本文报道了在手性膦过渡金属配合物催化下,以对溴甲苯与6-甲基-5-庚烯-2-溴化镁(5)经不对称交叉偶联反应合成(R)-(-)-和(S)-(+_)-α-姜黄烯的方法,光学收率分别为44%e.e和50.3%e.e.(R)-(-)-α-姜黄烯经Birch还原得到(R)-(-)-β-姜黄烯.对溴苯甲醚经不对称交叉偶联、Birch还原等反应合成了(S)-(+)-γ-姜黄烯.  相似文献

在二氯甲烷中,化合物(p-cymene)Ru2(μ-Se2)(S2C2B10H10)2 (1)与1-乙炔基环己醇反应得到加成产物(p-cymene)-Ru2(μ-Se2)(S2C2B10H10)2 (R1C=CR2) [R1 =H,R2=(cyclo-C6H10)(OH)(2);R1=(cyclo-C6H10)(OH),R2=H(3)].化合物2和3在氯仿中加热回流可脱水分别生成4和5,二者在甲苯中进一步加热回流可实现相互转化.所有化合物中,炔烃碳碳三键选择性地加成在两个不同的(S2C2B10H10)2-配体的硫原子[S(2)和S(3)]上,从而使混合价双钌中心RuⅡ/RuⅣ(18e/16e)转变为单一价态的RuⅡ/RuⅡ (18e/18e),得到进一步稳定的配合物.所有化合物通过元素分析、质谱、核磁共振进行了表征,并解析了化合物2的X衍射单晶结构.  相似文献

以4-乙基苯胺1为原料,经Sandmeyer反应得5-乙基靛红2;2经水合肼还原得到5-乙基-2-吲哚酮3;N,N-二甲基甲酰胺与三氯氧磷先形成Vilsmeier-Haack试剂,再与化合物3反应,合成2-氯-5-乙基-3-乙酰吲哚4;以丙酮为溶剂,对化合物4烃基化,以78.5%-95.6%的收率得到N-取代-2-氯-5-乙基-3-乙酰吲哚5a-5e。其中化合物4、5a-5e均未见文献报道,它们的结构均通过红外光谱、核磁共振氢谱(碳谱),质谱等确认。  相似文献

以(R)/(S)-4-苄基-2-噁唑烷酮为手性助剂,采用不对称合成方法制备了18个具有光学活性的2-甲氧羰基-4-氟苯基-1,5-苯并硫氮杂卓类化合物9a~9i和14a~14i,经HPLC分析e.e.值较为理想;通过核磁共振谱、红外光谱和高分辨质谱表征了其结构,通过单晶X射线衍射法确定化合物9h的相对构型;用抑菌圈法测试了目标化合物对新生隐球菌的抑菌活性.研究结果表明,由S型手性助剂诱导不对称合成的杂卓对新生隐球菌的抑制作用普遍高于由R型手性助剂诱导合成的杂卓及外消旋体.测试了抑菌活性较好的化合物14a~14f的最小抑菌浓度(MIC)和最小杀菌浓度(MFC),发现其对新生隐球菌的MIC和MFC均低于抗真菌药物氟康唑.  相似文献

用 ( -) -4 R-苯基唑烷酮为手性诱导试剂 ,N-端保护的 N-乙酰氯与 1 ,5 -苯并硫氮杂反应合成了 5个具有光学活性的 1 ,5 -苯并硫氮杂 -β-内酰胺衍生物 ,并通过 1 H NMR,MS,IR,[α]2 0D 和元素分析对此类化合物进行了表征 ,用 X射线衍射法确定了产物 b的立体结构 .晶体 X射线衍射分析结果表明 ,化合物 b属单斜晶系 ,P2 1 2 1 2 1 空间群 ,晶胞参数 :Mr=5 5 3.0 5 ,a=1 .2 2 93( 2 ) nm,b=2 .6 0 2 6 ( 5 ) nm,c=1 .0 1 4 6 ( 2 ) nm,β=90°,V=3.2 4 6 1 nm3 ,Z=4 ,Dc=1 .31 2 g/ cm3 ,F ( 0 0 0 ) =1 34 4 ,R1 =0 .0 5 84 ,w R2 =0 .1 1 4 0 .该化合物分子中七元杂环为类椅式构象 ,四元环上连接的两个取代基在环的同侧  相似文献

用自制的(S,S)-Salen Co(Ⅲ)OAc催化剂水解动力学拆分外消旋环氧氯丙烷得到高光学纯的(S)-3-氯-1,2-丙二醇和较高光学纯的(R)-环氧氯丙烷。 以(S)-3氯-1,2-丙二醇为手性原料和4-(2-甲氧基乙基)苯酚缩合,再与氯化亚砜反应得环状亚硫酸酯,最后和异丙胺反应得(S)-美托洛尔,光学纯度大于99%。 另外以(R)-环氧氯丙烷为手性原料和4-(2-甲氧基乙基)苯酚反应,再与异丙胺作用得到(S)-美托洛尔,光学纯度大于92%。 (S)-美托洛尔的总收率为53.9%,结构经IR、1H NMR、13C NMR和MS测试技术确证。 该路线原料利用率高,拆分后的2种产物均能用于目标化合物的合成。  相似文献

