Chinese Spring Tal kr phlb plants were developed by hybridizing Tal common wheat and Chinese Spring phlb mutant and backcrosses. When Chinese Spring Tal kr phlb plants were crossed with some wild species of Triticeae, much more seeds of wide hybridization can be obtained without artificial emasculation and pollination because of the existence of Tat and kr genes. The homoeologous chromosomes between common wheat and the wild species of Triticeae can also pair and exhibit cross-over in the hybrid F1 plants because of the existence of the phlb gene. So that more alien desirable genes can be transferred to common wheat rapidly. Therefore, Chinese Spring Tal kr phlb plants become a powerful tool for wide hybridization and transferring alien desirable genes to common wheat.  相似文献

Barley yellow dwarf virus (BYDV) is one of the most serious wheat diseases in China. So far no resistance has been described in common wheat. A certain level of BYDV resistance was found in thirteen Triticeae species. Thinopyrum intermedium, two octoploids derived from TH. intermedium/wheat, Zhong 4 awnless and TAF46, and one disomic addition line, L1 derived from TAF46, showed good resistance to BYDV by enzyme linked immunosorbent assay (ELISA). Two wheat/TA. intermedium translocation lines, CPI 119880 and CPI 119899, showing good BYDV resistance were developed from L1 by using both CSph mutant and tissue culture. It is found that their BYDV resistance was controlled by a single dominant gene. Two cDNA probes pEleAcc3 and pPJN8 (E1-T1) were screened for detecting Th. intermedium DNA in wheat background. A specific band for the DNA of Th. intermedium and its derivatives was found in Southern hybridization. The proline biosynthetic pathway and Pro genes in Saccharomyces cerevisiae have just begun to be studied recently. In our laboratory, Pro2 gene of S. cerevisiae had been cloned in yeast. As described in this paper, yeast Pro3 gene was also cloned, which can complement yeast Pro3 mutants, and be expressed efficiently in E. coli. The high activities of this gene product, L-pyrroline-5-carboxylate (P5C) reductase, can be detected in both organisms. The activity of the Pro3 gene product in multiple copy plasmids is not higher than that of single copy genes in chromosomes in both yeast and E. coll. The preliminary characterization of the gene is also reported.  相似文献

Chronic exposure of the gray, short-tailed oppossum, Monodelphis domestica to ultraviolet radiation (UVR) induces mesenchymal tumors of the cornea. High molecular weight DNA samples from 6 UVR-induced corneal tumors were assayed for their ability to transform NIH 3T3 cells to tumorigenicity. NIH 3T3 cells transfected with DNA from 5 of the corneal tumors produced 14 tumors in nude mice. Cell lines were established from these tumors. DNA from 13 of 14 tumor cell lines contained repetitive opossum DNA sequences. Southern blot analysis revealed that DNA from 3 of 4 cell lines derived from tumorigenic NIH 3T3 cells transfected with DNA from a single oppossum tumor contained opossum Ki-ras oncogene sequences in addition to the murine Ki-ras gene. Northern blot analysis of mRNA from a mouse tumor cell line containing opossum Ki-ras gene sequences showed mRNA species identical in size to opossum Ki-ras mRNA, as well as murine Ki-ras mRNA species. These results suggest that an activated Ki-ras oncogene was present in one of the original opossum corneal tumors tested. Thus, activation of Ki-ras may play a role in the development of UVR-induced corneal tumors in Monodelphis domestica. Further characterization of ras oncogenes in these opossum tumors may provide information on the molecular mechanisms by which UVR induces corneal tumors in this species.  相似文献

An in vitro-vivo technique for establishment of cell lines on murine leukemia has beendeveloped. Using this method, suppressive T lymphoblastic leukemia L7811-85, L7212-85, non-T, non-B lymphocytic leukemia L1210-86, B lymphocytic leukemia P 388-86 and Friend erythroleu-kemia FLCL cell lines have been established. Incidence of leukemia with these cell lines was 100%. Along with the increase of genera-tions of cell lines, cell growth accelerated, generation time shortened and cloning efficienciesrose. A following up electron microscopic observation on L7811-85 and L7212-85 showed thatthe virus particles were "A" particles in original cells. When they became cell lines in vitro,virus particles increased and transformed into typical "C" particles with budding. An inhibitory activity relevant to leukemic cells on proliferation of leukemic cells hasbeen observed in the supernatant of L7811-85 medium and was regarded as an "autocrine".  相似文献

Theoretical results of normal coordinate analysis of polybenzamide performed with two conformational isomerized models, the cis trans and the all-trans conformation, have been adopted to examine a number of infrared spectra of films of the polymer treated in a subsequent annealing process and a cooling process. It shows, in this investigation, that two sets of infrared spectral profiles around 1400 cm~(-1) and 900 cm~(-1) behave quite attractively. Their spectral behaviours have been correlated to the conformational defect variation in samples, and reasonably predicted by the normal coordinate calculation.  相似文献

This paper proposes the approximate methods of calculation for research on inhomogeneous finite strain fields in geologic bodies by means of the "Principal Axis" theory of continuum mechanics. The methods are to obtain the principal strain orientations at finite points in the earth's crust based on the information and data provided by the actual strain measurements of deformation marked objects in the field, or by the research on crystalline fabrics in rocks, and then to obtain two sets of smooth orthogonal principal strain trajectories through mathematical treatments. A network composed of two sets of orthogonal curves shows the deformation character of the rock, and correspondent strain components satisfy compatibility conditions. The curvatures of curves are used to describe the compatibility conditions in this paper. An analytic solution of a strain field is obtained when the two sets of lines can be simulated by analytic function; the magnitudes of principal strains at every point may be obtained b  相似文献

