Herein, we report the use of γ-valerolactone (GVL) and N-formylmorpholine (NFM) as DMF substitutes in polystyrene based SPPS. The solubility of selected amino acids and coupling reagents were studied in GVL and NFM, followed by their use in the successful synthesis of Aib-enkephalin pentapeptide (H-Tyr-Aib-Aib-Phe-Leu-NH2) and Aib-ACP decapeptide (H-Val-Gln-Aib-Aib-Ile-Asp-Tyr-Ile-Asn-Gly-NH2).  相似文献

A solution phase method was adopted for the synthesis of proline-containing cyclic pentapeptide 2 and total synthesis of naturally occurring cyclic heptapeptide Reniochalistatin B 3. For the synthesis of 3, both divergent and convergent strategies were used to improve the overall yield from 12 to 25%. Different N and C terminal modified linear analogs and congeners of 2 and 3 were synthesized. Both cyclic peptides 2 and 3 and their linear analogs/congeners were evaluated for anti-cancer activity against HeLa cell line, among which pentapeptide 2?h and hexapeptide 3n with N-terminal protected hexafluoroisopropyl carbamates (HFIPC) interestingly showed higher cytotoxicity with an IC50 of 2.73 and 4.3?µM, respectively compared to their Boc-protected analogs 2a (IC50 20?µM) and 3c (IC50 38.51?µM) and cyclic peptides 2 (>100?µM) and 3 (47?µM). These results were further validated by biological experiments such as colony formation and wound healing assays.  相似文献

Crown ethers1 and4 of the 18-crown-6 type containing two glucose units have been oxidised by KMnO4 into mono- and dicarboxylic acid derivatives (5 and11), and derivatives with different lipophilicities of the above crown ethers, namely the acetyl, benzyl and butyl derivatives (8–10, 13, 14) and methyl esters (6 and12) have been synthesizedThe association constants (K a) with Li, Na, K and NH4 cations measured in CHCl3 indicate that complexing ability increases on introduction of carboxy groups, and selectivity changes in favour of the Na cation. These compounds were able to transport alkyl-ammonium salts through a CHCl3 liquid membrane, displaying, however, no chiral recognition ability.  相似文献

A new method for the preparation of peptide thioester by the post-solid phase peptide synthesis (SPPS) approach was developed. A series of N-alkyl cysteine derivatives were prepared and used as the C-terminus residue of the peptides prepared by the Fmoc SPPS. The synthetic peptides released from resin by TFA were readily converted to the peptide thioester in aqueous 3-mercaptopropionic acid (MPA) without significant side reactions.  相似文献

General methods for the preparation of a variety of pyridone and oxypyridine derivatives are described. 2‐,3‐,4‐Hydroxy pyridine and 2‐pyridinemethanol were alkylated with ethylene‐, propylene‐, and stryrene‐oxide and epichlorohydrin in the presence of different Lewis acids as a catalyst. The best yield of 3a was achieved in the presence of CdI2/BF3 · OEt2. The corresponding pyridone derivatives (3ad, 7ad) were obtained from the reaction of 2‐and 4‐hydroxypyridine with oxiranes in good yields, whereas oxypyridine derivatives (5ad, 9a,b) were obtained from reactions of 3‐hydroxypyridine and 2‐pyridinemethanol with oxiranes. Chlorohydrines (3d, 5d, 7d) were easily converted to corresponding epoxy derivatives (10, 11, 12) in basic medium; then amino alcohols (1317) were obtained from the reaction of these epoxy derivatives with amines.  相似文献

Abstract

Lactoquinomycin C (1) and Lactoquinomycin D (2), two new medermycin derivatives together with six known compounds (38) were isolated from the rice solid fermentation of the marine-derived Streptomyces sp. SS17A. The intriguing structural features of the Lactoquinomycin D (2) which contains 5,14-epoxidation is relatively rare in medermycin derivatives. Compounds 1 and 2 exhibited no cytotoxicity against PC-3 and HCT-116 cancer cell lines, whereas compound 3 showed stronger cytotoxicity with IC50 values of 0.02 and 0.04?μM, respectively. A structure-activity relationship was observed for the cytotoxicity of the medermycin derivatives with a γ-lactone unit is required for significant cytotoxicity based on compounds 13.  相似文献

