比较了13个酵母菌株催化合成2-苯乙醇的活性,确定酿酒酵母菌株的催化活性较高.通过单因素和正交实验对酵母催化合成2-苯乙醇的培养基组成进行了优化,采用优化后的催化合成培养基,2-苯乙醇浓度可达4.654g/L,摩尔产率为0.63.实验进一步采用Box-Behnken中心组合设计和响应面分析,建立了2-苯乙醇合成浓度与培养基主要组分之间的二次多项式回归模型,该模型具有较高的准确性,可作为生物催化法合成2-苯乙醇培养基的最优化预测模型.由响应面分析及实验验证得出,在最优培养基条件下,2-苯乙醇浓度可达4.815g/L.  相似文献

利用吸附法,将圆柱状假丝酵母脂肪酶固定于4种疏水性不同的载体上,固定化酶的活性及稳定性随载休疏水性的增大而增大。用YGW-C6H5作为载体,在有机溶剂-水双液相体系中催化萘普生甲酯的不对称水解,反应120h转化率为24.72%,产品的对映体过量值为94.82%。  相似文献

本文用ICP-AES法顺序测定了铝合金中的主要成份如Ti、V、Zr、Mn和次要成分如Si、Fe、Mg。采用正交设计法优化了仪器和操作参数。研究了样品产生的条件干扰。本法简单、极大地提高了分析速度。方法的相对标准偏差在1.7%~2.8%。  相似文献

本文叙述了近十年来我国对正烷烃酵母研究的进展。我们用的菌种是热带假丝酵母Y-17,本所实验厂的生产规模是年产一百吨酵母。发酵罐为6米~3的内或外循环空气提升式,两级连续发酵,50%废水循环利用。生产能力为2克/升·小时。酵母产品的化学分析,包括蛋白质、氨基酸、脂肪、核酸、维生素等,都证明该产品为一优良蛋白饲料。有毒物质含量(如苯骈(a)芘、重金属)符合国际标准。动物试验(包括用大白鼠、鸡、猪的营养试验,慢性毒性试验,传代试验)证明该正烷烃酵母为安全可靠的优良饲料。我们也研究了酵母的核酸和脂肪的综合利用、并筛选了一株高产新菌株球拟嗜烷烃酵母(新种)4B-2,使生产能力提高至2.8克/升·小时。  相似文献

将小波神经网络和遗传算法应用到2-(9-咔唑)-乙基氯甲酸酯衍生化氨基酸的胶束电动力学色谱分离优化。小波神经网络结合正交试验设计用于分离过程的多因素模型建立。以训练好的小波神经网络模型为目标函数,采用实数编码的遗传算法搜寻确定最佳分离条件,在此条件下分离得到的归一化分离度积与正交试验设计中最佳条件相比,提高了12.5%。  相似文献

建立了微波衍生化GC—MS测定食用植物油中的脂肪酸含量.并采用校正变换矩阵法对食用植物油的成分进行测定的方法。对微波衍生化的实验条件进行了优化.利用建立的校正模型对40个花生油、菜籽油、芝麻油、棉籽油、棕榈油的二元、三元、四元、五元人工合成样品进行了计算预测,所有样品的平均预测误差部小于5%。对8个实际样品进行了测定.其中有3个掺伪样品.所测调和食用油的结果与标签标示结果相符性较好。本方法可用于快速、准确地测定食用油中各成分含量或定性、定量鉴别掺伪成分。  相似文献

本文用正交设计优化茯苓皮的超声提取条件,并有效提取了茯苓皮药材内的主要成分三萜类物质,同时利用高效液相色谱法(HPLC)建立了提取物的液相指纹图谱。实验考察了不同提取溶剂、提取条件对提取工艺的影响;确立了最佳色谱条件,得到了茯苓皮中24种总提取物的指纹图;同时对10种不同产地的茯苓皮药材进行分析,标定了17个共有峰。研究结果表明,HPLC指纹图谱方法重现性好,稳定可靠,可用于控制茯苓皮中三萜成分的质量。  相似文献

