建立了以QuEChERS样品预处理方法、四级杆飞行时间串联质谱(Q-TOF MS)为检测手段,针对鱼肉中的四环素类、磺胺类、喹诺酮类、三苯甲烷类、雄激素、孕激素和糖皮质激素7类59种残留药物的快速筛查技术。通过正交实验得到优化条件为:Na2EDTA-Mcllvaine为缓冲溶液,乙腈为提取剂,净化剂无水MgSO4、十八烷基键合硅胶(C18)和N-丙基乙二胺(PSA)的用量分别为每毫升提取液加入25,12.5和6.25 mg。对Q-TOF质谱筛查出的阳性样品,再用LC-MS/MS对样品进行确证。本方法对59种药物的检出下限为0.5~5.3μg/kg。本方法快速、简单、可靠,可筛查的渔药种类覆盖面广,灵敏度满足水产品药物残留检测的要求,适用于鱼类水产品中残留药物的快速测定。  相似文献

采用生物素-亲和素法在微悬臂梁传感芯片上固定维埃克斯、沙林适配子,建立了压阻式微悬臂梁适配子传感器检测维埃克斯、沙林及动力学分析方法。传感器对维埃克斯检测的线性范围为2~60μg/L,线性回归方程为ΔUe=0.886C-1.039(n=5,R=0.984,p<0.001),检出限为2μg/L(S/N≥3);对沙林检测的线性范围为10~60μg/L,线性回归方程为ΔUe=0.716C-2.304(n=5,R=0.996,p<0.001),检出限为10μg/L(S/N≥3)。传感器具有良好的选择性和抗干扰能力,对毒剂类似物O-丁基甲基膦酰氯基本无响应。在此基础上,根据受体-配体结合特性与压阻式微悬臂梁传感器输出电压变化之间的关系,建立了传感器检测维埃克斯、沙林的反应动力学模型,根据拟合方程求出的传感器对不同浓度维埃克斯、沙林反应达到平衡时的响应电压(ΔUe)、响应时间(t0)均与实测值非常接近。  相似文献

A new thiosemicarbazone compound derived from 1-phenyl-3-methyl-4-(2-thenoyl) pyrazolone-5(HPMTP) and thiosemicarbazide has been synthesized and characterized by IR, HNMR, MS, elemental analysis, UV and single-crystal X-ray diffraction. The compound crystallizes in monoclinic system, Cc space group with a = 7.5925(8), b = 20.263(2), c = 11.4669(13) ,  = 107.825(8)°, V = 1679.5(3) 3, Z = 4, R = 0.0316 and wR = 0.0687. The results of antibacterial activity test against Escherichia coli and Bacillus subtilis indicate that the compound possesses the same antibacterial activity as the contrast(Norfloxacin). Theoretical study of the forming mechanism to the title compound at the RHF/6-31G(d) level shows that there are two steps. The distal amino group of thiosemicarbazide is added to the 4-carbonyl group of HPMTP which forms TM. Then a dehydration reaction occurs in TM and generates a stable product PC.  相似文献

Systematic study on the electronic/geometrical structures and the parity alternation effect of silicon-doped ternary cationic clusters HCnSi2+(n = 1 ~9) have been carried out at the coupled cluster level. The ground-state (G-S) isomers of the clusters have been defined. The C, chains of the G-S isomers display polyacetylene-like structures. The even-n cations are more stable than the odd-n ones. Such a trend of even/odd alternation has been elaborated based on concepts of the bond character, atomic charge, incremental binding energy, ionization potential, proton affinity and fragmentation energies of the systems. The findings accord with the relative intensities of HC,,Si2+ species recorded in the related mass spectrometric experiments.  相似文献