目的研究配体结构对NAMI-A衍生物水解机理、电化学性质的影响。方法制备了trans-[RuCl4(DMSO)(3-MePy)][(3-MePy)H](3-MePy=3-甲基吡啶,化合物1)和trans-[RuCl4(DMSO)(4-MePy)][(4-MePy)H](4-MePy=4-甲基吡啶,化合物2)。用UV、NMR、CV法研究化合物1、化合物2的水解机理-动力学、溶液稳定性及电化学性质。结果化合物1和化合物2与NAMI-A相似,在pH7.40的缓冲液中发生脱氯水解反应(Ⅰ氯水解及Ⅱ氯水解)(分步反应);在pH 5.00缓冲液中DMSO(二甲亚砜)及少量吡啶水解。测定各水解反应表观速率常数及半衰期、溶液稳定性及氧化还原电位。结论化合物1、化合物2的Ⅰ氯、Ⅱ氯及DMSO水解反应机理与NAMI-A相似,而且各水解速率与NAMI-A相差不大,即用甲基吡啶取代咪唑环,对NAMI-A衍生物的Ⅰ氯、Ⅱ氯及DMSO水解反应速率影响较小。化合物在酸性溶液中的稳定性明显高于中性溶液。  相似文献

Five laterally methyl-substituted pyridine-based derivatives of the title compounds (I 8I 16), with molecular formula 4-CnH2n+1O-C6H4COOC6H3(3-CH3)-N=N-C5H4N were prepared and their molecular formulae elucidated via elemental analyses, infrared, nuclear magnetic resonance and mass spectra. The number of carbon atoms in the alkoxy chain (n) varies between 8, 10, 12, 14, and 16 carbons. The newly prepared pyridine-based derivatives were investigated for their mesophase behaviour by differential scanning calorimetry and polarised optical microscopy; most of them were found to possess monotropic smectic C (SmC) mesophase. Two groups (A and B) of the 1:1 hydrogen-bonded associates, formed between each of the derivatives I 8– I 16 and two types of 4-substituted benzoic acids (II), were prepared and similarly characterised to investigate the effect of lateral methyl substitution on the central phenylene ring, as well as terminal polar substituents and alkoxy-chain length on the stability of the mesophases induced by intermolecular hydrogen bonding. In Group A complexes, mesomorphic 4-alkoxy benzoic acids, that carry the terminal n-alkoxy group of varying chain length, were used. The other series of complexes (Group B) is composed from the same pyridine-based derivatives and each of the non-mesomorphic 4-substituted benzoic acids that carries small compact polar groups, varying between CH3O, CH3, H, Cl, Br, and CN. All complexes prepared were investigated for their mesophase behaviour by differential scanning calorimetry and polarised optical microscopy and found to be purely smectogenic, possessing SmC as the only mesophase observed. The formation of the hydrogen-bonded complexes was confirmed by constructing their binary phase diagrams, which cover the whole range of concentration of the two complements.  相似文献

Series of new thiosemicarbazones was prepared and molecular studied as inhibitors of HCV 4WTG polymerase. Thus, the thiosemicarbazone derivatives (3a–k) were synthesized by two different ways, from reacting thiosemicarbazides with aldehydes and by one-pot three component reaction using ZnCl2/SiO2 as a catalyst under solvent free conditions. Molecular docking analysis of the synthesized products predicted that the thiosemicarbazone derivatives 3c, 3g, and 3k were the most promised as a highly inhibitors for HCV 4WTG polymerase in comparison with Sofosbuvir drug.  相似文献

