The 2,3,7,8-substituted polychlorinated dibenzo-p-dioxins (PCDDs) and polychlorinated dibenzofurans (PCDFs) are among the most toxic compounds known, and several sources of exposure to these chemicals should be monitored to protect human and environmental health. The current predominant method of analysis is too expensive and cumbersome, and comprehensive two-dimensional GC coupled to time-of-flight mass spectrometry (GC x GC--TOF-MS) has the potential to lower the costs and speed analysis of PCDD/Fs. In this study, GC x GC--TOF parameters were evaluated and optimized to yield complete separation of the 17 most important PCDD/F congeners from polychlorinated biphenyls (PCBs) interferences, and to attain the lowest detection limits. The optimization study entailed evaluation of oven temperature programs, column flow rates, ion source temperatures, electron ionization energy, data acquisition rate, and various GC x GC parameters, including modulation period, modulator temperature offset and hot pulse duration. After optimization, all 17 PCDD/Fs were separated in <60 min, and in particular, the critical pair of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) and pentachlorobiphenyl congener CB126 did not co-elute chromatographically. Accurate identification and determination of all analytes could be made using their deconvoluted full mass spectra. In GC x GC, the modulation period and start time were the most important factors that affected sensitivity and selectivity for the analysis of the PCDD/Fs. The modulation period should be < or = 4s to preserve separations achieved in one-dimensional GC, and the modulation start time was important to achieve one large slice and two smaller symmetrical slices of TCDD to maximize its detection sensitivity. After optimization, the method could identify 0.25 pg of TCDD with standard injection from its full mass spectrum.  相似文献

An automated direct sample introduction technique coupled to comprehensive two-dimensional gas chromatography-time of flight mass spectrometry (DSI-GC x GC/TOF-MS) was applied for the development of a relatively fast and easy analytical screening method for 17 polychlorinated dibenzo-p-dioxins/dibenzofurans (PCDD/Fs) and 4 non-ortho polychlorinated biphenyls (PCBs) in fish oil. Comparison of instrumental performance between DSI-GC x GC/TOF-MS and the traditional gas chromatographic high resolution mass spectrometric (GC-HRMS) method showed good agreement of results for standard solutions analyzed in blind fashion. Relatively high tolerance of the DSI technique for lipids in the final extracts enabled a streamlined sample preparation procedure that only required gel permeation chromatography (GPC) and solid-phase extraction (SPE) cleanup with graphitized carbon black. The sample size for the method was 2g of cod liver oil, which achieved limits of quantitation (LOQs) of 0.019-7.8 pg/g toxic equivalent quotients for the individual PCDD/Fs. Lower detection limits can be achieved by using larger sample size and scaling up the sample preparation procedure, but this adds to the labor, time, solvent consumption, and expense of the approach. However, the streamlined method yielded 0.94 pg/g and 2.3 pg/g LOQs for 2,3,7,8-tetrachloro dibenzofuran (TCDF) and 3,3',4,4',5-pentachloro biphenyl (CB126), which were sufficiently low for regulatory monitoring of 2g samples. Therefore, instead of congener specific analysis, this streamlined analytical screening method for TCDF and CB126 has the potential to monitor fish oil contaminated with dioxin and dioxin-like PCBs at or above current food safety limits. Acceptable recoveries for nearly all analytes at three different spiking levels in fish oil samples were achieved with good repeatability.  相似文献

The separation of 38 toxic and predominant polychlorinated biphenyl (PCB) congeners, 11 persistent halogenated pesticides, 1 brominated biphenyl (BB), and 8 polybrominated diphenyl ethers (PBDEs) has been optimized using comprehensive two-dimensional gas chromatography coupled to time-of-flight mass spectrometry (GC x GC-TOFMS). A thermal desorption-programmable temperature vaporization (TD-PTV) step was used for the injection. Different column sets were investigated, and a 100% dimethylpolysiloxane (15 m x 0.25 mm i.d. x 0.25 microm film thickness) narrowbore capillary column coupled to a high temperature (8% phenyl)-polycarborane-siloxane (2 m x 0.10 mm i.d. x 0.10 microm film thickness) microbore column set was selected. Of the 58 compounds investigated, only one pair of PCBs was not resolved. All other analytes were either baseline separated into the chromatographic plane or were virtually separated using the deconvolution capability of the TOFMS.  相似文献

