The phase behaviour of a hydrated (32% (w/w) water) mixture of saturated and unsaturated fatty acids and their soaps has been examined. By progressively increasing the proportion of oleic acid, it could be shown that this unsaturated component exists partly as a separate phase within the others. The unsaturated fatty acids form an HII phase at 25°C, whereas the saturated fatty acids form a fatty acid—soap crystal under the conditions employed. The results help to explain the role played by the fatty acids (and also cholesterol) within the lipid fraction of human stratum corneum. A feature of this study is a comparison of the results obtained by X-ray diffraction, differential scanning calorimetry and Fourier transform infrared spectroscopy.  相似文献

A rapid method for the detection and quantltatlve determlnation of unsaturated fatty acids by TLC is presented. After separation on HPTLC-RP18-plates, reaction wlth p-hydroxybenzaldehyde is carried out by a dipping procedure. Nanogram amounts of unsaturated fatty acids can be determined with good reproducibility.  相似文献

以2-氨基-2-甲基丙醇为脂肪酸的化学修饰试剂,将不饱和脂肪酸羧基化学修饰成含氮杂环,从而避免了链烯基中碳碳双键在EI源中的移动,并使质谱呈现显示双键位置的规范信息。通过解析不饱和脂肪酸2-氨基-2-甲基丙醇化学修饰产物的EI质谱图,得到了不饱和脂肪酸中碳碳双键的定位规则,从而为分析不饱和脂肪酸精细结构提供了手段。用脂肪酸2-氨基-2-甲基丙醇化学修饰气相色谱-EI质谱法分析了螺旋藻脂肪酸,鉴定出9种不饱和脂肪酸,确定了螺旋藻脂肪酸中碳碳双键的位置,结果与文献一致。  相似文献

The complexation of fatty acids (both saturated and unsaturated) with various cyclodextrins and cyclodextrin derivatives greatly modifies their properties. Inclusion complex formation — depending upon the type of host cyclodextrin — may result in protection against the environment, in improved water solubility and bioavailability. Thus lipid complexation enables the preparation of more reliable diagnostic reagents, better chromatographic separations and higher yields in biotechnological processes. The relevant literature is reviewed with particular emphasis on the practical utility of the molecular encapsulation of fatty acids with cyclodextrins.  相似文献

We investigated the effect of fatty acids such as stearic acid (SA, 18:0), oleic acid (OA, 18:1), eicosapentaenoic acid (EPA, 20:5), and docosahexaenoic acid (DHA, 22:6) on a dipalmitoylphosphatidylcholine (DPPC) bilayer by determining the phase transition temperature, fluorescence anisotropy of 1,6-diphenyl-1,3,5-hexatriene (DPH), and detergent insolubility. Treatment with unsaturated fatty acid broadened and shifted the phase transitions of the DPPC bilayer to a lower temperature. The phase transition temperature and the value of fluorescence anisotropy of DPH at 37 degrees C decreased progressively with increasing treatment amounts of unsaturated fatty acid. A large amount of the DPPC bilayer treated with unsaturated fatty acid was dissolved in Triton X-100, obtaining a low level of detergent insolubility. These modifications of the bilayer physical properties were most pronounced with DHA and EPA treatment. These data show that unsaturated fatty acids, particularly DHA and EPA, induce a marked change in the lipid bilayer structure. The composition of fatty acids in the DPPC bilayer was similar after treatment with various unsaturated fatty acids, suggesting that the different actions of unsaturated fatty acids are attributed to change in the molecular structure (e.g., kinked conformation by double bonds). We further explored the change in physical properties induced by fatty acids dispersed in a water-in-oil-in-water multiple emulsion and found that unsaturated fatty acids acted efficiently on the DPPC bilayer, even when incorporated in emulsion form.  相似文献

