Homogeneous chitosan films of various thicknesses (10-65 nm) were deposited on thin gold films through spin coating. The resulting interfaces were characterized using surface plasmon resonance (SPR), AFM, profilometry and cyclic voltammetry. The strong chelating properties of chitosan films to Fe(3+) were investigated using cyclic voltammetry. Through SPR measurements, an affinity constant between chitosan and Fe(3+) of 9.49 x 10(5) M(-1) was determined with a detection limit as low as 250 ppb.  相似文献

This article reports chemical stability studies of a gold film electrode coated with thin silicon oxide (SiOx) layers using electrochemical, surface plasmon resonance (SPR) and atomic force microscopy (AFM) techniques. Silica films with different thicknesses (d = 6.4, 9.7, 14.5, and 18.5 nm) were deposited using a plasma-enhanced chemical vapor deposition technique (PECVD). For SiOx films with d >/= 18.5 nm, the electrochemical behavior is characteristic of a highly efficient barrier for a redox probe. SiOx films with thicknesses between 9.5 and 14.5 nm were found to be less efficient barriers for electron transfer. The Au/SiOx interface with 6.4 nm of SiOx, however, showed an enhanced steady-state current compared to that of the other films. The stability of this interface in solutions of different pH was investigated. Whereas a strongly basic solution led to a continuous dissolution of the SiOx interface, acidic treatment produced a more reticulated SiOx film and improved electrochemical behavior. The electrochemical results were corroborated by SPR measurements in real time and AFM studies.  相似文献

Simplification and evaluation of a gold-deposited SPR optical fiber sensor.   总被引:1,自引:0,他引:1  
The structure of the sensing element of a gold-deposited optical fiber sensor was simplified and quantitative analyses of various alcohols with the sensor showed improvement of the performance. The sensor uses surface plasmon resonance (SPR) at the interface of a sample solution and a thin (10 - 70 nm) gold film deposited on half of the exposed core of the optical fiber. The sensor with a film thickness of 45 nm can detect a small change of 5.6 x 10(-5) refractive index (RI) units in the refractivity. The response time is less than 0.5 min and the relative standard deviation for measurements is less than or equal to 1%. A straight line with a correlation coefficient of 0.9995 was obtained below 10%, v/v in the calibration curve for methanol solutions of benzyl alcohol. The minimum of the response curve due to the maximum excitation of SPR in the refractivity range from 1.33 to 1.44 RI units shifts to a lower refractivity as the film becomes thicker. The response curves of the sensors were calculated from SPR theoretical equations while considering of the distribution in the thickness of the deposited gold films. The improvement in the performance of the sensor is discussed.  相似文献

Gold-deposited optical fiber sensors with film thicknesses from 30 to 60 nm were prepared, and the responses to a wide range of a refractivity (1.33-1.54 refractive index (RI) units) were investigated both experimentally and theoretically. The response curve of the sensor has two minima in the refractivity range from 1.33 to 1.44 and at 1.462 RI units. The former minimum is due to surface plasmon resonance (SPR) in the thin gold film, and shifts to a lower refractivity as the film becomes thicker. The response curves of the sensors with film thicknesses of 45 and 60 nm agreed well with those calculated from SPR theoretical equations. Morphology observations of the surfaces of deposited gold films on glass by atomic force microscopy (AFM) and a variation in resistance of the films with various thicknesses show the structure of the gold films. We concluded that the thin deposited gold films have many defects, and that the core of the gold-deposited optical fiber leaks light through the defects to the sample solution with the same refractivity (1.462 RI units) as that of the core.  相似文献

Evaporation of ultrathin (1.3-10 nm nominal thickness) gold films onto quartz or mica leads to the formation of a layer of rather uniform gold islands on the transparent support. The morphology of ultrathin gold island films of various thicknesses was studied by using atomic force microscopy (AFM) and scanning electron microscopy (SEM) imaging. The surface plasmon (SP) absorption characteristic of such films is highly sensitive to the surrounding medium, with the plasmon band changing in intensity and wavelength upon binding of various molecules to the surface. The binding process can be monitored quantitatively by measuring the changes in the gold SP absorption, by using transmission UV/Vis spectroscopy. The method, termed transmission surface plasmon resonance (T-SPR) spectroscopy, is shown to be applicable to both chemically and physically adsorbed molecules, in liquid or gas phase, with measurements carried out either ex situ or in situ (real-time measurements) using a variety of molecular probes. Binding to a preformed molecular layer on the Au surface produces a similar response, suggesting the possible use of T-SPR for selective sensing. The sensitivity of T-SPR spectroscopy in detecting molecular binding to the gold depends strongly on the film preparation conditions, and may be comparable to that obtained in surface plasmon resonance (SPR) sensing.  相似文献

