Acephate pesticide contamination in agricultural production has caused serious human health problems. Metal oxide semiconductor (MOS) gas sensor can be used as a portable and promising alternative tool for efficiently detection of acephate. In this study, hierarchical assembled SnO2 nanosphere, SnO2 hollow nanosphere and SnO2 nanoflower were synthesized respectively as high efficiency sensing materials to build rapid and selective acephate pesticide residues sensors. The morphologies of different SnO2 3D nanostructures were characterized by various material characterization technology. The sensitive performance test results of the 3D SnO2 nanomaterials towards acephate show that hollow nanosphere SnO2 based sensor displayed preferable sensitivity, selectivity, and rapid response (9 s) properties toward acephate at the optimal working temperature (300 °C). This SnO2 hollow nanosphere based gas sensor represents a useful tool for simple and highly effective monitoring of acephate pesticide residues in food and environment. According to the characterization results, particularly Brunauer-Emmett-Teller (BET) and Ultraviolet-Visible Spectroscopy (UV–vis), the obvious and fast response can be attributed to the mesoporous hollow nanosphere structure and appropriate band gap of SnO2 hollow nanosphere.  相似文献

Organophosphorus pesticides are commonly used in both agricultural and residential settings. The widespread use of these chemicals makes it almost impossible for humans to avoid exposure. In order to determine background human exposure, there is a need for fast, reliable, and sensitive analytical methods. We have developed a sensitive method to quantify specific biomarkers of the organophosphorus pesticides acephate, azinphos, chlorpyrifos, coumaphos, diazinon, isazofos, malathion, methamidophos, parathion and pirimiphos or their O,O-dimethyl analogues in human urine, as their selective metabolites or as the intact pesticide. Isotopically labeled internal standards were used for eight of the analytes. The use of labeled internal standards in combination with high-performance liquid chromatography electrospray ionization–tandem mass spectrometry provided a high degree of specificity. Repeated analysis of urine samples fortified with high and low concentrations of the analytes gave relative standard deviations (RSD) of less than 10% for the analytes with an isotopically labeled standard. Analytes without isotopically labeled standards had higher RSD. For all compounds except methamidophos and acephate, the recoveries were greater than 70%. The limits of quantification for most of the analytes were in the range of 0.1 to 1 ng/mL. We detected concentrations of most of these pesticides and/or their metabolites in urine samples from non-occupationally exposed persons using our method. Our frequencies of detection for the analytes measured ranged from 1% to 98%.  相似文献

A new, simple, precise and rapid high performance liquid chromatographic method was developed for the determination of meropenem in human serum, urine and pharmaceutical dosage forms. Chromatography was carried out on an LC(18) column using a mixture of 15 mM KH(2)PO(4):acetonitrile:methanol (84:12:4; v/v/v), adjusted to pH 2.8 with H(3)PO(4). The proposed method was conducted using a reversed-phase technique, UV monitoring at 307.6 nm and cefepime as an internal standard. The retention times were 5.98 and 7.47 min for cefepime and meropenem, respectively. The detector response was linear over the concentration range of 50-10,000 ng/mL. The detection limit of the procedure was found to be 22 ng/mL. The detection limit for meropenem in human plasma was 108.4 ng/mL and the corresponding value in human urine was 179.3 ng/mL. No interference from endogenous substances in human serum, urine and pharmaceutical preparation was observed. The proposed method is sufficiently sensitive for determination of the concentrations of meropenem and may have clinical application for its monitoring in patients receiving the drug.  相似文献

建立了一种基于液相色谱-串联质谱法(LC-MS/MS)定量分析微量有机磷农药残留的方法,并应用于烟草中农药残留物的定量检测。采用乙腈超声提取烟草中的有机磷农药残留,以甲醇-水(含0.1%乙酸铵)(体积比为95∶5)为流动相,经高效液相色谱分离,以串联质谱在多反应监测(MRM)模式下测定,在2.5 min内完成了甲胺磷、乙酰甲胺磷、乐果、敌百虫、毒死蜱5种常用有机磷农药的定量分析。5种农药在1~200 μg/L内的线性关系良好(r>0.998),平均回收率为77%~104%,检出限为1.0~5.0 μg/kg。  相似文献