Langmuir monolayers of cholesterol and various fatty acids, such as stearic, oleic, linoleic, α-linolenic, and arachidonic acids, spread at the air/water interface are investigated. The system of cholesterol and stearic acid is found to be immiscible, with only one collapse, occurring at the same surface pressure for all composition range. However, surface pressure (π) – area (A) isotherms of cholesterol/unsaturated fatty acids show a characteristic course with two collapse states. The pressure of the first collapse varies with the proportion of the components in the mixture, while the second collapse, occurring at the surface pressure characteristic of cholesterol alone, is independent of mole fraction of the investigated fatty acid. The application of the surface phase rule indicates that the unsaturated fatty acids/cholesterol mixtures are miscible up to the surface pressure corresponding to the first collapse. Negative values of the excess free energy of mixing in all composition ranges prove that the mixtures are stable. The interactions existing in mixtures of cholesterol and unsaturated fatty acids possessing even numbers of double bonds are strongest in the lower region of fatty acid proportion, and the results are consistent with the minimum values of the excess free energy of mixing, indicating the most stable mixtures. For cholesterol and unsaturated fatty acids with odd numbers of double bonds the behavior is different, and the strongest interactions occur in both low and high regions of mole fraction of an acid. The separation of fatty acids derivatised with Nile Blue (NB) by CE with detection using a red light-emitting diode (LED) was examined. NB was selected as the derivatisation agent due to its high molar absorption coefficient of 76,000 M(-1) cm(-1) at 633 nm, making it well suited for sensitive absorbance detection using a red 635 nm LED. NB-labelled fatty acids were separated by both MEKC using SDS micelles, i-PrOH and n-BuOH and by NACE in a number of solvents including MeOH, EtOH and ACN. The sensitivity of NACE was superior to MEKC, with detection limits of 5x10(-7)-7x10(-7) M obtained for each acid, approximately 20 times lower than the MEKC method. The NACE detection limits are approximately 100 times lower than previous reports on the separation of fatty acids by CE using indirect absorbance detection, ten times lower than using indirect fluorescence detection and are inferior only to those obtained using precapillary derivatisation and direct fluorescence detection. The efficiency of the NACE method was also superior to MEKC and allowed the separation of unsaturated fatty acids to be examined, although it was not possible to baseline-resolve linoleic (C18:2) and linolenic (C18:3) acids in a reasonable time. The method was used to analyse the fatty acid profile of two edible oils, namely sunflower and sesame oils, after alkali hydrolysis, where it was possible to identify both the saturated and unsaturated fatty acids in each sample.  相似文献

以2-氨基-2-甲基-丙醇为脂肪酸的化学修饰试剂,气相色谱一电子轰击质谱(GC-EI MS)分析葵子油脂肪酸。2-氨基-2-甲基-丙醇将脂肪酸羧基修改为含氮杂环,使在EI源中避免了链烯基中碳碳双键的移动。解析了葵子油脂肪酸2-氨基-2-甲基-丙醇化学修饰产物的EI质谱图,讨沦了烯酸中碳碳双键的定位规则,确定了葵子油脂肪酸中碳碳双键的位置。鉴定出葵子油6种脂肪酸,不饱和脂肪酸相对含量为89.41%,其中人体必需脂肪酸9,12-十八碳二烯酸含量占65.30%。本方法为不饱和脂肪酸中双键的定位提供了新的技术手段。  相似文献

采用索氏提取法提取油松籽中的油脂,得油率为42.9%;对油脂进行甲酯化处理后用气相色谱-质谱联用仪检测其中的脂肪酸组成及含量。实验结果表明,油松籽油中含有7种脂肪酸,分别为肉豆蔻酸10.38%、硬脂酸3.05%、油酸21.98%、亚油酸(13,16-十八碳二烯酸)3.53%、亚油酸(9,12-十八碳二烯酸)38.38%、亚麻酸20.06%和二十碳三烯酸2.62%,其中饱和脂肪酸含量为13%,不饱和脂肪酸含量为87%。  相似文献

Summary This report describes a modified method for the separation and analysis of polyunsaturated fatty acids such as 20:4, 20:5, and 22:6, using HPLC. The results show that these fatty acids are well separated from the saturated acids. Since the unsaturated fatty acids elute earlier than saturated acids, and this method does not require the fractionation of free fatty acids using thin layer chromatography, a necessary step for the gas chromatographic analysis, the recoveries of polyunsaturated fatty acids were significantly higher as compared to those from gas chromatography. Furthermore, HPLC and gas chromatographic methods gave identical results for the acyl chain composition of phosphatidylserine. The advantages of using HPLC over gas chromatography in determining the acyl chain composition of free fatty acids and phospholipids are also discussed.  相似文献

首次对小鼠眼球中的脂肪酸组成进行了气相色谱分析。小鼠眼球中的脂肪酸经氯仿-甲醇-盐酸混合液萃取,用甲醇-氢氧化钾法甲酯化,在25m×0.25mmPEG-20M熔融石英毛细管柱内进行分离,眼球中12种主要脂肪酸得到了定性和定量。测试结果提示:正常小鼠眼球中饱和脂肪酸含量较高,占全部脂肪酸含量的52.8%;不饱和脂肪酸含量较少,但以多不饱和脂肪酸居多,占全部脂肪酸含量的18.2%,不饱和脂肪酸与饱和脂肪酸的比值为0.6。可为有关眼疾病的治疗、预防及生化方面的深入研究提供有意义的信息。  相似文献

