For selective dissolution of Mn oxides as components of soil (easily reducible oxides) 0.05 mol L(-1) and 0.1 mol L(-1) NH2OH.HCl acidified to pH 2 and for dissolution of Fe oxides (crystalline form of the oxides) 0.25 mol L(-1) NH2OH.HCl in 25% acetic acid, 0.2 mol L(-1) oxalate buffer and 0.1 mol L(-1) ascorbic acid in oxalate buffer were tested. Dissolved elements were determined by ICP-MS and ICP-OES. The studies indicate that the use of 0.05 mol L(-1) NH2OH.HCl in nitric acid solution (pH 2) and 0.1 mol L(-1) ascorbic acid in 0.2 mol L(-1) oxalate buffer led to selective leaching of trace metals bound by Mn and Fe oxide phases in soil. Comparison of different extraction schemes indicates that the trace elements investigated are mainly bound to minerals consisting of crystalline Fe oxides and insoluble minerals (under the extraction conditions used). The studies also indicate that Mn oxides and organic matter retain a major mobile fraction of the elements investigated in the soil.  相似文献

贝类体内麻痹性贝类毒素的提取方法研究   总被引:3,自引:1,他引:2  
采用浓度系列为0.04、0.07、0.10、0.15、0.20、0.25、0.30、0.40、0.50、0.70、1.0 mol/L的HCl和HAc溶液作为提取液,分别取10 mL提取液与10 g栉孔扇贝性腺混合,在沸水浴中加热5 min提取麻痹性贝类毒素(PSP);同时采用0.3 mol/L HAc和0.2 mol/L HCl,于冰水浴中进行超声波提取麻痹性贝类毒素5~30 min。提取完成后将混合物于4℃冷冻离心机内离心5 min(3500 r/min),取上清液并以0.1 mol/L NaOH或5 mol/L HCl调整至pH为2.0~4.0。经超滤膜过滤后的提取液以高效液相色谱柱后衍生荧光检测法进行毒素分析,研究毒素组分间的转化关系和提取效率,并与超声波提取法进行了比较。结果表明,采用0.04~0.25 mol/L HCl和0.04~1.0 mol/L HAc从贝肉中提取PSP毒素,各毒素组分浓度差异不大,当HCl浓度大于0.25 mol/L时,N-磺酰氨甲酰基类毒素C1浓度急剧降低,HCl浓度大于0.5 mol/L时,N-磺酰氨甲酰基类毒素C2和GTX5浓度急剧降低,三者在酸度过大的情况下分解或转化为膝沟藻毒素-2(GTX2),膝沟藻毒素-3(GTX3)和石房蛤毒素(STX)。在相同浓度酸的情况下,超声波提取液中C1毒素的浓度显著低于沸水浴提取法,但C2的浓度略高于沸水浴提取液。  相似文献

The kinetics and mechanism of troilite oxidation by H(2)O(2) was studied at temperatures of 25 and 45 degrees C. Solutions within the range 0.1-0.85 mol L(-1) H(2)O(2) in HClO(4) (0.01-0.1 mol L(-1)) were used as dissolution media. The experimental amount of dissolved iron was plotted versus t(n), with n ranging from 0.25 to 1.55. The theoretical interpretation of this dependence suggests that the troilite oxidation involves several processes: Both experimental results and theoretical considerations illustrate the importance of temperature, pH, and [H(2)O(2)] for the kinetics and mechanisms of troilite oxidation. The amounts of dissolved iron strongly increase with temperature and [H(+)], whereas an increase of H(2)O(2) concentration seems to reduce the troilite oxidation. The reaction orders with respect to [H(+)] are variable, pointing out notable modifications of reaction mechanism with experimental conditions. The estimated value E(a)=25.4+/-0.9 k J mol(-1) ([H(2)O(2)]=0.4 mol L(-1) and pH 1) points to dissolution kinetics controlled by a mix regime of surface reaction and diffusion.  相似文献