对淀积在玻璃衬底上厚度约60 nm的金银合金溅射薄膜进行硝酸腐蚀脱银处理, 得到纳米多孔金薄膜. 利用自建的波长检测型表面等离子体共振(SPR)传感装置研究了腐蚀时间对纳米多孔金薄膜SPR特性的影响, 结果发现纳米多孔金薄膜与水溶液接触后在400-900 nm光谱范围内不具有SPR效应, 而当薄膜置于空气中时会产生明显的传播等离子体共振吸收峰, 其共振波长随腐蚀时间增加逐渐红移. 纳米多孔金薄膜在空气气氛中的SPR效应使其能够用于原位监测气相分子在孔内的吸附, 还可对在液相中吸附的生化分子进行离位测试. 本文对L-谷胱甘肽、L-半胱氨酸、2-氨基乙硫醇三种含巯基的生化小分子在纳米多孔金薄膜内的吸附进行了离位分析, 结果表明与传统的致密金薄膜SPR芯片比较, 纳米多孔金薄膜对这些分子显示出更高的灵敏度和更低的检测下限, 这归功于多孔金的大比表面积使其能够吸附大量的生化小分子. 实验还对乙醇蒸气在纳米多孔金薄膜内的吸附进行了原位监测, 发现吸附平衡所用时间较长, 约为160 min.  相似文献

The use of Au/SiO(x) interfaces for the investigation of DNA hybridization using electrochemical impedance spectroscopy (EIS) and surface plasmon resonance (SPR) simultaneously is demonstrated. Standard glass chemistry was used to link single-stranded DNA (ss-DNA) on aldehyde-terminated Au/SiO(x) interfaces. The layer thickness and amount of grafted oligonucleotides (ODNs) were calculated from SPR on the basis of a multilayer system of glass/Ti/Au/SiO(x)/grafted molecule. Capacitance and resistance values of the modified interface before and after hybridization were calculated from EIS data using an equivalent circuit and allowed the affinity rate constant, K(A) = 4.07 x 10(5) M(-1), to be determined. The EIS results were comparable to those obtained by SPR hybridization kinetics recorded in parallel.  相似文献

We report a simple and rapid method for the deposition of amorphous silica onto a gold surface. The method is based on the ability of lysozyme to mediate the formation of silica nanoparticles. A monolayer of lysozyme is deposited via non-specific binding to gold. The lysozyme then mediates the self-assembled formation of a silica monolayer. The silica formation described herein occurs on a surface plasmon resonance (SPR) gold surface and is characterized by SPR spectroscopy. The silica layer significantly increases the surface area compared to the gold substrate and is directly compatible with a detection system. The maximum surface concentration of lysozyme was found to be a monolayer of 2.6 ng/mm(2) which allowed the deposition of a silica layer of a further 2 ng/mm(2). For additional surface functionalization, the silica was also demonstrated to be a suitable matrix for immobilization of biomolecules. The encapsulation of organophosphate hydrolase (OPH) was demonstrated as a model system. The silica forms at ambient conditions in a reaction that allows the encapsulation of enzymes directly during silica formation. OPH was successfully encapsulated within the silica particles and a detection limit for the substrate, paraoxon, using the surface-encapsulated enzyme was found to be 20 microM.  相似文献

Bilayered silver/gold films (gold deposited on top of the silver film) were used as substrates for electrochemical surface plasmon resonance spectroscopy (EC-SPR). EC-SPR responses of electrochemical deposition/stripping of copper and redox-induced conformation changes of cytochrome c immobilized onto self-assembled monolayers preformed at these substrates were measured. Influence of the Ag layer thickness and the double-layer capacitance on the EC-SPR behavior was investigated. The results demonstrated that the bilayered Ag/Au metal films produce a sharper SPR dip profile than pure Au films and retain the high chemical stability of Au films. Contrary to the result by the Fresnel calculation that predicts a greater fraction of Ag in the bilayered film should result in a greater signal-to-noise ratio, the EC-SPR sensitivity is dependent on both the Ag/Au thickness ratio and the chemical modification of the surface. Factors affecting the overall SPR sensitivity at the bilayered films, such as the film morphology, potential-induced excess surface charges, and the adsorbate layer were investigated. Forming a compact adsorbate layer at the bilayered film diminishes the effect of potential-induce excess surface charges on the SPR signal and improves the overall EC-SPR sensitivity. For the case of redox-induced conformation changes of cytochrome c, the SPR signal obtained at the bilayered silver/gold film is 2.7 times as high as that at a pure gold film.  相似文献

A novel method to prepare surface plasmon resonance(SPR) sensor chips based on grafted imprinted polymer is explored. Benzophenone photografting system is used to grow molecularly imprinted polymer(MIP) films from the modified surface of gold substrate.The surface morphology and thickness of MIP films were investigated by scanning electronic microscope(SEM).The adsorption properties of sensor chip were studied by SPR spectroscopy.The results demonstrate that nano-MIP films can be constructed on the surface of gold substrate with the good adsorption of template molecules.  相似文献

