The main objective of the present work is to study the influence of heat treatment on the esophageal cancer detection using the diffuse reflectance (DR) spectral intensity ratio R540/R575 of oxygenated hemoglobin (HbO2) absorption bands to distinguish the epithelial tissues of normal human esophagus and moderately differentiated esophageal squamous cell carcinoma (ESCC) at different heat treatment temperature of 20, 37, 42, 50, and 60°C, respectively. The DR spectra for the epithelial tissues of the normal esophagus and ESCC in vitro at different heat-treatment temperature in the wavelength range 400–650 nm were measured with a commercial optical fiber spectrometer. The results indicate that the average DR spectral intensity overall enhancement with concomitant increase of heat-treatment temperature for the epithelial tissues of normal esophagus and ESCC, but the average DR spectral intensity for the normal esophageal epithelial tissues is relatively higher than that for ESCC epithelial tissues at the same heat-treatment temperature. The mean R540/R575 ratios of ESCC epithelial tissues were always lower than that of normal esophageal epithelial tissues at the same temperature, and the mean R540/R575 ratios of the epithelial tissues of the normal esophagus and ESCC were decreasing with the increase of different heat-treatment temperatures. The differences in the mean R540/R575 ratios between the epithelial tissues of normal esophagus and ESCC were 13.33, 13.59, 11.76, and 11.11% at different heat-treatment temperature of 20, 37, 42, and 50°C, respectively. These results also indicate that the DR intensity ratio R540/R575 of the hemoglobin bands is a useful tool for discrimination between the epithelial tissues of normal esophagus and ESCC in the temperature range from room temperature to 50°C, but it was non-effective at 60°C or over 60°C.  相似文献

采用氧合血红蛋白吸收带的平均漫反射光谱比率(R540/R575)对离体的人正常胃、未分化胃腺癌、胃鳞状细胞癌和低分化胃腺癌的上皮粘膜组织进行鉴别诊断.实验采用带积分球附件的分光光度计测量组织样品在400 nm~600 nm的漫反射光谱.实验结果表明,在相同的波长范围内,人的未分化胃腺癌、胃鳞状细胞癌和低分化胃腺癌的上皮粘膜组织的漫反射光的强度较人正常胃的上皮粘膜组织的漫反射光强度要显著地低的,尤其是在氧合血红蛋白的吸收带540 nm和575 nm波长处,人正常胃、未分化胃腺癌、胃鳞状细胞癌和低分化胃腺癌的上皮粘膜组织的平均漫反射光谱比率分别是(94.25±2.4)%,(109±3.0)%,(1032±2.8)%和(98.6±2.6)%.  相似文献

采用氧合血红蛋白在540和575 nm吸收带的漫反射光谱比率(R540/R575)对人浅表性膀胱癌进行了鉴别诊断,实验采用带积分球附件的分光光度计获取组织的漫反射光谱。结果表明:正常膀胱的粘膜/粘膜下层组织的漫反射光谱比率(R540/R575)在组织离体后2 h的平均值为111%,3 h的平均值为107%,4 h的平均值为104%,5 h的平均值为102%,而浅表性的膀胱癌的粘膜/粘膜下层组织的漫反射光谱比率(R540/R575)在组织离体后2 h的平均值为98.4%,3 h的平均值为95.5%,4 h的平均值为93.1%,5 h的平均值为91.6%,正常膀胱与浅表性膀胱癌的粘膜/粘膜下层组织在组织离体后2,3,4和5 h的漫反射光谱比率(R540/R575)均有显著性的差异(p<0.05),分别为12.6%,11.5%,10.9%和10.4%。这结论为快速、低成本、非入侵的浅表性膀胱癌的光活检提供了一种新的方法。  相似文献