New [3:3]hexakisadducts 3 , 6 , and 7 have been synthesized by a stepwise addition of two tripodal malonate tethers to pristine [60]fullerene. For the first time, [3:3]hexakisadducts of two sets of asymmetrically substituted malonate addends could be prepared as single in/out isomers by following this synthetic protocol. Thereby, four spherically defined addend zones I–IV were created with various similar degrees of latitude on the fullerene sphere. Four amphiphilic [3:3]hexakisadducts 12 , 14 , 18 , and 19 were prepared with different relative arrangements of both the hydrophilic and lipophilic moieties in the addend zones I / II and III / IV , respectively.  相似文献

Various fused-heterocyclic-derivatives containing thiazolopyridine moieties has been synthesized by allowing 5-aminothiazolo[3,2-a]pyridine derivative 1 to undergo annulations reactions with different reagents under different-reaction conditions. The biological assessment of compounds 2 , 11 , 14 , 15 , and 19 showed remarkable antimicrobial activities. In addition, selected derivatives of the products were screened for their anticancer activities against two tumor cell lines using MTT assay and the results showed that some of these compounds have potent cytotoxic effect, as concluded from their IC50 values. Meanwhile, compounds 3a , 7 have exhibited very strong potency as anticancer candidates. Thiazolopyridine structures have been confirmed as a useful lead compounds for the development of new anticancer agents. Molecular docking showed that,-some of the synthesized compounds more suitable inhibitor against-ALR2 with farther alteration in future.  相似文献

In addition to one known compound, 3beta,8alpha-dihydroxyguaian-4(15),10(14),11(13)-trien-6,12 olide (8-desacylcynaropicrin) (3), two new sesquiterpene lactones have been isolated from the aerial parts of Crepis cameroonica. By means of spectroscopic analysis, the structures and relative configurations of the new compounds were established as 3beta,9beta-dihydroxyguaian-4(15),10(14),11(13)-trien-6,12 olide (1) and 8alpha-hydroxy-4alpha(13),11beta(15)-tetrahydrozaluzanin C (2). The in vitro antimicrobial spectrum of pure compounds and crude extracts are also reported.  相似文献

In this paper, superhigh reproductive rate strains of MEV with titre more than HA8192* or TCID50 log9.7 10 have been achieved both by cultivation in cell lines with different susceptibility to MEV and by isolating and identifying in field by the author. The systematic tests proved that S18 and L12 strains of MEV are the best strains for vaccine preparation. In this study, the best means for the tissue cultivation of MEV and the most advanced technological process for the production and detection of serum-free cell-cultured MEV fluids with super-high HA titre in batches in large quantities have been established for the first time. Optimum conditions for MEV inactivation were determined, and safe and effective inactivated vaccines with mineral oil or A1(OH)3 gel adjuvant were successfully prepared with serum-free cell-cultured MEV fluids. This paper describes an efficient algorithm based on a new concept called gene team for detecting conserved gene clusters among an arbitrary number of chromosomes. Within the clusters, neither the order of the genes nor their orientation need be conserved. In addition, insertion of foreign genes within the clusters are permitted to a user-defined extent. This algorithm has been implemented in a publicly available TEAM software that proves to be an efficient tool for systematic searches of conserved gene clusters. Abstract— The effectiveness of photodynamic therapy against P-glycoprotein ATPase activity in multidrug-resistant cells was studied. Chinese hamster ovary AUXB1 (drug-sensitive) and CR1R12 (multidrug-resistant) cell lines were compared with respect to uptake of 14C-polyhematoporphyrin and porphyrin photosensitization. Phototoxicity of Photofrin® was similar in both cell lines, and no major differences in uptake or efflux of 14C-polyhematoporphyrin were observed. Porphyrin photosensitization in vitro of CR1R12 cells or isolated plasma membranes from these cells caused inhibition of P-glycoprotein ATPase activity. Application of porphyrin photosensitization at a sublethal level to CR1R12 cells resulted in a small but significant increase in adriamycin-induced cytotoxicity. The hydrophobic "picket-fence" porphyrin, meso -tetrakis-( o -propionamidophenyl)porphyrin,α,α,α,β-isomer, was more inhibitory toward P-glycoprotein ATPase activity than the two less hydrophobic porphyrins tetraphenylporphine tetrasulfonate and Photofrin®.  相似文献

Microarrays have been widely used to identify differentially expressed genes. One related problem is to estimate the proportion of differentially expressed genes. For some complex diseases, the amount of differentially expressed genes may be relatively small and these genes may only have subtly differential expressions. For these microarray data, it is generally difficult to efficiently estimate the proportion of differentially expressed genes. In this study, I propose a likelihood-based method coupled with an expectation-maximization (E-M) algorithm for estimating the proportion of differentially expressed genes. The proposed method has favorable performances if either (i) the P values of differentially expressed genes are homogeneously distributed or (ii) the proportion of differentially expressed genes is relatively small. In both of these situations, I showed through simulations that the proposed method gave satisfactory performances when it was compared to other existing methods. As applications, these methods were applied to two microarray gene expression data sets generated from different platforms.  相似文献

A comparison of flame temperatures measured by the well-known spectroscopic “slope” method for Fe thermometric species residing in an isothermal, optically-thin environment shows that a range of 150°K is obtained when several different sets of recommended transition probabilities are used for the determinations. A similar lack of self-consistency is also shown by the temperatures obtained from two groups of Fe lines when the same set of transition probabilities is employed.