设计了一种利用立体选择性相反的两种脂肪酶催化二次动力学拆分,由外消旋萘普生甲酯制备高光学纯度(S)-萘普生的方法。理论曲线预测,与简单的酶促动力学拆分反应相比,二次动力学拆分可以明显提高高光学纯度产物的产率.根据几种脂肪酶在微水/异辛烷双液相反应体系中不对称水解萘普生甲酯的立体选择性和对映体比率(E),首先选用R选择性的固定化南极假丝酵母脂肪酶(Novozym 435)对外消旋萘普生甲酯进行第一次拆分,然后选用S选择性的柱状假丝酵母脂肪酶(CRL)对S过量的剩余底物萘普生甲酯在同样的反应体系中进行第二次拆分.该二次拆分反应对映体过量值为96.8%的(S)-萘普生产率达19.9%.  相似文献

采用气相色谱-质谱(GC—MS)法对1,3-二氟苯溴化产物定性分析,采用了峰面积归一化法对产物进行定量分析,并通过正交设计结合多指标综合评价法优化合成条件。结果表明,通过GC—MS监测,对正交实验Z值结果分析得到最佳制备工艺:四氯化碳为溶剂,反应摩尔比(间二氟苯与液态溴)2:1,5℃下反应12h。产物纯度高于99.50%  相似文献

 研究了在高压CO2条件下利用粗状假丝酵母Candida valida CICC1444活性细胞催化苯乙酮不对称还原合成(R)-α-苯乙醇. 高压CO2条件可对粗状假丝酵母细胞的活性产生一定的影响,但反应的选择性反而有所提高,证实了高压CO2作为该反应环境是可行的. 考察了底物浓度、细胞用量、反应温度和压力等因素对反应的影响. 结果表明,这些因素对反应产率有很大的影响,在最佳反应条件下,反应底物苯乙酮的转化率和产物α-苯乙醇的产率均可达90%以上,对映体过量值(ee)保持在80%左右. 与常压静置条件下的反应相比,高压CO2条件下粗状假丝酵母催化苯乙酮不对称还原反应的转化率和产率分别提高了20%和40%左右,对映体选择性也提高了5%~10%.  相似文献

Results of lipase production by a soil microorganism, expressed in terms of lipolytic activities of the culture were modeled and optimized using artificial neural network (ANN) and genetic algorithm (GA) techniques, respectively. ANN model, developed based on back propagation algorithm, were highly accurate in predicting the system with coefficient of determination (R 2) value being close to 0.99. Optimization using GA, based on the ANN model developed, resulted in the following values of the media constituents: 9.991 ml/l oil, 0.100 g/l MgSO4 and 0.009 g/l FeSO4. And a maximum value of 7.69 U/ml of lipolytic activity at 72 h of culture was obtained using the ANN-GA method, which was found to be 8.8% higher than the maximum values predicted by a statistical regression-based optimization technique-response surface methodology.  相似文献

Clavulanic acid is a naturally occurring antibiotic produced by Streptomyces clavuligerus. The present work reports on clavulanic acid production by Streptomyces clavuligerus MTCC 1142 using one-factor-at-a-time and L25 orthogonal array. The one-factor-at-a-time method was adopted to investigate the effect of media components (i.e., carbon source, nitrogen source and inoculum concentration) and environmental factors such as pH for clavulanic acid production. Production of clavulanic acid by Streptomyces clavuligerus was investigated using seven different carbon sources (viz. glucose, sucrose, modified starch, rice-bran oil, soybean oil, palm oil, and glycerol) and six different nitrogen sources (viz. peptone, yeast extract, ammonium chloride, ammonium carbonate, sodium nitrate and potassium nitrate). A maximum yield of 140 μg/mL clavulanic acid was obtained in the medium containing soybean oil as a carbon source and yeast extract as nitrogen source. Subsequently, the concentration of soybean flour, soybean oil, dextrin, yeast extract and K2HPO4 were optimized using L25 orthogonal array method. The final optimized medium produced 500 μg/mL clavulanic acid at the end of 96 h as compared to 140 μg/mL before optimization. Synthesis of precursor molecules as a metabolic driving force is of considerable importance in antibiotic synthesis. Attempts to increase the clavulanic acid synthesis by manipulating the anaplerotic flux on C3 and C5 precursors by supplementing the medium with arginine, ornithine, proline, valine, leucine, isoleucine, pyruvic acid and á-ketoglutarate were successful. Supplementing the optimized medium with 0.1 M arginine and 0.1 M leucine increased the yield of clavulanic acid further to 1100 μg/mL and 1384 μg/mL respectively.  相似文献

