The thermal and structural properties of binary blends of Nylon-6 (N6) and a chemically related biopolymer, Bombyx mori silk fibroin (SF), are reported in this work. Homopolymers and blends, in composition ratios of N6/SF ranging from 95/05 to 70/30, were investigated by thermogravimetric (TG) analysis, differential scanning calorimetry (DSC), Fourier transform infrared (FTIR) spectroscopy and wide angle X-ray scattering (WAXS). Silk fibroin typically degrades at temperatures just above 210°C, which occurs within the melting endotherm of N6. In TG studies, the measured mass remaining was slightly greater than expected, indicating the blends had improved thermal stability. No beta sheet crystals of SF were detected by FTIR analysis of the Amide I region. Strong interaction between N6 and SF chains was observed, possibly as a result of formation of hydrogen bonds between N6 and SF chains. DSC analysis showed that the addition of SF to N6 caused a decrease in the crystallization temperature, the melting temperature of the lowest melting crystals and the crystallinity of N6. Furthermore, the α-crystallographic phase dominates and the γ-crystallographic phase was not observed in N6/SF blends, in contrast to the homopolymer N6, which contains both phases. We suggest that the addition of SF might result in changes of the chain extension of N6, which lead to the appearance of α-rather than γ-phase crystals.  相似文献

Bis‐β‐ketoimine ligands of the form [(CH2)n{N(H)C(Me)?CHC(Me)?O}2] (LnH2, n=2, 3 and 4) were employed in the formation of a range of gallium complexes [Ga(Ln)X] (X=Cl, Me, H), which were characterised by NMR spectroscopy, mass spectrometry and single‐crystal X‐ray diffraction analysis. The β‐ketoimine ligands have also been used for the stabilisation of rare gallium hydride species [Ga(Ln)H] (n=2 ( 7 ); n=3 ( 8 )), which have been structurally characterised for the first time, confirming the formation of five‐coordinate, monomeric species. The stability of these hydrides has been probed through thermal analysis, revealing stability at temperatures in excess of 200 °C. The efficacy of all the gallium β‐ketoiminate complexes as molecular precursors for the deposition of gallium oxide thin films by chemical vapour deposition (CVD) has been investigated through thermogravimetric analysis and deposition studies, with the best results being found for a bimetallic gallium methyl complex [L3{GaMe2}2] ( 5 ) and the hydride [Ga(L3)H] ( 8 ). The resulting films ( F5 and F8 , respectively) were amorphous as‐deposited and thus were characterised primarily by XPS, EDXA and SEM techniques, which showed the formation of stoichiometric ( F5 ) and oxygen‐deficient ( F8 ) Ga2O3 thin films.  相似文献

Currently, there are no reliable biomarkers available that can aid early differential diagnosis of reactive arthritis (ReA) from other inflammatory joint diseases. Metabolic profiling of synovial fluid (SF)—obtained from joints affected in ReA—holds great promise in this regard and will further aid monitoring treatment and improving our understanding about disease mechanism. As a first step in this direction, we report here the metabolite specific assignment of 1H and 13C resonances detected in the NMR spectra of SF samples extracted from human patients with established ReA. The metabolite characterization has been carried out on both normal and ultrafiltered (deproteinized) SF samples of eight ReA patients (n = 8) using high-resolution (800 MHz) 1H and 1H─13C NMR spectroscopy methods such as one-dimensional 1H CPMG and two-dimensional J-resolved1H NMR and homonuclear 1H─1H TOCSY and heteronuclear1H─13C HSQC correlation spectra. Compared with normal SF samples, several distinctive 1H NMR signals were identified and assigned to metabolites in the 1H NMR spectra of ultrafiltered SF samples. Overall, we assigned 53 metabolites in normal filtered SF and 64 metabolites in filtered pooled SF sample compared with nonfiltered SF samples for which only 48 metabolites (including lipid/membrane metabolites as well) have been identified. The established NMR characterization of SF metabolites will serve to guide future metabolomics studies aiming to identify/evaluate the SF-based metabolic biomarkers of diagnostic/prognostic potential or seeking biochemical insights into disease mechanisms in a clinical perspective.  相似文献

