(E)-N-芳基-3-[2-(8-羟基喹啉基)-乙烯基]咔唑的合成、抗氧化活性及促进鼠骨髓间质干细胞增殖的研究

设计合成了3-[2-(8-羟基喹啉基)-乙烯基]-N-对甲苯基咔唑(8)和3-[2-(8-羟基喹啉基)-乙烯基]-N-对甲氧苯基咔唑(9)两个新的化合物, 用IR, MS, 1H NMR和元素分析确认其结构. 并利用DPPH•方法, 超氧阴离子自由基( )法, 羟基自由基HO•法和噻唑蓝比色法(MTT法)分别测定了目标产物的抗氧化活性和调控鼠骨髓间质干细胞(MSCs)的作用. 结果表明, 这两种化合物对DPPH•自由基、超氧阴离子自由基和羟基自由基具有较强的抗氧化活性, 化合物9在低浓度时对鼠骨髓间质干细胞增殖有很好的促进作用.     

Nitration of 2- and 4-[2-(2-furyl)vinyl]thiazole derivatives

Nitration of 4-methyl-2-[2-(nitro-2-furyl)vinyl]thiazole with a mixture of concentrated nitric and sulfuric acids leads to 4-methyl-5-nitro-2-[2-(3,5-dinitro-2-furyl)vinyl]thiazole. Under the same conditions 2-methyl- and 2-acetamido-4-[1-R-2-(5-nitro-2-furyl)vinyl]thiazoles (R=CH₃, Cl) are nitrated in the 3 position of the furan ring, 2-amino-4-[1-chloro-2-(5-nitro-2-furyl)vinyl]thiazole is nitrated in the 5 position of the thiazole ring and 2-acetamido-5-nitro-4-[2-(2-furyl)vinyl]thiazole undergoes profound changes. Under the influence of a mixture of of nitric acid and acetic anhydride the latter compound is converted quantitatively to the 5-nitro derivative (with respect to the furan ring), whereas 4-[2-(5-nitro-2-furyl)vinyl]thiazole derivatives do not undergo reaction.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 3, pp. 314–317, March, 1977.     

[Z]-1-[2-(三芳基锡基)乙烯基]环辛醇的合成和性质

本文用三苯基氢化锡、三对甲苯基氢化锡作为锡氢化试剂与1-乙炔基环辛醇进行反应, 合成了两个有机锡化合物: [Z]-1-[2-(三苯基锡基)乙烯基]环辛醇(1)和[Z]-1-[2-(三对甲苯基锡基)乙烯基]环辛醇(2), 并测定了1的晶体结构。1和2分别与ICl, Br2, I~2反应, 得到六个有机锡一卤化物和三个有机锡二卤化物(3-11)。有机锡二卤化物6和一卤化物5与KOH乙醇溶液反应, 分别得到相应的锡氧化物和锡氢氧化物(12, 13)。有机锡二卤化物8分别与含氮双齿配体[2,2'-联吡啶(Bipy),5-硝基-1,10-邻菲罗啉(Nphen),8-羟基喹啉(Oxin)]反应, 得到三个相应的配合物(14-16)。十六个新化合物通过元素分析、锡含量测定、IR、^1H NMR测定对其结构进行了表征, 同时提出了1和2的生成反应历程。

通过三苄基氢化锡与1-乙快基环庚醇发生加成反应，得到了一种有机锡化合物 ：(Z)-1-[2-(三苄基锡基)乙烯基]环庚醇(1)．化合物1与I2，Br2和IC1按1：1和1 ：2摩尔比投料进行卤化反应，得到6种构型保持的有机锡一卤化物2-4及二卤化物 5-7．有机锡一卤化物3与KOH乙醇溶液反应，得到了相应的有机锡氢氧化物8．以上 8种新化合物均通过熔点测定、元素分析、锡含量测定、红外光谱、核磁共振氢谱 对其结构进行了表征．并对其中的有机锡化合物1，3，4，6，7进行了体外抗P388 血癌活性测定，其中有机锡化合物4，6，7表现出强效．     