Comprehensive two-dimensional gas chromatography with isotope-dilution time-of-flight mass spectrometry (GC × GC-IDTOFMS) was used to measure polychlorinated dibenzo-p-dioxin (PCDD), polychlorinated dibenzofuran (PCDF), and coplanar polychlorinated biphenyl (cPCB) concentrations in ash, sediment, vegetation, and fish samples. The GC × GC capability was achieved by using a quad jet, dual stage, thermal modulator. Zone compression of the GC peaks from modulation resulted in a significant increase of the signal intensity over classical GC-IDTOFMS. The GC × GC column set used an Rtx-Dioxin 2 phase as the first dimension (1D) and an Rtx-500 as the second dimension (2D). The chromatographic separation of the 17 PCDD/Fs and the 4 cPCBs was attained in 1D except for 2,3,7,8-TCDD and CB126 for which deconvoluted ion currents (DIC) were required to be reported separately. The Rtx-500 phase separated the bulk matrix interfering compounds from the target analytes in 2D. The instrumental limit of detection (iLODs) was 0.5 pg for 2,3,7,8-TCDD. The calibration curves showed good correlation coefficients for all the compounds investigated in the concentration range of 0.5–200 pg. GC × GC-IDTOFMS results compared favorably to those from conventional isotope-dilution one-dimensional gas chromatography-high resolution mass spectrometry (GC-IDHRMS). The comprehensive mass analysis of the TOFMS further permitted the identification of other contaminants of concern in the samples.  相似文献

Polychlorinated naphthalenes (PCNs) are candidates for inclusion in the Stockholm Convention on persistent organic pollutants. PCNs are structurally and toxicologically similar to 2,3,7,8-tetrachlorodibenzo-p-dioxin (2,3,7,8-TCDD) and its analogues. Intake in food is considered to be an important human exposure pathway for PCNs. In this preliminary study, cheese and butter samples were analysed for PCNs, polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs) and polychlorinated biphenyls (PCBs) using an isotope dilution gas chromatography high-resolution mass spectrometry method. The aim of this study was to evaluate the PCN concentrations in the cheese and butter samples and to compare them with the PCDD, PCDF and PCB concentrations. The PCN concentrations were 5.6–103 pg g?1 of wet weight in the seven cheese samples tested and 5.0–199 pg g?1 of wet weight in the seven butter samples tested. The mass concentrations of lower chlorinated congeners were greater than those of the higher chlorinated congeners. Congeners of CN45/36, CN27/30 and CN33/34/37 were much more abundant than other congeners found in tetrachlorinated PCNs. Congeners of CN51, CN66/67 and CN73 were determined to be the predominant congeners in penta-, hexa- and heptachlorinated homologs, respectively. The PCNs contributed around 5% of the total PCN, PCDD, PCDF and PCB toxic equivalence (TEQ) values. CN73 was found to be the dominant PCN congener and contributed more than 40% to the PCN TEQ value. Congeners CN66/67, CN69 and CN63 were also found at relatively high levels. The PCB congener CB118 was the predominant congener (by mass-based concentration) of the 12 dioxin-like PCBs (dl-PCBs). The PCBs contributed 53.8% of the total TEQ, and congener CB126 contributed more than any other compound that was analysed to the total TEQ. The PCDDs and PCDFs contributed 11.6% and 29.7% of the total TEQ values, respectively.  相似文献