Specially synthesized amphiphilic resorcinol calixarene (resorcarene 2) molecules have been deposited as Langmuir-Blodgett (LB) films on a variety of substrates including hydrophobically treated glass slides, silicon and gold-coated glass slides. A value of 1.9 nm2 is obtained for the area per molecule from measurements of pressure/area isotherms of the floating layer. Optical absorption studies within the ultraviolet-visible frequency region have been performed on these molecules in both LB films and in solution of resorcarene 2 in chloroform, containing 10% ethanol. Molecular aggregation in the form of dimerization is believed to take place during film formation. Further analysis has been carried out for Langmuir-Blodgett films of resorcarene 2 by using Fourier transform infra-red spectroscopy, low-angle X-ray diffraction and surface plasmon resonance (SPR) techniques. The monolayer thickness of 1.6 nm found from SPR measurements is consistent with that from other experimental observations.  相似文献

A new simpler concept about the signal amplification of surface plasmon resonance (SPR) that is based on the utilization of mesoporous silica thin films is demonstrated. The stepwise construction of a novel kind of self-assembled organic/inorganic multilayers based on multivalent supramolecular interactions between guest-functionalized dendrimers and host-modified gold nanoparticles has been developed, yielding supramolecular layer-by-layer assembly. The deposition process was monitored by surface plasmon resonance spectroscopy. Further characterization of the multilayer films was performed by means of UV/vis absorption spectroscopy, which showed a linear increase in absorption with the number of bilayers. The growth of the gold nanoparticle plasmon absorption band corresponded to approximately a dense monolayer of gold nanoparticles per bilayer. Ellipsometry and atomic force microscopy (AFM) scratching experiments were used to measure the development of the film thickness with the number of bilayers, confirming linear growth and a thickness increase of approximately 2 nm/bilayer.  相似文献

Biotin-capped gold nanoparticles assembled on flat gold with volume fraction f are studied by surface plasmon resonance (SPR) spectroscopy and atomic force microscopy (AFM) in order to estimate the dielectric function of the gold nanoparticles based on the Maxwell-Garnett (MG) theory. The complex dielectric function (epsilon',epsilon') of the spherical nanoparticles at three representative wavelengths in the vis-near-IR region, i.e., lambda = 543, 632.8, and 1152 nm, is estimated for a surface homogeneously covered with nanoparticles in order to discuss the wavelength dependence of the dielectric function. The SPR response of a surface covered with particles in 2D aggregates is also analyzed. The experimental SPR curve of the particle aggregates deviates from the theoretical predictions, suggesting dipole interactions between particles.  相似文献

Through electrostatic layer-by-layer (LBL) assembly, negatively charged citrate-stabilized platinum nanoparticles (PtNPs) and positively charged [tetrakis(N-methylpyridyl)porphyrinato] cobalt were alternately deposited on a 4-aminobenzoic acid-modified glassy carbon electrode and also on indium tin oxide substrates, directly forming the three-dimensional nanostructured materials. Thus-prepared multilayer films were characterized by UV--visible spectroscopy, surface plasmon resonance (SPR) spectroscopy, atomic force microscopy (AFM), and cyclic voltammetry. Regular growth of the multilayer films is monitored by UV--visible spectroscopy and SPR spectroscopy. AFM provides the morphology of the multilayer films. The PtNPs containing multilayer films exhibit high electrocatalytic activity for the reduction of dioxygen with high stability. Rotating disk electrode voltammetry and rotating ring-disk electrode voltammetry demonstrate that the PtNP-containing multilayer films can catalyze an almost four-electron reduction of O(2) to H(2)O in an air-saturated 0.5 M H(2)SO(4) solution. Furthermore, the electrocatalytic activity of the films could be further tailored by simply choosing different cycles in the LBL process or more specifically the amount of the assembly components in the films. The high electrocatalytic activity and good stability for dioxygen reduction make the PtNP-containing multilayer films potential candidates for the efficient cathode material in fuel cells.  相似文献