Regioselective oxidative coupling of 4-hydroxystilbenes: synthesis of resveratrol and epsilon-viniferin (E)-dehydrodimers

Treatment of 5-[2-(4-hydroxyphenyl)vinyl]benzene-1,3-diol (resveratrol) with an equimolar amount of silver(I) acetate in dry MeOH at 50 degrees C for 1 h followed by chromatographic purification with a short silica gel column allowed the isolation of its (E)-dehydrodimer, 5-[5-[2-(3,5-dihydroxyphenyl)vinyl]-2-(4-hydroxyphenyl)-2,3-dihydrobenzofuran-3-yl]benzene-1,3-diol, as a racemic mixture in high yield. The present method was applicable to the oxidative dimerization of 4-hydroxystilbenes such as trans-styrylphenol and 5-[6-hydroxy-2-(4-hydroxyphenyl)-4-[2-(4-hydroxyphenyl)vinyl]-2,3-dihydrobenzofuran-3-yl]benzene-1,3-diol (epsilon-viniferin) leading to the corresponding 2-(4-hydroxyphenyl)-2,3-dihydrobenzofurans possessing various types of biological activities.     

Photochemical cycloaddition of mono-, 1,1-, and 1,2-disubstituted olefins to a chiral 2(5H)-furanone. Diastereoselective synthesis of (+)-lineatin

The photochemical [2 + 2] cycloaddition of (S)-4-methyl-5-O-pivaloyloxymethyl-2(5H)-furanone, 5, to vinyl acetate, vinyl pivalate, tert-butyl vinyl ether, 1,1-diethoxyethylene, and (Z)- and (E)-1,2-dichloroethylene has been studied. A practical synthesis of (+)-lineatin from 5 has been developed via the functionalized cyclobutane 6.     

Synthesis and NLO properties of new trans 2-(thiophen-2-yl)vinyl heteroaromatic iodides

The synthesis and characterisation of new trans 2-(thiophen-2-yl)vinyl pyridinium, imidazolium and quinoilinium iodides is reported together with their solvatochromic shifts and EFISH characterization. 2-{(E)-2-[5'-(dibutylamino)-2,2'-bithien-5-yl]vinyl}-1-methyl pyridinium and quinolinium iodides display high μ.β(vec) values up to 1200 × 10(-48) esu. The promising non-linear optical (NLO) properties of this new family of chromophores, which can be further improved by the design of highly efficient systems exploiting the donor and acceptor properties of both heteroaromatic rings and substituents, make them suitable candidates for second harmonic generation imaging with interesting biological applications.     

Synthesis and antifungal activities of a series of (1,2-disubstituted vinyl)imidazoles.

A series of vinylimidazoles containing a hetero atom such as sulfur or oxygen at a beta-position of the vinyl group was prepared and the antifungal activities were tested. It was found that sulfur-substituted derivatives such as (E)-1-[2-(methylthio)-1-[2-(pentyloxy)phenyl]ethenyl]-1H-imidazole (5a-5) and (E)-1-[1-[2-(hexyloxy)phenyl]-2-(methylthio)ethenyl]-1H-imidazole (5a-6) showed excellent antifungal activities against dermatophytes and yeast cells. The stereochemistry of the hydrochloride salt of 5a-5 was determined by X-ray crystallography. The structure-activity relationships were discussed.     

Synthesis and crystal structure of (E)-[(2-cyclohexenyl) vinyl]triaryltin

(E)-[(2-Cyclohexenyl)vinyl]triaryltin (aryl=phenyl, p-tolyl) were synthesized and characterized by elemental analysis, IR, 1H NMR spectroscopy. The crystal structure of (E)-[(2-cyclohexenyl)vinyl]triphenyltin was also determined by the X-ray diffraction. Experiments show that this compound has the conjugated system, which makes it have possible bioactivity and anti-tumor activity.     

