The present study is aimed at providing a new short-wavelength near-infrared (NIR) spectroscopic method for the nondestructive quantitative analysis of ciprofloxacin hydrochloride in powder via artificial neural networks (ANNs). For this purpose, the NIR spectra of 90 experimental powder samples in the range 700–1100 mm were analyzed. Four different pretreatment methods—first-derivative, second-derivative, standard normal variate (SNV), and multiplicative scatter correction (MSC)—were applied to three sets of the NIR spectra of the powder samples. Among all of the ANN models, the first-derivative model is found to be the best. The results presented here demonstrate that the short-wavelength NIR region is promising for the fast and reliable determination of the major components in pharmaceuticals. The degree of approximation as an evaluation criterion prevents the overfitting phenomenon occurring in ANNs. The text was submitted by the authors in English.  相似文献

A new method orthogonal projection to latent structures (O-PLS) combined with artificial neural networks is investigated for non-destructive determination of Ampicillin powder via near-infrared (NIR) spectroscopy. The modern NIR spectroscopy analysis technique is efficient, simple and non-destructive, which has been used in chemical analysis in diverse fields. Be a preprocessing method, O-PLS provides a way to remove systematic variation from an input data set X not correlated to the response set Y, and does not disturb the correlation between X and Y. In this paper, O-PLS pretreated spectral data was applied to establish the ANN model of Ampicillin powder, in this model, the concentration of Ampicillin as the active component was determined. The degree of approximation was employed as the selective criterion of the optimum network parameters. In order to compare the OPLS-ANN model, the calibration models that using first-derivative and second-derivative preprocessing spectra were also designed. Experimental results showed that the OPLS-ANN model was the best.  相似文献

A method for quantitative analysis of phenoxymethylpenicillin potassium powder on the basis of near-infrared (NIR) spectroscopy is investigated by using orthogonal projection to latent structures (O-PLS) combined with artificial neural network (ANN). Being a preprocessing method, O-PLS can remove systematic orthogonal variation from a given data set X without disturbing the correlation between X and the response set y. In this paper, O-PLS method was applied to preprocess the original spectral data of phenoxymethylpenicillin potassium powder, and the filtered data was used to establish the ANN model. In this model, the concentration of phenoxymethylpenicillin potassium as the active component was determined. The degree of approximation was employed as the selective criterion of the optimum network parameters. In order to compare with O-PLS-ANN model, the calibration models that use the original spectra and different preprocessing methods (first-derivative, second-derivative, standard normal variate (SNV) and multiplicative scatter correction (MSC)) of the spectra were also designed. Experimental results show that O-PLS-ANN model is the best.  相似文献

The application of the second most popular artificial neural networks (ANNs), namely, the radial basis function (RBF) networks, has been developed for quantitative analysis of drugs during the last decade. In this paper, the two components (aspirin and phenacetin) were simultaneously determined in compound aspirin tablets by using near-infrared (NIR) spectroscopy and RBF networks. The total database was randomly divided into a training set (50) and a testing set (17). Different preprocessing methods (standard normal variate (SNV), multiplicative scatter correction (MSC), first-derivative and second-derivative) were applied to two sets of NIR spectra of compound aspirin tablets with different concentrations of two active components and compared each other. After that, the performance of RBF learning algorithm adopted the nearest neighbor clustering algorithm (NNCA) and the criterion for selection used a cross-validation technique. Results show that using RBF networks to quantificationally analyze tablets is reliable, and the best RBF model was obtained by first-derivative spectra.  相似文献

ABSTRACT

This paper demonstrates the usefulness of near-infrared (NIR) spectra and artificial neural network (ANN) in nondestructive quantitative analysis of pharmaceuticals. Real data sets from near-infrared reflectance spectra of analgini powder pharmaceutical were used to build up an artificial neural network to predict unknown samples. The parameters affecting the network were discussed. A new network evaluation criterion, the degree of approximation, was employed. The overfitting was discussed. Owing to the good nonlinear multivariate calibration nature of ANN, the predicted result was reliable and precise. The relative error of unknown samples was less than 2.5%  相似文献

