用伊文思蓝(Evans blue, EB)作荧光探针研究了氨苄青霉素(Ampicillin, A)对牛血清白蛋白(Bovine serum albumin, BSA)的竞争反应. 伊文思蓝与牛血清白蛋白作用, 使牛血清白蛋白荧光发生猝灭, 根据Stern-Volmer方程及荧光寿命研究了荧光猝灭的类型及机理. 结果表明, 猝灭类型为静态猝灭, 即伊文思蓝和牛血清白蛋白形成了一种稳定的复合物. 伊文思蓝与牛血清白蛋白的结合常数KBSA-EB=1.122×106 L/mol, 结合点数n=0.9935, 并确定了EB和BSA之间的热力学常数及作用力类型. 当加入氨苄青霉素后, 牛血清白蛋白的相对荧光强度恢复. 这表明氨苄青霉素与伊文思蓝对牛血清白蛋白发生了竞争反应. 探讨了该竞争反应的相关机理, 求出了伊文思蓝与氨苄青霉素的结合常数为KEB-A=7.131×105 L/mol.  相似文献

用荧光光谱法研究了生理酸度条件下,头孢噻肟对牛血清白蛋白,Cu(Ⅱ)对牛血清白蛋白以及Cu(Ⅱ)对头孢噻肟和牛血清白蛋白荧光光谱特性的影响。结果表明:Cu(Ⅱ)和头孢噻肟均可使牛血清白蛋白的荧光强度发生静态猝灭,并且在Cu(Ⅱ)存在下,头孢噻肟对牛血清白蛋白的荧光猝灭作用显著增强。根据荧光猝灭双倒数图计算头孢噻肟和牛血清白蛋白的结合常数为3.11×104L/mol,结合位点数为1.03;二元配合物Cu(Ⅱ)与牛血清白蛋白之间的结合常数为1.13×103L/mol,结合位点数为0.74。  相似文献

利用荧光光谱研究了脱落酸(Abscisic acid)与伊文思蓝(Evans blue)的相互作用。用266 nm激发光激发时,伊文思蓝的最大发射波长为396 nm。伊文思蓝与脱落酸相互作用,使伊文思蓝发生荧光猝灭,根据Stern-Volmer方程研究了荧光猝灭的类型及机理,实验证明脱落酸与伊文思蓝发生的静态猝灭,即脱落酸和伊文思蓝形成了一种稳定的复合物。伊文思蓝的相对荧光强度与脱落酸的浓度线性相关,线性方程为F0/F=0.78153+3.9153×104c,线性相关系数为0.9976。通过实验求算了脱落酸与伊文思蓝的结合点常数KABA-EB=1.838×105L/mol和结合点数n=1.171,由此建立了测定植物中脱落酸的新方法。  相似文献

用荧光光谱法、分光光度法研究了水溶液中左西孟旦与牛血清白蛋白(BSA)的相互结合反应。研究表明:左西孟旦对BSA的内源荧光有较强的猝灭作用且该猝灭作用属于静态荧光猝灭作用。得出了反应的结合常数(KA=1.48×106L/mol)和结合位点数(n=1.14)。根据Frster非辐射能量转移机理,求算了给体(BSA)与受体(左西孟旦)间距离r=2.9 nm和能量转移效率E=0.33。  相似文献

研究了尿嘧啶猝灭伊文思蓝(Evans blue)的荧光猝灭机理,并根据尿嘧啶对伊文思蓝荧光的猝灭作用,建立了测定尿嘧啶的新方法.当用253 nm激发时,伊文思蓝的最大发射波长位于387 nm,线性回归方程为:c=11.85I0/IF-12.30,相关系数为0.999 0,线性范围为0.2 ~20 mg/L,检出限为0.14 mg/L,相对标准偏差(RSD)为3.6%.求得伊文思蓝与尿嘧啶的形成常数为K=4.88×103 L/mol,试验了pH、干扰离子、放置时间、核酸水解产物等对测定的影响.  相似文献

本文用自制L-天冬氨酸修饰电极(PLA/GCE),采用循环伏安法研究了牛血清白蛋白(BSA)与异烟肼(INH)的相互作用,并与荧光光谱法、紫外-可见光谱法进行了比较。使用循环伏安法和荧光光谱法,测得异烟肼与牛血清白蛋白的结合常数K分别为1.544×10~4、1.479×10~4 L/mol,结合位点数均接近1.1。实验测得异烟肼对牛血清白蛋白是静态猝灭。异烟肼的浓度与牛血清白蛋白的荧光强度的降低在2.5×10~(-7)~4.5×10~(-4) mol/L范围内呈线性关系,检出限为1.0×10~(-7) mol/L。BSA的浓度与异烟肼的氧化峰电流的下降在1.0×10~(-9)~5.0×10~(-5) mol/L范围内呈线性关系,检出限为5.0×10~(-10) mol/L。该方法可用于样品中异烟肼和牛血清白蛋白的测定。  相似文献

利用荧光光谱和紫外-可见吸收光谱研究了在缓冲溶液中不同温度下α-硫辛酸(ALA)与牛血清白蛋白(BSA)的相互作用。结果表明,ALA对BSA的内源荧光猝灭为静态猝灭过程,猝灭常数KSV分别为4.65×103L/mol(26℃)和4.46×103L/mol(37℃)。依据Frster非辐射能量转移机制,得到给体(BSA)-受体(ALA)间的结合距离r=2.90 nm,能量转移效率E=5%。测定了该反应在不同温度下的结合常数KA=4.31×103L/mol(26℃),4.27×103L/mol(37℃),以摩尔比1∶1结合。根据不同温度下的结合常数确定了相互作用过程的热力学参数,并根据热力学参数确定了ALA与BSA之间作用力主要是静电作用。  相似文献

