A simple, fast and reliable method, using high performance anion chromatography with pulsed amperometric detection, had been developed for the analysis of neomycin in water samples. The elution and separation were carried out with an isocratic mobile phase, containing 10 mmol/L NaOH. The influence of the concentration and pH of the mobile phase on the separation and detection was investigated. A quadruple-potential waveform used for the detection was optimized. The detection limit of neomycin was down to 0.027μg/mL. The linearity of neomycin calibration curve ranged from 0.050 to 0.505μg/mL with correlation coefficient of 0.9997. R.S.D. In this research, the effect of gamma irradiation on the inactivation of Escherichia coli O157:H7 (ATCC 33150), Staphylococcus aureus (ATCC 2392) and Salmonella typhimurium (NRRL 4463) inoculated into Tekirdag meatballs was investigated. The meatball samples were inoculated with pathogens and irradiated at the absorbed doses of 1, 2.2, 3.2, 4.5 and 5.2 kGy. E. coli O157:H7 count in 1 kGy irradiated meatballs stored in the refrigerator for 7 days was detected to be 4 log cfu/g lower than the count in nonirradiated samples (p<0.05). S. aureus counts were decreased to 4 log cfu/g after being exposed to irradiation at a dose of 1 kGy. Although it was ineffective on elimination of S. typhimurium, irradiation at a dose of 3.2 kGy reduced E. coli O157:H7 and S. aureus counts under detectable values in the meatballs. However, none of the test organisms were detected in the samples after irradiation with 4.5 kGy doses.  相似文献

A new method for the determination of antithrombotic activity of egg white protein hydrolysate(EWPH)was developed using amicroplate reader.Reaction was carried out at 37℃ and pH 7.2 with fibrinogen concentration 0.1%.Microplate reading wasconducted at 405 nm.Inhibition rate of EWPH on thrombin activity showed linearity(R~2=0.9971),when the inhibition rate was inthe range of 10-90%.The lower limit of detection(LLD,at 99.7%probability)and the biological limit of detection(BLD,at 99.7%probability)of the method were 10.643 and 40 mg/mL,respectively.The repeatability standard deviation(R.S.D.)was 1.08%.Thestandard deviation of the method was 0.027 AT-U.  相似文献

Three kinds of new layered perovskite compounds with Ruddlesden-Popper(R-P)phase,Ag_xNa_(2-x)La_2Ti_3O_(10)(x=0.2,0.3 and 0.5),were synthesized by an ion-exchange reaction of Na_2La_2Ti_3O_(10)with AgNO_3 solution.The structures of the compounds were characterized by EDX and XRD,and their antibacterial activity and light-resistance property were evaluated.The results indicated that the molecular formula of Ag_xNa_(2-x)La_2Ti_3O_(10)(x=0.2,0.3 and 0.5)was confirmed,and that the crystalline structure of Na_2La_2Ti_3O_(10)was not obviously affected by exchange of silver ion.The minimum inhibitory concentrations(MICs)of Ag_(0.3)Na_(1.7)La_2Ti_3O_(10)against Escherichia coli(E.coli),Staphylococcus aureus(S.aureus)were 180μg/mL and 240μg/mL, respectively,while its discoloration was not observed after 24 h light ageing test.  相似文献

Pt-nanoparticle-coated gold nanoporous film (PGNF) was synthesized via a simple nonpolluting approach and PGNF modified electrode was also constructed successfully for the rapid measurement of Escherichia coli (E. coli) in this work. Scanning electron microscopy (SEM) and energy dispersive spectroscopy (EDS) images showed that the resulting PGNF electrode had highly ordered arrangement and large surface area. Furthermore, the electrochemical characteristics of the PGNF electrode were investigated by cyclic voltammetry (CV) and amperometric i-t curve. The PGNF electrode showed excellent electrocatalytic activity to E. coli and the current responses were in good linear from 2 × 101 cfu/ml to 1 × 106 cfu/ml with the detection limit of 10 cfu/ml (S/N = 3) without pretreatment. The high sensitivity, wider linear range and good reproducibility make this PGNF a promising candidate for portable amperometric E. coli sensor.  相似文献