样品采用索氏抽提,抽提液依次经酸性硅胶床、多段混合硅胶柱和凝胶渗透色谱柱(GPC)净化后,用Florisil硅藻土柱分离出样品中的二噁英(PCDD/Fs)和类二噁英多氯联苯(dioxin-like PCBs),采用同位素稀释法和气相色谱/高分辨质谱联用仪(GC/HRMS)测定了其中的17个2,3,7,8-氯取代二噁英类化合物和12个类二噁英多氯联苯。结果表明,用该法分析二噁英和多氯联苯标准溶液,平行4次的分析结果为:RSD(PCDD/Fs)<8.9%,RSD(PCBs)<11.4%;回收率可达60%-105%。PCDD/Fs和PCBs的检出限分别为0.1-0.8 pg/g和0.05-0.6 pg/g。应用本方法成功测定了沉积物、淤泥、土壤和飞灰中的二噁英和类二噁英多氯联苯,并计算出它们的毒性当量。  相似文献

The chemical-activated luciferase expression (CALUX) assay is a reporter gene assay that detects dioxin-like compounds based on their ability to activate the aryl hydrocarbon receptor (AhR) and thus expression of the reporter gene. In this paper, the CALUX assay was examined for its application in the screening of polychlorinated dibenzo-p-dioxins (PCDDs), dibenzofurans (PCDFs) and dioxin-like polychlorinated biphenyls (dioxin-like PCBs) in retail fish. The sample extracts were cleaned up on a sulfuric acid-silica gel column followed by an activated carbon column, and the AhR activity of the separated PCDD/F and dioxin-like PCB fractions was determined using the assay. The quantitative limit for 2,3,7,8-tetrachlorodibenzo-p-dioxin (2,3,7,8-TCDD) was 0.98 pg ml(-1) (0.19 pg assay(-1) in the standard curve, corresponding to 0.16 pg g(-1) of CALUX-based toxic equivalency (2,3,7,8-TCDD equivalents) in the tested sample. Recovery tests in which dioxins were added to fish samples resulted in acceptable recoveries (77-117%). The CALUX assay performed well in the analysis of dioxins in fish samples and a comparative study revealed a strong correlation between the CALUX assay and high-resolution gas chromatography-high-resolution mass spectrometry analysis for the determination of PCDD/Fs (r = 0.89) and dioxin-like PCBs (r = 0.91) in retail fish (n = 22). These data revealed that the CALUX assay would be a useful screening method for PCDD/Fs and dioxin-like PCBs in retail fish.  相似文献

Gas chromatography coupled to ion trap tandem mass spectrometry (CG-MS-MS) has been evaluated for the analysis of polychlorinated dibenzo-p-dioxins (PCDDs) and dibenzofurans (PCDFs) and dioxin-like polychlorinated biphenyls (dl-PCBs) in vegetable oil samples containing different concentration levels (0.2-6 pg WHO-TEQ g(-1) for both PCDD/Fs and dl-PCBs) of the 29 toxic congeners of PCDD/F and dioxin-like PCBs. The effect of potential interfering compounds such as polychlorinated naphthalenes (PCNs), polychlorinated biphenyls (PCBs) and polychlorinated diphenylethers (PCDEs) on the quantification of mono-ortho PCBs has been investigated. In addition, the influence of the clean-up procedure on the final determination by CG-MS-MS was studied, showing that the quality of the results depends to a great extent on this analytical step. Quality parameters have been established and good precisions (CV: 3-19%) and low limits of detection for PCDD/Fs (0.04-0.20 pg g(-1) oil) and dl-PCBs (0.08-0.64 pg g(-1) oil) were obtained. The method was validated by a comparison of the CG-MS-MS results with those obtained by GC-HRMS.  相似文献

A simple and reliable method for the determination of polychlorinated biphenyls (PCBs) from mono- to octachlorobiphenyls in fish oil for dietary supplement is described. The method combines Florisil clean up and headspace solid-phase microextraction on 65 microm polydimethylsiloxane-divinylbenzene (PDMS-DVB). Analyte detection was carried out using GC-time-of-flight mass spectrometry (GC-TOF-MS). Fifty three PCB congeners including the seven indicator PCBs (IUPAC Nos. 28, 52, 101, 118, 138, 153 and 180) were analyzed. Under optimal conditions, the method detection limit (MDL) of each congener in the range from 0.8 to 31 ng/g was found. A certified reference material (BCR-349) was analyzed and it showed good agreement with the certified data.  相似文献