A rapid and simple high-performance liquid chromatographic method with photodiode array detection was developed for the separation and the simultaneous determination of phenytoin and dextromethorphan in human urine. Analysis was performed in less than 4.5 min in isocratic mode on a reversed-phase C18 column (5 microm; 150 x 4.6 mm) using a mobile phase composed of acetonitrile-buffer phosphate 0.01 M (60:40, v/v) adjusted to pH 6.0, at 1 mL/min flow rate and UV absorbance at 210 nm. The elution order of analytes was dextromethorphan (DXM), Internal Standard (IS), and phenytoin (PHT). Calibration curves were linear in the 7.5-25 microg/mL range for PHT and in the 10-30 microg/mL range for DXM. Spike recoveries for urine samples prepared at three spiking levels ranged from 97.8 to 102.3% for PHT and from 94.8 to 100.4% for DXM. The detection limit (LOD) values ranged from 0.08 microg/mL for PHT to 0.5 microg/mL for DXM. The quantitation limit (LOQ) values ranged from 0.3 microg/mL for PHT to 1.6 microg/mL for DXM. The sample preparation method involves a rapid and simple procedure based on solid-phase extraction using a C18 reversed-phase column. Validation of the optimised method was carried out according to the ICH guidelines. The method developed in this study allows the reliable simultaneous analysis of PHT and DXM, drugs that were never quantified together in previously reported analytical methods. The described method has the advantage of being rapid and easy and it could be applied in therapeutic monitoring of these drugs in human urine of epileptic patients.  相似文献

A method was developed for the determination of molybdenum (Mo) in human urine by direct dilution of the sample in doubly distilled water with 1% HNO3 (v/v) and inductively coupled mass spectrometry (ICP-MS). In and Y were used as internal standards. Since (98)Mo provides a higher sensitivity, it was chosen as the reference isotope. The influence of different factors, such as sample dilution, HNO3 concentration and the stability of the analyte were evaluated. The detection limit (LOD) was assessed at 0.2 microg/L Mo, while the lower limit of quantification (LOQ) was 0.6 microg/L. Recoveries ranged between 97.2 and 100.7% from solutions containing from 10 to 50 microg/L Mo. Linear calibration curves were generated from 2.1 and 52.1 microg/L with coefficients of variation (CV ) ranging from 1.62 to 3.56%. In order to establish reference values (RV) for molybdenum, the procedure presented here was used to determine Mo in the urine of a population group living in Tuscany, Italy.  相似文献

The occurrence of pesticides in vegetables such as tomato poses an increasing attention to their potential risk to human health. In this study, acephate and its metabolite methamidophos were investigated for their transport and degradation in tomato cultivated in spiking nutrient solutions containing acephate at 1 mg/L. Acephate was taken up and transported via xylems with the concentrations of 41–145 μg/L in xylem sap at 2 and 8 cm above the roots in control treatment. The residue levels of methamidophos derived from acephate were in a range of 0–4.21 μg/L, being 0.00–3.73% of the parent acephate. The influence of three plant growth regulating substances, 1-naphthylacetic acid (NAA), glucose and phlorizin, on the transport and degradation of acephate and methamidophos in tomato was evaluated. All of these substances had positive effects on the transport of acephate and methamidophos in tomato xylem system. The NAA or glucose treatment promoted the degradation of acephate to methamidophos; however, with the addition of phlorizin, the concentrations of acephate were reduced in comparison to that under glucose treatment, suggesting that phlorizin would cause an inhibitory effect on the transport of acephate in tomato plant. These results may indicate an effective approach to reduce acephate and methamidophos residues in tomato.  相似文献

A simple liquid chromatography (LC) method was developed for determination of the therapeutic level of mizoribine in human plasma. After precipitation of plasma proteins with 6% perchloric acid, mizoribine was determined by LC with spectophotometric detection. The peak height for mizoribine was linearly related to its concentrations, which ranged from 0.09 to 3.13 microg/mL. Therefore, the limit of quantitation was considered to be 0.09 microg/mL. The accuracy was 104.96-107.37%. The intra- and interday relative standard deviation values were in the range of 1.10-3.25%. The detection limit was 0.025 microg/mL, defined as a signal-to-noise ratio of 3. The plasma concentrations of mizoribine were not related to the dosage. Because mizoribine was mainly excreted in the urine, the plasma concentrations of mizoribine might be affected by a change in renal function. Therefore, the mizoribine concentration in blood should be monitored and the dosage adjusted, depending on the condition of renal function. It was suggested that the present method may be applied well in the therapeutic drug monitoring for mizoribine.  相似文献