Ultrathin gold films prepared by evaporation of sub-percolation layers (typically up to 10 nm nominal thickness) onto transparent substrates form arrays of well-defined metal islands. Such films display a characteristic surface plasmon (SP) absorption band, conveniently measured by transmission spectroscopy. The SP band intensity and position are sensitive to the film morphology (island shape and inter-island separation) and the effective dielectric constant of the surrounding medium. The latter has been exploited for chemical and biological sensing in the transmission localized surface plasmon resonance (T-LSPR) mode. A major concern in the development of T-LSPR sensors based on Au island films is instability, manifested as change in the SP absorbance following immersion in organic solvents and aqueous solutions. The latter may present a problem in the use of Au island-based transducers for biological sensing, usually carried out in aqueous media. Here, we describe a facile method for stabilizing Au island films while maintaining a high sensitivity of the SP absorbance to analyte binding. Stabilization is achieved by coating the Au islands with an ultrathin silica layer, ca. 1.5 nm thick, deposited by a sol-gel procedure on an intermediate mercaptosilane monolayer. The silica coating is prepared using a modified literature procedure, where a change in the reaction conditions from room temperature to 90 degrees C shortened the deposition time from days to hours. The system was characterized by UV-vis spectroscopy, ellipsometry, XPS, HRSEM, AFM, and cyclic voltammetry. The ultrathin silica coating stabilizes the optical properties of the Au island films toward immersion in water, phosphate buffer saline (PBS), and various organic solvents, thus providing proper conditions where the optical response is sensitive only to changes in the effective dielectric constant of the immediate environment. The silica layer is thin enough to afford high T-LSPR sensitivity, while the hydroxyl groups on its surface enable chemical modification for binding of receptor molecules. The use of silica-encapsulated Au island films as a stable and effective platform for T-LSPR sensing is demonstrated.  相似文献

The distance dependence of the localized surface plasmon (SP) extinction of discontinuous gold films is a crucial issue in the application of transmission surface plasmon resonance (T-SPR) spectroscopy to chemical and biological sensing. This derives from the usual sensing configuration, whereby an analyte binds to a selective receptor layer on the gold film at a certain distance from the metal surface. In the present work the distance sensitivity of T-SPR spectroscopy of 1.0-5.0 nm (nominal thickness) gold island films evaporated on silanized glass substrates is studied by using coordination-based self-assembled multilayers, offering thickness tuning in the range from approximately 1 to approximately 15 nm. The morphology, composition and optical properties of the Au/multilayer systems were studied at each step of multilayer construction. High-resolution scanning electron microscopy (HRSEM) showed no apparent change in the underlying Au islands, while atomic force microscopy (AFM) indicated flattening of the surface topography during multilayer construction. A regular growth mode of the organic layers was substantiated by X-ray photoelectron spectroscopy (XPS). Transmission UV-visible spectra showed an increase of the extinction and a red shift of the maximum of the SP band upon addition of organic layers, establishing the distance dependence of the Au SP absorbance. The distance sensitivity of T-SPR spectroscopy can be varied by using characteristic substrate parameters, that is, Au nominal thickness and annealing. In particular, effective sensitivity up to a distance of at least 15 nm is demonstrated with 5 nm annealed Au films. It is shown that intensity measurements, particularly in the plasmon intensity change (PIC) presentation, provide an alternative to the usually measured plasmon band position, offering good accuracy and the possibility of measuring at a single wavelength. The present distance sensitivity results provide the basis for further development of T-SPR transducers based on receptor-coated Au island films.  相似文献

In this work, we investigated the fabrication of surface plasmon resonance (SPR) nanosensor using gold nanoparticles (AuNPs) chemisorbed onto self assembled monolayer of 10-(3-amino phenoxy) decane-1-thiol on gold substrate. The fabrication process of SPR nanosensor was characterized using different techniques such as infrared reflection-absorption spectra (IRRAS), xX-ray photoelectron spectroscopy (XPS), and atomic force microscope (AFM). The fabricated SPR nanosensor was used for detection of Cu2+ in an aqueous solution using surface plasmon resonance refractometer. The results confirm the fabrication of new SPR nanosensor. The fabricated SPR nanosensor showed a good activity toward the detection of Cu2+. The detection of Cu2+ in an aqueous solution using the fabricated SPR nansensor was enhanced in the presence of gold nanoparticles.  相似文献

Temperature-sensitive hydrogel films were synthesized by electron beam irradiation of poly(vinyl methyl ether) (PVME) on silicon (Si/SiO(2)) substrates and gold (Au) coated glass slides. The temperature-dependent swelling behavior of the films in aqueous solution was characterized by in situ spectroscopic ellipsometry and a combination of surface plasmon resonance (SPR) and optical waveguide spectroscopy (OWS). The results of both techniques are compared. The suitability of both techniques for the characterization of the swelling behavior of thin hydrogel films is demonstrated. The volume swelling degree in the swollen state decreases with increasing radiation dose D. This is explained by the fact that the number of formed polymeric radicals, and hence cross-linking density, increases with D. Above the phase-transition temperature, the swelling degrees were independent of D, slightly above 1. The swelling/deswelling process was fully reversible and is mainly directed perpendicular to the substrate surface. The phase-transition temperature was determined to be T(cr) approximately 33 degrees C. However, T(cr) slightly decreases with increasing D and increasing film thickness d.  相似文献