The pharmaceutical industry faces increasing regulatory pressure to optimize quality control. Content uniformity is a basic release test for solid dosage forms. To accelerate test throughput and comply with the Food and Drug Administration's process analytical technology initiative, attention is increasingly turning to nondestructive spectroscopic techniques, notably near-infrared (NIR) spectroscopy (NIRS). However, validation of NIRS using requisite linearity and standard error of prediction (SEP) criteria remains a challenge. This study applied wavelet transformation of the NIR spectra of a commercial tablet to build a model using conventional partial least squares (PLS) regression and an artificial neural network (ANN). Wavelet coefficients in the PLS and ANN models reduced SEP by up to 60% compared to PLS models using mathematical spectra pretreatment. ANN modeling yielded high-linearity calibration and a correlation coefficient exceeding 0.996.  相似文献

A new acetolactate synthase (ALS)-inhibiting herbicide, propyl 4-(2-(4,6-dimethoxypyrimidin-2-yloxy)benzylamino)benzoate (ZJ0273), was applied to oilseed rape (Brassica napus L.) leaves in different leaf positions. Visible/near-infrared (Vis/NIR) spectroscopy was investigated for fast and non-destructive determination of ALS activity and protein content in rapeseed leaves. Partial least squares (PLS) analysis was the calibration method with comparison of different spectral preprocessing by Savitzky-Golay (SG) smoothing, standard normal variate (SNV), first and second derivative. The best PLS models were obtained by first-derivative spectra for ALS, whereas original spectra for soluble, non-soluble and total protein contents. Simultaneously, certain latent variables (LVs) were used as the inputs of back-propagation neural network (BPNN) and least squares-support vector machine (LS-SVM) models. All LS-SVM models outperformed PLS models and BPNN models. The correlation coefficient (r), root mean square error of prediction (RMSEP) and bias in validation set by LS-SVM were 0.998, 0.715 and 0.079 for ALS, 0.999, 33.084 and 1.178 for soluble protein, 0.997, 42.773 and 6.244 for non-soluble protein, 0.999, 59.562 and 7.437 for total protein, respectively. The results indicated that Vis/NIR spectroscopy combined with LS-SVM could be successfully applied for the determination of ALS activity and protein content of rapeseed leaves. The results would be helpful for further on field analysis of using Vis/NIR spectroscopy to monitor the growing status and physiological properties of oilseed rape.  相似文献

In the construction of a neural network, most attentions have been paid to the selection of the architecture, the selection of the learning parameters and the network validation while the selection of input variables shared little. This study focused on the selection of input variables by various data pre-treatment for constructing ANN models. The results showed that the validation results differed from each other when different data-pretreatment methods combined with near-infrared spectroscopy (NIRS) to build a model using artificial neural network (ANN) for quality control of paracetamol in coldrex. And wavelet coefficients after orthogonal signal correction (OSC) in the ANN models reduced RMSEP by up to 77% compared to ANN models using derivatives combined with PCA pretreatment. The selection of input variables has potent to improve the calibration ability of ANN, and the model can be used for pressure reduction of quality control in the pharmaceutical industry.  相似文献

Near-infrared (NIR) spectroscopy was used in simultaneous, non-destructive analysis of antipyriine and caffeine citrate tablets. Principal component artificial neural networks (PC-ANNs) were used to construct models for the analytes, using the testing set for external validation. Four pretreated spectra, namely, first-derivative, second-derivative, standard normal variate (SNV) and multiplicative scatter correction (MSC) spectra led to simplified and more robust models than conventional spectra. In PC-ANNs models, the spectra data were analyzed by principal component analysis (PCA) firstly. Then the scores of the principal compounds (PCs) were chosen as input nodes for input layer instead of the spectra data. The artificial neural networks (ANNs) models using the spectra data as input nodes were also established, which were compared with the PC-ANNs models. The result shows the SNV model of PC-ANNs multivariate calibration has the lowest training error and predicting error. The concept of the degree of approximation was introduced and performed as the selective criterion of the optimum network parameters.  相似文献