An extended study of seven fused silica capillary gas chromatographic (GC) columns has been conducted with regard to separation of international toxic equivalent factor (I-TEF) isomers (tetra- through octa-chlorinated at 2,3,7,8 positions) of polychlorinated dibenzo-p-dioxins and polychlorinated dibenzofurans (PCDDs/PCDFs) from closely co-eluted other isomers using high-resolution gas chromatography-high-resolution mass spectrometry (HRGC-HRMS). The data are explicated in mass chromatograms of Series 5 GC columns from a variety of manufacturers (Varian CP-Sil 8 CB LowBleed/MS, Phenomenex ZB-5UMS, Agilent HP-5MS, Restek Rtx-5MS, Supelco Equity-5, J&W Scientific DB-5 and DB-5MS), according to relative retention times, and 2,3,7,8-substituted isomer concentrations for each of the columns tested. Results showed differences between 5% phenyl methyl silicone and 5% silphenylene (Si-arylene) silicone polymer type GC stationary phases in separation of 2,3,7,8-substituted PCDDs/PCDFs from closely co-eluted isomers. The separation differences for Si-arylene type columns resulted in lower toxic equivalence (TEQ) values compared to the siloxane-based columns. Because of differences in product nomenclature and manufacturing practices by various manufacturers, incorrect assumptions and comparisons may be made regarding the interchangeability of these columns for PCCD/PCDF separations. The data presented are the most comprehensive to date and provide a valuable addition to operational criteria for the standard US Environmental Protection Agency methods 1613b and 8290.  相似文献

The combination of narrow-bore capillary gas chromatography with bench-top quadrupole mass spectrometric detection was evaluated for the determination of polychlorinated biphenyls (PCBs). The qualitative and quantitative performances of the system are illustrated by several analyses (PCB standards and human milk extracts). Capillary columns with different internal diameters (0.10, 0.18 and 0.22 mm, respectively) were compared for their ability to separate PCB congeners and the analysis time. Short run times (less than 7 min) were sufficient for complete separation of PCB congeners on a 0.10-mm internal diameter (I.D.) capillary column without any loss of resolution when compared with a 0.22 mm I.D. column. Good qualitative and quantitative data acquisition was possible with quadrupole mass spectrometer for run times of 8 min, but incomplete peak reading was observed when run times were reduced to 3-4 min. Selected ion monitoring and dwell times of 10 ms are necessary to obtain detection limits for individual PCB congeners as low as 0.4 pg microl(-1) for standard solutions and 0.2 ng g(-1) fat for milk extracts. By using cold splitless injection, relatively high volumes (1 microl) for narrow-bore capillaries could be injected without any peak distortion.  相似文献

A method for the high-throughput analysis of human serum for the 38 most prevalent polychlorinated biphenyls (PCBs) based on the use of fast gas chromatography-isotopic dilution time-of-flight mass spectrometry (GC-IDTOFMS) is presented. The chromatographic separation time was 8 min. The separation of the congeners was carried out either chromatographically or analytically using the mass spectral deconvolution capability of the TOFMS. The instrument and the method limits of detection (LODs) were 0.5 pg microL(-1) and 20 pg microL(-1), respectively, which is not as good as the one achieved using high resolution mass spectrometry (HRMS) but allows the detection and quantification of the prevalent PCBs present in real human serum samples. The dynamic range covered 3 orders of magnitude. The comparison with the high resolution mass spectrometry (HRMS) reference method (28 min) was good and some separation improvements have been observed. This method allows the analysis of 100 samples per day per instrument.  相似文献

Past years, many efforts have been dedicated to the development of alternative analytical methods for the measurement of dioxins in various types of matrices. Polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs), and polychlorinated biphenyls (PCBs) are compounds that are present in samples at part-per-billion (ppb) or part-per-trillion (ppt) level. Their measurement requires the use of very sensitive analytical methods. Gas chromatography (GC) coupled to quadrupole ion storage mass spectrometry (QISTMS), fast GC (FGC) coupled to time-of-flight mass spectrometry (TOFMS) and comprehensive two-dimensional gas chromatography (GC x GC) coupled to TOFMS are the more promising tools challenging the reference GC high resolution mass spectrometry (HRMS) based on sector instruments. We report herein some of the advances we achieved in the past years in our laboratory on the development of alternative measurement methods for those compounds.  相似文献

