紫杉醇治疗小鼠S180肉瘤的体内31P MRS研究   总被引:5,自引:0,他引:5  
为了研究紫杉醇治疗小鼠S180肉瘤的31P MRS参数的变化及这些参数的变化是否早于常规观察的瘤体积的变化.方法:利用表面线圈31P NMR方法,研究小鼠皮下接种的S180肉瘤.结果:用药48h后给药组的PCr/Pi、β-NTP/Pi、PME/β-NTP比值与对照组有显著性差别(P<0.05),而肿瘤体积在给药组和对照组之间无显著性差别,31P MRS参数的变化早于瘤体积变化.结论:小鼠S180肉瘤31P MRS不仅可以给出定量的结果,而且可以较早地显示出治疗的效果.  相似文献

1H Magnetic resonance imaging and 31P magnetic resonance spectroscopy (MRS) have been carried out in experimental rodent filariasis, i.e., Acanthocheilonema viteae infection in the rodent host, Mastomys coucha. The T2-weighted image of the infected host shows fine hyperintense thread like structures of adult filariid nests in the cervical region. 31P MRS of normal and infected hosts, localized over the same region of interest, show seven major peaks corresponding to phosphomonoesters (including glucose-6-phosphate, fructose-6-phosphate, fructose-1-6-diphosphate, phosphorylcholine, and adenine monophosphate or AMP), inorganic phosphate, glycerophosphorylcholine, phosphoenolpyruvate, phosphocreatine and nucleoside di- and tri-phosphates. Concentrations of phosphomonoesters (PMEs) are higher in the normal rodent compared with the infected ones. In vivo 31P MRS provides a non-invasive assessment of tissue bioenergetics and phospholipid metabolism.  相似文献

31P-MR-Spectroscopy was performed in 28 patients with focal (n = 23) and diffuse (n = 5) liver disease and in 18 healthy volunteers. The spectra were obtained with a whole body scanner operating at 1.5 T by using a surface coil. To get T1-weighted 31P-spectra a short TR of 600 msec was taken, because T1-weighted spectra of focal liver disease were more significantly different from spectra from healthy volunteers than density weighted ones. The VOI from patients with focal superficial alterations showed a mean volume of 172 ml, with diffuse liver disease 196 ml, and from volunteers 158 ml. Focal tumors filled up the VOI on an average of 70%. This investigation demonstrated that PME/β-ATP- and PDE/β-ATP-ratios were sensitive indicators for focal liver disease. As a result of this study we could establish a significant increase of PME/β-ATP- (0.75 ± 0.30) and PDE/β-ATP-ratios (1.68 ± 0.62) in patients with superficial focal liver metastases (n = 19) compared to the control group (PME/β-ATP: 0.49 ± 0.17, PDE/β-ATP: 1.24 ± 0.24; t-test: p < 0.02). Patients with a hemangioma (n = 1), liver infarction (n = 1), empyema of gallbladder (n = 1) and a hepatic involvement by a malignant lymphoma (n = 1) showed a similar increase of PME/β-ATP and/or PDE/β-ATP. Up to now spectral changes seemed to be non-specific. The ratios of 31P metabolites of the cirrhoses (n = 4) and the fatty liver (n = 1) did not show any characteristic changes versus the volunteers.  相似文献

测定了Langendorff灌流大鼠和兔心脏的31P NMR谱.观测到PCr、ATP、SP、Pi以及PME和GPC等含磷代谢物的共振峰,各谱峰之间实现了很好的分辨.在3~5min的累加时间内测得的图谱具有较好的S/N,可用于以上含磷代谢物的定量测定,以高能含磷化合物ATP和PCr相对峰强度以及冠脉流量的变化对灌流大鼠心脏代谢的稳定性进行了考察,结果表明,在本实验条件下大鼠心脏的能量储存至少可稳定2h,用所建方法以3min的时间间隔对同一大鼠心脏缺血21min及再灌注12min过程中心脏的31P NMR谱进行了连续跟踪测定,并初步观测了缺血及再灌注过程中心肌细胞内ATP、PCr与Pi之间的消长关系.  相似文献

利用表面线圈31P NMR研究了小鼠S180肉瘤生长过程中能量代谢和磷脂类变化的特点.结果发现:随着肿瘤体积的增大,(1)Pi和PME升高;(2)PCr降低,在肿瘤体积较大时常检测不到;(3)β-NTP(通常用来表示ATP的量)变化较小;(4)PDE波动性较大;(5)PCr/Pi和β-NTP/Pi比值均下降,且PCr/Pi比β-NTP/Pi下降得快;(6)PME/β-NTP比值升高;(7)肿瘤pH下降,且与PCr/Pi、β-NTP/Pi或(PCr+β-NTP)/Pi比值有相关性.讨论了与这些参数变化相关联的生物学意义.  相似文献