采用水酶法制备虹鳟鱼骨油,单因素分析法优化虹鳟鱼骨酶解的工艺条件,考察了料液比、pH值、酶解时间、酶解温度、加酶量5个因素对鱼油提取率的影响,采用气相色谱-质谱联用 (GC-MS) 技术对鱼骨油的脂肪酸组成和含量进行了分析鉴定.结果表明,在55℃、pH 7.5、酶解时间为3 h、料液比为1∶1、加酶量为2000 U/g的条件下,利用碱性蛋白酶提取的虹鳟鱼骨油中的油脂含量最高.GC-MS分析结果表明,虹鳟鱼骨油中主要成分是不饱和脂肪酸,含量为脂肪酸总量的80.4% (w/w),其中单不饱和脂肪酸和多不饱和脂肪酸分别约占不饱和脂肪酸的76.9%和23.1% (w/w), DHA和EPA的总量为3.4% (w/w).本研究优化了虹鳟鱼油的提取技术,对虹鳟鱼油的主要挥发性物质进行了分析鉴定,初步确定了其中对鱼油风味起主要贡献的物质,对鱼油产品的分析与鉴别具有参考价值.  相似文献

Response surface methodology (RSM) was employed for optimization of medium components and cultural parameters in cost effective cane molasses based medium for attaining high yield of succinic acid. The important factors obtained by “one-variable-at-a-time-approach” (cane molasses, corn steep liquor, sodium carbonate, and inoculum density) were further optimized by RSM. The optimum values of the parameters obtained through RSM (cane molasses 12.5%, corn steep liquor 7.5%, and sodium carbonate 25 mM) led to almost double yield of succinic acid (15.2 g/l in 36 h) as against “one-variable-at-a-time-approach” (7.1 g/l in 36 h) in 500-ml anaerobic bottles containing 300-ml cane molasses based medium. Subsequently, in 10-l bioreactor succinic acid production from Escherichia coli was further improved to 26.2 g/l in 30 h under conditions optimized through RSM. This fermentation-derived succinic acid will definitely help in replacing existing environmentally hazardous and cost-intensive chemical methods for the production of succinic acid.  相似文献

In this paper, we present the results of simultaneous screening of eight gibberellins (GAs) in coconut (Cocos nucifera L.) water by MEKC directly coupled to ESI-MS detection. During the development of MEKC-MS, partial filling (PF) was used to prevent the micelles from reaching the mass spectrometer as this is detrimental to the MS signal, and a cationic surfactant, cetyltrimethylammonium hydroxide, was added to the electrolyte to reverse the EOF. On the basis of the resolution of the neighboring peaks, different parameters (i.e., the pH and concentration of buffer, surfactant concentrations, length of the injected micellar plug, organic modifier, and applied separation voltage) were optimized to achieve a satisfactory PF-MEKC separation of eight GA standards. Under optimum conditions, a baseline separation of GA standards, including GA1, GA3, GA5, GA6, GA7, GA9, GA12, and GA13, was accomplished within 25 min. Satisfactory results were obtained in terms of precision (RSD of migration time below 0.9%), sensitivity (LODs in the range of 0.8-1.9 microM) and linearity (R2 between 0.981 and 0.997). MS/MS with multiple reaction monitoring (MRM) detection was carried out to obtain sufficient selectivity. PF-MEKC-MS/MS allowed the direct identification and confirmation of the GAs presented in coconut water (CW) sample after SPE, while, the quantitative analysis of GAs was performed by PF-MEKC-MS approach. GA1 and GA3 were successfully detected and quantified in CW. It is anticipated that the current PF-MEKC-MS method can be applicable to analyze GAs in a wide range of biological samples.  相似文献

In this study, the production and optimization of extracellular lipase from Kluyveromyces marxianus IFO 0288 was investigated by using optimized nutritional and cultural conditions in a yeast medium containing glucose as the carbon source in fully aerobic batch fermentation (150?rpm). The influence of four fermentation parameters (type of lipidic source, initial culture pH, temperature, and length of fermentation) on growth and lipase production was investigated and evaluated using the conventional ??one variable at a time?? approach and response surface methodology. An 18-fold increase in lipase production during 65?h of fermentation was obtained with optimized nutritional (0.5?% olive oil) and cultivation (pH?6.5, 35?°C) conditions by employing the conventional optimization method. By applying the response surface methodology technique the initial pH value of 6.4 and temperature of 32.5?°C were identified as optimal and led to further improvements (up to 18-fold) of extracellular lipase production. The results provide, for the first time, evidence that K. marxianus has the potential to be used as an efficient producer of extracellular lipase with prospective application in a variety of industrial and biotechnological areas.  相似文献

