binding capacities (Kb), Stern-Volmer quenching constant (Ksv), and related Gibbs free energies (ΔGb). The in-depth mechanism revealed that the binding mode in SD–surfactants combinational system is spontaneous and quenching exists in static mode initiated by ground-state complex formation. We believe that the true knowledge of host–guest interaction mechanisms concerning model membrane with entrapped moiety can help in better understanding of molecular recognition in related phospholipid membrane models. 相似文献

Some new derivatives 7‐chloro‐2‐[2‐(2,6‐dichlorophenyl)amino]benzyl‐3‐[4‐(2‐substituted phenyl‐4‐oxo‐ thiazolidin‐3‐yl)phenyl]sulfonamido‐quinazolin‐4(3H)‐ones 5a – 5l were synthesized from 2‐[2‐(2,6‐dichloro‐phenyl)amino]phenyl acetic acid via acid chloride, benzoxazinone, amino quinazolin‐4(3H)‐one and Schiff base formation. The synthesized compounds were screened for in vitro antibacterial and antifungal activities by broth micro dilution method. Some of the Schiff base as well as 4‐thiazolidinone derivatives showed promising antibacterial activity while pronounced antifungal activity was observed against C. albicans. 相似文献

建立了加速溶剂萃取(ASE)-高效液相色谱-串联质谱联用法同时检测鱼肉中22种抗生素药物残留的分析方法。样品经ASE提取,HLB固相萃取柱净化后进入高效液相色谱-串联质谱仪分析。对ASE的萃取条件进行优化,并采用XTerraMSC18柱对药物进行分离,以甲醇-乙腈(体积比1∶1)为色谱流动相A,以0.3%(体积分数)甲酸水溶液(含0.1%甲酸铵,pH=2.9)为流动相B。22种喹诺酮、磺胺和大环内酯类抗生素药物在加标水平为20、100μg/L时的回收率分别为72%~120%与66%~114%,相对标准偏差(RSD,n=5)分别为1.9%~16%与0.7%~10%,方法的检出限为0.02~0.6μg/kg。结果显示所建立的方法精密度好,准确度高,可满足同时对鱼肉样品中多种喹诺酮、磺胺和大环内酯类抗生素残留进行定性及定量分析的要求。 相似文献

建立固相萃取–高效液相色谱法测定地表水中磺胺嘧啶、磺胺二甲嘧啶、磺胺氯哒嗪、醋磺胺甲恶唑4种磺胺类抗生素。样品采用HLB柱进行萃取富集,流动相为甲醇–水(体积比为20∶80),流量为0.5 m L/min,用SPD检测器检测,检测波长为270 nm;采用外标法定量。4种磺胺类抗生素质量浓度在4~160 ng/L范围内与色谱峰面积的线性关系良好,相关系数不低于0.998 2,方法检出限为1.0~1.7 ng/L,样品加标回收率为75.2%~97.4%,测定结果的相对标准偏差为2.8%~6.1%(n=7)。该方法操作简便,灵敏度高,可用于地表水中磺胺类抗生素的检测。 相似文献