The relaxation time T1 values and nuclear Overhauser enhancement factor for 31P signal were determined in model solutions of metabolites ATP, PCr and Pi, and AMP at two frequencies and in H2O and 2H2O solutions. The data were analyzed to resolve the contribution of different relaxation mechanisms. A knowledge of NOE is important in the light of recent applications of double resonance methods to enhance the sensitivity of in vivo 31P spectroscopy. The results show that chemical shift anisotropy is the dominant mechanism for 31P in ATP at the high field, whereas the dipolar interaction mechanism is the main feature for the 31P relaxation of PCr and Pi. The dipolar mechanism responsible for NOE originates from interactions of solvent water with 31P moiety. Implications for in vivo spectroscopy are indicated.  相似文献

利用表面线圈探头测得了小鼠大脑,大腿肌肉和肝脏的体内31P NMR谱图。观察到PCr,ATP,PME,PDE,Pi等化合物的共振峰,并确定了各谱峰的化学位移。由大脑的Pi化学位移计算得到其细胞内pH值为7.15.用差谱方法解决了因定位不准而造成的肝脏谱图不纯。观察了小鼠大脑在缺氧状态下的代谢变化过程。当O2浓度降至2%时,PCr开始下降,降到1%时,Pi明显升高。从空气变化到1%的O2含量,致使小鼠死亡,小鼠大脑的pH值从7.16降到6.46。  相似文献

31P-NMR spectroscopy is widely used for studies of phospholipid liposomes, a commonly used model of a biological membrane. For the correct analysis of 31P-NMR spectra of the liposomes it is necessary to take into account that they are deformed by the magnetic field of the spectrometer. The liposomes become ellipsoidal and this affects the lineshape of the spectrum. In the present communication we suggest a new analytical formula for modeling of 31P-NMR spectra of the prolate phospholipid liposomes. The formula assumes a Lorentzian broadening function and exactly ellipsoidal shape of the liposomes. Based on the formula a program called P-FIT is designed for the practical analysis of the experimental multicomponent spectra of the prolate liposomes. The versatility of the program developed in a Mathematica environment is demonstrated by simulations of a number of 31P-NMR spectra with different complexity.  相似文献

31P核磁共振(NMR)观察了人淋巴瘤白血病细胞系Molt-4的31P谱.NMR测试温度为37℃、20℃和4℃时,随着测试时间增长,细胞31P谱中ATP峰降低,无机磷峰升高,细胞31P谱的变化随测试温度降低依次减缓.实验温度为4℃,从收集细胞至开始NMR累加在30min内,NMR累加在60min内,获得活细胞的稳定的31P谱.根据细胞内无机磷的化学位移数值,对细胞内pH值进行了测定.  相似文献

The primary goal of this study was to establish a rigorous approach for determining and comparing the NMR detection sensitivity of in vivo 31P MRS at different field strengths (B0). This was done by calculating the signal-to-noise ratio (SNR) achieved within a unit sampling time at a given field strength. In vivo 31P spectra of human occipital lobe were acquired at 4 and 7 T under similar experimental conditions. They were used to measure the improvement of the human brain 31P MRS when the field strength increases from 4 to 7 T. The relaxation times and line widths of the phosphocreatine (PCr) resonance peak and the RF coil quality factors (Q) were also measured at these two field strengths. Their relative contributions to SNR at a given field strength were analyzed and discussed. The results show that in vivo 31P sensitivity was significantly improved at 7 T as compared with 4 T. Moreover, the line-width of the PCr resonance peak showed less than a linear increase with increased B0, which leads to a significant improvement in 31P spectral resolution. These findings indicate the advantage of high-field strength to improve in vivo 31P MRS quality in both sensitivity and spectral resolution. This advantage should improve the reliability and applicability of in vivo 31P MRS in studying high-energy phosphate metabolism, phospholipid metabolism and cerebral biogenetics in the human at both normal and diseased states noninvasively. Finally, the approach used in this study for calculating in vivo 31P MRS sensitivity provides a general tool in estimating the relative NMR detection sensitivity for any nuclear spin at a given field strength.  相似文献

为评价维拉帕米(Ver)防治心脏缺血-再灌注损伤的作用,采用31P核磁共振(3lP NMR)技术对大鼠心肌缺血前,缺血过程中及缺血后高能磷化物的含量及细胞内pH (pHi)的变化过程进行了动态跟踪测定,离体心脏于37℃下平衡灌流30min,停止灌流30min,再灌注30min.灌流液中始终含有Ver (0.2μmol·L-1).Ver可使再灌注后心脏的冠脉流量有较高程度的恢复,3lP NMR测定显示Ver可使心脏产生代谢上的改善作用.缺血10min后对照组心脏即检测不到磷酸肌酸(PCr),而Ver组心脏PCr仍保持在缺血前的20%.缺血过程中治疗组比未治疗组心脏ATP下降减缓,至缺血结束时心肌ATP分别为缺血前的53%和34%.再灌注后两组心肌的ATP均未回升,但Ver使PCr的恢复显著提高(P<0.05),给药心脏PCr/Pi(无机磷酸盐)无论在缺血前或再灌注阶段,都非常显著(P<0.01)地高于对照组心脏.Ver还可显著减轻缺血过程中的酸中毒并防止再灌注后心肌出现严重酸化的区域.  相似文献