The cationic complex [(JohnPhos–Au)3(acetylide)][SbF6] (JohnPhos=(2-biphenyl)di-tert-butylphosphine, L1) has been characterised structurally and features an acetylide–trigold(I)–JohnPhos system; the trinuclear–acetylide unit, coordinated to the monodentate bulk phosphines, adopts an unprecedented μ,η121 coordination mode with an additional interaction between distal phenyl rings and gold centres. Other cationic σ,π-[(gold(I)L1)2] complexes have also been isolated. The reaction of trimethylsilylacetylene with various alcohols (iPrOH, nBuOH, n-HexOH) catalysed by cationic [AuIL1][SbF6] complexes in CH2Cl2 at 50 °C led to the formation of acetaldehyde acetals with a high degree of chemo- and regioselectivity. The reaction mechanism was studied, and several organic and inorganic intermediates have been characterised. A comparative study with the analogous cationic [CuIL1][PF6] complex revealed different behaviour; the copper metal is lost from the coordination sphere leading to the formation of cationic vinylphosphonium and copper nanoparticles. Additionally, a new catalytic approach for the formation of this high-value cationic vinylphosphonium has been established.  相似文献

Abstract

In the study, we treated C6 rat glioma cells with 25?mg/ml Dulcitol for 24?h. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assays were used to detect cellular growth. The measurements of the superoxide dismutase (SOD), malondialdehyde (MDA) and catalase (CAT) were used to assess oxidative stress level. Western was performed to detect the autophagy and apoptosis expression. The data showed that Dulcitol significantly decreased the cell viability, upregulated the Bax level in mitochondria and the Cytochrome C level in cytoplasm, and downregulated anti-apoptotic protein Bcl-xl. Moreover, it enhanced MDA level, reduced CAT and SOD activities, decreased LC3-II/LC3-I ratio, and increased P62 expression. However, rapamycin increased autophagy level and cell viability, and decreased ROS in Dulcitol treated C6 cells. Moreover, Dulcitol inhibited the glioma growth and enhanced survival in vivo. These results suggest that Dulcitol evidently increase cellular ROS levels and apoptosis in glioma cells, which can be significantly regulated by autophagy.  相似文献

Recently, Bombyx mori silk fibroin (SF) has been shown to be a suitable material for vascular prostheses for small arteries. In this study, we developed a softer SF graft by coating water-dispersed biodegradable polyurethane (PU) based on polycaprolactone and an SF composite sponge on the knitted SF vascular graft. Three kinds of 13C solid-state nuclear magnetic resonance (NMR), namely carbon-13 (13C) cross-polarization/magic angle spinning (MAS), 13C dipolar decoupled MAS, and 13C refocused insensitive nuclei enhanced by polarization transfer (r-INEPT) NMR, were used to characterize the PU-SF coating sponge. Especially the 13C r-INEPT NMR spectrum of water-dispersed biodegradable PU showed that both main components of the non-crystalline domain of PU and amorphous domain of SF were highly mobile in the hydrated state. Then, the small-diameter SF artificial vascular grafts coated with this sponge were evaluated through implantation experiments with rats. The implanted PU-SF-coated SF grafts showed a high patency rate. It was confirmed that the inside of the SF grafts was covered with vascular endothelial cells 4 weeks after implantation. These results showed that the water-dispersed biodegradable PU-SF-coated SF graft created in this study could be a strong candidate for small-diameter artificial vascular graft.  相似文献

Insoluble complexes are formed in acidic aqueous media when poly(acrylic acid) (PAA) and poly-(vinylbenzo-18-crown-6) (P18C6) or polyvinylbenzoglymes are mixed. Complex formation results from hydrogen bonding between carboxyl groups and crown ether- or glyme–oxygen atoms as well as from hydrophobic interactions. The precipitation is pH dependent and was determined as a function of the ratio PAA to P18C6 or to polyglyme at different HCl concentrations in 10?4M solutions of polycrown or polyglyme. Precipitation is nearly quantitative in 0.01N HCl. The compositions of PAA/P18C6 precipitates were determined as a function of the initial PAA/P18C6 ratio in solution. The complexes with P18C6 can be solubilized in acidic media when crown-complexable cations (K+, Cs+, Ba2+) are added, but the charged P18C6 reprecipitates in basic solution as a polysalt complex with the PAA–polyanion. More stable PAA–P18C6 complexes in the form of fibers can be obtained by interfacial complex formation. Poly(methacrylic acid) is less effective as a complex former.  相似文献