采用DNA和蛋白质吸收带的Kubelka-Munk光谱函数对离体的人正常胃、未分化胃腺癌、胃鳞状细胞癌和低分化胃腺癌的上皮组织进行鉴别诊断。实验采用带积分球附件的分光光度计获取组织的漫反射光谱。结果表明:在250~650 nm,人胃的上皮组织的癌变导致未分化胃腺癌、胃鳞状细胞癌和低分化胃腺癌等的上皮组织的Kubelka-Munk光谱函数f(r∞)及其对数log[f(r)]的平均值在DNA吸收带(260 nm)处都有非常显著性的差异,其差异分别为68.5%(p<0.05),146.5%(p<0.05),282.4%(p<0.05)和32.4%(p<0.05),56.0%(p<0.05),83.0%(p<0.05)。人胃的上皮组织的癌变导致未分化胃腺癌、胃鳞状细胞癌和低分化胃腺癌等的上皮组织的Kubelka-Munk光谱函数f(r)及其对数log[f(r)]的平均值在蛋白质的吸收带(280 nm)处也都有非常显著性的差异,其差异分别为86.8%(p<0.05),262.9%(p<0.05),660.1%(p<0.05)和34%(p<0.05),72.2%(p<0.05),113.5%(p<0.05)。人胃的上皮组织的癌变导致未分化胃腺癌、胃鳞状细胞癌和低分化胃腺癌等的上皮组织的Kubelka-Munk光谱函数f(r)及其对数log[f(r)]的平均值在β-胡萝卜素吸收峰(480 nm)处也有着显著的差异性,其差异分别为59.5%(p<0.05),73%(p<0.05),258.9%(p<0.05)和118.7%(p<0.05),139.2%(p<0.05),324.6%(p<0.05)。可见,人胃上皮组织的癌变导致其DNA、蛋白质和β-胡萝卜的含量都发生了显著的变化。这结论为快速、低成本、非入侵的胃癌的光活检及鉴别诊断胃癌提供有益参考。  相似文献

Low-temperature (4.2–130 K) photoluminescence spectra of HgI2 crystals have been measured in the 540–700 nm region. An analysis of the characteristics (intensity vs temperature and excitation power relations, afterglow times, excitation spectra) of the 560, 620, and 635 nm emission bands suggests the following assignments: the 560 nm band is due to radiative annihilation of excitons bound to mercury vacancies, and the “red” emission originates from recombination of free (620 nm) and donor-localized (635 nm) electrons with a hole-filled acceptor level. The energies of the corresponding donor and acceptor levels have been estimated. New emission bands at 540, 545, and 575 nm have been discovered, and their origin discussed. Fiz. Tverd. Foot ulceration due to diabetes mellitus is a major problem affecting 12–25% of diabetic subjects in their lifetime. An untreated ulcer further gets infected which causes necrosis leading to amputation of lower extremities. Early identification of risk factors and treatment for these chronic wounds would reduce health care costs and improve the quality of life for people with diabetes. Recent clinical investigations have shown that a series of factors including reduced oxygen delivery and disturbed metabolism have been observed on patients with foot ulceration due to diabetes. Also, these factors can impair the wound healing process. Optical techniques based on diffuse reflectance spectroscopy provide characteristic spectral finger prints shed light on tissue oxygenation levels and morphological composition of a tissue. This study deals with the application of diffuse reflectance intensity ratios based on oxyhemoglobin bands (R542/R580), ratios of oxy- and deoxy-hemoglobin bands (R580/R555), total hemoglobin concentration and hemoglobin oxygen saturation between normal and diabetic foot ulcer sites. Preliminary results obtained are found to be promising indicating the application of reflectance spectroscopy in the assessment of foot ulcer healing.  相似文献

本文用FT-Raman光谱方法研究了40例胃癌与胃正常组织,所得的光谱经统计处理的发现,肿瘤光谱中水、蛋白质有关的OH(NH)伸缩振谱带,C=O伸缩振动,H-O-H变角振动谱带明显强于正常组织的光谱,造成这一现象的原因可能是水与蛋白相互作用及氢键结构在正常组织和肿瘤组织中不同所致。  相似文献