采用荧光光谱、紫外光谱对吡柔比星与牛血清白蛋白的相互作用进行了研究。结果表明,吡柔比星和牛血清白蛋白可形成基态配合物导致牛血清白蛋白的内源荧光猝灭,猝灭机理主要为静态猝灭和非辐射能量转移。通过计算获得了二者在不同温度下的结合常数及结合位点数。根据热力学参数判断吡柔比星与牛血清白蛋白之间的作用力主要为范德华力和氢键。根据Frster非辐射能量转移理论确定了吡柔比星和牛血清白蛋白的作用距离。研究了不同金属离子存在下对吡柔比星与牛血清白蛋白结合常数及结合位点数的影响。  相似文献

用溶胶-凝胶法以磷钼酸(MPA)的镍盐溶液水解钛酸四丁酯制备了NiPMo/TiO2催化剂.使用ICP、 XRD、 TG-DTA、 IR、 TPD-MS和微反应技术研究了催化剂的化学组成、热稳定性、化学吸附性质和催化反应性能.杂多钼酸盐与TiO2通过O2-在TiO2表面发生了键合.在623 K下,杂多阴离子仍保持原有的Keggin结构.CO2在Lewis酸位Ni(Ⅱ)和Lewis碱位Ni-O-Mo的桥氧协同作用下生成CO2卧式吸附态Ni(Ⅱ)←O-(CO)←(O--Ni).丙烯有多种吸附态在催化剂上吸附.在563 K、 1 MPa和空速1500 h-1的反应条件下,丙烯的摩尔转化率为3.2%,产物MAA选择性为95%.  相似文献

A new and simple synthesis of novel N-protected methyl 5-substituted-4-hydroxypyrrole-3-carboxylates, which exist in equilibrium with their 4-oxo tautomers, has been developed in two steps starting from N-protected α-amino acids. The key intermediates are enaminones, which can also be isolated, characterized, and used for the construction of other functionalized heterocycles, before they spontaneously decompose to pyrrole products. 4-Hydroxypyrroles are prone to partial aerial oxidation but can be efficiently alkylated or reduced to stable polysubstituted pyrrolidine derivatives.  相似文献

The chemoselectivity in the intramolecular CH insertion of various diazosulfonamides has been experimentally studied. The results reveal that the aliphatic 1,4-, 1,5-, or 1,6-C(sp3)?H insertions of diazosulfonamides are not accessible, while the aromatic 1,5-C(sp2)?H insertion can be realized specifically by adjusting the diazo-adjacent group. In addition, the general chemoselectivities in the intramolecular CH insertions of diazosulfonyl compounds are summarized. Generally, diazosulfones undergo both aromatic 1,5-C(sp2)?H and aliphatic 1,5- and 1,6-C(sp3)?H insertions, while diazosulfonates undergo aliphatic 1,5- and 1,6-C(sp3)?H insertions. However, diazosulfonamides only undergo aromatic 1,5-C(sp2)?H insertion.  相似文献

The Langevin paramagnetic theory can’t describe the relation between magnetization of ferrofluids and applied magnetic field. The structuralization of ferrofluids, which is considered the main influence factor of the magnetization, is regarded. The part of magnetization works is deposited when the structure is forming. This action influences the magnetization of ferrofluids directly or indirectly. On the base of the “compressing” model, the Langevin function that usually describes the magnetization of ferrofluid is modified, and a well-fitted curve is obtained. An equation of the relation between the equivalent volume fraction after being “compressed” and the intensity of magnetic field is discovered, which approximately describes the process of magnetization. The relation between the approximate initial susceptibility and the volume fraction can be obtained from modified formula.  相似文献

The highly regioselective Buchwald–Hartwig amination at C-2 of the cheap and readily accessible reagent, 2,4-dichloropyridine with a range of anilines and heterocyclic amines is described. This new methodology is robust and provides a facile access to 4-chloro-N-phenylpyridin-2-amines on 0.25 mol scale. These intermediates undergo a further Buchwald–Hartwig amination at higher temperature to enable rapid exploration of the chemical space at C-4 and to provide a library of 2,4-bisaminopyridines.  相似文献

An efficient iodine-mediated oxidative Pictet-Spengler reaction in dimethyl sulphoxide (DMSO) using terminal alkynes as the 2-oxoaldehyde surrogate for the synthesis of aryl (9H-pyrido[3,4-b]indol-1-yl)methanones is described. The scope of the protocol includes the total synthesis of Fascaplysin, Eudistomins Y1 and Y2. The methodology is extended for preparing pyrrolo[1,2-a]-quinoxaline and indolo[1,5-a]quinoxaline derivatives. The utility of 1-aroyl-β-carbolines was demonstrated by performing palladium-catalyzed β-carboline directed ortho-C(sp2)-H functionalization of the phenyl ring with thiomethyl (SMe) group using DMSO as source and for accessing 4-aryl-canthin-6-ones.  相似文献

In this Letter, we described a facile method for constructing fused bicyclic 1-arylpyrazol-5-one ring system. We employed various methylene-containing carboxylic acids as the substrates and proved that the pyrazolone ring closure requires activated methylene group in intermediate II. Accordingly, a series of structurally diversified, fused bicyclic 1-arylpyrazol-5-ones was prepared in moderate to high yields using the requisite substrates.  相似文献

Regiospecific and direct imidation of the methyl C(sp3)–H bond of thioanisoles is realized under mild and metal-free conditions with N-fluorobis(benzenesulfonyl)imide as an oxidant and nitrogen source. Proposed mechanism suggests that thionium ion intermediates and a Pummerer-type reaction are involved. The imidation has advantages such as high step-economy, excellent functionality tolerance, and regiospecificity, giving structurally diverse imidation products.  相似文献