An innovative reversed-phase high-performance liquid chromatographic method is validated for the simultaneous determination of rofecoxib and celecoxib in human plasma. The internal standard is 4-n-pentyl-phenyl-acetic acid. Good chromatographic separation is achieved using a Zorbax SB-CN (5 microm) analytical column operated at room temperature and mobile phase consisting of acetonitrile and water containing 0.1M potassium dihydrogen orthophosphate buffer adjusted to pH 2.4 with 85% orthophosphoric acid (42:58, v/v). UV detection is performed at 254 nm, and the flow rate is maintained at 1.0 mL/min. Plasma samples are extracted into an organic solvent (1-chlorobutane) and evaporated under an air flow. The calibration curve for rofecoxib is linear over the range of 10 to 500 microg/L, and the celecoxib calibration curve is linear over the range of 20 to 2000 microg/L. The lower limit of quantitation for rofecoxib and celecoxib is 10 and 20 microg/L, respectively, using 1.0 mL of human plasma. The validation data show that the assay is sensitive, accurate, specific, and reproducible for the determination of rofecoxib and celecoxib. This method is therefore appropriate for pharmacokinetic studies to quantitate these therapeutic agents in patients with arthritis conditions.  相似文献

Health-care workers handling antineoplastic agents may be exposed to extremely low doses of these drugs. Very sensitive and specific analytical methods are therefore needed for biological monitoring. The aim of this study was to develop and validate a method for trace level determination of doxorubicin, epirubicin, daunorubicin and idarubicin in human urine, using epi-daunorubicin as an internal standard. Solid-phase extraction (SPE) was used for sample preparation. Urine samples were loaded onto Bond Elut C18 cartridges. The analytes were eluted in methylene chloride/2-propanol (1:1, v/v) and then evaporated to dryness. The residue was reconstituted with the mobile phase prior to high-performance liquid chromatography/tandem mass spectrometry (HPLC/MS/MS) analysis. Quantitation of each analyte was performed using the multiple reaction monitoring (MRM) method. The urine assay was linear over the range 0.1-2.0 microg/L, with a lower limit of quantification (LLOQ) of 0.10 microg/L for doxorubicin and epirubicin, and 0.03 microg/L for daunorubicin and idarubicin. The respective limits of detection (LODs) were 0.04 and 0.01 microg/L. The precision and accuracy of the assay were determined on three different days. The within-series precision was found to be always less than 13.9% for all the analytes. The overall precision expressed as relative standard deviation (RSD) was always less than 10.6%. The recovery of anthracyclines was assessed at two concentrations of the range tested (0.1 and 2.0 microg/L) and it ranged from 87.7% (daunorubicin) to 102.0% (doxorubicin) and from 79.1% (daunorubicin) to 90.7% (idarubicin) for the lower and the higher level, respectively, with a RSD always less than 9.1%. The uncertainty of the present assay was also evaluated and the combined uncertainty was always less than 20% over all the days of the validation study. This is the first method that makes use of LC/MS/MS for the biological monitoring of occupational exposure to anthracyclines.  相似文献

A simple, sensitive, and accurate method for the determination of propranolol in human urine has been developed based on solid-phase microextraction (SPME) followed by GC-flame ionization detection (FID). The sol-gel 5,11,17,23-tetra-tert-butyl-25,27-dihydroxy-26,28-diglycidyloxycalix[4]arene/hydroxy-terminated silicone oil (diglycidyloxy-C[4]/OH-TSO) fiber was prepared to accommodate to the harsh extraction conditions. It possesses excellent alkali-proof ability and retains its extraction characteristics intact even after treatment with highly alkaline (4 mol/L) NaOH solution. Direct chemical bonding of the coating to the fiber surface provides it with excellent solvent resistance and the introduction of calixarene enhances its thermal stability. The newly developed sol-gel calixarene coating was effectively used for the extraction of propranolol in human urine. No interference with the determination of propranolol was observed from the urine components. Standard curves were linear in the range 50-5000 microg/L for headspace-SPME (HS-SPME) and 25-25000 microg/L for direct-SPME (Dir-SPME) with correlation coefficients better than 0.9999. The detection limit was 0.275 microg/L for HS-SPME and 0.193 microg/L for Dir-SPME. The method was validated using standard addition methodology and recovery values were between 91.4 and 117% for both the sampling modes with the RSDs less than 6% at different concentration levels in the linear ranges. The results obtained by both the sampling modes were feasible, and no significant differences between them regarding accuracy, precision, and detection limits were seen.  相似文献

建立了高效液相色谱(HPLC)测定猪可食性组织中咪唑苯脲残留的方法。猪组织经β-葡萄糖苷酶水解后,用1 mol/L盐酸提取,再用正己烷-异戊醇(3:2, v/v)混合溶剂萃取净化。以乙腈和0.0075 mol/L戊烷磺酸钠水溶液(含0.1%三乙胺,pH 3.0)作为流动相,经C18反相色谱柱分离后用紫外检测器检测。结果表明: 该方法在咪唑苯脲含量为10~10000 μg/L范围内呈现良好的线性关系,相关系数大于0.999;空白组织中加标样品的检出限(LOD)为10 μg/kg,定量限(LOQ)为20 μg/kg。在定量限、最高残留限量(MRL)、2倍MRL添加水平下,不同组织的平均回收率为80.04%~110.32%,相对标准偏差为0.82%~10.00%。表明该检测方法简单、灵敏,适用于猪组织中咪唑苯脲残留的定量检测。  相似文献

