以2,4,6-三羟基苯乙酮和2,6-二羟基苯乙酮为原料, 分别通过甲基保护酚羟基、 苯甲酰氯酰化、 Bake-Venkataraman重排、 异戊烯基化、 酸催化关环及EtSLi脱去甲基等6步反应, 以高收率完成了天然5,7-二羟基-3-异戊烯基黄酮(1a, 收率80.6%)和5-羟基-3-异戊烯基黄酮(1b, 收率84.9%)的全合成, 所有化合物均经 1H NMR 和 13C NMR表征确定. 通过密度泛函理论方法对目标产物(1a和1b)的生物活性进行了预测. 结果表明, 3位异戊烯基侧链的存在能大大增强化合物相应的生物活性, 而且是化合物生物活性增强必需的取代基. 另外, 目标产物1a的生物活性高于产物1b, 归因于黄酮类化合物分子中A环上的7-OH属增效基团, 起到增强生物活性的作用, 化合物1a分子中A环上有7-OH, 而化合物1b分子中则无该基团. 本合成方法对其它3-烃基黄酮类天然化合物的合成具有潜在的适用性, 所预测的生物活性结果为3-烃基黄酮类化合物的构效关系研究奠定了基础.  相似文献

用胺甲基树脂与5-氟-2-羟基苯甲醛,5-氟-3-氯-2-羟基苯甲醛,5-氟-3-甲基-2-羟基苯甲醛反应合成了3种新颖的高分子担载席夫碱和它们Cr(Ⅲ)配合物,并用元素分析、磁化率、IR、1H NMR、UV-Vis、TG/DTA对其进行了表征。就一些革兰氏阳性菌(芽孢杆菌属腊状芽孢杆菌(Bacillus cereus sp),单核细胞增生李斯特菌4b(Listeria monocytogenes 4b),藤黄微球菌(Micrococcus luteus),金黄色葡萄球菌(Staphylococcus aureus)表皮葡萄球菌(Staphylococcus epidermis))和革兰氏阴性菌(布鲁氏菌(Brucella abortus),大肠杆菌(Escherichia coli),恶臭假单胞菌SP(Pseudomonas putida sp.),痢疾志贺氏菌10(Shigella dysenteria type 10),伤寒杆菌H(Salmonella typhi H)和酵母菌(念珠菌)(yeast(Candida albicans))用井扩散法对高分子担载席夫碱和它们Cr(Ⅲ)配合物的生物学活性进行了筛选。  相似文献

以2-巯基-5-(2-羟基苯基)-1,3,4-噻二唑为原料,经硫醚化、肼解、腙化反应合成了9个5-(2-羟基苯基)-1,3,4-噻二唑-2-硫基乙酰腙化合物,其结构由1H NMR,13C NMR,IR,MS表征和元素分析,并初步研究了目标化合物的抑菌活性.结果表明它们大多数具有优良的抑菌活性,芳香醛-5-(2-羟基苯基)-1,3,4-噻二唑-2-硫基乙酰腙(4a~4h)比2-丁烯醛-5-(2-羟基苯基)-1,3,4-噻二唑-2-硫基乙酰腙(4i)有更好的抑菌活性.  相似文献

以系列羟基卟啉为原料,用对甲苯磺酰肼还原,再用四氯对苯醌氧化,制得二氢卟吩(2a~2d);以二环己基碳二亚胺为脱水剂,2与5-氟尿嘧啶-1-基乙酸经酯化反应合成了4个新型二氢卟吩-5-氟尿嘧啶化合物(3a~3d),总收率分别为6.9%, 6.9%, 7.0%和7.6%,其结构经UV-Vis, 1H NMR, IR和MS表征。  相似文献