A sensitive method for simultaneous determination of six phenolic whitening agents,including arbutin, phenol,resorcinol,hydroquinone,kojic acid,and salicylic acid in cosmetics has been developed using micellar electrokinetic capillary chromatography with amperometric detection(MECC-AD).Effects of several factors,such as the pH value and concentration of running buffer,potential applied to the working electrode,separation voltage,and injection time were investigated to obtain optimum conditions for separation and detection.With a 75 cm long fused-silica capillary tube,well-defined separation of six phenolic compounds was achieved in 10mmol/L SDS/40 mmol/L H3BO3-Na2B4O7 running buffer(pH 9.0).Good linear relationship was obtained for each analyte over three orders of magnitude with correlation coefficients(r2) between 0.9985 and 0.9994,and the detection limit(S/N=3) ranged from 0.04μg/mL to 0.45μg/mL The proposed method has been successfully applied for the determination of phenolic whitening agents in real cosmetic samples with satisfactory results,providing an alternative monitoring method for cosmetics safety regulation.  相似文献

This study evaluated effect of gamma irradiation on survival of Salmonella Typhimurium and Staphylococcus aureus on lettuce and damage of cell envelope. S. Typhimurium and S. aureus were inoculated on red leaf lettuce, and they were irradiated at 0, 0.5, 1, 1.5, 2, 2.5, and 3 kGy, and the samples were then stored at 7 and 25 °C for 7 days. Survival of S. Typhimurium and S. aureus were enumerated on xylose lysine deoxycholate agar and Baird–Parker agar, respectively. D10 value (dose required to reduce 1 log CFU/leaf) was calculated, and kinetic parameters (maximum specific growth rate; μmax and lag phase duration; LPD) were calculated by the modified Gompertz model. In addition, cell envelope damage of the pathogens was observed by scanning electron microscope (SEM) and transmission electron microscope (TEM). D10 values were 0.35 and 0.33 kGy for S. Typhimurium and S. aureus, respectively. During storage at 7 °C, S. Typhimurium and S. aureus had significant (P<0.05) growth only on non-irradiated samples up to about 2.5 and 4 log CFU/leaf at 0.42 and 1.28 log CFU/leaf/day of μmax, respectively. At 25 °C, cell counts of S. Typhimurium and S. aureus on the samples irradiated at 0 and 0.5 kGy increased (P<0.05) up to 3–6 log CFU/leaf. The μmax of both pathogens were higher in 0 kGy (1.08–2.27 log CFU/leaf/day) and 0.5 kGy (0.58–0.92 log CFU/leaf/day), and LPDs ranged from 1.53 to 3.14 day. SEM and TEM observations showed that cells irradiated at 1.5 and 3 kGy showed disrupted cell membrane. These results indicate that gamma irradiation could be a useful decontamination technology to improve food safety of lettuce by destroying cells of S. Typhimurium and S. aureus.  相似文献

A protein-based electrochemical sensor for hydrogen peroxide (H2O2) was developed by an easy and effective film fabrication method where spinach ferredoxin (Fdx) containing [2Fe–2S] metal center was cross linked with 11-mercaptoundecanoic acid (MUA) on a gold (Au) surface. The surface morphology of Fdx molecules on Au electrodes was investigated by atomic force microscopy (AFM). Electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV) were employed to study the electrochemical behavior of adsorbed Fdx on Au. The interfacial properties of the modified electrode were evaluated in the presence of Fe(CN)63?/4? redox couple as a probe. From CV, a pair of well-defined and quasi-reversible redox peaks of Fdx was obtained in 10 mM, pH 7.0 Tris–HCl buffer solution at ?170 and ?120 mV respectively. One electron reduction of the [2Fe-2S]2+ cluster occurs at one of the iron atoms to give the reduced [2Fe-2S]+. The formal reduction potential of Fdx ca. ?150 mV (vs. Ag/AgCl electrode) at pH 7.0. The electron-transfer rate constant, ks, for electron transfer between the Au electrode and Fdx was estimated to be 0.12 s?1. From the electrochemical experiments, it is observed that Fdx/MUA/Au promoted direct electron transfer between Fdx and electrode and it catalyzes the reduction of H2O2. The Fdx/MUA/Au electrode displays a linear increase in amperometric current for increasing concentration of H2O2.The sensor calibration plot was linear with r2 = 0.998 with sensitivity approximately 68.24 μAm M?1 cm?2. Further, the effect of nitrite on the developed sensor was examined which does not interfere with the detection of H2O2. Finally, the addition of H2O2 on MUA/Au electrode was observed which has no effect on amperometric current.  相似文献