A simultaneous steam distillation-solvent extraction (SDE) procedure was used for determining polychrlorinated biphenyls, polychlorinated dibenzo-p-dioxins and polychlorinated dibenzofurans (PCBs, PCDDs and PCDFs) at sub-ppb levels in water samples. Recoveries of 39.8–138.7% and a standard deviation of lower than 10% were achieved for the individual coplanar PCB and the 2,3,7,8-substituted PCDD/F congeners. SDE is a fast, clean, inexpensive and reproducible procedure for the determination of individual PCBs, PCDDs and PCDFs in waters at very low concentrations.  相似文献

The principles, practicability and potential of comprehensive two-dimensional (2D) gas chromatography coupled to a rapid-scanning quadrupole mass spectrometer (GC x GC-qMS) for the analysis of complex flavour mixtures in food, allergens in fragrances and polychlorinated biphenyls (PCBs) were studied. With a scan speed of 10,000 amu/s, monitoring over a mass range of up to 200 atomic mass unit (amu) can be achieved at an acquisition frequency of 33 Hz. Extending this mass range and/or increasing the data acquisition frequency results in a loss of spectral quality. Optimal parameter settings allow, next to unambiguous identification/confirmation of target compounds on the basis of high-quality mass spectra, fully satisfactory quantification (three to four modulations per peak) with linear calibration plots and detection limits in the low-pg level. The potential of time-scheduled data acquisition to increase the effective mass range within one GC x GC run was also explored. The analyses, with baseline separation of the flavours, allergens and PCB target compounds, took less than 30 min.  相似文献

Polychlorinated dibenzo-p-dioxins and dibenzofurans in crude extracts of fly ash and flue gas from municipal waste incinerators were quantified using a comprehensive multidimensional gas chromatograph (GC x GC) coupled to a high-resolution time-of-flight mass spectrometer (HR-TOFMS). For identification and quantification, we developed our own program to prepare 3D chromatograms of selected mass numbers from the data of the GC x GC/HR-TOFMS. Isolation of all congeners with a TCDD toxic equivalency factor from the other isomers by only one injection was confirmed. The instrumental detection limit of TCDD on the GC x GC/HR-TOFMS was 0.9 pg by the relative calibration method. Quantification of these substances in the crude extracts was achieved by direct injection to the GC x GC/HR-TOFMS. The results agree with the values obtained using a generic gas chromatography/high-resolution mass spectrometry (GC/HRMS) system. It was confirmed that measurement by high-resolution TOFMS and GC x GC effectively reduces interference from other chemicals.  相似文献

The applicability of 13 different GC columns (Agilent HP-5MS, Restek Rtx-5MS, Rtx-Dioxin2, Supelco Equity 5, SP-2331, Varian VF-5MS, CP-Sil 8 CB LowBleed/MS, J&W Scientific DB-5, DB-225, DB-XLB, DB-5MS, Phenomenex ZB-5MS, and ZB-5UMS) for US Environmental Protection Agency (EPA) methods 1613b, 8290 and European Standard Method EN 1948 for measurement of polychlorinated dibenzo-p-dioxins (PCDDs) and polychlorinated dibenzofurans (PCDFs) has been evaluated for the separation of all International Toxic Equivalent Factor (I-TEF) isomers (tetra- through octachlorinated at 2,3,7,8 positions) from closely eluting isomers using high-resolution gas chromatography/high-resolution mass spectrometry (HRGC/HRMS). The relative performance data are compared based on mass chromatograms using a "visualization approach", absolute retention times, 2,3,7,8-substituted, total dioxins and furans concentrations, as well as TEQ comparisons. None of the columns tested were able to separate all 17 I-TEFs from other co-eluted isomers. Our data indicate that all I-TEFs isomers can be fully differentiated from closely eluting isomers using either of two sets of non-polar and polar stationary phase combinations. One set consists of DB-5 (HP-5MS, Rtx-5MS, Equity-5) and DB-225 GC columns and another set would have a combination of DB-5MS (ZB-5MS, VF-5MS, CP-Sil 8 CB LowBleed/MS) with SP-2331. However, depending on the source of PCDDs/PCDFs a laboratory could choose a single GC column that separates the 2,3,7,8-substituted congeners that contribute most significantly to the overall TEQ. These data are the most comprehensive to date, provide a valuable addition to operational criteria for the standard EPA methods 1613b, 8290, European Standard Method EN 1948 and will allow researches to compare data generated according to the different compliance analytical procedures.  相似文献