依据邻羟基二苯醚及三唑类化合物的抗菌特性及生物活性叠加原理, 将邻羟苯基和1,2,4-三唑分子片断有机结合, 设计合成了12个新型3-取代硫基-5-(1-羟基苯基)-4H-1,2,4-三唑类化合物. 首先, 水杨酸甲酯与水合肼反应生成水杨酰肼, 水杨酰肼再与硫氰酸铵和盐酸反应, 生成5-(1-羟基苯基)-4H-1,2,4-三唑-3-硫酮(3), 最后在碱性条件下化合物3与取代苯乙酮、氯苄和碘甲烷发生烷基化反应生成目标化合物, 化合物结构经 1H NMR及IR等表征确认. 抑菌测试结果表明, 当化合物质量分数为0.01%时, 目标化合物对白色念珠菌和大肠杆菌的抑菌率高达90%, 具有强抑菌活性; 对金黄色葡萄球菌的抑菌率高达80%, 具有一定的抑菌活性.  相似文献

2/3-取代硫基-5-邻羟基苯基唑类化合物的合成及抑菌活性   总被引:1,自引:0,他引:1  
根据生物活性叠加原理,将"邻羟基苯基"和"唑类杂环"分子片断合理组合,设计合成了三个系列12种新型2/3-取代硫基-5-邻羟基苯基唑类化合物5a~7d.水杨酸甲酯在乙醇中与水合肼反应生成水杨酰肼,水杨酰肼与二硫化碳或硫氰酸铵和盐酸反应,生成5-邻羟基苯基-1,3,4-噁二唑-2-硫酮(2),5-邻羟基苯基-1,3,4-噻二唑-2-硫酮(3)和5-邻羟基苯基-4H-1,2,4-三唑-3-硫酮(4),最后在碱性条件下与(取代)卤代苯乙酮发生烷基化反应生成目标化合物.目标化合物的结构经1HNMR,IR和元素分析等表征确认.抑菌测试表明,质量浓度为0.01%时,对白色念珠菌、大肠杆菌的抑菌率高达92%以上,具有强抑菌活性;对金黄色葡萄球菌抑菌率高达82%以上,具有较强的抑菌活性;这表明目标化合物对不同菌株具有广谱抑菌活性,是一类极具潜力的抗真菌、抗革兰氏阴性菌化合物.构效分析表明,苯乙酮环上取代基的类型对化合物抑菌活性有重要影响,引入Cl,Br等卤原子,能显著增强化合物的抑菌活性,而引入CH3供电基团,能降低其抑菌活性.  相似文献

以二环己基碳二亚胺为脱水剂,meso-四(对羟基苯基)卟啉与5-氟尿嘧啶-1-基乙酸反应,合成了一种新型meso-四[4-(5-氟尿嘧啶-1-基乙酰氧基)苯基]卟啉化合物(A),产率12.7%;将其与乙酸锌反应得到其锌配合物(B),产率30.8%。 通过1H NMR、IR、MS、UV-Vis及元素分析确证了卟啉化合物A及其锌配合物B的结构。  相似文献

以(-)-α-蒎烯为原料合成了系列新型4-芳亚甲基-2-羟基-3-蒎酮类化合物.(-)-α-蒎烯经选择性氧化得到(+)-2-羟基-3-蒎酮;在碱催化作用下,(+)-2-羟基-3-蒎酮与苯甲醛、对甲基苯甲醛、对甲氧基苯甲醛、对羟基苯甲醛、对氯苯甲醛、对硝基苯甲醛和糠醛等芳香醛缩合,得到系列光学活性4-芳亚甲基-2-羟基-3-蒎酮类化合物a~g.采用1H NMR,13C NMR,GC-MS和FT-IR等分析手段对合成所得4-芳亚甲基-2-羟基-3-蒎酮类化合物的结构进行了表征,考察了它们的紫外吸收特性及光稳定性.结果表明,化合物a,b,e对UVB具有良好的吸收性能;而化合物c,d,f,g兼具长波紫外线(UVA)和中波紫外线(UVB)的吸收性能.化合物a~g的光稳定性顺序为d>c>e>f>b>a>g.  相似文献