Cyclic cyano derivatives 3 were obtained by the reaction of Me3SiCN / ZnI2 with the cyclic ketones 2 which gave the trimethylsilyloxy nitriles 4. C60与亚氨基二乙酸甲酯[NH(CH2COOMe)2]的光化学反应制得2,5-双(甲氧羰基)富勒烯吡咯烷(1), 产率为52% (基于已反应的C60). C60吡咯烷衍生物1与氯乙酸甲酯(ClCH2COOCH3)的N-烃基化反应, 在微波辐射、无溶剂及相转移条件下, 得到2,5-双(甲氧羰基)-N-(甲氧羰基)甲基富勒烯吡咯烷(2), 产率47%(基于C60吡咯烷衍生物1). C60吡咯烷衍生物12用NaH, CH3OH水解后, 经盐酸酸化得相应的二羧酸衍生物3和三羧酸衍生物4, 产率分别为65%和53% (基于C60吡咯烷衍生物12). C60吡咯烷衍生物14的结构由1H NMR, 13C NMR, IR, FAB-MS和元素分析证实. 用电导法测定了C60吡咯烷二羧酸3和三羧酸4钠盐的临界聚集浓度(CAC), 分别为3.58×10-4 mol/L (3)和3.33×10-4 mol/L (4). 这一结果被C60吡咯烷衍生物34, 在临界聚集浓度(CAC)附近的UV-Vis光谱的特征变化所支持. 透射电子显微镜(TEM)和静态光散射(SLS)等方法也被用于检测C60吡咯烷衍生物34在临界聚集浓度(CAC)时的聚集行为, 结果显示, C60吡咯烷衍生物3在缓冲溶液中(0.001 mol/L NaCO3-NaHCO3), 其聚集体粒径的大小(Rg≈21 nm)不同于C60吡咯烷衍生物4 (Rg≈23 nm). C60吡咯烷二羧酸3钠盐的临界聚集浓度(CAC)比C60吡咯烷三羧酸4钠盐的临界聚集浓度(CAC)大, 聚集体粒径大小的不同, 表明C60单加成衍生物加成基团中羧基(COOH)数目的多少对其聚集行为的影响. 用化学发光法分别检测了C60吡咯烷二羧酸3和C60吡咯烷三羧酸4在缓冲溶液(0.05 mol/L NaCO3-NaHCO3)中对邻苯三酚自氧化产生的超氧阴离子(O2·)的清除活性. C60衍生物34对 O2· 的清除呈现有明显的剂量效应, 但当超过一定浓度时(3: ~3.50×10-4 mol/L, 4: ~3.25×10-4 mol/L), 清除效率出现转折, 并下降. 这一现象与电导率测定时出现的CAC现象相一致, 进一步证实了C60吡咯烷二羧酸3和C60吡咯烷三羧酸4在较高浓度的水溶液中有聚集的倾向, 也说明了C60吡咯烷二羧酸3和C60吡咯烷三羧酸4的聚集会影响其清除超氧阴离子(O2·)的活性.  相似文献

By introducing three pyridyl-1H-tetrazole derivatives to Keggin–Ag-(1,10-phenanthroline) system, five new polyoxometalate-based inorganic–organic hybrid compounds with different dimensionalities and structures, [Ag3(4-ptz)2Na(phen)(HPMo12O40)]·2H2O (1), [Ag4(3-ptz)2(phen)2(HPMo12O40)] (2), [Ag4(3-ptz)2(phen)2(HVW12O40)] (3), [Ag3(3-bptzb)(phen)2(PMo12O40)] (4) and [Ag3(3-bptzb)(phen)2(VW12O40)] (5) (phen = 1,10-phenanthroline, x-ptzH = 5-(x-pyridyl)-1H-tetrazole (x = 3, 4), 3-bptzb = 1,4-bis(5-(3-pyridyl)-tetrazolyl)butane), have been synthesized under hydrothermal conditions. Compound 1 is a 1-D belt architecture constructed from an Ag-4-ptz metal–organic chain and two Ag–PMo12O40–Na–phen inorganic–organic chains. When 3-ptzH was introduced as secondary ligand in 2 and 3 instead of 4-ptzH, two isostructural architectures were obtained, which contain a 1-D zigzag Ag-3-ptz chain with phen ligands hanging on both sides of the chain. The Keggin anions connect adjacent Ag-3-ptz chains to construct 2-D networks. When the flexible 3-bptzb was selected, isostructural 4 and 5 with 2-D networks were obtained, which were constructed from binuclear subunits and Keggin–Ag inorganic chains. The influence of three pyridyl-1H-tetrazole derivatives as secondary ligands on the dimensionalities and architectures of 15 was discussed. The electrochemical and photocatalytic properties of the compounds were investigated.  相似文献

An efficient one-pot synthesis of 6-amino-4-(2-chloroquinolin-3-yl)-3-methyl-2, 4-dihydro-pyrano[2,3-c]pyrazole-5-carbonitrile derivatives (4a–f)(5a–f) by three component reactions of 2-chloroquinolin-3-carbaldehyde derivatives, malanonitrile, and 3-methyl pyrazolin-5-one derivatives catalyzed by L-proline in ethanol medium under mild conditions is established. The synthesized compounds were evaluated for antimalarial activity and the LC50/LC90 values were described. Compounds 4d, 5d, and 5f exhibits good antimalarial activity when compared to other pyrano[2,3-c]pyrazole scaffolds.  相似文献