A simple, rapid, and sensitive method was developed for detection and quantitation of lysine (Lys) in various biological samples by isocratic liquid chromatography (LC). Samples containing Lys and other amino acids were derivatized with 9-fluorenylmethyl chloroformate (FMOC-CI). The mobile phase used for isocratic elution was 50 mmol/L sodium acetate buffer (pH 4.20)-acetonitrile (43 + 57, v/v). Lys was detected with a UV detector at 265 nm. The derivatized Lys eluted from a LiChrospher 100 RP-18 (150 x 4.0 mm id) column at a retention time of 5.6 min. The limit of detection was 0.73 mumol/L (signal-to-noise [S/N] ratio, 3:1), and the limit of quantitation was 2.37 mumol/L (S/N ratio, 10:1). Lys recoveries from fortified biological samples were > 97.5%. Average Lys contents found in rumen fluid samples collected before the morning feeding and at 2.0, 4.0, and 6.0 h after feeding were 4.26, 3.34, 3.58, and 3.82 mumol/L, respectively. The hydrolysate of a sample of mixed rumen microorganisms collected before the morning feeding was determined to contain 1.372 mumol/mg microbial nitrogen in the form of Lys. The Lys concentrations of human plasma, goat plasma, human urine, and goat urine were 140.0, 102.0, 58.0, and 32.0 mumol/L, respectively.  相似文献

建立了反相高效液相色谱测定血浆中单硝酸异山梨酯浓度的方法。样品在碱性条件下经二氯甲烷提取后,用C18柱进行分离,以H2O(用0.03 mol/L氨水调pH至7.8)-乙腈(体积比为80∶20)为流动相,对乙酰氨基酚为内标,于230 nm处测定。结果表明,单硝酸异山梨酯的质量浓度为20~1000 μg/L时其峰面积与内标峰面积之比与单硝酸异山梨酯浓度有良好的线性关系。方法的最低检测质量浓度为12 μg/L,平均回收率为(97.11±2.45)%~(104.34±2.17)%,日内测定的相对标准偏差(RSD)≤2.52%,日间测定的RSD≤5.21%。  相似文献

A direct, rapid and selective method for the quantitative determination of the ethylenethiourea (ETU) in human urine has been validated and is reported in the present study. It allows the accurate quantification of ETU in this complex matrix without the use of any internal standard as the sample cleanup is effective enough for the removal of interferences that could lead to ion suppression in the electrospray ionization (ESI) source. This simple and rapid purification system, based on the use of a Fluorosil phase of a BondElut column followed by a liquid-liquid extraction procedure, achieves mean extracted recoveries, assessed at three different concentrations (2.5, 10.0, and 25.0 microg/L), always more than 85%. High-performance liquid chromatography (HPLC) with positive ion tandem mass spectrometry, operating in selected multiple reaction monitoring (MRM) mode, is used to quantify ETU in human urine. The assay is linear over the range 0-50 microg/L, with a lower limit of quantification (LOQ) of 1.5 microg/L and a coefficient of variation (CV) of 8.9%. The lower limit of detection (LOD) is assessed at 0.5 microg/L. The overall precision and accuracy were determined on three different days. The values for within- and between-day precision are < or = 8.3 and 10.1%, respectively, and the accuracy is in the range 97-118%. The relative uncertainties for the LOQ and QC concentrations have been estimated to be 18 and 8%, respectively. The assay was applied to quantify ETU in human urine from growers that regularly handle ethylenebisdithiocarbamate pesticides in large crop plantations. The biological samples were collected at the start and end of the working day, and the ETU urine levels were found to vary between 1.9 and 8.2 microg/L.  相似文献

A simple, selective, sensitive, accurate, and precise method was developed for determination of metformin hydrochloride (MF) in human urine using RP-HPLC. The method depends upon using an octylsilyl (C8) 5 microm particle size column at ambient temperature with mobile phase consisting of 33 mM sodium dihydrogen phosphate containing 6.38 mM hexanesulfonic acid sodium salt and adjusted to apparent pH 3.0 with phosphoric acid-acetonitrile (93 + 7, v/v) at a flow rate of 1.5 mL/min. Quantitation was achieved with UV detection at 231 nm based on peak area with a linear calibration curve over the concentration range of 0.01-50 microg/mL. The proposed method was applied to the determination of the urinary excretion pattern of MF (the cumulative amounts excreted were calculated without pretreatment of the urine sample) and for determination of the dissolution pattern of MF tablets. The proposed method was completely validated according to the U.S. Food and Drug Administration guidelines.  相似文献