A simple and fast procedure is proposed for the extraction of iron (Fe), copper (Cu), and nickel (Ni) in 16 varieties of sunflower seed oil samples using an ultrasonic bath. The experimental parameters of the ultrasonic-assisted extraction (UAE) method were optimized to improve the sensitivity and detect the metals at trace levels in minimum time. Conventional wet acid digestion method was used for comparative purposes. The optimum recovery of all 3 metals was obtained by UAE for 7 min, while the separation of aqueous and organic phases after extraction using centrifugation (UAE-2) required 3 min, as compared to the conventional equilibration method (UAE-1) that required 90 min. The respective recoveries of Cu, Fe, and Ni obtained with UAE-2 were in the range of 95.8-97.5, 93.5-98.3, and 95.6-98.2%, respectively, for different varieties of sunflower oil samples. Accuracy was determined by the standard addition method. Under the optimum operating conditions, the limits of detection obtained from the standard addition curves were 21.7, 20.4, and 35.6 ng/mL for Fe, Cu, and Ni, respectively. The fact that all varieties of sunflower oil contain significant amounts of Fe, Cu, and Ni indicates the deterioration of sunflower oil quality immediately after extraction from seeds, which poses a threat to oil quality and human health.  相似文献

ABSTRACT: The optimization processes of photo degradation are complicated and expensive when it is performed with traditional methods such as one variable at a time. In this research, the condition of ortho-cresol (o-cresol) photo degradation was optimized by using a semi empirical method. First of all, the experiments were designed with four effective factors including irradiation time, pH, photo catalyst's amount, o-cresol concentration and photo degradation % as response by response surface methodology (RSM). The RSM used central composite design (CCD) method consists of 30 runs to obtain the actual responses. The actual responses were fitted with the second order algebraic polynomial equation to select a model (suggested model). The suggested model was validated by a few numbers of excellent statistical evidences in analysis of variance (ANOVA). The used evidences include high Fvalue (143.12), very low P-value (<0.0001), non-significant lack of fit, the determination coefficient (R2 = 0.99) and the adequate precision (47.067). To visualize the optimum, the validated model simulated the condition of variables and response (photo degradation %) by a few number of three dimensional plots (3D). To confirm the model, the optimums were performed in laboratory. The results of performed experiments were quite close to the predicted values. In conclusion, the study indicated that the model is successful to simulate the optimum condition of o-cresol photo degradation under visible-light irradiation by manganese doped ZnO nanoparticles.  相似文献

In this article, we explore the efficiency of using a coupled genetic algorithm (GA) and density functional theory (DFT) based strategy to evaluate probable structures of (H(2) O)(n) F(-) micro-clusters, with n = 1 - 6. We use the stochastic optimization technique of GA to arrive at structures of the cluster systems and once the structures are obtained, do a DFT calculation with the optimized coordinates from the GA calculation as input to get the infra-red spectrum of all the systems. The results of our work closely resembles the pure quantum chemical results obtained by Baik et al. (J Chem Phys 1999, 110, 9116-9127).  相似文献

Genetic algorithms (GA) were used to solve one of the multidimensional problems in computational chemistry, the optimization of force field parameters. The correlation between the composition of the GA, its parameters (p(c), p(m)) and the quality of the results were investigated. The composition was studied for all combinations of a Simple GA/Steady State GA with a Roulette Wheel/Tournament Selector using different values each for crossover (0.5, 0.7, 0.9) and mutation rates (0.01, 0.02, 0.05, 0.10, 0.20). The results show that the performance is strongly dependent on the GA scheme, where the Simple GA/Tournament Selector yields the best results. Two new MM3 parameters were introduced for rhenium compounds with coordination number four (204) and coordination number five (205), the formal oxidation states of rhenium ranging from +V to +VII. A manifold of parameters (Re-C, N, O, S) was obtained by using a diverse set of CSD structures. The advantage of the GA vs. UFF calculations is shown by comparison of several examples. The GA optimized parameters were able to reproduce the geometrical data of the X-ray structures.  相似文献