高强度PAMPS-PAAm互穿网络凝胶及其溶胀性能   总被引:1,自引:0,他引:1  
通过考察不同单体浓度或离子强度下凝胶的力学性能和溶胀特性,对聚2-丙烯酰胺-2-甲基丙磺酸(PAMPS)与聚丙烯酰胺(PAAm)形成的互穿网络凝胶的高强度性能和作用机理进行了研究.结果表明:PAMPS-PAAm互穿网络凝胶的力学强度对c(AMPS)存在一个最佳值(1 mol/L),且随c(AAM)的增大而显著增大(0.5~4 mol/L).当c(AMPS)=1 mol/L、c(AAM)=4 mol/L时,互穿网络凝胶的最大抗压强度达6.46 MPa;改变凝胶体系内水的离子强度,PAMPS-PAAm凝胶在0.25 mol/kg离子强度时的抗压强度与纯水状态下相比增加了29%.  相似文献

The extraction of U(VI), Ce(IV), La(III), Nd(III), Sm(III), and Y(III) from an aqueous solution of Na2CO3 (0.25 mol/L) resulting from oxidative dissolution of U(IV) in the presence of H2O2 into a solution of methyltrioctylammonium carbonate (0.25 mol/L) in toluene. It was found that βU(VI)/Ln(III) values vary from ~8 to 3290 as the O : W ratio changes from 2 : 1 to 10 : 1, while βU(VI)/Ce(IV) varies from ~1.5 to 10, which allows for the extraction separation of U(VI) from Ce(IV) in a 8- to 10-stage counter-current extraction cascade and from Ln(III) in 2- to 3-stage cascade under the same conditions.  相似文献

利用亲和毛细管电泳法对牛血清白蛋白(BSA)与加替沙星(GT)间的结合反应及其相互作用做了初步探索,并应用淌度比(M)作为指标测定了两者的结合常数。以20 mmol/L pH 7.4的磷酸盐缓冲液作为运行缓冲液,分别以GT和BSA作为添加剂,另一组分为进样样品,内标为二甲基甲酰胺,于214 nm波长下检测。两种测定条件下得到的结合常数分别为4.4×104 L/mol和4.2×104 L/mol,与传统的荧光淬灭法测得的结果基本一致。该方法具有简单、高效的优点。  相似文献

AP5280 is an N-(2-hydroxypropyl)methacrylamide (HPMA) copolymer to which are linked tetrapeptide side chains containing bioactive platinum complexes at their C-terminal sides. We have developed and validated a rapid and sensitive analytical assay for the determination of total platinum concentrations in plasma, and free platinum of an AP5280 origin in plasma ultrafiltrate (PUF), of subjects dosed with AP5280. The total platinum levels were determined by use of graphite-furnace atomic-absorption spectrometry (GF-AAS) with Zeeman correction after appropriate dilution of the plasma sample with plasma-hydrochloric acid 0.2 mol L(-1) (1:5) as diluent. The limit of quantitation of this assay is 0.25 micromol L(-1) platinum in plasma. Linear calibration curves were obtained over the concentration range 0.25-5.0 micromol L(-1). Accuracy was between 87.7% and 104.2% and precision was 15.3% at the lowest concentration and less than 14% for all other levels tested. Accuracy and precision were thus in accordance with generally accepted criteria for analytical methods. Analysis of samples obtained from patients receiving AP5280 demonstrated the applicability of the described assay.Analysis of free platinum in PUF was performed by use of a previously validated and reported assay from our institute in which the same instrumental method is used.  相似文献

The bidentate ligand diformylhydrazine (OHC-HN-NH-CHO), DFH, combines with iron(II) and iron(III) in alkaline media in the pH range 7.3-9.3 to form an intensely colored red-purple iron(III) complex with an absorption maximum at 470 nm. Beer's law is obeyed for iron concentrations from 0.25 to 13 microg mL(-1). The molar absorptivity was in the range 0.3258x10(4)-0.3351x10(4) L mol(-1) cm(-1) and Sandell's sensitivity was found to be 0.0168 microg cm(-2). The method has been applied to the determination of iron in industrial waste, ground water, and pharmaceutical samples.  相似文献