Two multiresidue methods were developed for the determination of 15 pesticides (organochlorines, organphosphorus compounds, pyrethroids, and fungicides) in medicinal herbs Isatis indigotica Fort. and its formulations. The analytical procedure is based on ultrasonic assisted extraction and liquid-liquid extraction (LLE). After solvents were added, the raw material or granule sample was sonicated in an ultrasonic water bath and then centrifuged, filtered, and cleaned up by LLE. The infusion sample was extracted with petroleum ether by LLE. The pesticide residues were determined by capillary gas chromatography with electron-capture or flame photometric detection. Recoveries with the method at concentrations between 0.4 microg/kg and 10 mg/kg ranged from 70.2 to 119.5% for raw material, 73.2 to 105.1% for granule formulation, and 72.8 to 113.3% for infusion formulation. The relative standard deviation values were <20% for all of the pesticides studied. The pesticide detection limits were within the ranges 0.3-0.5 microg/L for endosulfan, 3-7.5 microg/L for pyrethroids, 0.7-32.5 microg/L for organophosphorus pesticides, and 0.1-0.6 microg/L for the other pesticides. The proposed methods are simple and rapid and provide simultaneous determination of pesticide residues in Isatis indigotica Fort. with acceptable recoveries and repeatability and an adequate limit of determination.  相似文献

以4-氯-7-硝基苯并-2-氧杂-1,3-二唑(NBD-Cl)为柱前衍生试剂,建立了胶束电动色谱-激光诱导荧光检测法测定肌松弛药巴氯芬(BAL)的新方法。经过实验条件的优化,采用15 mmol/L硼砂、20 mmol/L十二烷基硫酸钠、10%(v/v)乙腈、pH 9.75的缓冲体系,在分离电压为17.5 kV、柱温为25 ℃的条件下,压力进样3.45 kPa(0.5 psi)×3 s,巴氯芬及其内标物的衍生产物在7 min内实现较好的基线分离,线性范围为0.025~25 mg/L,相关系数为0.9999,检出限(S/N=3)和定量限(S/N=10)分别为0.90 μg/L和6.25 μg/L。该方法被应用于巴氯芬制剂及加入巴氯芬对照品的尿液样品分析,回收率范围分别为101.6%~107.9%和107.0%~109.6%。该方法有望应用于巴氯芬药物制剂的质量监控以及为巴氯芬药物代谢的研究提供辅助手段。  相似文献

建立了对牛奶中氯霉素的残留量进行检测的高效液相色谱-荧光检测方法。氯霉素还原后在温和条件下与荧光胺发生衍生化反应,采用十八烷基键合硅胶固定相,以乙腈/四氢呋喃/0.02 mol/L醋酸钠-醋酸缓冲液(pH 6.0)(体积比为16∶8∶76)为流动相,流速1.0 mL/min,柱温40 ℃,荧光检测激发波长为410 nm,发射波长为508 nm。在上述实验条件下,氯霉素检测的线性范围为0.4~800 μg/L (r2=0.9999),检出限为0.2 μg/L。当空白样品中氯霉素添加水平为2~40 μg/L时,该方法的回收率为66.6%~92.8%,相对标准偏差为4.5%~9.4%。该方法适用于牛奶中氯霉素痕量残留的监测,具有干扰小、选择性好、灵敏度高等优点。  相似文献

In the present work, we have developed a simple and rapid liquid chromatography/mass spectrometry (LC/MS) method for the identification and quantification of vitamin B5 in human urine. Urine was spiked with vitamin B5 internal standard, hopantenic acid (HOPA), and then diluted with the LC mobile phase prior to its analysis by LC/MS. The quantification was performed in single ion monitoring mode. The calibration curve was linear (r2 = 0.999) between 0.25 to 10 microg/mL. With a limit of detection of 0.1 microg/mL the method was sensitive enough to determine low levels of vitamin B5 in urine. The overall quantitative efficiency of the method was evaluated by spiking urine samples with four different concentrations of vitamin B5; the intra-assay coefficient of variation was below 5% and the recoveries were between 96 to 108%. The results of the present study show that the proposed method is selective and sensitive enough for the quantification of vitamin B5 in urine.  相似文献