Stereospecific preparation of (Z)- and (E)-2,3-difluoro-3-stannylacrylic ester synthons and a new, efficient stereospecific route to (Z)- and (E)-2,3-difluoroacrylic esters

[reaction: see text] The (Z)-2,3-difluoro-3-stannylacrylic ester is readily prepared from (Z)-1,2-difluorovinyltriethylsilane via stereospecific stannyl/silyl exchange with KF/(Bu3Sn)2O or Bu3SnCl in DMF at 70 degrees C. The corresponding (E)-2,3-difluoro-3-stannylacrylate is prepared by stereospecific carbonylation of (E)-1,2-difluorovinyl iodide followed by low temperature/in situ stannylation of the resultant (Z)-2,3-difluoroacrylic ester. With Cu(I) iodide and Pd(PPh3)4 catalysis, the (Z)- and (E)-stannylacrylate esters readily couple with aryl iodides and vinyl bromides, as well as 2-iodothiophene, at room temperature to stereospecifically produce the respective (E)- and (Z)-2,3-difluoro-3-aryl substituted acrylic esters or conjugated dienes in high yields.     

Synthesis and Characterization of (Z)-1-[2-(Triarylstannyl)vinyl]-1-Cyclohexanols and their Arylhalostannyl Derivatives

1-Ethynyl-1-cyclohexanol reacts with a triaryltin hydride Ar₃SnH (Ar = phenyl, p-tolyl) and generates the corresponding (Z)-1-[2-(triarylstannyl)vinyl]-1-cyclohexanol. The product obtained reacts with one or two equivalents of halogen (ICl or I₂) to form the associated (Z)-1-[2-(diarylhalostannyl)-vinyl]-1-cyclohexanol or (Z)-1-[2-(aryldihalostannyl)vinyl]-1-cyclohexanol, respectively. All compounds were characterized by elemental analysis, ¹H, ¹³C, ¹¹⁹Sn NMR and Mössbauer spectroscopy.     

The synthesis and transformations of ethyl (Z)-2-[2,2-bis(ethoxycarbonyl)vinyl]amino-3-dimethylaminopropenoate,a new reagent in the synthesis of heterocyclic compounds

Ethyl (Z)-2-[2,2-bis(ethoxycarbonyl)vinyl]amino-3-dimethylaminopropenoate (5) , a new reagent in the synthesis of heteroaryl substituted β-amino- α,β- -dehydro—amino acid derivatives and some fused hetero-cyclic systems, was prepared from ethyl N-2,2-bis(ethoxycarbonyl)vinylglycinate (3) and N,N-dimethyl-formamide dimethyl acetal (4) . The substitution of the dimethylamino group in the compound 5 with heterocyclic amines produced ethyl 2-[2,2-bis(ethoxycarbonyl)vinyl]amino-3-heteroarylaminopropenoates 7a-f and, in some instances, [2,2-bis-(ethoxycarbonyl)vinyl]aminoazolo- or -azinopyrimidine derivatives 8g-k.     

Synthesis,structure, and luminescent properties of 2-[2-(9-anthryl)vinyl]quinolines

Previously unknown 2-[2-(9-anthryl)vinyl]quinolin-8-ol and 2-[2-(9-anthryl)vinyl]-8-methoxyquinoline were synthesized by condensation of 8-hydroxy(methoxy)-2-methylquinoline with 9-anthraldehyde in acetic anhydride, as well as by the Wittig reaction. The product structure was determined on the basis of their 1H NMR, IR, UV, and mass spectra and quantum-chemical calculations. 2-[2-(9-Anthryl)vinyl]-8-methoxyquinoline showed luminescence with a quantum yield φ of 0.25, which was considerably higher than that of its 8-hydroxy analog (φ = 0.067).     

4-[(E)-2-(3,4,5-三甲氧基苯基)乙烯基]苯氧基羧酸酯的合成及抗肿瘤活性研究

二苯乙烯的多种衍生物表现出一系列的生理活性,如3,4,5.三羟基二苯乙烯(白藜芦醇)具有预防心脏病,抑制血小板凝聚,调控脂质和脂蛋白代谢,抗氧化及对癌症的治疗等作用[1-3].     