Raman spectroscopy is a molecular vibrational spectroscopic technique that is capable of optically probing the biomolecular changes associated with neoplastic transformation. The purpose of this study was to apply near‐infrared (NIR) Raman spectroscopy for differentiating dysplasia from normal gastric mucosa tissue. A total of 65 gastric mucosa tissues (44 normal and 21 dysplasia) were obtained from 35 patients who underwent endoscopy investigation or gastrectomy operation for this study. A rapid NIR Raman system was utilized for tissue Raman spectroscopic measurements at 785‐nm laser excitation. High‐quality Raman spectra in the range of 800–1800 cm−1 can be acquired from gastric mucosa tissue within 5 s. Raman spectra showed significant differences between normal and dysplastic tissue, particularly in the spectral ranges of 850–1150, 1200–1500 and 1600–1750 cm−1, which contained signals related to proteins, nucleic acids and lipids. The diagnostic decision algorithm based on the combination of Raman peak intensity ratios of I875/I1450 and I1208/I1655 and the logistic regression analysis yielded a diagnostic sensitivity of 90.5% and specificity of 90.9% for identification of gastric dysplasia tissue. This work demonstrates that NIR Raman spectroscopy in conjunction with intensity ratio algorithms has the potential for the noninvasive diagnosis and detection of precancer in the stomach at the molecular level. The metabolic end products from cells/tissues that are released into the circulating blood stream and any changes in their level because of pathological conditions may be used as markers in disease diagnosis. Raman spectroscopy has been exploited to characterize the biomolecules present in the blood plasma of clinically confirmed normal group, premalignant (Oral Sub Mucous Fibrosis) and malignant (Oral Squamous Cell Carcinoma) at 784.15 nm. Raman spectral signatures show relatively less intense Raman bands of phenylalanine, lipid and antioxidant beta carotene but higher intense bands for proteins, DNA base components and amino acids (tyrosine and tryptophan) for malignant group than that of normal group. However premalignant group possess high intense Raman bands for amino acids (tyrosine and tryptophan) at 830, 1020 and 1620 cm−1 and protein peaks at 913, 978 and 1646 cm−1 when compared to that of malignant and normal group. Principal component analysis coupled with linear discriminant analysis (PCA‐LDA) yielded a diagnostic sensitivity of 96.3% and 91.2%, and a specificity of 80.0% and 96.7% in the classification of normal from premalignant and normal from malignant, respectively. Diffuse reflection spectra of biotissues in vivo and transmission and reflection coefficients for biotissues in vitro are measured over 300–800 nm. These data are used to determine the spectral absorption and scattering indices and the scattering anisotropy factor for stomach mucous membranes under normal and various pathological conditions (chronic atrophic and ulcerous defects, malignant neoplasms). The most importan tphysiological (hemodynamic and oxygenation levels) and structural-morphological (scatterer size and density) parameters are also determined. The results of a morphofunctional study correlate well with the optical properties and are consistent with data from a histomorphological analysis of the corresponding tissues.  相似文献

利用共焦显微拉曼光谱研究了胃炎组织和胃溃疡组织的粘膜细胞。结果表明,与胃炎组织细胞比较,溃疡组织细胞中胞嘧啶的振动峰781 cm-1相对较弱,而腺嘌呤和胸腺嘧啶793,823cm-1处的峰强有所增加; 对应于α螺旋结构蛋白质酰胺Ⅰ和酰胺Ⅲ带的1654和1230~1270cm-1的强度均相对减小,色氨酸特征峰1332cm-1和苯丙氨酸特征峰1003cm-1强度有所降低,同时与色氨酸残基微环境变化密切相关的1554频移到1556cm-1,且强度有所增加;脂类的特征谱线1073 cm-1向高波数频移到了1078cm-1, 对应于CH2扭转振动的1303cm-1与来自于CH面内变形振动的1268cm-1强度比值减小。上述变化表明两种疾病组织细胞中的核酸,蛋白质和脂类在成分、结构和构型上都存在明显差异;研究发现利用C—H弯曲振动的谱线1449cm-1和蛋白质酰胺Ⅲ带的谱线1660cm-1的强度比值A1449/A1660区分两种疾病具有较好的准确性。  相似文献

