1,3-二苄基咪唑啉-2-酮-顺-4,5-二羟酸经脱水、单酯化,折分成分(4S,5R)-顺1,3-二苄基-5-甲氧基羧基-2-氧代咪唑啉-4-羧酸,再经还原环合,硫代成(3aS,6aR)-1,3-二苄基-四氢-4H-噻吩并[3,4-d)咪唑-2,4(1H)-二酮(7),后者经格氏反应、脱水、还原、裂解环合一锅合成(3aR,8aS,8bS)-1,3-二苄基-2-氧代十氢咪唑[3,4-d]噻吩并[1,2-a]锍翁溴化物(11),继而经缩合、开环、水解、脱羧、脱苄而得d-生物素(1),总收率为14.5%。 相似文献

以二异丁基氢化铝使(3aS,6aR)-1,3-二苄基-四氢-4H-噻吩并[3,4-d]-咪唑-2,4(1H)-二酮(1)高立体选择性地还原成(3aS,4S,6aR)-1,3-二苄基-4-羟基-四氢-1H-噻吩并[3,4-d]-咪唑-2(3H)-酮(2),再经缩合,Witting烯化成(3aS,6aR)-1,3-二苄基-四氢-1H-噻吩并[3,4-d]咪唑-2(3)-酮-4-烯戊酸(4),后者经催化氢转移还原、脱苄即得d-生物素,以化合物1计算总产率为42%. 相似文献

通过合成得到了一系列新颖的苯并[b]氧杂棘-5(2H)-酮类化合物单晶,结构经~1HNMR,~(13)CNMR和HREIMS确证,E-7-[(2,5-二羟基苯基)甲亚基]氨基-3,4-二氢-苯并[b]氧杂棘-5(2H)-酮(8k)和E-7-[(2,3,4-三羟基苯基)亚基]氨基-3,4-二氢-苯并[b]氧杂棘-5-(2H)-酮(8n)的结构经单晶X衍射方法进一步确证.系统测试了化合物对蛋白酪氨酸激酶(PTKs),如ErbB1,ErbB2,c-Met,ALK,FGFR1,RET和KDR等的抑制活性,结果表明含有邻苯二酚片段的化合物对蛋白酪氨酸激酶具有显著的抑制活性,其中E-7-[(3,4-二羟基苯基)甲亚基]氨基-3,4-二氢-苯并[b]氧杂棘-5(2H)-酮(8i)对ErbB1和ErbB2的IC50分别为1.0和0.33μmol/L,8n对RET的IC_(50)为0.7μmol/L,7-[(3,4-二羟基苯基)甲基]氨基-2,3,4,5-四氢-苯并[b]氧杂棘-5-醇(10b)对ErbB2的IC_(50)为1.02μmol/L. 相似文献

Seventeen novel 3-methyl-4-oxo-3,4-dihydroimidazo[5,1-d][1,2,3,5]tetrazine-8-carboxylate and -carboxamide derivatives were synthesized and evaluated for their growth inhibition in seven human solid tumor and a human leukemia HL-60 cell lines. Compound IVa showed more activity than the other compounds and the positive control temozolomide. In the presence of 40 μg/mL of IVa, the survival rate of all tested tumor cells was less than 10%. Esters displayed more potent antitumour activity than amides and temozolomide against HL-60 cells. These compounds also exhibited considerably enhanced water-solubility. 相似文献

Ethyl 5-amino-3-trifluoromethyl-1H-pyrazole-4-carboxylate (1) was efficiently synthesized via the condensation of ethyl cyanoacetate and trifluoroacetic anhydride, followed by chloridization and the condensation with aqueous hydrazine. Its unique reactivity was exploited for the synthesis of trifluoromethylated pyrazolo[1,5-a]pyrimidine (5) and pyrazolo[5,1-d][1,2,3,5]tetrazine-4(3H)-ones (8a-j). Among them, 5 was firstly found to be a novel fluorescent molecule that might be exploited as an attractive fluorophore for possessing many binding sites, and its fluorescence intensity was obviously stronger than its methyl analogue. 8 were found to be a new chemical class of potential monocotyledonous Echinochloa crus-galli L. Beauv inhibitors, and were more active than their methyl analogues. 相似文献

A novel, efficient, microwave assisted route for the synthesis of 6-substituted-2,3,4-trihydropyrimido[1,2-c]-9,10,11,12-tetrahydrobenzo[b]thieno[3,2-e]pyrimidines in good yields has been developed. The intermediates, 2-substituted-4-[3-hydroxy(propyl-1-amino)]5,6,7,8-tetrahydrobenzo[b]thieno[2,3-d]pyrimidines were obtained by irradiating 2-substituted-4-chloro-5,6,7,8-tetrahydrobenzo[b]thieno[2,3-d]pyrimidines with 1-amino-propanol under basic conditions in a microwave oven. 4-Chlorothieno[2,3-d]pyrimidines were synthesized by microwave irradiation of equimolar mixture of 4-hydroxythieno[2,3-d]pyrimidines and phosphorus oxychloride. The final compounds were screened for antibacterial activity by Kirby Bauer's method using amicacin as the standard against various gram positive and gram negative bacteria. All the compounds showed antibacterial activity comparable with the standard. 相似文献

Some 3-substituted-5,6-dihydro[1,8]naphthyrido[3,2-c][1,8]naphthyridines (V) were obtained by the condensation of 7-substituted-2,3-dihydro-1,8-naphthyridin-4-(1H)ones (IV) with 2-aminonicotinaldehyde. These Mannich bases is also possible to prepare by reaction of 7(5)-hydroxymethyl-3-methyl-4,7(5)dihydro-3H-imidazo[4,5-d][l,2,3]triazin-4-one
with amines. The compounds obtained exist in solutions as mixtures of N5 and N7 isomers, and the fraction of the latter grows with the polarity of the solvent. 相似文献