A Bacillus subtilis isolate was shown to be able to produce extracellular protease in solid-state fermentations (SSF) using soy cake as culture medium. A significant effect of inoculum concentration and physiological age on protease production was observed. Maximum activities were obtained for inocula consisting of exponentially growing cells at inoculum concentrations in the range of 0.7–2.0 mg g−1. A comparative study on the influence of cultivation temperature and initial medium pH on protease production in SSF and in submerged fermentation (SF) revealed that in SSF a broader pH range (5–10), but the same optimum temperature (37°C), is obtained when compared to SF. A kinetic study showed that enzyme production is associated with bacterial growth and that enzyme inactivation begins before biomass reaches a maximum level for both SF and SSF. Maximum protease activity and productivity were 960 U g−1 and 15.4 U g−1 h−1 for SSF, and 12 U mL−1 and 1.3 U mL−1 h−1 for SF. When SSF protease activity was expressed by volume of enzyme extract, the enzyme level was 10-fold higher and the enzyme productivity 45% higher than in SF. These results indicate that this bacterial strain shows a high biotechnological potential for protease production in solid-state fermentation.  相似文献

Two new steroidal alkaloids, neoverapatuline ( 1 ) and (1β,3α,5β)‐1,3‐dihydroxyjervanin‐12‐en‐11‐one ( 2 ), together with the four known compounds, veratramine ( 3 ), rubijervine ( 4 ), veratrosine ( 5 ), and veratroylzygadenine ( 6 ), were isolated from the roots and rhizomes of Veratrum nigrum L. Their structures were established through combined analyses of physicochemical properties and spectroscopic evidence. All compounds 1 – 6 were tested for their cytotoxicities in vitro against the human glioma cell line SF188.  相似文献

Reaction of [Ru6C(CO)16]2− with an excess of AgX (X = Cl, Br or I) affords heteronuclear clusters of formula [{Ru6C(CO)16Ag2X}2]2− in 80% yield, which for X = I and X = Br/Cl were crystallographically characterised. The formation of the cluster was followed in solution using electrospray ionisation mass spectrometry (ESI-MS), which revealed the presence of a wide range of clusters with the general formula [{Ru6C(CO)16} x Ag y X z ](2x−y+z)− where x = 1 or 2, y = 1, 2, 3 or 4 and z = 0, 1 or 2. The high yield of the product despite the evident complicated solution speciation is attributed to selective crystallisation of the observed compound driving the equilibrium toward this product.  相似文献

Silk fibroin (SF) with good biocompatibility and degradability has great potential for tissue engineering. However, the SF based scaffolds lack the electroactivity to regulate the myogenic differentiation for the regeneration of muscle tissue, which is sensitive to electrical signal. Herein, a series of electroactive biodegradable scaffolds based on SF and water‐soluble conductive poly(aniline‐coN‐(4‐sulfophenyl) aniline) (PASA) via a green method for skeletal muscle tissue engineering are designed. SF/PASA scaffolds are prepared by vortex of aqueous solution of SF and PASA under physiological condition. Murine‐derived L929 fibroblast and C2C12 myoblast cells are used to evaluate cytotoxicity of SF/PASA scaffolds. Moreover, myogenic differentiation of C2C12 cells is investigated by analyzing the morphology of myotubes and related gene expression. These results suggest that electroactive SF/PASA scaffolds with a suitable microenvironment, which can enhance the myogenic differentiation of C2C12 cells, have a great potential for skeletal muscle regeneration.  相似文献