The phytochemical investigation of extracts from the roots of Sophora tonkinensis yielded three new isoprenylated flavanones, the tonkinochromanes F (2), G (3), and H (5), along with four known compounds. A new xanthone named cratoxylumxanthone A (1), together with five known compounds: dulcisxanthone B (2), alpha-mangostin (3), beta-mangostin (4), 2-geranyl-1,3,7-trihydroxy-4-(3-methylbut-2-enyl)xanthone (5) and tectochrystin (6), was isolated from Cratoxylum cochinchinense stems. The structure of new compound was characterized by 1D and 2D NMR techniques. The isolated compounds showed free radical scavenging against DPPH and lipid peroxidation inhibition.  相似文献

Two new lactones (1, 2), descurainolide A and B, and one new aryl-8-oxa-bicyclo[3,2,1]-oct-3-en-2-one (3), descurainin, together with five known compounds (4-8), were isolated from the seeds of Descurainia sophia (L.) From the stems of Croton micans Sw., five new 3,4-seco-ent-kaurene dimers: micansinoic acid (1), isomicansinoic acid (2), and the dimethyl (3), monomethyl (4) and monoethyl ester (5) of micansinoic acid were isolated. The structures of the new compounds were elucidated by spectroscopic data interpretation, mainly 1D and 2D NMR experiments and MS. Four 3'-hydroxy-4'-methoxy-isoflavonoids: calycosin-7-O-beta-D-glucopyranoside (1), calycosin (2), pratensein-7-O-beta-D-glucopyranoside (3), and pratensein (4) were isolated from Astragalus membranaceus var. mongholicus, among which compound 4 was obtained from this plant for the first time. The solvent effect obscuring (1)H signal patterns of B ring of compounds 1-4 was reported to avoid mis-assignments. Previously reported NMR data of compounds 1 and 2 were corrected based on 1D and 2D NMR experiments.  相似文献

Two new cytochalasins, 18‐deoxycytochalasin Q ( 1 ) and 21‐O‐deacetylcytochalasin Q ( 2 ), together with four known analogues, cytochalasin Q ( 3 ), 19,20‐epoxycytochalasin Q ( 4 ), 21‐O‐deacetyl‐19,20‐epoxycytochalasin Q ( 5 ), and cytochalasin D ( 6 ), were isolated from the fungus Xylaria sp. SCSIO156 originated from the South China Sea marine sediment. The structures of 1 and 2 were elucidated by MS and 1D‐ and 2D‐NMR data analyses, and comparison with known compounds. The known compounds 3 – 6 were identified by comparison of their MS and NMR data with those reported in the literature. In the in vitro antitumor assay, compounds 2 – 6 showed mild cytotoxicity against three tumor cell lines (MCF‐7, SF‐268, and NCI‐H460).  相似文献

Three new C-3 dehydrated bufadienolides were isolated from the venom of Bufo bufo gargarizans. Their structures were elucidated as 5β,12β-12,14-dihydroxy-11-oxobufa-3,20,22-trienolide (1), 5β,12β- 12,14-dihydroxy-11 -oxobufa-2,20,22-trienolide (2), and 5β,12β-12,14-dihydroxy-11 -oxobufa-2,20,22- trienolide (3) on the basis of extensive spectroscopic analysis, especially 1D NMR and 2D NMR data. In addition, all three compounds were tested for their cytotoxic activities against A549 and HepG2 cancer cell lines. Compounds 2 and 3 showed significant cytotoxicities with IC50 values less than 10 μmol/L on both cancer cells.  相似文献

Two new phenyl glycosides, mangliesides A and B (1, 2), a new ionol glycoside, manglieside C (3), two new lignan glycosides, mangliesides D and E (4, 5), were isolated from the leaves of Manglietia phuthoensis, along with two known lignans, 3-methoxymagnolol (6) and obovatol (7). Their structures were established by means of 1D and 2D NMR, electrospray ionization (ESI)-MS and HR-ESI-MS experiments. Among them, compounds 2 and 5 significantly (p<0.05) increased the growth and differentiation of osteoblastic MC3T3-E1 cells in vitro.  相似文献