Fast analysis of thiocyanate by ion-pair chromatography using a silica-based monolithic column and direct conductivity detection was carried out. Chromatographic separation was performed on a Chromolith Speed ROD RP-18e using tetrabutylammonium hydroxide (TBA)-phthalic acid-acetonitrile as eluent. The effects of eluent concentration, eluent pH value, column temperature and flow rate on retention time of thiocyanate were investigated. The optimized chromatographic conditions for the determination of thiocyanate were as follows: 0.25 mmol/L TBA-0.18 mmol/L phthalate-7% acetonitrile (pH 5.5) as eluent, column temperature of 30 ℃, and flow rate of 6.0 mL/min. Retention time of thiocyanate was less than 1 min under the conditions. Common anions (Cl^-, NO3 , SO42 and I^- ) did not interfere with the determination of thiocyanate. Detection limit (S/N = 3) of thiocyanate was 0.96 mg/L. Calibration graph between peak area and the concentration of thiocyanate was linear in the range of 2.0- 100.0 mg/L. Relative standard deviation (RSD) of chromatographic peak area was 1.4% (n = 5). This method has been applied to the determination of thiocyanate in ionic liquids. Recoveries of thiocyanate after spiking were 100.5%.  相似文献

A novel method for the simultaneous determination of benserazide and levodopa using capillary electrophoresis(CE)- chemiluminescence(CL)has been developed.Under the optimal conditions,the detection limits(S/N=3)were 1.85μg/mL for benserazide and 0.12μg/mL for levodopa.This method was successfully applied to the determination of benserazide and levodopa in Medopar tablet,the results showed that the detected values are in accordance with those by official methods.  相似文献

A flow injection irreversible biamperometric method for the determination of chlorogenic acid is described.The proposedmethod is based on the electrochemical oxidation of chlorogenic acid at pretreated platinum electrode and the reduction ofpermanganate at another electrode to form an irreversible biamperometric detection system.Under the external potential difference(△E)of 0 V,in the 0.05 mol/L sulfuric acid,chlorogenic acid can be determined over the range 0.8-120 mg/L with a samplemeasurement frequency of 80 samples/h.The detection limit is 0.18 mg/L.The proposed method exhibits the satisfactoryreproducibility with a relative standard derivation(R.S.D.)of 2.21%for 19 successive determinations of 40 mg/L.  相似文献

The crude extracts and isolated compounds of Piper porphyrophyllum (Piperaceae) were evaluated for antibacterial and anti-inflammatory activities. The ethyl acetate extract and 3′,4′,5,7-tetramethoxyflavone exhibited the highest activity against Staphylococcus aureus giving values of MIC = 62.5 and 250 μg/mL, respectively. 5-Hydroxy-7-methoxyflavanone and 4′,5-dihydroxy-3′,7-dimethoxyflavone were active against Pseudomonas aeruginosa, both with MIC value 125 μg/mL. The hexane extract and 4′,5-dihydroxy-3′,7-dimethoxyflavone gave the highest anti-inflammatory activity in in vitro quantitative lipoxygenase inhibition assay with inhibitory activity of (IE) 99.72% and 91.81%, respectively.  相似文献

The threat to public health from bacterial infections has led to an urgent need to develop simpler, faster and more reliable bacterial detection methods. In this work, we developed a universal dual-recognition based sandwich fluorescence resonance energy transfer (FRET) sensor by using specific aptamer-modified quantum dots (Aptamer-QDs) as energy donor and lectin concanavalin A (Con A) modified gold nanoparticles (Con A-AuNPs) as energy acceptor to achieve rapid and sensitive detection of Escherichia coli (E. coli) within 0.5 h. In the presence of the target E. coli, the energy donor of Aptamer-QDs and acceptor of Con A-AuNPs were close to each other, causing changes of FRET signals. Based on the constructed FRET sensor, a linear detection range of from 102 cfu/mL to 2 × 108 cfu/mL with the detection limit of 45 cfu/mL for E. coli was achieved. Furthermore, the FRET sensor was applied to detect E. coli in the milk and orange juice with the detection limit of 300 cfu/mL and 200 cfu/mL, respectively and recovery rate from 83.1% to 112.5%. The strategy holds great promise in pathogenic bacteria detection due to its rapid and sensitivity.  相似文献

An amperometric method for the rapid detection of Escherichia coli (E. coli) by flow injection analysis (FIA) using an IrO2–Pd chemically modified electrode (CME) was developed in this paper. The method is based on a good marker β-d-galactosidase which was found in E. coli strains. β-d-galactosidase was produced by the induction of isopropyl β-d-thiogalactopyranoside (IPTG) and released from E. coli cells through the permeabilization of both polymyxin B nonapeptide and lysozyme to E. coli cells wall. The released β-d-galactosidase could catalyze the hydrolysis of the substrate p-aminophenyl β-d-galactopyranoside (PAPG) in the culture medium to produce 4-aminophenol which was proportional to the concentration of E. coli. Hence, E. coli could be detected by the determination of 4-aminophenol. An IrO2–Pd CME, which showed high sensitivity in determination of 4-aminophenol, was prepared as the electro-detector in FIA. The amplified response current of 4-aminophenol obtained at the IrO2–Pd CME was linear with the concentration of E. coli ranging from 2.0 × 102 to 1.0 × 106 cfu/mL, the detection limit of this method to E. coli was 150 cfu/mL and the complete assay could be performed in 3 h.  相似文献