Three new thiacalix[4]arene derivatives, 5,11,17,23-tetra-tert-butyl-25,27-di(2-hydroxyethoxy)-26,28-dihydroxythiacalix-{}[4]arene (2), 5,11,17,23-tetra-tert-25, 26,27,28-tetrakis[(methylcarboxyl)methoxy]thiacalix[4]arene (3),5,11,17,23-tetra-tert-butyl-25,26,27,28-tetrakis(2-hydroxy-1-propanoxy)thiacalix[4]arene (4), were synthesized for the first time. The coordination properties of thiacalix[4]arene(1) and its derivatives (2 and 4) were investigated by detecting the interactions betweenthese compounds and two palladium complexes, cis-[Pd(en)(H2O)2]2+ and cis-[Pd(dtco-3-OH)(H2O)2]2+, by means of electrospray ionization mass spectrometry (ESI-MS) technique.  相似文献

Two new Pb(II) coordination polymers, [Pb(3-pdip)(L1)]?·?0.5H2O (1) and [Pb(4-pdip)(L2)] (2) [L1?=?heptane diacid, L2?=?hexane diacid, 3-pdip?=?2?-(3-pyridyl)imidazo[4,5-f]1,10-phenanthroline, 4-pdip?=?2?-(4-pyridyl)imidazo[4,5-f]1,10-phenanthroline], have been obtained from hydrothermal reactions of Pb(II) nitrate with two flexible fatty carboxylic acids and two phenanthroline derivatives. Single-crystal X-ray diffraction analysis reveals that 1 is a 1-D staircase-like double chain coordination polymer extended by binuclear [Pb2N4O8] subunits and pairs of L1 ligands. Compound 2 exhibits a 2-D (4,4) network based on bi-metallic [Pb2N6O8] units and (L2)2/(4-pdip)2 double linkers. Furthermore, adjacent chains of 1 and layers of 2 are extended into 3-D supramolecular networks by π–π stacking interactions between aromatic rings of phenanthroline groups. The organic carboxylic acids with different lengths show great influence on the shapes of Pb(II)-carboxylate double chains. Phenanthroline derivatives possessing different sites of N-donors (3-pyridyl and 4-pyridyl) determine the final coordination frameworks of 1 and 2. Photoluminescence and thermal stabilities of 1 and 2 were also investigated.  相似文献

[Co(CN)2(H3Cdta)] · 2H2O (I) and Na2[Co(CN)(Cdta)] · 3H2O (II) (Cdta4− is the 1,2-cyclohexanediaminetetraacetate) were synthesized. In I, the two cyanide ions and the Cdta nitrogen atoms are located in the equatorial plane, the carboxyl groups connected to different N atoms are in axial positions; in addition one of them, like two other uncoordinated carboxyl groups, is protonated. In II, Cdta is a pentadentate ligand coordinated in the cis-equatorial mode. UV excitation of the dicyano complexes at the ligand-to-metal charge transfer band gives organic derivatives of cobalt(III) stable enough for chromatographic isolation. Photolysis of II in neutral medium gives aqua(cyclohexanediaminetriacetato)cobalt (III) complexes as the final products.  相似文献

Abstract

Selected 4-oxybenzaldehyde and 2,2-dioxybiphenyl cyclotriphosphazene derivatives were synthesized via substitution reactions through tailored control. The reactions of cyclotriphosphazene with 4-oxybenzaldehyde and 2,2-dioxybiphenyl gave the following synthesized derivatives: one mono-substituted open-chain compound, N3P3Cl5(O2C7H5) (1, 69%); mono spiro, N3P3Cl4(O2C12H8) (2, 91.1%); non-gem tri-substituted, N3P3Cl3 (O6C21H15) (3, 17%); dispiro, N3P3Cl2(O4C24H16) (4, 92.3%); penta-substituted, N3P3Cl(O10C35H25) (5, 92%);hexa-substituted, N3P3(O12C42H30). Most of these derivatives (1–6) are obtained with good yield (up to 97%), This work provides a simple and available approach to obtain versatile cyclotriphosphazene derivatives, which is expected to further promote the use of HCCP as phosphorus platform for the construction of multi-functional materials.  相似文献