Treatment of 1,8‐bis(diphenylphosphino)naphthalene (dppn, 1 ) with stoichiometric amounts of sulfur or selenium in toluene at 80 °C selectively afforded the diphosphine monochalcogenides 1‐Ph2P(C10H6)‐8‐P(:S)Ph2 (dppnS, 2 a ) and 1‐Ph2P(C10H6)‐8‐P(:Se)Ph2 (dppnSe, 2 b ). The 31P{1H} NMR spectrum of 2 b showed an unusually large 5J(P–Se) value, which indicates a significant through‐space coupling component. The monosulfide acted as a bidentate P,S‐ligand towards platinum(II) ( 3 a ), whereas the corresponding monoselenide complex ( 3 b ′) lost elemental selenium with formation of the previously reported complex [PtCl2(dppn)‐P,P′] ( 3 ). Treatment of dppnSe with [(nor)Mo(CO)4] (nor = norbornadiene) led to formation of [(dppnSe)Mo(CO)4P,Se] ( 3 b ). Solutions of the latter slowly deposited Se with formation of [(dppn)Mo(CO)4P,P′] ( 4 ) which was also obtained by independent synthesis from 1 and [(nor)Mo(CO)4]. All isolated new compounds were characterised by a combination of 31P, 1H, 13C and 77Se ( 2 b ) NMR spectroscopy, IR spectroscopy, mass spectrometry and elemental analysis. Single‐crystal X‐ray structure determinations were performed for dppnSe ( 2 b ), [PtCl2(dppnS)‐P,S] ( 3 a ), [(dppnSe)Mo(CO)4P,Se] ( 3 b ) and [(dppn)Mo(CO)4P,P′] ( 4 ). In 2 b steric effects cause the naphthalene ring to be distorted and force the phosphorus atoms by 65 and 59 pm to opposite sides of the best naphthalene plane. In the metal complexes 3 a , 3 b and 4 the phosphino‐phosphinochalcogenyl systems act as bidentate ligands through the P and the chalcogen atoms. The naphthalene systems are again distorted. The two independent molecules of 4 differ in their conformations.  相似文献

The chemistry and short lifetimes of metal-based anti-cancer drugs can be turned into an advantage for direct injections into tumors, which then allow the use of highly cytotoxic drugs. The release of their less toxic decomposition products into the blood will lead to decreased toxicity and can even have beneficial effects. We present a ternary VV complex, 1 ([VOL1L2], where L1 is N-(salicylideneaminato)-N′-(2-hydroxyethyl)ethane-1,2-diamine and L2 is 3,5-di-tert-butylcatechol), which enters cells intact to induce high cytotoxicity in a range of human cancer cells, including T98g (glioma multiforme), while its decomposition products in cell culture medium were ≈8-fold less toxic. 1 was 12-fold more toxic than cisplatin in T98g cells and 6-fold more toxic in T98g cells than in a non-cancer human cell line, HFF-1. Its high toxicity in T98g cells was retained in the presence of physiological concentrations of the two main metal-binding serum proteins, albumin and transferrin. These properties favor further development of 1 for brain cancer treatment by intratumoral injections.  相似文献

In search for antitumor metal-based drugs that would mitigate the severe side-effects of cisplatin, Ru(II) complexes are gaining increasing recent interest. In this work, we report on the synthesis, characterization (1H- and 13C-NMR, FT-IR), and cytotoxicity studies of two new half-sandwich organometallic Ru(II) complexes of the general formula [Ru(η6-arene)(XY)Cl](PF6) where arene?=?benzene or toluene and XY?=?bidentates: dipyrido[3,2-a:2′,3′-c]phenazine (dppz) or 2-(9-anthryl)-1H-imidazo[4,5-f][1,10]phenanthroline (aip), which are bound to Ru(II) via two phenanthroline-N atoms in a characteristic “piano-stool” configuration of Ru(II)-arene complexes—as confirmed by vibrational and NMR spectra. In addition, cytotoxic studies were performed for similar half-sandwich organometallic [Ru(η6-p-cymene)(Me2dppz)Cl]PF6 complex (Me2dppz = 11,12-dimethyl-dipyrido[3,2-a:2′,3′-c]phenazine). This study is complemented with elaborate modeling with density functional theory (DFT) calculations, which provided insight into reactive sites of Ru(II) structures, further detailed by molecular docking on the B-DNA dodecamer, which identified binding sites and affinities: most pronounced for the [Ru(η6-benzene)(aip)Cl](PF6) in both A-T and G-C regions of the DNA minor groove. Cytotoxic activity was probed versus tumor cell lines B16, C6, and U251 (B16 mouse melanoma, C6 rat glioma, U251 human glioblastoma) and non-tumor cell line HACAT (HACAT normal human keratinocytes).  相似文献

A series of novel 6-substituted 2-(benzo[4,5]imidazo[1,2-c]quinazolin-5(6H)-yl)acetic acids were synthesized and characterized by 1H, 13C, 19F NMR, 1H-1H-COSY, 1H-13C-HSQC, NOESY, LC-MS, IR, and elemental analysis. The mitotic toxicity of the synthesized compounds was determined according to the Allium test procedure. The 2-(6-(pentafluorophenyl)benzo[4,5]imidazo[1,2-c]quinazolin-5(6H)-yl)acetic acid inhibited mitotic spindle formation, which resulted in significant cytotoxic effect for meristematic cells of Allium cepa l . roots. In a preliminary antimicrobial evaluation, only Streptococcus pyogenes and Candida albicans were slightly susceptible to some of the synthesized compounds.  相似文献