In this paper,we developed and validated a simple,sensitive,and selective high-performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS) method to identify and measure the following prohibited substances that may be found in cosmetic products:minoxidil,hydrocortisone, spironolactone,estrone,canrenone,triamcinolone acetonide and progesterone.Chromatographic separation was performed on a Waters Symmetry C18(100 mm×2.1 mm,3.5μm particle size) with a gradient elution system composed of 0.2%(v/v) formic acid aqueous solution and methanol containing 0.2%(v/v) formic acid at a flow rate of 0.3 mL/min.The substances were detected using a triple quadrupole mass spectrometer in the multiple reaction monitoring mode with an electrospray ionization source.All of the calibration curves showed good linearity(r > 0.999) within the tested concentration ranges.The limit of detection was <25 pg.The relative standard deviations for intraday precision for each of the prohibited substances were <3.5%at two concentration levels(2μg/g,10μg/g). The relative recovery rate for each of the prohibited substances ranged from 91.8%to 111%at three concentration levels(0.1μg/g,2μg/g,10μg/g),including the limit of quantification.In conclusion,we have developed and validated a method that can identify seven prohibited substances in cosmetic products.  相似文献

A sensitive flow injection chemiluminescence method has been developed for the detection of resveratrol in red wine based on the fact that resveratrol can greatly enhance chemiluminescence reaction between KMnO_4 and HCHO in sulfuric acid medium. Analytes were pre-concentrated on solid sorbents (C_(18) solid-phase extraction cartridges).Under the optimum conditions,the proposed method allows the measurement of resveratrol over the range of 1.32×10~(-8) to 1.32×10~(-5) mol/L with a detection limit of 3.30×10~(-9) mol/L,and the relative standard deviation for 1.32×10~(-5) mol/L resveratrol (n=11) is 3.8%.This method has been successfully applied for the determination of the resveratrol in red wine.Furthermore,the possible reaction mechanism was also discussed.  相似文献

Determination of the total tannin of white and red rind pomegranate (Punica granatum L.) has been carried out by colorimetry method using reagent 1,10 phenantroline. This method is based on reduction of iron (III) into iron (II) by tannin at temperature 800C for 20 min. Then the formed of iron (II) was reacted with 1,10 phenantroline to form orange red colour complex that could be measured by spectrophotometer visible at maximum absorption wavelength of 508 nm. The limit of detection (LOD) and the limit of quantitation (LOQ) obtained were 0.34 μg/mL and 1.14 μg/mL, respectively. This result was found to be linier with R value of 0.9984; accuracy as percent recovery was 84.69 ± 0.85% and coefficient of variant (KV) was 1.003% for white rind pomegranate while red rind pomegranate percent recovery was 84.38 ± 0,45% and coefficient of variant (KV) was 0.53%. The total tannin of white rind pomegranate was 18.28 ± 0.072%b/b and red rind pomegranate was 17.33 ± 0.081%b/b  相似文献

A new, low power capacitively coupled plasma microtorch (30 W, 13.56 MHz, 0.5 L min?1 Ar) was investigated in conjunction with commercially available microspectrometers for the simultaneous multielemental analysis by atomic emission spectrometry of liquid samples without desolvation. Emission spectrum is simpler than in ICP-AES, the resonance lines are the most intense, so that a microspectrometer with FWHM of at least 1.5 nm is satisfactory for the record. The deviation from the Boltzmann distribution for Fe I has demonstrated the departure from the LTE in plasma. The non-spectral matrix effects of Li, Na, K, Ca and Mg on analytes emission are depressive and depend on matrix volatility, ionization potential of the interferent and analyte excitation energy. The detection limits (μg mL?1) are in the range 0.003 (Li) and 1.5 (Mn). The use of the standard additions method allowed the simultaneous determination of elements in environmental certified reference materials with overall recovery of 95 ± 10% and relative standard deviation of 1.7–8.2%. Compared to the traditional ICP, the microtorch has a simple construction, runs at low argon flow and can be integrated in a portable system suitable for in-situ simultaneous determination of elements.  相似文献