Synthesis and transformations of 2-(2-furyl)-and 2-[β-(2-furyl)vinyl]imidazo[4,5-b]phenazines

The corresponding 2-(2-furyl)- and 2-[-(2-furyl)vinyl]imidazo[4,5-b]phenazines were obtained by the condensation of 2,3-diaminophenazine with furfural and 2-furylacrolein and with their 5-bromo and 5-nitro derivatives. The phenazines were also synthesized by the reaction of 2-methylimidazo[4,5-b]phenazine with furfural and 5-bromo- and 5-nitrofurfural. The alkylation, acetylation, and nitration of the compounds obtained, as well as replacement of the halogen in the furan ring by a nitro group, were studied.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 10, pp. 1425–1428, October, 1971.     

Synthesis and transformations of 2-(2-furyl)-and 2-[β;-(2-furyl)vinyl]naphth[1,2-d]imidazoles

The corresponding 2-(2-furyl)naphth[1,2-d]imidazoles were obtained by heating Schiff bases, prepared from 1,2-naphthalenediamine and furfural and 5-bromo- and 5-nitrofurfurals, in nitrobenzene. 2-[-(2-Furyl)vinyl]naphth[1,2-d]imidazoles were synthesized by condensation of 2-methylnaphth[1,2-d]imidazole with furfural and 5-bromo- and 5-nitrofurfural. The methylation, nitration, and acetylation of the compounds obtained were studied, and the replacement of the bromine atom in the furan ring by a nitro group was also investigated.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 11, pp. 1555–1557, November, 1972.     

Synthesis,halogenation and structural characterization of (Z)-1-[2-(triphenylstannyl)vinyl]-1-cyclododecanol

Synthesis and characterisation of (Z)-1-[2(triphenylstannyl)vinyl]-l-cyclododecanol, c-(CH₂)₁₁C(OH)CH=CHSnPh₃, are reported, together with its halogenation by I₂, Br₂ and ICI to yield derivatives of the types c-(CH₂)₁₁C(OH)CH=CHSnPhs_?nX_n (n=1, 2; X=I, Br, Cl, respectively). The molecular structures of two compounds have been determined by X-ray diffraction analysis. The tin atom exhibits a distorted tetrahedral geometry in the crystal of (Z)-l-[2–(triphenylstannyI)vinyl]-l-cyclododecanol, but a trigonal bipyramidal geometry in the monoiodo-derivative (Z)-l-[2]-(diphenyliodo-stannyl)vinyl)-1-cyclododecanol and other derivatives, in which significant intramolecular coordinative interaction HO→Sn ia observed. And the formation of a five-membered tin containing ring is significant for their antitumour activities.     

Structure and spectral property of 8-(2-(5-(4-methylphenyl)-2-thienyl)vinyl)-10,10-dimethyl-10H-pyrido[1,2-a]indolium perchlorate

The title compound 8-(2-(5-(4-methylphenyl)-2-thienyl)vinyl)l-10,10-dimethyl-10H-pyrido[1,2-a]indolium perchlorate (3) has been synthesized by cycloaddition and condensation reactions, and characterized by elemental analysis, IR, ¹H NMR, and single-crystal X-ray diffraction. The crystal structure analysis exhibits that the molecule of 3 possesses good coplanarity, and the three rings (10H-pyrido[1,2-a]indolium, thienyl and phenyl) and vinyl moiety can make up a large conjugated system. Its UV-Vis and fluorescence spectra were measured, and found that it displays larger maximum absorptions and emission wavelengths in comparison with 8-(4-methylphenyl)vinyl analogue.     

5-二氰亚甲基-4-二环丙亚甲基-3-[1-(2, 5-二甲基-3-呋喃基)亚乙基]四氢呋喃-2-酮的合成

报道俘精酸酐类化合物(E/Z)4-二环丙亚甲基-3-[1-(2, 5-二甲基-3-呋喃基)亚乙基]四氢呋喃-2, 5-酮的拆分, 及(E)和(Z)-5-二氰亚甲基-4-二环丙亚甲基-3-[1-(2, 5-二甲基-3-呋喃基)亚乙基]四氢呋喃-2-酮 4(E)和4(Z)的合成, 并对它们的光致变色特性进行了初步研究。