The stability constants of the [Cs(DB18C6)]+ complex (DB18C6 is dibenzo-18-crown-6, L) in hydrophobic ionic liquids (room-temperature ionic liquids, RTIL) trioctylmethylammonium salicylate ([TOMA][Sal]), tetrahexylammnoium dihexylsulfosuccinate ([THA][DHSS]), and 1-butyl-3-methylimidazolium bis[(trifluoromethyl)sulfonyl]imide ([BMIM][N(Tf)2], as well as of the [Cs(18C6)2]+ complex in [BMIM][N(Tf)2], were measured by 133Cs NMR in the temperature range 27–57°C. The changes in the enthalpy and entropy of complex formation were determined. A linear correlation was revealed between logK 1 and the extraction factor logD CsDB18C6 for the cesium extraction from an aqueous solution into the RTIL.  相似文献

In the present study, a series of chiral C2‐symmetric ferrocenyl based binuclear η6‐benzene‐Ru(II) complexes bearing diphenylphosphinite and diisopropylphosphinite moieties have been synthesised. The new binuclear η6‐benzene‐Ru(II)‐phosphinite complexes were characterised based on nuclear magnetic resonance (1H, 13C, 31P–NMR), FT‐IR spectroscopy and elemental analysis. Then, these complexes have been screened as catalytic precursors in the transfer hydrogenation of acetophenone with 2‐propanol as both the hydrogen source and solvent in the presence of KOH. The corresponding optically active secondary alcohols were obtained in excellent conversion rates between 96 and 99% and moderate to good enantioselectivities (up to 78% ee). The complex 5 was the most efficient catalyst among the four new complexes investigated herein.  相似文献

A conductance study of the interaction between Pb2+ ion and 18-crown-6 (18C6), benzo-18-crown-6 (B18C6), dicyclohexyl-18-crown-6 (DC18C6), aza-18-crown-6 (A18C6), diaza-18-crown-6 (DAI8C6), dibenzopyridino-18-crown-6 (DBPy18C6), and dibenzyldiaza-18-crown-6 (DBzDA18C6) in acetonitrile–dimethyl sulfoxide mixtures was carried out at various temperatures. The formation constants of the resulting 1:1 complexes were determined from the molar conductance–mole ratio data and found to vary in the order DA18C6 > A18C6 > DBzDA18C6 > DC18C6 > 18C6 > B18C6 > DBPy18C6. The enthalpy and entropy of complexation reactions were determined from the temperature dependence of the formation constants. In all cases, the resulting complexes are enthalpy stabilized, but entropy destabilized. A linear relationship is observed between log K f of different complexes and mole fraction of acetonitrile in the solvent mixtures. The TS 0 vs. H 0 plot of all thermodynamic data obtained shows a fairly good linear correlation indicating the existence of an enthalpy–entropy compensation in the complexation reactions.  相似文献

Titanium dioxide (TiO2) is thought to be a photocatalytic agent excited by UV light. Our aim was to investigate the photocatalytic antitumor effect of water-dispersed TiO2 nanoparticles on C6 rat glioma cells and to evaluate the treatment responses by the spheroid models. Water-dispersed TiO2 nanoparticles were constructed by the adsorption of chemical modified polyethylene glycol (PEG) on the TiO2 surface (TiO2/PEG). Each monolayer and spheroid of C6 cells was coincubated with various concentrations of TiO2/PEG and subsequently irradiated with UV light. Damage of the cells and spheroids was evaluated sequentially by staining with the fluorescent dyes. The cytotoxic effect was correlated with the concentration of TiO2/PEG and the energy dose of UV irradiation. More than 90% of cells were killed after 13.5 J cm−2 of UV irradiation in the presence of 500 μg mL−1 TiO2/PEG. The irradiated spheroids in the presence of TiO2/PEG showed growth suppression compared with control groups. In TiO2/PEG-treated spheroids, the number of Annexin V-FITC-stained cells gradually increased during the first 6 h, and subsequently propidium iodide-stained cells appeared. The results of this study suggest that newly developed photoexcited TiO2/PEG have antitumoral activity. Photodynamic therapy utilizing this material can be a clue to a novel therapeutic strategy for glioma.  相似文献