利用傅里叶变换红外光谱对宫颈鳞癌(17例)、宫颈腺癌(5例)及宫颈正常组织(13例)进行了分析。结果显示,18个谱带在各类组织中的出现概率都在80%以上,显示其在组织中的特征性;这3类组织红外光谱的相对吸收峰吸光度差异主要表现在1 080,1 238,1 314,1 339,1 397,1 454,1 541,1 647,2 854, 2 873,2 926和2 958 cm-1处的比较上。傅里叶变换红外光谱仪可用于区分宫颈鳞癌、腺癌及宫颈正常组织,以该仪器为基础的方法有望成为宫颈癌筛检及临床诊断的新方法。  相似文献

采用基于小波变换的光谱去噪和背景扣除预处理技术,对32例(其中:13例正常,19例癌变)胃粘膜组织拉曼光谱进行分析,克服了手动背景扣除的一些缺点,并观察到一些新的光谱特征。基于此,给出能对所有正常组织和癌变组织进行有效分类的特征量和判据。通过研究这些特征量与胃粘膜组织癌变发展阶段之间的定量关系,可望实现胃癌的早期诊断。  相似文献

Experimental-computational spectra of the permittivity and characteristic losses –Im–1 for energies in the range 5–21 eV at a temperature of 4.2 K and theoretical spectra of and –Im–1 of a fluorite crystal are resolved into elementary transition bands. The parameters of transition bands (energies of their maxima E i, band halfwidths H i and areas S i, and oscillator forces f i) are determined. 本文采用傅里叶变换红外光谱法与中红外光纤及ATR探头联用,首次实现了肿瘤组织的在体原位检测,即在有血液循环的活体组织中获得了具有很好信噪比的中红外光谱。实验结果表明在体原位的正常和肿瘤组织的光谱特征同我们先前液氮冰冻样品以及新鲜离体检测的结果是相似的,即符合液氮和新鲜离体样品研究中所得到的鉴别癌症与正常组织光谱变化规律,例如图1所列出的肠癌的原位在体光谱,与离体癌变组织光谱相似,这一创新结果表明中红外光纤-红外光谱仪联用可能发展成一种外科手术中无损、快速、准确的在体检测的方法。  相似文献

本文用傅里叶变换红外显微光谱对一系列胃肠道(食道、胃、结肠)肿瘤组织和相应正常组织的冰冻切片进行了研究。结果显示,所有肿瘤组织的光谱基本一致。而正常组织的光谱按1400-950cm^-1区域的特点可分为三类。对光谱中蛋白质酰胺I带曲线拟合的结果表明,正常组织和肿瘤组织中蛋白质的二级结构有着很大的差别。  相似文献

Nanoparticles of the II-V semiconductor (ZnP2) were prepared and investigated. ZnP2 nanoparticles were incorporated into zeolite Na-X matrix. Absorption, diffuse reflection (DR) and photoluminescence (PL) spectra of ZnP2 nanoparticles were measured at the temperature of 77 K. Five bands B1-B5 are observed in both the DR and PL spectra demonstrating the blue shift from the line of free exciton in bulk crystal. We attribute the B1-B5 bands to five stable nanoparticles with size less than the size of zeolite Na-X supercage. We observed Stokes shift of the PL bands with respect to the absorption bands. This dependence of this Stokes shift on the particle size is nonmonotonic.  相似文献

应用傅里叶变换红外光谱(FTIR) 法对184例不同病理状态的胃镜下活检组织样品进行检测和分析,结果表明,慢性浅表性胃炎的基本谱图特征包括:较强的酰胺Ⅱ带;1 460 cm-1处的峰强高于1 400 cm-1处的峰强。典型的胃癌组织的谱图特征包括:较低的酰胺Ⅱ带;1 460 cm-1处的峰强低于1 400 cm-1处的峰强;1 300 cm-1的峰强明显增加。慢性萎缩性胃炎呈现出部分胃癌的光谱特征。这一结果表明,应用FTIR可以对胃镜下活检的小块组织进行鉴别诊断。  相似文献