Three new rare cyclopiane diterpenes(1-3),together with thirteen known compounds(4-16),were isolated and identified from a sea sediment-derived fungus Penicillium sp.TJ403-2.The planar and relative structures of compounds 1-3 were elucidated by HRESIMS,one-and two-dimensional NMR analyses,and their absolute configurations were further established by X-ray crystallography experiment.Compounds 1-3 were evaluated for the anti-inflammatory activity against LPS-induced NO production,and compound 1 showed notable inhibitory potency with an IC50 value of2.19±0.25μmol/L,which was three fold lower than the positive control indomethacin(IC50=8.76±0.92μmol/L).Further Western blot and immunofluorescence experiments demonstrated its mechanism of action to be that 1 inhibited the NF-κB-activated pathway,highlighting it as a promising starting point for the development of new anti-inflammatory agents.  相似文献

An improved HPLC method was developed for the determination of piperacillin and tazobactam in human plasma and pharmacokinetic study in Chinese healthy volunteers. Piperacillin and tazobactam in human plasma were extracted by solid-phase extraction and separated on a C(18) column and detected at 220 nm. The mobile phase for piepracillin consisted of 0.01 mol/L sodium dihydrogen phosphate (pH = 4.65) and acetonitrile (71:29, v/v), and that for tazobactam was 0.05 mol/L sodium dihydrogen phosphate (pH = 4.45) and methanol (90:10, v/v). The method was linear in the range 0.25-320.00 microg/mL for piperacillin (r(2) = 0.995) and 0.25-64.00 microg/mL for tazobactam (r(2) = 0.994). The lower limit of quantification of both compounds was 0.25 microg/mL. The intra- and inter-day precisions of piperacillin and tazobactam at three concentrations were all less than 9.2% and accuracies were within the range 97.0-108.0%. The method was used to investigate the pharmacokinetics of piperacillin and tazobactam in 12 volunteers who were intravenously given a dosage of 1.25, 2.50 and 3.75 g in three periods. The results showed that piperacillin sodium-tazobactom sodium (4:1) for injection in Chinese people fits linear dynamics, and the administred dosage can be adjusted with therapeutic effect.  相似文献

A simple new chemiluminescent, CL, method is described for the determination of fluoroquinolones such as: ciprofloxacin (CF), norfloxacin (NF), and ofloxacin (OF). This method is based on the measurement of terbium(iii) emission. This emission follows an energy transfer to the uncomplexed terbium(iii) ions from the excited products of fluoroquinolone oxidations. Under optimum conditions, calibration graphs were obtained for 2 × 10(-8)-2 × 10(-6) mol L(-1) of NF; 3 × 10(-8)-2 × 10(-6) mol L(-1) of CF and 4 × 10(-7)-5 × 10(-5) mol L(-1) of OF. The detection limits are 7 × 10(-9) mol L(-1) norfloxacin, 1 × 10(-8) mol L(-1) ciprofloxacin and 1.5 × 10(-7) mol L(-1) ofloxacin. The method was successfully applied to the determination of these drugs in pharmaceutical formulations.  相似文献

采用线性循环溶出伏安法和差分脉冲溶出伏安法对磺胺嘧啶在电活化玻碳电极上的电化学行为进行了研究。玻碳电极在PBS溶液中(pH 7.0),用恒电位法在1.7 V阳极氧化400 s,在B-R缓冲溶液中,磺胺嘧啶在1.02V(vs.Ag/AgCl)处有一良好的氧化峰,在0.02~0.25 V/s范围内,其氧化峰电流与扫描速率呈良好线性关系,表明电极过程为受吸附控制的不可逆过程。差分脉冲溶出伏安法的氧化峰电流(Ipa)与磺胺嘧啶浓度1×10-6~1×10-4mol/L范围内呈良好的线性关系(r=0.9977),检出限为8.7×10-7mol/L(S/N=3)。方法已用于分析磺胺嘧啶片剂的分析。  相似文献