The framework structure of As-synthesized A1PO4-14 has been investigated with a combination of different one-dimensional 27Al and 31P solid state NMR techniques and 27Al/31P double resonance methods. The results are found to be fully consistent with the assumed structural model. 27Al MAS and DOR experiments at three different magnetic field strengths together with simulations show the presence of two tetrahedral sites, one pentacoordinated and one octahedral aluminum site. The 27Al quadrupolar coupling constants and the 31P isotropic chemical shifts of the tetrahedral sites correlate well with tetrahedral shear-strain parameters and mean P-O-A1 bond angles, respectively. These correlations allow one to assign all of the NMR resonances to specific T-sites in the proposed framework structure. The assignments are then further confirmed by the application of three different two-dimensional heteronuclear correlation methods (i.e., 27Al → 31P TEDOR, CP, and INEPT) which reveal the connectivities between AlOx and PO4 polyhedra. The two-dimensional INEPT experiment is applied here for the first time in the solid state.  相似文献

Application of spectroscopic imaging in epilepsy   总被引:8,自引:0,他引:8  
Functional and anatomical neuroimaging has had a dramatic effect on the evaluation of patients for seizure surgery. The demonstration by PET that the epileptogenic focus has interictal metabolic abnormalities has allowed a greater number of patients to come to seizure surgery, with fewer of these patients requiring intracranial electrode evaluations. Metabolic changes have also been demonstrated utilizing single voxel and whole brain 1H and 31P MRS imaging techniques with the interictal focus characterized by increased Pi, pH, and decreased PME and NAA. These findings can be used to accurately lateralize temporal lobe as well as frontal lobe epilepsy. Furthermore, there is evidence that these findings can be used to localize the seizure focus with the changes specific for the epileptogenic region; although, more diffuse changes both ipsilaterally and contralaterally have been seen. In patients with anterior hippocampal seizure foci the pH is significantly alkaline only in the ipsilateral hippocampus, whereas the increased Pi and decreased PME can be seen throughout the ipsilateral temporal lobe. When compared to controls the contralateral hemisphere is acidotic. Decreased NAA concentrations as well as NAA/Cr ratios have been demonstrated in the epileptogenic region in temporal and frontal lobe epilepsy. The decreased NAA has been correlated with the severity of cell loss, and may be a more sensitive measure than qualitative or quantitative measures of the hippocampal atrophy; however, the NAA decrease is more widespread than just the epileptogenic focus but may be maximal at the site of seizure initiation. In preliminary work, NAA maps of deviation from normality have suggested the maximal change to coincide with the epileptogenic region. These results suggest that in focal epilepsy there is abnormal metabolic activity throughout the brain detectable by MRS, with patterns of metabolic asymmetry that are useful for seizure localization.  相似文献

The 31P(3He, d)32S reaction has been studied with high resolution at 12 MeV bombarding energy. A detailed level scheme for 32S has been determined up to an excitation energy of 9.5 MeV revealing several previously unobserved states. The lp values and absolute spectroscopic factors extracted from a DWBA analysis of the experimental deuteron angular distributions have provided information on the wave functions for the T = 0 states as well as for the T = 1 isobaric analogue states of 32P. A comparison between the present data and those of previous experiments is made, and the results are discussed in terms of existing theoretical work in this mass region. Information on Coulomb displacement and symmetry energies is also extracted.  相似文献

Two patients affected by severe Alzheimer's disease (AD) were investigated by MRI and image-guided 31P MRS. In one case, 1H MRS was additionally performed. In both cases the diagnosis of AD was confirmed, post mortem, by the pathologist. The spectral parameters of the 31P MR spectra were estimated by fitting the 31P MR signals in the time domain. Our 31P MRS results suggest that it is possible to detect the membrane catabolism, as indexed with the level of PDE resonances visible in in vivo 31P MRS, at least in severe AD cases. The 1H spectrum from AD brain showed a marked decrease of NAA signal respect to choline.  相似文献

While liquid-state 29Si NMR of phosphorus-bearing organosilicon compounds with more than one phosphorus per molecule can take advantage of the presence of J-coupling nJ(31P29Si) for purposes of structural assignment from J-coupling patterns, conventional 29Si CP/MAS spectra of such molecular solids do not reveal structural details in a straightforward manner. For such compounds it is necessary to obtain 29Si CP/MAS spectra under conditions of simultaneous 1H- and 31P-high power decoupling in order to derive reliable 29Si chemical shift information. 29Si CP/MAS NMR spectra, obtained with and without 31P high power decoupling during the acquisition time, of several organosilicon compounds containing SixPy (x = 1 −10, y = 1 −10) moieties are reported.  相似文献