Comprehensive two-dimensional gas chromatography (GC x GC) with electron-capture detection (ECD) has been optimized for the separation of seventeen 2,3,7,8-substituted polychlorinated dibenzo-p-dioxins and dibenzofurans and 12 dioxin-like polychlorinated biphenyls, with emphasis on the selection of the first- and second-dimension, commercially available, columns. When eight second-dimension columns were subsequently combined with a 100% methylpolysiloxane stationary phase (DB-1) in the first dimension to create orthogonal conditions, a complete separation of all congeners with different TEF values was obtained with two column combinations, DB-1 x VF-23 and DB-1 x LC-50. When other types of first-dimension columns were used (and orthogonality was partly sacrificed), a DB-XLB column combined with 007-65HT, VF-23 and LC-50 was found to provide a complete separation of all 29 priority congeners. Next, the potential of these three column combinations for real-life analysis was preliminarily studied. With a spiked and fractionated milk extract, DB-XLB x LC-50 was found to be the most powerful column combination, because of the good separation of the 29 priority congeners from each other as well as from the matrix constituents. Quantitative performance (close to three-order linearity; LODs, 30-150 fg injected; R.S.D.s, 1.5-6.5% (n = 10)) was satisfactory.  相似文献

A sample mixture of polychlorinated biphenyls (PCBs) was measured by gas chromatography/multiphoton ionization/time-of-flight mass spectrometry (GC/MPI/TOF-MS) using four types of laser sources. When a fourth harmonic emission (266 nm) of a picosecond Nd:YAG laser (1064 nm) was utilized, highly chlorinated PCBs larger than hepta-CBs were not observed. A fifth harmonic emission (213 nm) of the picosecond Nd:YAG laser allowed the measurement of PCBs from di-CBs to octa-CBs, and the limit of detection (LOD) was several pg for each component of PCBs. The LOD for the total amount of PCBs, which was calculated using the protocol provided by the Ministry of the Environment, Japan, was 1000 pg. The signal intensity of the congeners with chlorine atoms at the ortho positions (non-coplanar PCBs) was enhanced by using the fifth harmonic emission. When the fourth harmonic emission remaining after fifth harmonic generation was simultaneously used, the LOD for total PCBs was improved to 667 pg. The PCB sample was also measured using a third harmonic emission (267 nm) of a femtosecond Ti:sapphire laser (800 nm), providing an LOD of 677 pg. Thus, the two-color beam (266/213 nm) of a picosecond Nd:YAG laser had a comparable, or even slightly superior, performance to the more expensive femtosecond Ti:sapphire laser.  相似文献

采用高分辨气相色谱/高分辨质谱(HRGC/HRMS)定量测定了底泥中的17种2,3,7,8位多氯代二噁英和呋喃(PCDD/Fs) ,并测定了四至八氯取代的二噁英和呋喃总量。样品经加速溶剂萃取,然后通过流体控制系统(FMS)自动过硅胶柱、氧化 铝柱和碳柱净化,最后浓缩。以HRGC/HRMS电压选择离子检测模式对样品中的PCDD/Fs进行了定性分析,采用同位素稀释 技术定量,该方法可精确定量到pg/g水平。结果表明该方法分析的17种二噁英和呋喃异构体的检出限可达0.1 pg/g。同 位素标准的回收率为49.8%~85.3%,样品中各异构体的回收率为93.2%~115.6%。该方法不但满足国际标准的要求,还大 大提高了分析速度,使分析周期从原来的2周缩短到2 d以内。  相似文献