The decomposition of unsaturated fatty acids and tocopherols in 10 commercial edible oils during deep frying was investigated. The dominant tocopherol in oils rich in polyunsaturated fatty acids (PUFAs) was γ-tocopherol, except for natural perilla oil (δ-tocopherol dominant), and the main tocopherol in oleic acid-rich oils was α-tocopherol. The PUFA-rich oils had higher tocopherol contents than the oleic acid-rich oils. Both the reduction rate of total unsaturated fatty acid (TUFA) and total tocopherol (TToc) were linear with frying time (t). The decomposition rate of TToc is faster than that of TUFA since the slope values obtained from fitting equations (Y?=?k t) kTToc (1.520–14.483) were obviously larger than for kTUFA (0.155–0.270). By establishing a dynamic decomposition index, unsaturated fatty acids and tocopherol in oils showed dynamic decomposition over multiple frying cycles. The obtained results showed that decomposition characteristics of oils are related to their fatty acid compositions.  相似文献

深黄被孢霉催化转化十八醇合成不饱和脂肪酸在优化转化条件下所得产物分析表明,干细胞中含油脂47.5%,蛋白质31.5%,碳水化合物28.9%.十八醇转化率为67.2%,油脂选择性为30.6%,产率20.6%.油脂中脂肪酸的选择性为:棕榈酸4.00%,棕榈油酸0.81%,硬脂酸0.95%,油酸20.75%,亚油酸1.95%,亚麻酸2.19%.催化转化条件下的研究表明,底物浓度越低,油脂和不饱和脂肪酸的选择性越好,当十八醇质量分数为0.5%时,油脂和不饱和脂肪酸的选择性达到最大值;培养基pH为中性时,获得了最大的油脂选择性和收率;33℃时醇转化率达最大值88%,23℃油脂产率达最大值9.7%.Mg2+对不饱和脂肪酸的生成有显著影响,当培养基中[Mg2+]=25mmol/L时,获得了最大的油脂选择性(25.7%)和产率(17.6%).  相似文献

A method is described for the determination of fatty acids in dried sweat spot and plasma samples using gas chromatography with flame ionization detection. Plasma and dried sweat spot samples were obtained from a group of blood donors. The sweat was collected from each volunteer during exercise. Sweat was spotted onto collection paper containing butylated hydroxytoluene. Fatty acids were derivatized with acetyl chloride in methanol to form methyl esters of fatty acids. The fatty acids in dried sweat spot samples treated with butylated hydroxytoluene and stored at –20°C were stable for 3 months. Our results indicate that sweat contains, among fatty acids with short chain, also fatty acids with long chain and unsaturated fatty acids. Linear relationships between percentage content of selected fatty acids in dried sweat spot and plasma were observed.  相似文献

用Whitsett法提取18只正常足月妊娠分娩的胎盘合体滋养细胞质膜(SPM),采用Landon法提取脂质,使用气相色谱法和丁二酸二乙二醇酯(DEGS)填充柱分离测定SPM的脂肪酸,结合气相色谱/质谱定性,将13种碳数大于14的脂肪酸进行了定性和定量。定量出的脂肪酸占出峰物质总量的74.1%,其中饱和脂肪酸为30.64%,不饱和脂肪酸为43.46%。  相似文献

Capillary zone electrophoresis was investigated for the separation of free fatty acids as an alternative to well established techniques such as GC or HPLC. The analysis was performed with indirect UV detection in a counterelectroosmotic flow mode using a diethylbarbiturate carrier electrolyte at a pH between 10 and 11 in a mixed aqueous-organic solvent. Separation of saturated and unsaturated fatty acids could be achieved after bromination of the double bonds. Problems with wall adsorption of fatty acids could be overcome by increasing the temperature and using a high concentration of a zwitterionic reagent to inactivate the silica surface. Increased sensitivity could be achieved after preconcentration on alumina. The method was applied to the determination of free fatty acids in dairy products. 建立了一种基于气相色谱-质谱的拟靶向代谢组学分析方法对大米中脂肪酸进行分析,共检测到16种脂肪酸,并研究了不同大米中脂肪酸的轮廓差异。以提取到饱和脂肪酸和不饱和脂肪酸的总量为评价指标,比较了6种提取方法及4种提取溶剂对脂肪酸提取效率的影响。将该方法用于5种不同大米(稻花香、吉星、金浪子、农大、状元)中脂肪酸的分析,发现稻花香大米中脂肪酸轮廓与其他4种均有较大差异;而金浪子与农大、状元间脂肪酸差异也较大,与吉星脂肪酸轮廓较为相似。该方法简单,有较好的稳定性和准确性,可为大米品质和营养价值改善研究提供基础数据。  相似文献