The inclusion complexes of four ligands binding to cyclodextrins (CDs) were studied by electrospray ionization mass spectrometry (ESI-MS) and the dissociation constants of the complexes were obtained. The 1:1 stoichiometric inclusion complex was found in the system of CD and fenbufen or aspirin. The obtained KD values of the inclusion complexes of fenbufen binding to alpha-CD and to beta-CD are 4.38x10(-4) mol L(-1) and 2.12x10(-4) mol L(-1), respectively. The KD values of the inclusion complexes of alpha-CD-aspirin and beta-CD-aspirin are 3.33x10(-4) mol L(-1) and 1.83x10(-4) mol L(-1), respectively. A non-linear least squares regression method was applied to validate the results which were consistent with each other. For the system of tetracycline hydrochloride and CD, the 1:1 and 1:2 stoichiometric inclusion complexes were found in the mass spectra. The KD,1 and KD,2 values of the 1:1 and 1:2 stoichiometric inclusion complexes of alpha-CD and tetracycline hydrochloride are 4.47x10(-4) mol L(-1) and 6.51x10(-4) mol L(-1), respectively, and those of beta-CD and tetracycline hydrochloride are 2.26x10(-4) mol L(-1) and 8.57x10(-4) mol L(-1), respectively. For the system of norfloxacin and CD, besides the 1:1 and 1:2 inclusion complexes, the 1:3 stoichiometric inclusion complex was also found. The KD,1, KD,2 and KD,3 of alpha-CD and norfloxacin inclusion complexes are 4.61x10(-4) mol L(-1), 6.05x10(-4) mol L(-1) and 1.45x10(-3) mol L(-1), respectively. The three KD values of beta-CD and norfloxacin are 1.96x10(-4) mol L(-1), 4.93x10(-4) mol L(-1) and 1.15x10(-3) mol L(-1), respectively.  相似文献

In this paper, water-soluble CuInS(2) ternary quantum dots (QDs) modified by mercaptopropionic acid (MPA) were directly synthesized by hydrothermal method. Ascorbic acid (AA) can induce the fluorescence enhancement of MPA-capped CuInS(2) QDs and can be used for the detection of AA. Under the optimized conditions, the relationship between the fluorescence intensity of the CuInS(2) QDs and AA concentration was linear in the range of 0.25-200 μmol?L(-1). Most relevant molecules and physiological ions had no effect on the detection of AA. The fluorescence intensity of CuInS(2) QDs enhanced by a certain amount of AA could be reduced in the presence of folic acid (FA) and thus can be used for the detection of FA with the linear range of 0.2-100 μmol?L(-1). Compared with previous reports, the established approach utilized a simple, sensitive, and selective strategy to develop the QDs probe based on fluorescence enhancing and quenching phenomena without complicated immobilization.  相似文献

A biosensor based on stearic acid-graphite powder modified with sweet potato (Ipomoea batatas (L.) Lam.) tissue as peroxidase source was constructed and applied in organic solvents. Several parameters were studied to evaluate the performance of this biosensor such as stearic acid-graphite powder and tissue composition, type and concentration of supporting electrolyte, organic solvents, water/organic solvent ratio (% v/v) and hydrogen peroxide concentration. After selection of the best conditions, the biosensor was applied for the determination of hydroquinone in cosmetic creams in methanol. At the peroxidase electrode hydroquinone is oxidized in the presence of hydrogen peroxide and the radical formed was reduced back electrochemically at -180 mV vs Ag/AgCl (3.0 mol L(-1) KCl). The reduction current obtained was proportional to the concentration of hydroquinone from 6.2 x 10(-5) to 1.5 x 10(-3) mol L(-1) (r = 0.9990) with a detection limit of 8.5 x 10(-6) mol L(-1). The recovery of hydroquinone from two samples ranged from 98.8 to 104.1% and an RSD lower than 1.0% for a solution containing 7.3 x 10(-4) mol L(-1) hydroquinone and 1.0 x 10(-3) mol L(-1) hydrogen peroxide in 0.10 mol L(-1) tetrabutylammonium bromide methanol-phosphate buffer solution (95:5% v/v) (n = 10) was obtained.  相似文献