A new near-neutral pH near-infrared (NIR) fluorescent probe utilizing a fluorophore–receptor molecular framework that can modulate the fluorescence emission intensity through a fast photoinduced electron transfer process was developed. Our strategy was to choose tricarbocyanine (Cy), a NIR fluorescent dye with high extinction coefficients, as a fluorophore, and N-methylpiperazine (MP) as a receptor. The pH titration indicated that MP-Cy can monitor the minor physiological pH fluctuations with a pKa of ~7.10 near physiological pH, which is valuable for intracellular pH researches. The probe responds linearly and rapidly to minor pH fluctuations within the range of 3.05–7.10 and exhibits strong dependence on pH changes. As expected, the real-time imaging of cellular pH and the detection of pH in situ was achieved successfully in living HepG2 cells by this probe. It is shown that the probe effectively avoids the influence of autofluorescence and native cellular species in biological systems and meanwhile exhibits high sensitivity, good photostability, and excellent cell membrane permeability.  相似文献

The luminescent nanoparticles were prepared by encapsulating the [LnL4]? (Ln = Eu, Tb; L = BTFA, HFAA, TTFA, TFAA) complexes anion into the silicon framework. We firstly synthesized a series of novel siloxy-bearing lanthanide complex precursor, and then encapsulated them into the silica sphere by a modified Stöber process. As a result, four europium and two terbium tetrakis β-diketonate complexes functionalized silica sphere nanoparticles were obtained and characterized in detail using Fourier transform infrared spectra, X-ray diffraction, scanning electronic microscope, thermogravimetric analysis, luminescence excitation and emission spectroscopy, luminescence lifetime measurements, and diffuse reflectance UV–Vis spectroscopy. The result shows that these luminescent nanoparticles maintain the distinctive luminescence character of lanthanide chelate including broad excitation spectra, line-like emission spectra, high quantum efficiency, and long luminescent lifetime, which makes them great potential application in the synthesis of luminescent nanoparticle.  相似文献

Anisotropic polystyrene/poly(styrene-co-divinylbenzene) (PS/P(S-DVB)) protrusion particles with various morphologies such as eyeball-like, snowman-like, and raspberry-like were synthesized using a modified seeded polymerization method by dynamically controlling and stabilizing the phase separation. The effects of swelling agent, crosslinker, and monomer concentrations on the particle morphologies were studied. Using the PS/P(S-DVB) protrusion particles as templates, anisotropic silica (SiO2) hollow microspheres were fabricated facilely. The obtained anisotropic silica hollow spheres had a potential application in rapid waste removal and detoxification extraction with a very simple procedure.  相似文献

建立了同时检测蜂胶中16种磺胺及2种林可胺类药物残留的方法,以1.0 mol/L HCl溶液为提取液,经阳离子固相萃取小柱净化富集后,高效液相色谱-串联质谱仪分析。采用Agilent Polaris C18色谱柱(100 mm×2.0 mm,5μm),以甲醇和0.1%甲酸为流动相梯度洗脱,质谱模式为电喷雾正离子监测。该方法前处理简单,分别以13C6-磺胺二甲基嘧啶、13C6-磺胺甲噁唑为磺胺类药物内标、以D3-林可霉素为林可胺类药物内标进行定量,磺胺和林可胺类药物的线性范围均为1.0~50μg/L,相关系数均在0.99以上,方法定量限为10.0μg/kg,在10,20和40μg/kg 3个水平做添加回收,回收率范围为69.5%~114.6%,相对标准偏差小于10%。  相似文献

Presently, little is known of how the inter-organelle crosstalk impacts cancer cells owing to the lack of approaches that can manipulate inter-organelle communication in cancer cells. We found that a negatively charged, enzyme cleavable peptide (MitoFlag) enables the trafficking of histone protein H2B, a nuclear protein, to the mitochondria in cancer cells. MitoFlag interacts with the nuclear location sequence of H2B to block it from entering the nucleus. A protease on the mitochondria cleaves the Flag from the MitoFlag/H2B complex to form assemblies that retain H2B on the mitochondria and facilitate H2B entering the mitochondria. Adding NLS, replacing aspartic acid by glutamic acid residues, or changing the l - to d -aspartic acid residue on MitoFlag abolishes the trafficking of H2B into mitochondria of HeLa cells. As the first example of the enzyme-instructed self-assembly of a synthetic peptide for trafficking endogenous proteins, this work provides insights for understanding and manipulating inter-organelle communication in cells.  相似文献