Temperature-constrained cascade correlation networks (TCCCNs) were used to identify powdered rhubarbs based on their near-infrared spectra. Different network configurations that used multiple network models with single output (Uni-TCCCN) and single networks with multiple outputs (Multi-TCCCN) were compared. Comparative studies were made by using Latin-partitions and leave-one-out cross-validation methods. Results showed that multiple networks with single output predicted generally better than single network with multiple outputs. Better results with TCCCN models were obtained compared with conventional back propagation neural networks (BPNNs). The effects of parameters on correct identification and parameter optimizations were discussed in detail. With optimized neural network training parameters, NIR spectra from powdered rhubarb samples were classified by a TCCCN model with 100% accuracy.  相似文献

Wafers with varying concentrations of diphenhydramine hydrochloride (DPH-HCl) as active pharmaceutical ingredient (API) were prepared and their near infrared (NIR) and Raman spectra recorded. The purpose of this study was to compare the suitability of these two vibrational spectroscopic techniques for the quantification of DPH-HCl in pharmaceutical wafers. Partial least squares (PLS1) calibration models with different data pretreatments were tested. Both NIR and Raman spectroscopy proved to be suitable to predict DPH-HCl contents at lower concentration ranges. At higher concentrations, interference by crystallization processes was observed. For investigating the general applicability of the quantification methods, two commercially available products were examined.  相似文献

It is important to monitor quality of tobacco during the production of cigarette. Therefore, in order to scientifically control the tobacco raw material and guarantee the cigarette quality, fast and accurate determination routine chemical of constituents of tobacco, including the total sugar, reducing sugar, Nicotine, the total nitrogen and so on, is needed. In this study, 50 samples of tobacco from different cultivation areas were surveyed by near-infrared (NIR) spectroscopy, and the spectral differences provided enough quantitative analysis information for the tobacco. Partial least squares regression (PLSR), artificial neural network (ANN), and support vector machine (SVM), were applied. The quantitative analysis models of 50 tobacco samples were studied comparatively in this experiment using PLSR, ANN, radial basis function (RBF) SVM regression, and the parameters of the models were also discussed. The spectrum variables of 50 samples had been compressed through the wavelet transformation technology before the models were established. The best experimental results were obtained using the (RBF) SVM regression with gamma=1.5, 1.3, 0.9, and 0.1, separately corresponds to total sugar, reducing sugar, Nicotine, and total nitrogen, respectively. Finally, compared with the back propagation (BP-ANN) and PLSR approach, SVM algorithm showed its excellent generalization for quantitative analysis results, while the number of samples for establishing the model is smaller. The overall results show that NIR spectroscopy combined with SVM can be efficiently utilized for rapid and accurate analysis of routine chemical compositions in tobacco. Simultaneously, the research can serve as the technical support and the foundation of quantitative analysis of other NIR applications.  相似文献

The near-infrared(NIR) diffuse reflectance spectroscopy was used to study the content of Berberine in the processed Coptis. The allocated proportions of Coptis to ginger, yellow liquor or Evodia rutaecarpa changed according to the results of orthogonal design as well as the temperature. For as withdrawing the full and effective information from the spectral data as possible, the spectral data was preprocessed through first derivative and multiplicative scatter correetion(MSC) according to the optimization results of different preprocessing methods. Firstly, the model was established by partial least squares(PLS); the coefficient of determination(R2) of the prediction was 0.839, the root mean squared error of prediction(RMSEP) was 0.1422, and the mean relative error(RME) was 0.0276. Secondly, for reducing the dimension and removing noise, the spectral variables were highly effectively compressed via the wavelet transformation(WT) technology and the Haar wavelet was selected to decompose the spectral signals. After the wavelet coefficients from WT were input into the artificial neural network(ANN) instead of the spectra signal, the quantitative analysis model of Berberine in processed Coptis was established. The R^2 of the model was 0.9153, the RMSEP was 0.0444, and the RME was 0.0091. The values of appraisal index, namely R^2, RMSECV, and RME, indicate that the generalization ability and prediction precision of ANN are superior to those of PLS. The overall results show that NIR spectroscopy combined with ANN can be efficiently utilized for the rapid and accurate analysis of routine chemical compositions in Coptis. Accordingly, the result can provide technical support for the further analysis of Berberine and other components in processed Coptis. Simultaneously, the research can also offer the foundation of quantitative analysis of other NIR application.  相似文献