A highly sensitive spectrofluorimetric method is proposed for determination of human serum albumin (HSA) and some quinolone drugs. Using quinolones-terbium (Tb3+) complex as a fluorescent probe, in the buffer solution of pH 7.8, HSA can remarkably enhance the fluorescence intensity of the quinolones-Tb3+ complex at 545 nm and the enhanced fluorescence intensity of Tb3+ ion is in proportion to the concentration of HSA and quinolone drugs. Optimum conditions for the determination of HSA were also investigated. The linear ranges and limits of detection are 8.0 x 10(-9) to 8.0 x 10(-8) mol L(-1), 4.20 x 10(-9) mol L(-1) (for HSA); 1.0 x 10(-6) to 4.0 x 10(-6) mol L(-1), 1.87 x 10(-8) mol L(-1) (for norfloxacin) and 1.0 x 10(-7) to 1.0 x 10(-6) mol L(-1), 4.82 x 10(-8) mol L(-1) (for enoxacine), respectively. This method is simple, practical and relatively free interference from coexisting substances, as well as much more sensitive than most of the existing assays.  相似文献

A column chromatographic method has been developed for the separation and determination of cerium(Ⅲ) using poly[dibenzo-18-crown-6]. The separation was carried out in L-valine medium. The adsorption of cerium(Ⅲ) was quantitative from 1×10-1 to 1×10-4 mol/L L-valine. Amongst the various eluents, 1.0-8.0 mol/L hydrochloric acid, 1.0-8.0 mol/L hydrobromic acid, 1.0-8.0 mol/L perchloric acid, 1.0-2.0 mol/L sulfuric acid and 4.0-5.0 mol/L acetic acid, were found to be the efficient eluents for cerium(Ⅲ). The capacity of poly[dibenzo-18-crown-6] for cerium(Ⅲ) was (0.428±0.01) mmol/g. The method was applied to the separation of cerium(Ⅲ) from associated elements link uranium(Ⅵ) and thorium(Ⅳ). It was also applied for the determination of cerium(Ⅲ) in geological samples. The method is simple, rapid and selective with good reproducibility (approximately±2% ).  相似文献

An analytical method was developed for determination of p-hydroxyphenylphenylhydantoin enantiomers in rat liver microsome by using reversed-phase high-performance liquid chromatography. A 50 mm C(8) column was used as the analytical column. The mobile phase was made up of 8.8 mmol/L beta-cyclodextrin, 0.25 mol/L urea and 0.05 mol/L ammonium acetate in water. The assay was linear from 2.05 to 410.0 micromol/L for each enantiomer. The limits of detection and of quantitation for the method were 0.90 and 2.05 micromol/L for each enantiomer, respectively. The analytical method afforded average recoveries of 93.59 +/- 2.75% and 94.72 +/- 1.78% for S- and R-p-hydroxyphenylphenylhydantoin, respectively. The method allowed study of the in vitro glucuronidation of p-hydroxyphenylphenylhydantoin in rat liver microsomal incubates. The stereoselectivity of p-hydroxyphenylphenylhydantoin phase II metabolism was observed.  相似文献

A high-performance liquid chromatographic method with electrochemical detection (HPLC-ED) at a boron-doped diamond film electrode with preliminary separation and preconcentration by solid-phase extraction (SPE) has been developed for the determination of 1-hydroxypyrene (1-HP) in human urine. 1-HP is among the most widely used biomarkers of exposure to polycyclic aromatic hydrocarbons. Optimal HPLC-ED conditions have been found: mobile phase methanol-0.05 mol L(-1) phosphate buffer pH 5.0 (80:20, v/v), detection potential +1,000 mV versus Ag/AgCl (3 mol L(-1) KCl), and flow rate 0.8 mL min(-1). For SPE, LiChrolut(?) RP-18 E cartridges were used. The extraction yield was (87.0 ± 5.8)% (n = 5). The concentration dependence of 1-HP was measured in the concentration range from 0.01 to 10 μmol L(-1) (2.18-2,180 μg L(-1)) using methanolic solutions resulting from the SPE pretreatment of spiked human urine samples. The limit of detection (signal-to-noise ratio 3) and the limit of quantification (signal-to-noise ratio 10) of the biomarker were 0.013 μmol L(-1) (2.84 μg L(-1)) and 0.043 μmol L(-1) (9.39 μg L(-1)), respectively, which is sufficient for its determination in the urine of persons exposed to polycyclic aromatic hydrocarbons.  相似文献