手性元5-(R)-(1R,2S,5R)-孟氧基-3-溴-2(5H)-呋喃酮(3)与氮亲核试剂伯胺(4), 通过串联的不对称Michael加成/分子内亲核取代反应得到了具有两个新的手性中心的1R,5S-6-烷基-6-氮杂-2R-孟氧基-3-氧杂-4-氧代二环[3,1,0]己烷(5a5d), 产率41%~51%, e.e.≥98%. 后者经LiAlH4还原得到N-烷基-2,3-双(羟甲基)氮杂环丙烷(6a6d), 产率66%~91%. 化合物56通过元素分析, IR, 1H NMR, 13C NMR, MS以及X射线晶体分析, 测定了它们的化学结构及立体化学构型. 本文为N-烷基氮杂环丙烷类化合物的合成提供了一种有效途径.  相似文献

Synthesis of novel chiral compounds of purine and pyrimidine bases   总被引:2,自引:0,他引:2  
The physiologically active groups such as purine and pyrimidine bases are introduced to the asymmetric ynthesis. The optically pure compounds bearing purine and pyrimidine bases (5a—5e) were prepared via the asymetric Michael addition reaction of purines and pyrimidines as Michael donators with the chiral source 5-(R)-[(1R, 2S, 5R)-menthyloxy]-2(5H)-furanone (3a), which was prepared from the natural chiral auxiliary (-)-menthol. The synthetic method was studied in detail and the new compounds were identified on the basis of their analytical data and spectroscopic data, such as [α]_D~(20), IR, UV, ~1H NMR, ~(13)C NMR and MS. The absolute configuration of 5a was established by X-ray crystallography. The results provided an efficient synthetic route to chiral purines and pyrimidine analogues, and offered chiral sources for further research on the physiologically active compounds of chiral nucleotides.  相似文献

An asymmetric total synthesis of ent-(-)-roseophilin (1), the unnatural enantiomer of a novel naturally occurring antitumor antibiotic, is described. The approach enlists a room temperature heterocyclic azadiene inverse electron demand Diels-Alder reaction of dimethyl 1,2,4,5-tetrazine-3,6-dicarboxylate (7) with the optically active enol ether 6 bearing the C23 chiral center followed by a reductive ring contraction reaction for formation of an appropriately functionalized pyrrole ring in a key 1,2,4,5-tetrazine --> 1,2-diazine --> pyrrole reaction sequence. A Grubbs' ring closing metathesis reaction was utilized to close the unusual 13-membered macrocycle prior to a subsequent 5-exo-trig acyl radical-alkene cyclization that was used to introduce the fused cyclopentanone and complete the preparation of the tricylic ansa-bridged azafulvene core 32. Condensation of 32 with 33 under the modified conditions of Tius and Harrington followed by final deprotection provided (22S,23S)-1. Comparison of synthetic (22S,23S)-1 ([alpha](25)(D), CD) with natural 1 established that they were enantiomers and enabled the assignment of the absolute stereochemistry of the natural product as 22R,23R. Surprisingly, ent-(-)-1 was found to be 2-10-fold more potent than natural (+)-1 in cytotoxic assays, providing an unusually rewarding culmination to synthetic efforts that provided the unnatural enantiomer.  相似文献

A novel strategy for the synthesis of (R)- and (S)-alpha-methyl(alkyl)serine-containing peptides is presented. Using (S)-phenylalanine cyclohexylamide 6 as chiral auxiliary, the optically pure azlactones (R)- and (S)-2 were synthesized via a novel azlactone/oxazoline interconversion reaction (Figures 3 and 6). These azlactones constitute fully protected and activated synthetic equivalents of (R)- and (S)-alpha-methylserine and can be directly incorporated into peptides without further protective group manipulations. Like other alpha,alpha-dialkylated glycines, optically pure alpha-alkylserines can be used to stabilize beta-turn and alpha-helical conformations in short peptides.  相似文献

Optically active nano‐ and microparticles have constituted a significant category of advanced functional materials. However, constructing optically active particles derived from synthetic helical polymers still remains as a big challenge. In the present study, it is attempted to induce a racemic helical polymer (containing right‐ and left‐handed helices in equal amount) to prefer one predominant helicity in aqueous media by using emulsifier in the presence of chiral additive (emulsification process). Excitingly, the emulsification process promotes the racemic helical polymer to unify the helicity and directly provides optically active nanoparticles constructed by chirally helical polymer. A possible mechanism is proposed to explain the emulsification‐induced homohelicity effect. The present study establishes a novel strategy for preparing chirally helical polymer‐derived optically active nanoparticles based on racemic helical polymers.

Highly optically active 4-substituted-2(5H)-furanones 6a-6j were obtained in good yields with de≥98% by the tandem Michael addition/elimination reaction of chiral 3-bromo-2(SH)-furanone (4a), which was conveniently prepared starting from 2-furaldehyde under mild conditions. The products were identified on the basis of their satisfactory elemental analysis and spectroscopic data of IR, UV, ^1H NMR, ^13C NMR and mass spectra. The stereochemistry and absolute configuration of this type of compounds were established by the X-ray crystallographic study. The reaction provided a short and efficient synthesis of the interesting highly optically active 4-subsdtuted-2(5H)-furanones containing an active pyrimidine and a purine base group.  相似文献

设计并合成了2个新的手性源S-(一)-α-取代-2-羟基-5-甲基苄胺,从该手性源合成了4个光活性铜络合物,考察了它们对苯乙烯、1,1-二苯乙烯和2,4-二甲基-2,5-已二烯的不对称环丙烷化反应的光学诱导活性.结果表明,由S-(一)-1-(2-羟基-5-甲基苯基)-2-甲基丙胺与2-羟基萘甲醛缩合而得到的Schiff碱为配体的铜络合物的诱导活性最佳.  相似文献