Near-infrared (NIR) spectroscopy, in combination with chemometrics, enables nondestructive analysis of solid samples without time-consuming sample preparation methods. A new method for the nondestructive determination of compound amoxicillin powder drug via NIR spectroscopy combined with an improved neural network model based on principal component analysis (PCA) and radial basis function (RBF) neural networks is investigated. The PCA technique is applied to extraction relevant features from lots of spectra data in order to reduce the input variables of the RBF neural networks. Various optimum principal component analysis-radial basis function (PCA-RBF) network models based on conventional spectra and preprocessing spectra (standard normal variate (SNV) and multiplicative scatter correction (MSC)) have been established and compared. Principal component regression (PCR) and partial least squares (PLS) multivariate calibrations are also used, which are compared with PCA-RBF neural networks. Experiment results show that the proposed PCA-RBF method is more efficient than PCR and PLS multivariate calibrations. And the PCA-RBF approach with SNV preprocessing spectra is found to provide the best performance.  相似文献

The combination of infrared (MIR) and near-infrared (NIR) spectroscopy has been employed for the determination of important quality parameters of beers, such as original and real extract and alcohol content. A population of 43 samples obtained from the Spanish market and including different types of beer, was evaluated. For each technique, spectra were obtained in triplicate. In the case of NIR a 1 mm pathlength quartz flow cell was used, whereas attenuated total reflectance measurements were used in MIR. Cluster hierarchical analysis was employed to select calibration and validation data sets. The calibration set was composed of 15 samples, thus leaving 28 for validation. A critical evaluation of the prediction capability of multivariate methods established from the combination of NIR and MIR spectra was made. Partial least squares (PLS) and artificial neural networks (ANN) were evaluated for the treatment of data obtained in each individual technique and the combination of both. Different parameters of each methodology were optimized. A slightly better predictive performance was obtained for NIR-MIR combined spectra, and in all the cases ANN performs better than PLS, which may be interpreted from the existence of some non-linearity in the data. The root-mean-sqare-error of prediction (RMSEP) values obtained for the combined NIR-MIR spectra for the determination of real extract, original extract and ethanol were 0.076% w/w, 0.14% w/w and 0.091% v/v.  相似文献

The most common fraudulent practice in the vinegar industry is the addition of alcohol of different origins to the base wine used to produce wine vinegar with the objective of reducing manufacturing costs. The mixture is then sold commercially as genuine wine vinegar, thus constituting a fraud to consumers and an unfair practice with respect to the rest of the vinegar sector. A method based on near-infrared spectroscopy has been developed to discriminate between white wine vinegar and alcohol or molasses vinegar. Orthogonal signal correction (OSC) was applied to a set of 96 vinegar NIR spectra from both original and artificial blends made in the laboratory, to remove information unrelated to a specific response. The specific response used to correct the spectra was the extent of adulteration of the vinegar samples. Both raw and corrected NIR spectra were used to develop separate classification models using the potential functions method as a class-modeling technique. The previous models were compared to evaluate the suitability of near-infrared spectroscopy as a rapid method for discrimination between vinegar origin. The transformation of vinegar NIR spectra by means of an orthogonal signal-correction method resulted in notable improvement of the specificity of the constructed classification models. The same orthogonal correction approach was also used to perform a calibration model able to detect and quantify the amount of exogenous alcohol added to the commercial product. This regression model can be used to quantify the extent of adulteration of new vinegar samples.  相似文献

The present study aimed at providing a new method in sight into short-wavelength near-infrared (NIR) spectroscopy of in pharmaceutical quantitative analysis. To do that, 124 experimental samples of metronidazole powder were analyzed using artificial neural networks (ANNs) in the 780-1100 nm region of short-wavelength NIR spectra. In this paper, metronidazole was as active component and other two components (magnesium stearate and starch) were as excipients. Different preprocessing spectral data (first-derivative, second-derivative, standard normal variate (SNV) and multiplicative scatter correction (MSC)) were applied to establish the ANNs models of metronidazole powder. The degree of approximation, a new evaluation criterion of the networks was employed to prove the accuracy of the predicted results. The results presented here demonstrate that the short-wavelength NIR region is promising for the fast and reliable determination of major component in pharmaceutical analysis.  相似文献
